1.Clinical analysis of deep fungal infections in ICU hospital

ObjectiveTo investigate the clincal status and countermeasures for intensive care unit (ICU) hospital deep fungal infections.Methods108 cases of ICU hospital deep fungal infections patients were divided into comprehensive treatment group and converntional treatment group,54 cases in each group.The morbidity and treatment effects were observed.ResultsThe cure rate of observation group was 96.3%,significantly higher than 79.6%in control group.The observation group complication rate was 9.2%,significantly lower than the control group 20.4%,the difference was significant (P < 0.05 ).ConclusionIn the ICU nosocomial fungal infection treatment,the scientific and effective means of comprehensive prevention and control best practices,could significantly improve patient outcomes,improve treatment,reduce hospital stay and reduce complications.
2.Effect of Emotion Regulation on Anxiety in Patients Undergoing Functional Endoscopic Sinus Surgery under Acupuncture-drug Compound Anesthesia

Objective To observe the effect of emotion regulation on anxiety in patients undergoing functional endoscopic sinus surgery (FESS) under acupuncture-drug compound anesthesia.Method Ninety patients who were going to receive FESS were randomized into group A, B and C, 30 cases in each group. Acupuncture-drug compound anesthesia [electroacupuncture at Yingxiang (LI 20) and Yintang (GV 29)] was adopted in the three groups, ordinary doses of anesthetic drugs for group A and B (1% Tetracaine hydrochloride injection 100 mg+2% Lidocaine hydrochloride injection 0.1 g+Adrenaline hydrochloride injection 2 mL) and reduced doses for group C (1% Tetracaine hydrochloride injection 70 mg+2% Lidocaine hydrochloride injection 0.1 g+Adrenaline hydrochloride injection 2 mL). Conventional nursing was applied to group A, while specific emotion regulation was additionally adopted in group B and C during the perisurgical period. The three groups were evaluated by using Zung's Self-rating Anxiety Scale (SAS) prior to the surgery, 24 h and 48 h after the surgery, and the satisfaction ratings of the patients towards the surgery, anesthesia and nursing were compared.Result The SAS score 48 h after the surgery was significantly different from that before the surgery in group A (P<0.05). The SAS scores 24 h and 48 h after the surgery were significantly different from the score before the surgery in group B and C (P<0.05); the score 48 h after the surgery was significantly lower than that 24 h after the surgery in group B and C (P<0.05). 24 h and 48 h after the surgery, the SAS scores in group B and C were significantly different from those in group A (P<0.05). The satisfaction ratings of nursing in group B and C were significantly higher than the rating in group A (P<0.05). Conclusion Specific emotion regulation can effectively mitigate the anxiety of patients undergoing FESS under acupuncture-drug anesthesia, and improve the satisfaction rate of patients towards nursing.
3.Gallbladder emptying and gastrointestinal hormones in functional dyspepsia

Objective To study the role of plasma levels of gastrointestinal hormones and gallbladder kinetics in the pathogenesis of functional dyspepsia. Methods Thirty two patients with functional dyspepsia (FD) and 20 normal subjects as controls were studied. Gallbladder ejection fractions (GBEF) were determined with 99m Tc radionucleography, and plasma levels of motilin (MTL)、cholecystokinin (CCK)、 vasoactive intestinal peptide (VIP) and somatostatin (SS) were measured with radioimmunoassay. Results Both preprandial and postprandial GBEF and MTL levels were markedly lower in FD group than those in control group ( P 0.05), while the levels of postprandial CCK were significantly decreased in FD group ( P 0.05). Conclusion (1) Patients with FD might have a decreased level of preprandial and postprandial gallbladder emptying. The decrease of plasma levels of MTL、CCK and the elevation of VIP might be the cause of gallbladder slow emptying in FD. (2) Radionucleography is non invasive and safe. It can be used in the quantitative analysis and dynamic study of gallbladder and Oddi sphincter functions, and provide accurate, physiological results.
4.Effects of Sodium Aescinate on ET and CGRP Contents in Gerbils with Cerebral Ischemia/reperfusion In-jury

