Objective To explore the expressions of CD44, CD44V3, CD44V6 and PCNA and the relationship between the expressions and clinical pathologic changes of human osteosarcoma Methods The expressions of CD44, CD44V3, CD44V6 and PCNA in 15 osteochondromas and 40 osteosarcomas were detected immunohistochemically Results (1) The positive expression rates of CD44, CD44V3 and CD44V6 in osteosarcoma were markedly higher than those in osteochondroma (2) The positive expression rate of CD44V6 was higher than those of CD44 and CD44V3 in osteochondroma (3) The expression of CD44V6 was not involved in the pathological type or the prognosis in osteochondroma (4) The expressions of CD44 and CD44V6, particularly that of CD44V6, were closely involved in the PCNA positive rate Conclusion CD44V6 might act as a marker of malignancy of the bone tumors, but the relationship with the prognosis in malignant bone tumors waits to be further studied
2.Isolation of the drug-resistant Staphylococcus aureus strains from burn wound flora and analysis of norA genetic mutation

Objective To investigate the genetic mutation of the norA gene and its promotor from the wild-type drug-resistance Staphylococeus aureus(S.aureus)strains. Methods A total of 10 antibiotic-resistant S.aureus strains were isolated and screened from the burn wound for the sequencing and analysis of the nora gene and its promoter. Results There isolated 87 S.aureus strains from the burn wound flora,which were completely sensitive to vacomycin,highly sensitive to Quinupristin and Nitrofurantoin,but highly resistant to the other antibiotics,even up to91.7% of MRSA.There found the same point mutation(G→A) located at 1 349 sites of the norA gene coding region in all the S.aureus strains,saying that the amino acid was changed from Gly(glycin)to Asp(agpartic acid) in 291 sites.The resetpine reverse test showed that the MICs value of three antibiotics was lowered at various degrees in all 10strains.Conclusion NorA gene mutation is one of the mechanisms for antibiotic-resistance of S.aureus.
3.The expression of CD14 in rat Kupffer cells

AIM: To explore the expression of CD14 in rat Kupffer cells (KCs). METHODS: In rat KCs induced by LPS or the mediators from KCs induced by LPS,the changes of CD14 expression were measured by RT-PCR and immunohistochemistry.The expressions of TNF? mRNA?IL-6 mRNA or the concentrations of TNF??IL-6 were estimated by in situ hybridization and radioimmunoassay,respectively. RESULTS: LPS increased the expression of CD14 in KCs in a dose-dependent fashion (LPS,1 ?g/L-10 mg/L) and in a time-dependent fashion(0.5 h-24 h,peaked at 3-6 hours). While the expression of CD14 in KCs stimulated by the active mediators from KCs which had been exposed to LPS 1 hour were obviously increased. CONCLUSIONS: There was a close relationship between LPS or the active mediators from KCs induced by LPS and the expressions of CD14. It is implied that the increase in CD14 expression may be induced by LPS and the cytokines produced by KCs,it also reveals that there is a auto-regulated loop in CD14 expression.
4.Experimental study on early liver injury and expressions of TNF-alpha mRNA in burn rats with endotoxemia.

OBJECTIVE: To observe tissue distribution and cell localization of TNF-alpha mRNA and its protein and study their role in the pathogenesis of liver injury in burn rats. METHODS: An animal model of rats subjected to 20% TBSA III degree burns combined with intraperitoneal injection of lipopolysaccharide (LPS) was used for this experiment. The changes of hepatic morphology and functions and serum TNF-alpha content and expression and localization of liver TNF-alpha and TNF-alpha mRNA were determined with light microscope (LM) and electron microscope (EM), quantitative analysis, immunohistochemistry (IHC) and in situ hybridization (ISH). RESULTS: It showed that there were sinusoid reaction, KCs activation and degeneration, necrosis of HCs, and platelets aggregation, fibrins deposition and PMNs attachment in sinusoid. The activity of ALT was obviously elevated and ALB content was slightly decreased. The serum content of TNF-alpha showed peak at 3 hours. TNF-alpha was mainly localized in sinusoid endothelial cells (SECs) and Kupffer cells (KCs), and TNF-alpha mRNA was mainly distributed in KCs, polymorphonuclears neutrophils (PMNs) and macrophages (MPs). CONCLUSIONS: It suggests that TNF-alpha mRNA and its protein expression and localization are coincident with the pathological changes of liver injury. TNF-alpha is one of the key cytokines in the pathogenesis of liver injury in burn rats with endotoxemia.
5.Retrospective analysis on acupuncture in treatment of cerebral infarction evaluated with propensity score.

