1.Determination of Rhamnocitrin in Oxytropis falcate Bunge by HPLC

Objective To establish a method of HPLC for determination of rhamnocitrin in Oxytropis falcate Bunge.Methods The HPLC method was used.Chromatographic column:Lichrospher C18(4.6 mm? 250 mm,5 ?m) reversed-phase column;mobile phase:0.4% phosphoric acid in water-methanol(20∶80);flow rate:1.0 mL/min;detective wavelength:350 nm.Results Rhamnocitrin was liner at the range of 4~ 30 ?g(r =0.999 8),and its average recovery rate was 99.7%,RSD=1.4%.Conclusion The method was simple and accurate,so it can be used for the quality control of Oxytropis falcate Bunge.
2.A study on the amplification, rearrangement and deletion of ras-oncogenes in gastric cancerous and juxtacancerous tissues

The amplification,rearrangement and deletion of c-Ha-ras,K-ras and N-ras were studied in 33 cases of gastric cancerous and juxtacancerous tissues.It was found that the amplification rearangement,and deletion of c-Ha-ras were shown in 25.8% (8/33) of cases and the ampli-cation of K-ras in 3.8% (1/26).No such changes was found for N-ras.
3.A study on the activation of oncogene and inactivation of suppressor gene in human gastric cancer

The changes of several oncogenes and suppressor genes in the specimens of cancerous and juxtacancerous tissues of 42 cases of gastric cancer were studied with Southern blot hybridization and PCR-RELP method.The probes used were c-Ha-ras,K-ras,N-ras,N-myc,c-myc,hst,EGFR,c-erbB-2,p53 and Rb.Amplification,rearrangement,and deletion of c-Ha-ras were detected in 8/33 (25.8%) cases of gastric cancer (amplification or rearrangement of hst and c-erbB-2 in 11/42 (26.6%) and 12/42 (29.2%) cases respectively (amplification of EGFR in 21.4% of cas-esideletion or rearrangement of p53 and Rb in S/30 (30%) and 2/15 (13%) cases respectively) and amplification or rearrangement of N-ras (0/33),K-ras (1/26),N-myc (1/26),and c-myc (1/ 35) was only rarely encounted.The point mutation in codon 248 and 249 of p53 in gastric cancer was analyzed.2 cases of the 42 harbored point mutation in codon 248 of p53.These findings suggest that e-Ha-ras,hst,c-erbB-2,EGFR and p53 may be the hot point genes in the occurrence and development of gastric cancer.
4.A study on point mutation of ras oncogenes

It is known that no restriction endonuclease cleavage site exists at 12 codon in N-ras gene and at 12 and 13 codon in K-ras gene.Amismatched base was incorporated at the 3' end primer to creat a kind of restriction endonuclease cleavage site.Then it was possible for us to analyze the point mutation of the above mentioned codons with the polymerase chain reaction-restriction fragment length polymorphism.In this paper,the study of the point mutation at 12 and 61 codon in c-Ha-ras,at 12 codon in N-ras,and at 12 and 13 codon in K-ras was reported.
5.A study on p53 and Rb genes in gastric cancer and juxta-cancerous tissue

The deletion and rearrangement of p53 and Rb genes were studied in gastric cancer with Southern blot hybridization and the point mutation in 248 and 249 codons of p53 was studied with PCR-RFLP.It was found that deletion or rearrangement of p53 was seen in 9 cases out of 30 cases of gastric cancer and point mutation at 248 codon of p53 in 2 cases out of 42 cases,and deletion and rearrangement of Rb gene were seen in 2 cases out of 15 cases.
6.The clinical features and risk factors for invasive fungal infection in multiple myeloma

Objective To study the clinical features and risk factors of invasive fungal infection (IFI) in multiple myeloma ( MM) . Methods Three hundred and fifty-seven cases of MM were retrospectively analyzed for IFI, clinical features, complicating diseases, treatment of fungus and side effect of anti-fungal drugs. Results Forty-four cases ( 12. 3% ) of IFI were diagnosed. Three of them were diagnosed definitely, 8 clinically and 33 probably. Ten cases incurred IFI in (he induction therapy, 4 in platform, 27 in progress and 3 in the treatment with autologous stem cell transplantation. The lung was the commonest site of infection ( 50. 0% ) . The total effective rates of amphotericin B liposome, voriconazole, itraconazole, caspofungin and fluconazol were 83. 3% , 75. 0% , 78. 9% , 75. 0% and 57. 1% respectively (P= 0.493). In a multivariate analysis, independent factors significantly associated with IFI were diabetes (P=0.035, OR 2. 527, 95%CI 1.005-6.052), dialysis (P=0. 022,OR 2. 768, 95%CI 1. 161-6. 600), persistent agranulocytosis (P = 0.019, OR 3.215, 95% CI 1.200-7.407), broad-spectrum antibiotic therapy (P = 0.009,OR 3. 350,95% CI 1.353-8.295) and fludarabine treatment( P = 0. 001,0R 4. 669, 95% CI 1.813-12.023). Conclusions Patients with MM are in high risk of IFI. The lung is the commonest site of infection. The therapeutic effect was similar with itraconazole, voriconazole, caspofungin and amphotericin B liposome in MM patients with complicating IFI. The risk factors for IFI in MM were diabetes, dialysis, persistent agranulocytosis and the use of broad-spectrum antibiotics and fludarabine.
7.Detection and analysis of allergens in 859 patients with chronic urticaria in Chengdu