Objective:To investigate the effects of sodium aescinate on endothelin ( ET ) and calcitonin gene -related peptide ( CGRP) contents in the gerbils with cerebral ischemia/reperfusion injury ( CI/R) . Methods:The gerbil model of CI/R was prepared by bilateral common carotid arteries ligation for 10 min followed by 2-hour reperfusion. Sodium aescinate (i. p. , 10, 20 and 40 mg· kg-1 ) was administered once a day for 3 days before the operation and once every 1 h after the operation, respectively. The levels of ET and CGRP in brain tissue homogenate were determined by a radioimmunoassay method. Results:Sodium aescinate significantly in-hibited the level of ET (28. 69-37. 03 ng·L-1) in the brain tissue of gerbils with CI/R compared with the model group (P<0. 05 or P<0. 01), while showed no evident influence on CGRP (P>0. 05). Conclusion:Sodium aescinate has obvious protective effects a-gainst CI/R in gerbils, which may be due to its inhibitory action on ET levels in brain tissue.
5.Ruptured Achilles tendon repaired with hernia repair materials using a wrap method

BACKGROUND:The incidence of complications folowing conventional repairing for the ruptured tendon is up to 73%, such as delayed healing, adhesion, secondary rupture. These complications severely affect ankle joint function of the patients. OBJECTIVE:To reduce the complications folowing the repair and investigate the role of hernia repair materials in the repairing of ruptured Achiles tendon. METHODS:Eleven patients who underwent repair of ruptured Achiles tendon were retrospectively studied. Among them there were eight males and three females with the mean age of 42 years (26-58 years). Three cases were injured in left side and eight cases were in right side; four cases were open injury by sickle-induced damage and seven cases were closed injury by exercise-induced damage. The hernia repair materials after pruning was applied with a “wrap” method to strengthen the anastomosis repair of Achiles tendon after the conventional repair surgery had been completed. At the time of folow-up, al patients were observed for postoperative local appearance and function recovery, and assessed with Arner Lindholm standards. RESULTS AND CONCLUSION:The folow-up interval was 6-45 months (average 24.5 months). Two cases were lost to folow-up. According to the Arner Lindholm standards, the effects were excelent in seven cases (78%) and good in two cases (22%). Local appearance and function recovered wel postoperatively, and no complications occurred such as wound infection, secondary rupture and so on. A “wrap” method with hernia repair materials is suggested to strengthen the anastomosis repair of ruptured Achiles tendon after the conventional repair surgery is completed, it can significantly promote the healing of Achiles tendon ruptured end anastomosis, prevent Achiles tendon recurrent ruptures, local adhesion and infection.
6.Correlation between Glutathione S-transferase Polymorphisms and Sperm DNA Integrity in Male Patients with Idiopathic Infertile

Objective to investigate the correlation between glutathione S- transferase gene M1,t1 and P1(GStM1,GStt1 and GStP1)poly-morphism and sperm DNA fragmentation index(DFI)in male patients with idiopathic infertile. Methods the study included 246 male patients with idiopathic infertility. Polymerase chain reaction(PCR)and polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) were used to identify the genotype of GStM1,GStt1 and GStP1,respectively. Sperm DFI was analyzed by flow cytometry with acridine orange staining. Results the sperm DFI was significantly higher in GStt1 gene null type group than in GStt1 gene wild type group(P < 0.01);and there were no significant differences in GStM1 and GStP1 gene mutation type groups when comparing with the wild type group(P > 0.05). Conclusion GStt1 gene deletion was positive associated with the sperm DFI in male idiopathic infertile patients.
7.Therapeutic effect of granulocyte colony stimulating factor on CCl4 induced chronic liver injury in mice

Objective To observe the therapeutic effects of recombinant human granulocyte colony stimulating factor(rhG-CSF)on CCl4 induced chronic liver injury.Methods Male BALB/C mice were randomly allocated into treatment and control groups.The mice model were established by injection with daily for 7 days,while the control mice were received the same volumes of saline.The mice were sacrificed to get weight,liver mass and spleen mass.The count of CD34+ cells and Thy-1+ cells were analyzed by flow cytometry and immunohistochemical staining,respectively.Results The ratio of liver/spleen was 15.94±1.20 and 10.52±0.66 on day 8 and 15 in treatment group,respectively,while those were 7.14±1.68 and 8.31±1.71 in control group,respectively(all P value<0.05).But there was no significant difference in body weight and liver mass between two groups(P>0.05)The concentration of album in treatment group was raised rapidly on day 15.The concentrations of alanine aminotransferase (ALT),aspartate aminotransferase(AST),hyaluronic acid(HA)and laminin(LN)on day 30 were significantly lower in treatment group compared to control group(P<0.05).There was significant difference in score of liver fibrosis on day 30 between two groups(treatment group:5.49±2.16,control:8.74±1.86,P<0.05).The number of CD34+ cell and Thy-1+ in treatment group(on day 8:9.54±2.24 and 5.10±1.25 and on day 15:8.18±1.93 and 7.53±1.39,respectively)were higher than those in control group(on day 8:5.40±0.99 and 3.25±0.75;on 15 days:4.46±0.77 and 3.35±0.86,all P value<0.05).Conclusion The rhG-CSF may improve the reparation of chronic liver injury,and may provide a novel method in treatment of liver fibrosis.
8.Laboratory analysis in patients with severe acute respiratory syndrome after ten years of convalescence in Tianjin