The actual efficacy of acupuncture on cerebral infarction was explored in clinical practice. The retro spective cohort study was adopted to investigate 344 cases via inpatient's medical cases. According to whether acupuncture was received or not, an acupuncture group (207 cases) and a non-acupuncture group (137 cases) were divided. The matching method, regression method and weighting method of propensity score (PS) were adopted, and the efficacy on muscle strength was taken as effect index so that the specific impacts of acupuncture were ex plored on the muscle strength in the patients of cerebral infarction. Before matching, COX regression model and Logistic regression model were used. And PS hierarchical regression, PS inverse probability weighting method (IPTW) and PS standardized mortality weighting method (SMRW) were applied to the analysis on the relationship between the muscle strengthen changes and the total effective rate in the two groups. It was found that the efficacy in the acupuncture group was better than that in the non-acupuncture group, indicating the significant difference (P<0.05). Meanwhile, the rehabilitation therapy also brought the obvious impacts on the efficacy evaluation (OR=2.737, P=0.0055). After PS matching, the Logistic regression model was used to analyze whether acupuncture or rehabilitation therapy impacted the total effective rate of muscle strength. The results showed that the efficacy was impacted apparently with the rehabilitation therapy involved (OR=2.930, P=0.0247). Without the rehabilitation effect considered, the efficacy in the acupuncture group was better potentially than that in the non-acupuncture group, but without significant difference (OR=2. 235, P=0,058 7). All of these indicate that on the basis of routine treatment, without the effect of rehabilitation therapy considered, acupuncture improves in tenden cy of the muscle strength of the patients with cerebral infarction. However, it is expected to increase the study medical cases for further verification.

6.A double-blind RCT of the hemostatic effects of local packing of Nasopore combined with hemocoagulase injection for postoperative management of FESS.

OBJECTIVE:
To compare the hemostatic effects of local packing of Nasopore combined with hemocoagulase injection and local packing of Nasopore combined with saline injection for postoperative management of functional endoscopic sinus surgery by a double-blind, randomized control clinical trial.
METHOD:
Sixty-eight cases of chronic sinusitis needed functional endoscopic sinus surgery were randomly divided into the experimental group of 40 cases and control group of 28 cases, respectively. For the experimental group, 1 U of hemocoagulase dissolved in 0.5 ml saline was injected into Nasopore which was packed into the nasal cavity after operation. For the control group, 0.5 ml of saline was injected. The postoperative bleeding of the two groups were scored by visual analogue scale.
RESULT:
There was statistically significant difference between the bleeding VAS scores assessed 6 hours and the ones assessed 1, 2 and 3 days after the operation in the control group (P < 0.05). There was the statistically significant difference between the bleeding VAS scores of experimental group and control group assessed 6 h after the operation (P < 0.05).
CONCLUSION
The hemocoagulase may improve the hemostatic effect of Nasopore 6 hours after the operation by combined injection with Nasopore as nasal cavity packing.

