Objective To explore the relationship between genital endocrine hormones and sleep disorder in climacteric depression,and study the therapeutical functions of Bushen Tiaogan Qingxin Prescription.Methods Sixty patients were randomly divided into Bushen Tiaogan Qingxin prescription group(n=30) and HRT group(n =30).The effects were appraised by both Hamilton depression rating scale(HAMD) and sleep disorder scores before and after the treatment of 12 weeks.The values of blood E2,FSH,LH were measured.Results Decreasing rate of HAMD and sleep disorder improving rate were significant in Bushen Tiaogan Qingxin prescription group than HRT group(P

Objective: To investigate the clinical efficacy of needling Danzhong(CV 17) in the treatment of postpartum hypogalactia and provide clinical evidence for indications of the point. Methods: A multi-centre single-blind randomized controlled trial was carried out. Two hundred and seventy-six puerperal women with postpartum hypogalactia were randomly allocated into acupuncture group and herb group, and respectively treated for three consecutive days. The degree of mammary fullness, the amount of milk secreted, prolactin, baby weight, the frequency and volume of artificial feeding, the number of infant urination events, and the duration of baby crying were observed. The clinical curative effects on postpartum hypogalactia were compared. Results: Hypogalactia was effectively treated in both acupuncture and herb groups. There were statistically significant differences in degree of mammary fullness, amount of milk secreted, baby weight, the frequency and amount of artificial feeding, and the number of infant urination events between pretreatment and post-treatment, but no difference between the two groups. There was no significant difference in prolactin in the acupuncture group and there was a difference in prolactin in the herb group between pretreatment and posttreatment. Conclusion: Needling Danzhong(CV 17) can effectively promote lactation.
4.Hypermethylation in the promoter region inhibits LRRN3 expression and promotes proliferation of non-small cell lung cancer

Objective To study the role of leucine rich repeat neuronal 3 (LRRN3) in the prolif-eration of non-small cell lung cancer and its possible mechanism of expression regulation. Methods The expression of LRRN3 in non-small cell lung cancer was detected by real-time quantitative polymerase chain reaction ( qPCR) , immunohistochemistry and bioinformatics retrieval;A lung cancer cell line A549-LRRN3 with stable over-expression of LRRN3 was established by lentivirus over-expression technology;The effect of LRRN3 on the proliferation of non-small cell lung cancer was detected by methyl thiazolyl tetrazolium ( MTT) assay;Bioinformatics search for changes in methylation of LRRN3 promoter region and treatment of lung cancer cells by methyltransferase inhibitors to detect the effect of methylation on the regulation of LR-RN3 expression; Finally, bioinformatics search analyzes the correlation between LRRN3 and lung cancer prognosis. Results The mRNA expression of LRRN3 in clinical tissues of non-small cell lung cancer (n=12) was significantly lower than that of adjacent normal tissues (n=12) (P=0. 0014). The results of immunohistochemistry showed that the protein expression level of LRRN3 in non-small cell lung adenocar-cinoma was lower than that in normal tissues (P=0. 001), and the expression in non-small cell lung squa-mous cell carcinoma was also lower than that in normal tissues (P=0. 003). Overexpression of LRRN3 in-hibited the proliferation of tumor cells (P<0. 01), and the hypermethylation of LRRN3 in the promoter re-gion inhibited its transcriptional expression. LRRN3 was positively correlated with the survival prognosis of lung adenocarcinoma (P=5. 2e-09;HR=0. 48). Conclusions Hypermethylation in the promoter region of LRRN3 inhibits its transcriptional expression, thereby promoting the proliferation of lung cancer cells.
5.Effects of kidney-supplementing and blood-activating formula on Wnt/β-catenin signal pathway of stem cell of thin endometrium