Objective To analyze the allergens and sex ,age distribution of chronic urticaria in Chengdu .Methods Totally 859 patients with chronic urticaria were tested with 13 kinds inhaled allergens and 15 kinds of food allergens by skin prick test .Re‐sults The top 5 allergens were:dermatophagoides farinae、dermatophagoides pteronyssinus ,cockroach ,shrimp and sea‐crab .The positive rate of dermatophagoides farinae was 58 .7% ,dermatophagoides pteronyssinus which was 55 .1% took second place .No difference was found between sex ,more inhaled allergens were found positive than food allergens in both sex groups .The positive rate was higher in people younger than 60 .Conclusion Dermatophagoides farinae ,dermatophagoides pteronyssinus ,cockroach , shrimp and sea‐crab are the commonest allergens in Chengdu .The skin prick test is important in the individualized treatment of chronic urticaria and health education .It may also be helpful in the management of chronic allergic skin diseases .
8.Simultaneous Determination of Ferulic Acid and Paeoniflorin in Ankong Zhongzi Wan by Dual Wavelength HPLC

Objective To determine ferulic acid and paeoniflorin in Ankong Zhongzi Wan by HPLC under dual wavelength ultraviolet detection. Methods Ferulic acid and paeoniflorin were separated by Waters SymmetryShield-C18 column (4.6 mm × 250 mm, 5 μm) with gradient elution of acetonitrile-0.1%phosphoric acid as the mobile phase at a flow rate of 1.0 mL/min. The detection wavelength was 230 nm and 323 nm. Results The linear relationship of ferulic acid and paeoniflorin was good in the range of 0.058 2-0.582 4 μg (r=0.999 4) and 1.664-16.64 μg (r=0.999 6), and the average recovery rate was 97.77% (RSD=1.88%) and 98.84% (RSD=1.96%), respectively. Conclusion The method is accurate and quick for determining the two effective components in Ankong Zhongzi Wan, and can be used for its quality control.
9.Arsenic trioxide inhibits KM3 cell growth and induces apoptosis and potentiated by bortezomib

AIM: To observe if there is a synergistical effect on induction of apoptosis when arsenic trioxide alone or combination with bortezomib in KM3 cells. METHODS: KM3 cells were treated with arsenic trioxide alone or combined with bortezomib, the numbers of viable cells were determined by trypan blue exclusion. Cell growth inhibition was examined by MTT method. The cells were simultaneously stained with annexin V-FITC and PI and apoptosis was determined by bivariate flow cytometry using a FACScan. Reverse trascriptional-PCR (RT-PCR) method was used to examine the change of p65 mRNA and Western blotting to measure the expression of protein p65, p-p65, caspase-3, -8, -9, and poly ADP-ribose polymerase (PARP). RESULTS: Arsenic trioxide inhibited the cell growth and induced apoptosis. The mechanism was responsible for the activation of caspase-mediated induction of apoptosis. A synergistic effect of combination with bortezomib on apoptosis was observed. CONCLUSION: Arsenic trioxide inhibits KM3 cell growth and induces apoptosis with a synergistical effect when cotreated with bortezomib.
10.Characteristics of obesity in teenage with a family history of type 2 diabetes

A cross-sectional study was conducted on 1619 youngsters aged 13-15.They were divided into two groups:one with family history of diabetes (FHD~+) and another without a family history of diabetes (FHD~-).Measurements included height,weight,waist circumference (WC) and hip.FHD~+ group had significantly higher WC measurement,waist-to-hip ratio (WHR) and waist-to-height ratio (WHtR) when compared with FHD~-group (P <0.01).The abdominal obesity rate defined by WC measurement in FHD+ group was higher than that in FHD~-group (P<0.01).The rate of overweight and obesity defined by body mass index (BMI) were no significant difference between two groups.After adjusting the gender and age,logistic regression analysis showed that the odds ratios of WC alone and BMI+WC for FHD~+ were 2.029 and 1.364 (95% CI:1.211-3.400,1.043-1.784,P<0.05) respectively.Our data suggest that teenage with a family history of type 2 diabetes has a tendency of overweight characterized by the abdominal obesity.
