1.Contrast analyse of risk factors on intracerebral hemorrhage and coronary heart disease in Tongliao city in Innermongolia

Objective To investigate the distribution of rick factors between intracerebral hemorrhage(ICH) and coronary heart disease (CHD) in Tongliao city in Innermogolia. Methods Medical records(departments of medical neurology and cardiovascular internal medicine in 2003~2005) were randomly selected from 6 general hospitals above the second class in Tongliao city Innermogolia .All the risk factors of the cartain diseases were carryed on retrospective investigation analysis. Results All the survey index have significantly statistical difference in the basic data. Single-factor non-conditional Logistic regression analysis showed 10 risk factors including the gender, age, nation, smoking, hypertension, history of hypertension, alcohol drinking, glucose(GLU), history of diabetes and triglyceride(TG) have significantly statistical difference. These 10 risk factors were taken into multifactor stepwise regression model. 8 risk factors( gender, age, smoking, history of hypertension, history of diabetes,GLU,TG and hypertension) had significantly statistical difference. Conclusions Compared with CHD, the influence of age, smoking, history of diabetes and TG are lower, and the influence of the gender, the history of hypertension, GLU and hypertension are higher in ICH.
2.Regulation impact of vitamin C in DC function

Objective:To study the influence of vitamin C ( VC) on dendritic cells ( DC) ,and detect DC maturation,to provide a feasible method and thought for quickly preparating DC vaccines.Methods:Collected the peripheral blood (about 50 ml) from healthy volunteers,and isolated peripheral blood mononuclear cells with lymphocyte separation medium and obtain DC.With stimulating with different concentrations of VC for (24 h),then washed with PBS,and set up blank control group (V0).The expression of DC surface co-stimulating molecules CD80/86 and HLA-DR, CD40 was detected by flow cytometry.By setting the concentration gradient and time gradient, exciting optimal concentration and stimulating time of VC on DC, and analyzed the reasons of VC promoting DC maturation.Results:VC could effectively stimulate DC,CD80/86 expression had significantly increased contrast to the blank control group (V0).And the experiments show that VC’s best stimulating concentration was 1 mmol/L,and on the third day,the CD80/86 expression rate of VC group was (78.6±4.6) %,and blank control group V0 was (34.1±5.7) %.DC surface HLA-DR expression:VC (56.8± 4.4) %,blank control group V0 (25.4 ±4.7) %,the difference between two groups was statistically significant,P<0.05.CD40 and CD40L expression and results show that VC 2.5 mmol/L group of CD40 expression rate up to (59.3±3.7) %,while V0 group was only (11.1 ±2.4) %,that illustrate VC could significantly regulate CD40 expression on DC surface,but CD40L not reflect.Fluorescence mi-croscope results showed that DC’ s antigen catching ability was also significantly promoted.Conclusion:VC can significantly regulate DC maturity,and may up regulate CD40,thus promoting the express of CD80/86 and HLA-DR.When the concentration is 1 mmol/L,VC expresses the strongest regulation function on DC.

4.Characterization of a Novel AP2/EREBP Transcription Factor TSH1 Specifically Binding to GCC Elements

TSH1, a novel transcription factor in rice containing conserved AP2/EREBP domain, was screened from the EST database. The specific interaction of TSH1 and DNA elements was detected by two kinds of traditional methods, electronic mobility assay and yeast one-hybrid assay, which suggested that THS1 specifically binds to GCC element in vitro and in vivo. Furthermore, It was confirmed by atom force microscopy (AFM) that TSH1 binds to GCC element (core sequence GCCGCC) not DRE element (core sequence GCCGAC), and the single molecular force between TSH1 and GCC element was quantitatively measured. The molecular force of TSH1 with GCC element is (83.9 ?2.2) pN. This interaction can be observably blocked by the dissociate TSH1 protein in the solution. These above results proved that TSH1 recognizes and binds to GCC with specifically interaction, which also demonstrate the high sensitivity of the AFM measurements for the single base substitution of the GCC core sequence greatly reduced the binding affinity. Comparing these methods for identification the binding of transcription factor with its DNA element, it was considered that AFM is expected to be a sensitive and reliable method that offers valuable information for the characterization of transcription factors.
5.The effects of different amounts of iodine intake on the immune status of patients with Graves disease

