1.Expression and significance of high mobility group box 1 protein in BALF of patient with COPD

Objective To preliminarily investigate the effect of high mobility group box 1 (HMGB1) in the pathogenesis of chronic obstructive pulmonary disease (COPD) by detecting the HMGB1 level in bronchoalveolar lavage fluid(BALF) of the COPD patients and analyzing the relation between HMGB1 and airway limitation .Methods The research subjects were divided into the COPD group (stable COPD ,20 cases) and control group[20 cases of upper airway cough syndrome(UACS)] .All subjects were performed the bronchoscopic examination as well as bronchoalveolar lavage .The total cell counting and percentage of neutrophilic granulocytes in BLAF were detected and compared between the two groups .The HMGB1 ,IL‐1βand TNF‐αlevels in BLAF were detected by using ELISA and compared between the two groups .The relation between HMGB1 with IL‐1βand TNF‐αand the rela‐tion between HMGB1 with pulmonary function(FEV1% predicted value) in the COPD group was analyzed .Results The total cell counting and percentage of neutrophilic granulocytes of BLAF in the COPD group were higher than those in the control group (P<0 .01) .The levels of HMGB1 ,IL‐1β,TNF‐αof BALF in the COPD group were higher than those in the control group(P<0 .01 , P<0 .05 ,P<0 .01) .In the COPD group ,the HMGB1 level in BALF was positively correlated with the IL‐1βand TNF‐αlevels(r=0 .79 ,P< 0 .01 ;r= 0 .48 ,P< 0 .05) ,while negatively related with the pulmonary function value(FEV1% predicted value ,r=-0 .70 ,P<0 .01) .Conclusion HMGB1 participates in and promotes the occurrence and development of COPD airway inflamma‐tion and the HMGB1 level in BALF is correlated with the severity of COPD .
2.Relationship between the cervical vertebra sickness with partial upper limb edema and the operation to reduce the cervical vertebra pressure

[Objective]To discuss the relationship between partial upper limb edema in the cervical spondylosis patients and the decompression operation.[Method]Ten cervical spondylosis cases with partial upper limb edema were reviewed,including 7 cases of cervical spondylotic myelopathy,2 cases of cervical radiculopathy and 1 case of OPLL(ossification of the posterior longitudinal ligament).Seven cases were treated with subtotal vertebrectomy and anterior internal fixation,one with subtotal vertebrectomy and anterior discectomy,2 cases with posterior decompression and fusion.All the patients were available for follow-up to evaluate the decay of the edema.[Result]All the 10 patients' edema disappeared with the duration from one day to over half a year.The treatment of anterior surgery made patients' edema disappear quicker than that of posterior surgery.[Conclusion]The occurrence of the upper limb edema in the cervical spondylosis patients is related to do with the irritated sympathetic nerve.The edema can be reduced in short term after the decompression operation.

MicroRNAs(miRNAs) is a class of small endogenous noncoding RNAs with 22 nucleotides.They can negatively regulate gene expression by inhibiting target mRNAs′translation or degrading mRNAs .Up to now, it has been proved that miRNAs play an important role in cellular proliferation , apoptosis , cell differentiation , and embryonic development .In this article , mechanism and function of miRNAs , principal methodologies in the research of miRNAs and the latest development of miRNAs in early embryonic de -velopment are discussed .

7.Application of Diffusion-weighted MR Imaging and Diffusion Tensor Imaging in Brain Tumor Diagnosis and Evaluation

Objective To discuss the application of DWI and DTI in diagnosis and evaluation of brain tumor. Methods Combining with literatures, the application of DWI and DTI in brain tumor diagnosis, differential diagnosis, tumors classi- fication, preoperative evaluation, operation plan establishment, follow-up role of treatment were reviewed and evaluated. Results The DWI and DTI were available to brain tumor diagnosis, differential diagnosis and tumor grade. The DTI could be clearly shown the influence of tumor with white matter quality, it was important in operation plan establishment. The combination of intra-operative MRI, functional MRI and DTI was valuable for brain tumor resection. In the meantime, the treatment response of tumor and the changes of treatment-induced white matter were accurately assessed by DTI. Conclu- sion With the MRI hardware and software technology development, the DWI and DTI imaging information can be provided important value in brain tumor pathological changes diagnosis, evaluation, therapeutic planning and post treatment assess- ment.
8.CT and MR diagnosis of hepatic hydatidosis

