Objective:To observe the level of serm soluble E-selectin(sE-selectin) in patients with pulmonary tuberculosis and to study the clinical significance of it. Objective To observe the clinical effect ofHuangkui capsule combined with benzbromarone on the treatment of the patients with chronic uric acid nephropathy (CUAN).Methods A total of 60 patients were recruited and divided into the treatment group and the control group by using the random number table method, each group 30 patients. Two groups were treated with conventional treatment, such as low purine, low protein diet, quit smoking and drinking extra water. The control group was added with oral benzbromarone, and the treatment group was added with oralHuangkui capsule. All the treatments last 8 weeks. Theblood uric acid (BUA), serum creatinine (SCr), blood urea nitrogen (BUN), cystatin C (Cys-C), SOD, 24-hour urinary protein quantity and Urinary sediment red blood cell count were detected by the automatic biochemical analyser.The clinical effect was evaluated by the Rheumatoid arthritis (RA) quality of life questionnaire.Results Compared with the control group, the total effective rate in the treatment group was 86.64% (26/30), which was significantly higher than that of 63.33% (19/30) in the control group (χ2=7.264, P=0.001). After treatment, the BUA (273.52 ± 110.37μmol/Lvs. 331.28 ± 126.54μmol/L,Z=-2.543), BUN (6.24 ± 1.23 mol/Lvs. 8.16 ± 2.35 mol/L,Z=-2.680), SCr (90.37 ± 20.16μmol/Lvs. 110.38 ± 16.72μmol/L,Z=-2.534), Cys C (0.86 ± 0.51 vs. 1.03 ± 0.10,Z=-2.372) in the treatment group were lower than those of the control group (P<0.01); SOD (156.37 ± 32.04μmol/Lvs. 43.36 ± 31.52μmol/L,Z=-2.041) in the treatment group was higher than that in the control group (P<0.01); the 24 hours urinary protein (439.86 ± 250.41 mg/24hvs.897.69 ± 213.37 mg/24h,Z=-2.853), urine sediment RBC counts (50.31 ± 14.06 points/μlvs.213.47 ± 38.46 points/μl, Z=-2.106) in the treatment group were lower than those in the control group (P<0.01); the physiological function(43.14 ± 2.06 pointsvs.36.48 ± 3.21 points,Z=10.362), the psychological function (40.76 ± 3.28 points vs. 16.54 ± 3.71 points,Z=9.547), social function (40.74 ± 3.58 points vs. 33.04 ± 5.48 points,Z=6.034), healthy self-awareness (24.57 ± 1.97 points vs. 22.63 ± 3.43 points,Z=4.236) and total score (127.38 ± 6.43 points vs. 107.69 ± 13.57 points,Z=6.754) in the treatment group were higher than those in the control group (P<0.01) . Conclusions TheHuangkui capsule combined with benzbromarone could reduce CUAN patient's blood uric acid levels, protect renal function, and improve the quality of life.
3.The mechanism of direct relaxant effects of propofol on rabbit tracheal smooth muscle

Objective To investigate the possible mechanism of direct relaxant effects of propofol on tracheal smooth muscle.Methods Nine Japanese long-ear white rabbits of either sex, weighing 2-3 kg were sacrificed by air embolism. The trachea of each rabbit was removed and cut into six strips of (6-8) mm x (12-15) mm. The tracheal strips were suspended in Kreb' s solution, exposed to 95 % O2 and 5 % CO2 mixture and maintained at 37℃. One end of strip was fixed and the other was connected to a tension monitor through a tension transducer. The experiment was divided into six groups: group I propofol; group Ⅱ propofol + indothelin; group Ⅲp Ⅴ propofol + verapamil and group Ⅵ propofol + glibenclamide. In each group the tracheal strips were precontracted with acetylcholine (10-5 mol? L-1). The strips were then exposed to increasing concentrations of propofol (5 ? 10-3 , 10-2 ,2.5 ? 10-2 , 5 ? 10-2 , 10-1 mg?ml-1 ). At each concentration of propofol, the change in muscle tension was allowed to reach the steady state for 30 min. In group I no other drug was added to the Kreb' s solution besides propofol, while in group Ⅱ-Ⅵ before the effects of propofol were determined, the strips were pretreated with indothelin (10-4 mol? ml-1 ) (in group Ⅱ), L-NAME(10-4 mol?ml-1) (in group Ⅲ), methylene blue (10-5 mol?ml-1) (in group Ⅳ), verapamil (10-5 mol?ml-1) (in group Ⅴ) or glibenclamide (10-5 mol?ml-1) (in group Ⅵ). Results The three concentrations of propofol examined (2.5?10-2, 5?10-2 and 10-1 mg?ml-1) produced relaxation of isolated tracheal smooth muscle. In group Ⅱ , Ⅲ , Ⅳand Ⅴ pretreatment of tracheal strips with indothelin, L-NAME, methylene blue or verapamil did not significantly affect the relaxation of tracheal smooth muscle induced by propofol as compared with group Ⅰ. 4.Comparative Identification of Different Cultivated Varieties of Amomum villosum

[Objective] To compare and identify the different cultivated varieties of Amormun villosum for providing scientific evidence for the selection, breeding, standardization and industrialization. [Methods] Source appraisal, character identification and scanning electron microscope technique were applied to observe and compare the appearance of the original plant, the properties of the medicinal material and the outside characteristics of pollen of Amomum villosum in Yangchun, Guangdong province. [Results] The two cultivated varieties of Amomum villosum in this area, long-fruit and round-fruit types, are quite different in the appearance and the characteristics of pollen. [Conclusion] It is suggested that long-fruit Amomum villosum and round-fruit Amomum villosum should be the selected varieties for breeding.
5.Influence of Biological Pesticide on Fruting Rate and Yield of Amomum villosum Lour.