7.Activation of liver X receptors induced pancreatic β cell cycle arrest by up-regulating the expression of p27 protein

Objective To investigate the effects of liver X receptor (LXR) agonist on the proliferation of mouse pancreatic β cell line MIN6 cells.Methods The viability,changes of cell cycle,mRNA levels of S phase kinase associated protein 2 (Skp2) and p27,and protein levels of Skp2 and p27 in MIN6 cells treated with LXR agonist T0901317 were determined by the CCK-8 method,flow cytometry,real-time RT-PCR and western blot,respectively.Results The viability of MIN6 cells treated with 1 μmol/L,5 μmol/L and 10 μnol/L of T0901317 were (98.54 ±0.94)%,(87.03 ±0.93)% and (75.57 ± 1.85)% of the controls,respectively,and there was significant difference among them (F =301.90,P < 0.01).The percentages of G1 phase cells in the MIN6 cells treated with 0 μmol/L,1 μmol/L,5 μmol/L and 10 μmol/L of T0901317 were (35.93 ±2.25)%,(38.45 ±0.91)%,(45.46±1.34)% and (53.28 ± 1.14) %,respectively,and there was significant difference among them (F =80.83,P < 0.01).Similarly,the percentages of S phase cells in the MIN6 cells treated with 0 μmol/L,1 μmol/L,5 μmol/L and 10 μmoi/L of T0901317 were (52.87 ± 1.19) %,(48.65 ± 0.85) %,(36.31 ± 1.37) % and (31.45 ± 1.22) %,respectively,and there was also significant difference among them (F =221.30,P < 0.01).The protein levels of p27 in the MIN6 cells treated with 10 μmol/L of T0901317 (2.84 ± 0.14) were significantly higher than that in the controls (2.28 ± 0.10) (t =4.54,P < 0.05),while there was no significant difference in the mRNA levels of p27 between them (t =0.28,P > 0.05).However,10 μmol/L of T0901317 significantly decreased mRNA (0.52 ± 0.02,t =29.22,P < 0.01) and protein levels (0.98 ± 0.12 vs 1.89 ± 0.01,t =10.98,P < 0.01) of Skp2 in MIN6 cells.Based on the control siRNA transfection group as a reference (100%),the cell survival rates of the p27 siRNA transfection group,10 μmol/L of T0901317 treatment group and the intervention group (p27 siRNA transfection + T0901317 treatment) were (100.97 ± 1.08) %,(75.03 ± 1.83) % and (86.67 ± 2.45) %,respectively.There was no significant difference between the control siRNA and p27 siR-NA transfection groups (t =1.542,P > 0.05).Compared with the control siRNA transfection group,the cell survival rates of the T0901317 treatment group decreased (t =23.58,P < 0.01).There was also significant difference in the cell survival rates between the T0901317 treatment group and the intervention group (t =7.77,P < 0.01).Conclusion The activation of LXR may induce pancreatic β cell cycle arrest by up-regulating the expression of p27 and down-regulating the expression of Skp2.
8.Analysis of the effect of home quarantine on blood lipid and glucose of healthy adults during the COVID-19 epidemic times based on the data of physical examination