ObjectiveTo investigate the effects of different amounts of iodine intake on the cellular and humoral immune in Grave's disease (GD) patients.MethodsThe clinical GD cases were diagnosed by thyroid fine needle Cytology examination.Patients in GD group are divided into GD group Ⅰ and GD group Ⅱ based on the median of urine iodine.The blood levels of FT4,FT3,TSH,TPOAb,TGAb,TRAb and TNF-t were detected.The difference and association of these parameters between these groups were analyzed.ResultsThe TNF-αt level in GD Ⅰ group was higher than that of GD Ⅱ group( P > 0.05 ) ;The average level of TRAb of GD Ⅰgroup ( [ 1.4 ±0.2 ] U/L) were higher than that of GD Ⅱ group ( [ 1.2 ± 0.1 ] U/L) ( P < 0.05 ) ;The positive rates of TGAb and TPOAb of GD Ⅰ group were higher than that of GD Ⅱ group ( P < 0.05 ).The percentages of patients with high level of TGAb and TPOAb in GD Ⅰ group ( 78.9% 、84.2% ) were higher than that in GD Ⅱ group (50.0%,62.5% ) ( x2 =6.79,10.70,P <0.05 ) ; Analysis showed a linear positive correlation of TNF-αwith TRAb and TPOAb ( r is 0.489 and 0.563,P < 0.01 ).ConclusionIodine is an important factor to the development of Graves disease.Excessive iodine intake will exaggerate the GD condition and patients with GD should be controlled for iodine intake.
6.Effect of dominant negative Ⅰ?B? transfection on NF-?B pathway and cell motility in breast cancer

AIM:To observe the effect on NF-?B pathway and cell motility in breast cancer cell lines after transfection of dominant negative I?B? plasmid.METHODS:After stable transfection of mutant I?B? plasmid into highly metastatic breast cancer lines MDA-MB-231 and MDA-MB-435,we detected NF-?B binding activity by EMSA,cell growth ability by cell growth curve,colony forming test,and cell motility by millicell-PCF chamber.RESULTS:Constitutive activities in MDA-MB-231 and MDA-MB-435 were observed.Stable transfection of a dominant negative Ⅰ?B? resulted in downregulation of NF-?B binding activity,thus inhibited cell mobility without significant effect on cell growth.CONCLUSION:Cell migration ability is inhibited in highly invasive breast cancer cells by inhibition of NF-?B pathway in vitro.
7.Clinical study on coracoclavicular ligament reconstruction with hook plate fixation combined with wire anchors anatom

Objective To compare the effects of application of clavicular hook plate combined with wire anchors anatomical coracoclavicular ligament reconstruction and application of clavicular hook plate in the treatment of NeerII distal clavicle fracture and Tossy Ⅲtype~V acromioclavicular joint dislocation. Methods A retrospective analysis of the clinical data from June 2006 to June 2013. Total 73 cases patients suffered with Neer Ⅱtype distal clavicle fractures and Tossy Ⅲtype~V acromioclavicular joint dislocation. Of which , 41 cases were subjected to treatment with clavicular hook plate , 32 patients subjected to treatment of using clavicular hook plate combined with anchors .The incision length, operative time, postoperative complications, length of hospital stay and postoperative 1 month, 6 month shoulder VAS score of two groups were analyzed; the shoulder function of both groups after 1 month, 6 months were assessed by using Constant shoulder function assessment method. Results Surgical incision length and operational time between the two groups were statistically significant (P<0.05), while the amount of bleeding was not statistically significant. All patients were followed up . The two groups did not occur any complications such as loosening, decoupling, acromioclavicular joint dislocation and wound infections. Hospitalization time was 5~14 d (averaged 10d), no significant difference between two groups. 4 the shoulder Constant score and VAS scores showed no significant difference 1 months postoperation; 6 months after hook plate removed, VAS score and Constant shoulder score improved significantly in anchors hook plate group (P<0.05). Conclusion Anatomical coracoclavicular ligament reconstruction by application of hook plate combined with anchors is a good biomechanical model characterized with simple surgery , less trauma and good clinical outcomes , worthy of clinical application.