Objective To observe the CT and MR features of hepatic hydatid cyst, and to evaluate the diagnostic value of CT and MR. Methods CT and MR images were retrospectively analyzed in 42 patients with pathologically proved hepatic hydatid diseases, and the relative articles were reviewed. Results Hepatic echinococcosis granulosus occured in 85.71% patients, among which ring like pattern calcification, the ribbon calcification, multiple daughter vesicles and detached membranes within cyst were the main findings. Hepatic echinococcosis alveolaris occured in 14.29% patients, whereas the main finding included calcification, small vacuoles, peninsula sign, geographic sign and colliquative necrosis. For the diagnosis of hepatic hydatidosis, MR was better than CT in the judgement of the cyst wall, detached membranes and fibrous septum, but was inferior to CT in displaying calcification sign. Conclusion Hepatic hydatidosis can be diagnosed either with CT or MR, CT is superior to MR in integrated diagnosis of this disease.
9.Analysis and state of HCV genotype 6a infection

Objective: To investigate the infection state of hepatitis C virus genotype 6a in China.Methods: Three(95,126,150)HCV genotype 6a serum samples were identified by digesting 5′NCR with compound enzyme method.Then,HCV 5′NCR and NS5B fragments were amplified from these samples by RT-PCR assay and sequenced.The phylogenetic trees of the samples were analyzed and compared with 24 HCV complete gene sequences from GenBank.Results: The sequencing reports on 5′NCR showed "CA" bases in 3 serum samples(95,126,150) were inserted into-145 site,and the sequences of 3 serum samples had the highest homology with sequence Y12083(0.934,0.930,and 0.926,respectively).The results of the phylogenetic trees suggested these 3 serum samples belonged to HCV genotype 6a.The sequencing reports on NS5B showed the 3 serum samples also had the highest homology with HC-J4(0.934,0.930,and 0.926,respectively),and the results of the phylogenetic trees suggested these 3 serum samples belonged to HCV genotype 1b.To exclude the influence of amplification efficiency of primers,NS5B fragments were amplified by HCV genotype 6a specific primers and no amplification products appeared.Conclusion: There are different results of HCV genotype by analyzing 5′NCR and NS5B in 3 samples infected with HCV genotype 6a.It may be related with gene recombination.It suggests HCV genotype should be analyzed on more than two regions.
10.Utilization of Uracil-DNA glycosylase for combining reverse transcription and anti-contamination with polymerase chain reaction in hepatitis C virus

Objective: To develop a hepatitis C virus(HCV) reverse transcription-polymerase chain reaction (RT-PCR) assay using Uracil-DNA glycosylase (UDG) for amplicon contamination control and evaluate the temperature and UDG concentrations for anti-contamination. Methods: In this new HCV RT-PCR assay, reverse transcription, UDG anti-contamination and the first PCR were carried out at the same time. The layer candles were used to prevent the contamination in the second PCR. dU-DNA was used as quality control for UDG anti-contamination and templates to determine the sensitivity of the new HCV RT-PCR assay. HCV cDNA was detected by DNA enzyme immunoassay (DNA-EIA). Results: Complete degradation of amplicon DNA was observed on the conditions of 0.2 u UDG per reaction volume respectively at 37 ℃ and 42 ℃ for 40 min. The anti-contamination condition also could eliminate all detectible dU-DNA, including the highest concentration of amplicon DNA.The 1∶104 dilution of the HCV RNA sample containing 2.110?105copies/mL copies of RNA could be detected. Conclusion: Our results indicate that this new RT-PCR assay can control the contamination stringently and is sensitive as well.