Objective:To analyze the effect of home quarantine on blood glucose and lipids in healthy adults during the COVID-19 epidemic times.Method:From April 7, 2020 to May 1, 2020, 512 adults wereexamined in Beijing physical examination center, of which 87 adults aged olderthan 18, received community closed management or home quarantine from January 23, 2020 to March 30, 2020, and theadultshad physical examination in Beijing physical examination center in the same month of 2018 and 2019 were selected as controls. The change trend of blood glucose and blood lipid acrossthe three years was analyzed by one-way repeated measurement of variance, and the difference of blood lipid and blood glucose between two years of physical examination was further analyzed by Bonferroni method. Datawereanalyzed by gender.Result:Among the 87 subjects, 36 (41.4%) were male and 51(58.6%) were female. The average age was (46.3±13.2) years and ranged from 24 to 74. The difference of high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), triglyceride (TG), total cholesterol (TC) and fasting blood glucose (FBG) among 2018, 2019 and 2020 was statistically significant (all P<0.05), while differences in LDL-C, TC and FBG were statistically significant for males (all P<0.05) and HDL-C, LDL-C, TC and FBG were statistically significant for females (all P<0.05). HDL-C in 2018and 2020were significantly lower than that in 2019 [(1.27±0.29), (1.30±0.31) vs. (1.36±0.34) mmol/L], LDL-C in 2020was significantly higher than that in 2018and 2019 [(3.11±0.88) vs. (2.81±0.77), (2.84±0.71) mmol/L], TG in 2020 was significantly higher than that in 2019[(1.54±1.17) vs. (1.32±0.80) mmol/L], TC in 2019and 2020were significantly higher than that in 2018 [(4.88±0.94), (5.10±0.99) vs. (4.63±0.90) mmol/L], and the FBG in 2019and 2020were significantly lower than that in 2018 [(5.34±1.17), (5.44±1.58) vs. (5.84±1.70) mmol/L] (all P<0.05). The comparative analysis of men and women showed that the means of LDL-C, TG, TC and FBG of men were higher than those of women while the mean of HDL-C was lower than that of women in each year; compared with 2019, the increase of LDL-C and TC of men was obvious while the decrease of HDL-C of women was obvious (all P<0.05) in 2020. Conclusion:During the epidemic period of COVID-19, the home quarantine has adverse effects on blood glucose and lipid.
9.The role of LPS in the CD14 expression and the activation of Kupffer cells.

OBJECTIVETo explore the effect of LPS on the expression of CD14 and the activation of Kupffer cells (KCs).
METHODSRat KCs were isolated and cultured with LPS. Immunohistochemistry and RT-PCR methods were employed to determine the changes in the CD14 expression and the concentration of TNFalpha, IL-6 and NO in the supernatant of the cultured KCs with LPS.
RESULTS(1) The expression of CD14mRNA and the synthesis of CD14 protein in the KCs increased evidently when stimulated by various concentrations of LPS, and the CD14mRNA expression was correlated in dose-dependent manner with LPS levels. (2) The expression of CD14mRNA and the synthesis of CD14 protein in KCs induced by LPS (10 micro g/ml) increased significantly and peaked at 3 approximately 6 hours. (3) The expression of CD14mRNA and the synthesis of CD14 protein in freshly cultured KCs were obviously up-regulated by the active mediators produced by KCs after being stimulated by LPS. (4) The release of TNFalpha, IL-6 and NO from cultured KCs was evidently down-regulated by the addition of anti-CD14McAb in the presence of serum or by the addition of LPS in the absence of serum, but up-regulated by the concomitant addition of LPS and LBP.
CONCLUSION(1) The CD14mRNA expression and the protein synthesis in cultured KCs were closely related to LPS and the active mediators produced from the KCs.The increased CD14 expression was possibly caused by LPS, and the further increase of the expression might be closely correlated to the cytokines released from the KCs. (2) The KC activation by low concentration of LPS was CD14 dependent.

10.Surgical treatment of De Bakey type III aortic dissection.

OBJECTIVETo summarize the experience in surgical treatment of De Bakey type III aortic dissection and discuss the operative indications, techniques, and methods for spinal cord protection.
METHODSFrom January 1994 to January 2000, 49 patients with De Bakey type III aortic dissection were operated on. Of these patients, 35 were subjected to partial thoratical aorta replacement, 4 partial thoratical aorta replacement and plasty, 8 total thoratical aorta replacement, and 2 thoracoabdominal aorta replacement. In the 35 patients, the operations were performed by using the technique which preserved blood was transfused back by pump via the femoral artery.
RESULTSHospital mortality in this group was 6.1% (3/49). Eight patients received emergence operation, and 1 patient died. Paraparesis developed in one patient (2.0%). Other complications included re-exploration for bleeding in 3 patients, and wound infection in 5.
CONCLUSIONSThe rate of mortality and complication is high in patients with De Bakey type III aortic dissection. Appropriate operative techniques and methods of spinal cord protection are essential to resolve this problem.