Shiwei Dangguiyin(SWDGY)is mainly composed of Radix Angelicae Sinensis,Radix Adenophorae,Radix Notogenseng,Radix Bupleuri,etc. Oral administration of SWDGY could significantly inhibit the metatarsal swell- ing eaused by dimethylbenzene in rats,raise the pain threshold in hot-plate test and depress the torsive reaction caused by acetic acid in mice.In vitro SWDGY exerted bacteriostatic and bacteriocidal effects on Staphylococcus aureus,Bacil- lus pyocyaneus,Escherichia coli,Streptococcus A,B and C.It was shown that SWDGY possessed anti-inflammatory,analgesic and antiseptic effects in vitro.In mice LD_(50) of SWDGY by oral administration was more than 840g/kg.Affer cral adminstration in a daily dose of 189.Sg/kg continuously for one month in rats, no toxic reactions appeared,This dosage was 118.6 times as much as the clinical one.
10.Characteristics of microbiota of colonic lavage fluid from patients with diarrhea-predominant irritable bowel syndrome

Objective To investigate the characteristics of the microbiota of colonic lavage fluid from patients with diarrhoea-predominant irritable bowel syndrome (D-IBS).Methods From August 2014 to March 2015,at the Department of Gastroenterology Shanxi Provincial People's Hospital,39 patients with D-IBS were enrolled and during the same period 39 patients with self-reported symptoms such as occasional abdominal discomfort or abdominal pain but were excluded from the diagnosis of irritable bowel syndrome (IBS) were selected as control group.The colonic mucosal tissues and colonic lavage fluid of two groups of patients were collected.DNA was extracted from tissues and colonic fluid.Ten dominant bacteria,including Bacteriodes prevotella,Bacteroidetes,Clostridium,Bifidobacterium,Dialister pneumosintes,Enteropathogenic Escherichia coli,Faecalibacterium prausnitzii,Firmicutes,Lactobacillus and Salmonella,were analyzed by real-time fluorescence quantitative polymerase chain reacticn (RT-PCR).T test and Spearman correlation analysis were performed for statistical analysis.Results The numbers of Bacteriodes prevotella in colonic lavage fluid and the descending colon mucosal tissues of patients in D-IBS group were 3.096 ± 1.462 and 4.278 ± 0.290,respectively,which were lower than those of control group (4.842 ± 1.032 and 5.341 ± 0.433,respectively),and the differences were statistically significant (t =6.093 and 12.657,both P < 0.01).Correspondingly,the numbers of Bacteroidetes were 3.259 ± 0.607 and 3.698 ± 0.554,respectively,which were lower than those of control group (4.085 ± 0.504 and 4.339 ± 0.263,respectively),and the differences were statistically significant (t =6.538 and 6.528,both P < 0.01).The numbers of Clostridium were 5.254 ± 0.522 and 4.471 ± 0.690,which were lower than those of control group (5.949 ± 0.505 and 4.950 ± 0.604,respectively),and the differences were statistically significant (t =5.976,P < 0.0l;t =3.262,P =0.002).The number of Faecalibacterium prausnitzii from colonic lavage fluid of patients in D-IBS group was 2.238 ± 1.845,which was lower than that of control group (3.222 ± 1.450),and the difference was statistically significant (t =2.619,P =0.011).The Firmicutes (r =0.678,P =0.012),Lactobacillus (r =0.358,P =0.034),Bifidobacterium (r =0.418,P =0.042) and Faecalibacterium prausnitzii (r =0.568,P =0.022) from colonic mucosal tissues were all positively correlated with those from colonic lavage fluid.However,Salmonella (r =-0.638,P =0.012) and Enteropathogenic Escherichia coli (r =-0.343,P =0.025) were negatively correlated with those from mucosal tissues.Conclusions An imbalance of the microbiota in colonic lavage fluid is found in D-IBS patients.The numbers of Bacteriodes prevotella,Bacteroidetes,Clostridium and Faecalibacterium prausnitzii significantly decrease.
