1.IMMUNOHISTOLOGICAL PATTERN OF TGF-?1 IN CHICKENCALLUS IN EXPERIMENTAL FRACTURE HEALING

Objective To observe transforming growth factor-?1(TGF-?1) in chicken callus. Methods The right humerus fracture models of 16 New Roman hens, at the age of 6-month old, about 1.75kg in weight, were established under no germ operational condition. TGF-?1 of the tissue samples procured from callus on 3d,6d,10d and 15d of fracture healing were examined by immunohistochemical method. Results Histochemical study shows that the immunostaining of TGF-?1 exists in blood cells on 3d, connective tissue cells and extracellular matrix on 6d, cartilage on 10d and 15d. Toll-like receptor 5 (TLR5) is one of the pattern recognition receptors and recognizes the flagellin protein of bacteria.It activates innate immune responses and induces production of a series of cytokines.TLR5 functions as a bridge linking innate and adaptive immunities.It is known that TLR5 plays an important role in the occurrence and development of certain infectious diseases.This review summarizes the relationships of TLR5 polymorphisms with the development of infectious diseases and discusses the possible pathogenesis.
3.The Experimental Study on Liqihuatanquyu Formula(LF)Against NASH in Rats

Objective To investigate the effect of LF on blood biochemistry,cytokines in rats with NASH in order to approach possible mechanism of LF against NASH.Methods 50 male SD rats were randomly divided into the normal control group(10 rats),the model group(10 rats),the Essentinale group(10 rats)、the LF high dose group(10 rats)and the LF low dose group(10 rats).Except for the normal control group which was raised by standard animal feeds,others were all raised by high fat feeds to make NASH model and administered by Essentinale,high dose LF and low dose LF for 12 weeks.Hepatic lipid contents,plasma TG,CHO,HDL-C,LDL-C,ALT,AST and TNF-?,FFA,Leptin concentrations were evaluated,as were histopathological changes in the liver.Results In this NASH model,the liver FZ(liver weight /body weight),plasma CHO,LDL-C,ALT,AST and TNF-?,FFA,Leptin concentrations were higher than that in normal control group,but the levels of serum HDL-C and TG were reduced.In addition,the histopathological showed increased fatty droplets within hepatocytes and liver inflammation.The levels of serum ALT,AST,HDL-C,TNF-?,Leptin,FFA and hepatic CHO,TG contents in the LF high dose group and LF low dose group were reduced compared with that in the model group,while the levels of serum TG,HDL-C were enhanced remarkably.In addition,liver inflammation was improved evidently.Conclusions Treatment with LF might improve non-alcholic steatohepatitis,because LF inhibits the elevation in the level of plasma cytokine.
4.Construction, Screening and Antibody Structure Homology Modeling of Phage Single Chain Variable Fragment Library Against Ofloxacin

To construct a library of mouse single chain variable fragment (scFv) antibody against ofloxacin using phage display and recombinant antibody technique, specific anti-ofloxacin scFv was screened and 3D structure was homology modeling. Total RNA was extracted from hybridoma cell of ofloxacin mAb, and was used to amplify VH and VL gene by RT-PCR using random primer. Then they were linked by a DNA linker encoding (G1y4 Ser) 3 as VH-linker-VL sequence forming scFv by SOE(splicing by overlap extension) PCR. These fragments were inserted into phage T7 after double digestion and transformed with host bacteria BLT5403. 3 ×105 pfu / ml single chain antibody phage libraries were successfully constructed. Four positive phage scFv clones were screened by direct competitive ELISA after four times of enriched procedure in the order of adsorption-elution-amplificatio, 3D structure of specific scFv was homology modeling finally. This research lays a foundation for further massive expression of anti-ofloxacin scFv.
5.Discussion in nursing service and charge

Objective To discuss nursing service and charge in order to establish standard and perfect nursing cost accounting system. Methods Nursing charge was described through analyzing the nursing technology during treatment of long-term hospitalization patients and no charge of disposable items. Results Nursing work was nearly a no-reward technological service, the proportion and the standard of nursing charge was low, the charge of nursing items was incomplete, labor cost was neglected and nursing value could not be really embodied. Conclusions Nursing managers should strengthen the nursing cost management in order to establish perfect and standard nursing cost accounting system.
6.A case report of the ketogenic diet for severe cerebral complications after ependymoma operation

The ketogenic diet(KD) treatment program planned for a 2 years old and 5 months boy suffered from severe cerebral complications after ependymoma operation in the Children′s Hospital of Nanjing Medical University at December 2018 was analyzed retrospectively.The patient developed various complications, including cerebral hemorrhage, hydrocephalus and hernia after the operation of ependymoma, with coma, fever and convulsion, and 1 month of anti-infection and antiepileptic treatment was ineffective.After 2 weeks of 4∶1 KD, fever and convulsion were effectively relieved without obvious side effects.One month later, the patient regained consciousness, demonstrating that KD can be applied to treat severe cerebral complications after ependymoma operation.
7.Comparison of occurrence of postoperative cognitive dysfunction in elderly patients using different anesthetic methods