Objective To investigate the effects of Bushen Huoxue Fang kidney-supplementing and blood-activating formula(BSHXF)on Wnt/β-catenin signal pathway in stem cell of thin endometrium. Methods During the 7thto 9thday after ovulation,100 mg endometrial membrane samples of selected patients with thin endometrium were taken with disposable endometrial sampling device.After washing with saline,the samples was fixed in 4%polyformaldehyde and then separated to culture the endometrial stem cells,and drug serum was prepared.The cultured endometrium stem cells were divided into the control group,High,medium and low dose groups of kidney-supplementing and blood-activating formula (BSHXF)and Western medicine group.After intervention,β-catenin /TCF(T cytokine)and transcrip-tional activity were measured by using double Luciferase Report System;CD34/CD117 protein expression was examined with immunofluorescence; Oct-4 gene was examined with RT-PCR; and ABCG2 protein was measured by using Western blot.Results After transfecting the same dosage of plasmid,there was a significant difference among mid-dose BSHXF group, high-dose BSHXF group, Western medicine group,and control group(P<0.05).CD34/CD117, Oct-4 gene and ABCG2 protein had positive ex-pression in the endometrium stem cells.There was a significant difference in the expression of CD 34 pro-tein between BSHXF groups and control group(P<0.05).There was a significant difference between high-dose BSHXF group and Western medicine group(P<0.01).The expression of CD117 protein in the high-dose group was significantly different from that in the control group(P<0.05),and there was a significant difference between the Western medicine group and the control group(P<0.01).RT-PCR test showed that the expression rate of Oct-4mRNA were significantly different from the interventional groups and the control group(P<0.05).Western blot test showed that the expression rate of ABCG 2 protein in the interventional groups were significantly different from that in the control group(P<0.05). Conclusion Kidney-supplementing and blood-activating formula can increase the fluorescent activity of thin endometrial stem cells, thereby promote the activation of Wnt/β-catenin signal pathway, and in-crease the expression of CD34/CD117,OCT4 gene and ABCG2 protein.
6.Analysis of drug therapy in a patient with pulmonary infection secondary to suppurative thrombophlebitis

Objective To explore the role of pharmaceutical care in the treatment of patients with pulmonary infection secondary to suppurative thrombophlebitis. Methods The treatment of a patient diagnosed with pulmonary metastatic infection secondary to suppurative thrombophlebitis and the whole process of clinical pharmacists participating in the monitoring were analyzed retrospectively. The use of antibiotics was evaluated, and the experience of coagulation management in suppurative thrombophlebitis was explored. Results Based on the infection site, characteristics of septic thrombus, monitoring of vancomycin blood concentration, pharmacokinetics and pharmacodynamics characteristics of antibiotics, clinical pharmacists provided comprehensive pharmaceutical services for clinicians and patients in terms of anti-infection scheme adjustment, optimization of vancomycin individualized treatment, anticoagulant timing. Patient’s systemic infection and septic thrombus can be effectively controlled and which promotes the treatment of patients with suppurative thrombophlebitis. Conclusion Clinical pharmacists can play an important role in the treatment team of severe patients to improve the rational use of antibiotics.
7.Case analysis of acute eosinophilic pneumonia caused by recombinant human granulocyte stimulating factor (rhG-CSF)

Objective To investigate the role of clinical pharmacists in identifying adverse drug reactions (ADR), to draw clinical attention to the possibility of drug-induced lung injury caused by rhG-CSF, and distinguish them from infectious diseases. Methods A case of rhG-CSF induced acute lung injury was analyzed. After analyzing the relationship between rhG-CSF and acute eosinophilic pneumonia, exploring the possible mechanism, in combination with the patient's condition, the clinical pharmacist put forward the suggestion for the treatment of the disease. Results After receiving rhG-CSF, the patient's eosinophils increased, the pneumonia was aggravated, and the effect of anti-infection treatment was poor. Eosinophils pneumonia associated with rhG-CSF was considered. The patient's pulmonary symptoms improved after treatment with glucocorticoid in combination with withdrawal of antibiotics and antiviral drugs, and eosinophil returned to normal. Conclusion rhG- can cause rare eosinophilic pneumonia. The clinical pharmacist's participation in clinical treatment can help to identify drug-induced diseases, reorient the direction of treatment and ensure the success of clinical therapy.