Objective To investigate the changes of blood test indexes in patients with severe acute respiratory syndrome (SARS) after 10 years of rehabilitation, and to evaluate the effects of SARS on blood system. Methods Twenty-five cases of SARS confirmed in our hospital (SARS group) and 25 healthy personnel (control group) were included in this study. Values of blood routine test, liver and kidney function, blood lipids, blood glucose, thyroid function, tumor marker and T cell subset index were compared between two groups. Results The serum level of calcium was higher in SARS group than that of control group (mmol/L:2.32 ± 0.08 vs. 2.29 ± 0.08,t=2.072,P<0.05). The level of thyroxine was also higher in SARS group than that of control group (nmol/L: 119.95 ± 29.09 vs. 96.39 ± 14.26, t=3.681,P<0.05), but the results were in the normal range, there was no clinical significance. The level of neuron specific enolase (NSE) was higher in SARS group than that of control group (μg/L, 10.368 ± 3.684 vs. 3.282 ± 1.828, t=9.012, P<0.05), in which only 2 cases were slightly higher than the normal range, but did not exceed more than 1 times of the normal value. The imaging was only part of the old change, and there was no clinical significance. There were no significant differences in other results between two groups. Conclusion Virus of SARS cause no long-term effects on blood system, liver and kidney function, thyroid function and immunity in patients with SARS, and do not increase the probability of cancer.
9.Expression of Heat Shock Protein-70 in Paraquat-induced Pulmonary Inflammation Response

Objective To investigate the expression of heat shock protein-70(HSP70)in acute lung injury induced by paraquat poisoning.Methods A total of 72 Wistar rats were randomly divided into 3 groups(n=24 each group)and intraperitoneally injected with normal saline in group A,20 mg/kg paraquat in group B,and 40 mg/kg paraquat in group C.The lungs of rats were excised 1,3,6,12,and 24 hours after the injection,respectively.The expression of HSP70 in the lung tissue was detected by immunohistochemistry and Western blot.Results Paraquat induced acute lung injury in groups B and C.In group B,the expression of HSP70 increased at 2 hours,reached the peak at 6 hours,decreased at 12 hours,and returned to the baseline level at 24 hours.In group C,the expression of HSP70 increased at 1 hour,reached the peak at 3 hours,and returned to the baseline level at 24 hours.Conclusion Paraquat-induced acute lung injury may contribute to the enhanced expression of HSP70 in the bronchus,bronchiole,and alveolar epithelial cells,which indicates that HSP70 has a protective effect on lung injury.
10.Inhibitory effect of juglone on migration and invasion of glioma cell line U-87 MG

Objective To investigate the effects and mechanisms of pin1(peptidyl-prolyl isomerase 1) inhibitor ju?glone on migration and invasion in glioma cell line U-87 MG. Methods Glioma cells were treated with juglone at 0, 0.8, 1.6 and 3.2μmol·L-1. Wound-healing assay and invasion assay were performed to examine the inhibitory activity of ju?glone on glioma cell line U-87 MG. Western blot was used to analyze the protein expression of β-catenin, VEGF, MMP-2 and MMP-9. Results The wound-healing assay showed that the wound-healing rate in juglone-treated groups was 46.04%±6.25%and 30.05%±13.35%at concentrations of 1.6μmol·L-1 and 3.2μmol·L-1 , repectively. Juglone treat?ment significantly reduced the wound-healing rate compared with controls (P<0.05). Transwell invasion assay showed that the number of invaded cells in juglone–treated groups was 103.67 ± 5.69 and 77.33 ± 7.77 at the concentrations of 1.6μmol·L-1 and 3.2μmol·L-1 , respectively. Juglone treatment significantly reduced cell invasion compared with con?trols (P<0.05). Treatment with juglone significantly down-regulated the expression levels ofβ-catenin, VEGF, MMP-2 and MMP-9 in U-87 MG cells in a dose-dependent manner. Conclusion The present data suggests that juglone has a significant inhibitory action on cell migration and invasion through down-regulation of theβ-catenin and its downstream VEGF, MMP-2 and MMP-9 protein expressions in glioma cell line U-87 MG.