Objective To compare the occurrence of postoperative cognitive dysfunction (POCD) in elderly patients using differentanesthetic methods.Methods Ninety-three ASA Ⅱ or Ⅲ patients,aged ≥ 65 yr,weighing 45-67 kg,scheduled for artificial femoral head replacement,were randomly divided into 2 groups:general anesthesia (group G,n =47) and combined spinal-epidural anesthesia group (group S-E,n =46).In group G,anesthesia was induced with iv injection of midazolam 0.1 mg/kg,propofol 2 mg/kg,fentanyl 3-5μg/kg,and vecuronium 0.1 mg/kg,and maintained with continuous infusion of propofol 2-3 mg· kg-1 · h-1,intermittent iv boluses of fentanyl 1 μg/kg and vecuronium 0.04 mg/kg and inhalation of 1.5%-2.0% isoflurane.In group S-E,hyperbaric 0.5 % ropivacaine 2 ml was injected into the subarachnoid space over 20 s,the patients were kept in the original position for 15 min,the level of anesthesia was simultaneously adjusted to below T8 on the operated side,and 0.5 % ropivacaine 3-5 ml was injected into the epidural space when needed during operation.Cognitive function was assessed by mini-mental state examination at 24 h before anesthesia and 24 and 72 h after operation.Venous blood samples were collected for determination of plasma amyloid-beta levels by ELISA.Results Compared with group G,the incidence of POCD at 24 h after operation and level of plasma amyloid-beta were significantly decreased in group S-E (P < 0.05).Conclusion Elderly patients are more likely to develop POCD under general anesthesia than under combined spinal-epidural anesthesia.
8.MR Imaging of Synovial Lesions in Early Rheumatoid Arthritis

Objective To assess the diagnostic value of MR imaging of the wrist synovitis in early rheumatoid arthritis.Methods 60 wrists in 30 patients underwent MR imaging examination,including T1WI,T2WI,STIR and contrast-enhanced fat-suppressed 3D fast-gradient-echo sequence.MRI manifestations were analysed in comparison with clinical and laboratory test data.Results All changes including synovial hyperplasia and pannus formation(30 cases),articular cartilage destruction(21 cases),bone erosion(16 cases),joint effusion(25 cases) and tendon lesion of wrists could be showed clearly by MRI.The signal intensity of the pannus before and after administration of contrast agent and the enhanced curve of the pannus could be used to assess the lesions whether or not activation.Conclusion MRI is helpful in diagnosing early rheumatoid arthritis.
9.Silicosis:The Evaluation of Pulmonary Function Damage with Multi-slice Spiral CT

Objective To investigate the value of MSCT in evaluating pulmonary functional changes in silicosis.Methods 56 cases of silicosis and 10 healthy people(as control group) underwent inspiratory and expiratory MSCT scans and pulmonary functional test one week later.The CT findings including silicotic nodules,large opacity,emphysema,reticular opacities,bronchiectasis and air trapping were recorded and graded subjectively on CT images.Air retention and emphysema were quantified using the software of Pulmo.CT scores were correlated with spirometric findings by using spearman rank correlative analysis in comparison with pulmonary functional index.Results The scores of CT features except for reticular shadows were of significant difference between silicotic group and control group,but not between simple silicotic group and complex silicotic group.The scores of air retention and emphysema on CT were significantly negative correlation.The reticular shadows were of positive correlation with FEF 75%,FEF 50% and FEV1,but on correlation with FEV1/FVC.There were no correlation between the scores of silicotic nodules,bronchiectasis,large shadows and pulmonary functional test.There was obvious correlation between air retention,the scores of emphysema on CT and the index of pulmonary function in obstruction of small airway.Conclusion MSCT is of important value in evaluating the damage of pulmonary function in silicosis.
10.Effect of Xinxibao on the humoral immunologic functions of patients with rotavirus enteritis

Objective To discuss the effect of Xinxibao to rotavirus enteritis sufferers' humoral immunologic functions. Methods 260 cases rotavirus enteritis sufferers' immunoglobulin and its alexin were measured. Except conventional therapy, the treatment group plus the Xinxibao to cure, and observe two sufferer teams' clinical curative effect and the change of immunologic function. Results The acute stage sufferers' blood serum IgG, IgM, IgA, C3 were lower obviously, C4 didn't have obvious change, the activity ratio of the treatment group(81.6 % ) is obviously higher than the control group (63.5 % ). At the same time, the treatment group IgG, IgM, IgA, C3 rose obviously than before,C4 had no change. Conclusion The rotavirus enteritis sufferers' immunologic functions are disorder,Xinxibao is useful to shorten the treatment of the sufferers, and strengthen sufferers' immunologic functions in the restoration stage.