[Objective] To observe the influence of different kinds of biological pesticide on the fruiting rate and yield of Amomum villosum Lour. (AVL) . [Methods] Medicinal plant field experiment method was used to observe the influence of biological pesticide A (highly effective immuno-biologic bactericide, 75.76mg?m-2 for one time) and its 1000-fold, 800-fold and 500-fold diluent on the fruiting rate and yield of AVL. Meanwhile, the influence of biological pesticide B (edible oligosaccharide, 30.30mg?m-2 for one time) and biological pesticide C (deguelin emulsion, 1.89mg?2m-2 for one time) was also observed. [Results] The three biological pesticides, as well as the diluents of biological pesticide A increased the fruiting rate and yield of AVL, the influence of the moderate-concentration biological pesticide A being the greatest. [Conclusion] Proper application of biological pesticide is one of the important ways to raise the yield of AVL.
6.Determination of alliin in mice plasma by HPLC

Objective: To develop HPLC method for determination of alliin concentration in mice plasma. Methods: The plasma samples were extracted with methanol. The analysis involved a ODS 1 column as stationary phase and distilled water as mobile phase. The flow rate was 0.5mL?min -1 , UV detection wavelength was at 220nm. 5 Fu was used as the internal standard. Results: The calibration curve was linear over a range from 14.8?g?mL -1 to 148.1?g?mL -1 with a correlation coefficient of 0.9992. Conclusion: The method is economic, simple, sensitive and accurate.
7.A case of hidrotic ectodermal dysplasia with pseudo-ainhum and detection of connexin gene mutations

Objective To detect the mutations in connexin genes in a family with hidrotic ectodermal dysplasia(HED)complicated by pseudo-ainhum.Methods Peripheral blood samples were collected from a 20-year-old patient with HED complicated by pseudo-ainhum,and from his unaffected sister.Total DNA was extracted from these samples,and PCR was performed to amplify the partial coding region of GJB2,GJB5 and GJB6 genes.Subsequently.PCR products were bidirectionally sequenced in both subjects.Results No mutation was detected in GJB5 or GJB6 gene in either subjects.Two mutations (V27I and V37I)were detected in the GJB2 gene in the patient but not in his sister.Conclusion The mutation in the GJB6 gene may be absent in patients with HED;there might be other genes involved in the pathogenesis.
8.Prognostic analysis of primary gastrointestinal diffuse large B-cell lymphoma

Objective To assess the clinical characteristics of primary gastrointestinal diffuse large B-cell lymphoma (PGI-DLBCL) and its treatment and prognosis. Methods Twenty patients diagnosed with PGI-DLBCL were admitted to hospital between 2003 and 2007. The clinical characteristics and tumor molecular model of PGI-DLBCL as well as therapeutic methods were retrospectively studied. The factors that related to survival and prognosis were statistically analysed.Results The overall survival (OS) time of the patients were from 42 months to 52 months,while the progression-free survival (PFS) time were from 37months to 47 months. The International Prognostic Index (IPI) score (0~2 or >2) played a reminding role in prognosis of the disease. Tumor molecular model was no effect on prognosis [that was no significant difference between germinal center B-cell-like (GCB) type and non-GCB type]. The efficacy of Rituximab in combination with CHOP chemotherapy (R-CHOP) in treatment of PGI-DLBCL was similar to CHOP chemotherapy alone, whereas surgical intervention might prolong survival period. Conclusions The biological characteristics of PGI-DLBCL is so particular that the most therapeutic method, which need to be further studied, would be different from DLBCL in other position.
9.Approved effect of hydrochloric-acid arotinolol on trembling symptom of patients with essential tremor: 30 cases analysis

BACKGROUND: Essential tremor is a kind of dyskinesia disease with a tendency to heredity. β-adnephrin receptor blocker is mainly used for treatment, but the dosage was great, patients could not persist in treatment. Hydrochloric-acid arotinolol, which characterizes by little side effect, is a β-receptor blocking agent combined with blocking effect of α receptor, and is an ideal drug to treat essential tremor in Japan.OBJECTIVE: To observe the reliability and curative effect of hydrochloric-acid arotinolol on essential tremor among Chinese people.DESIGN: Case analysis.SETTING: Neurological Departments of Beijing Hospital of Ministry of Public Health and General Hospital of Beijing Military Area Command of Chinese PLA.PARTICIPANTS: Totally 30 patients with essential tremor, 19 males and 11 females, aged from 21 to 74 years, with course of 1-40 years, average of (15.8±12.6) years, were selected from Neurological Departments of Beijing Hospital of Ministry of Public Health and General Hospital of Beijing Military Area Command of Chinese PLA from April 1999 to December 2000.METHODS: Tremor degree of upper limb, caput and lower limb, degree of dysfunction of hand, tremor degree of hand during writing and circling, and chief complaint of pain caused by tremor were scored before medication and 2, 4 and 6 weeks after medication. Scores were determined as 1 point, 2 points and 3 points according to mild, moderate and severe degree respectively. Evaluated criteria: ① Obviously effective: Differences were more than 7 points before and after treatment. ② Effective: Differences were 6.5-3 points before and after treatment. ③ A little effective: Differences were 3-1.5 points before and after treatment. ④ No changes: Differences were from 1 to -1 points before and after treatment. ⑤ Worse: Differences were less than -1.5 points before and after treatment. Meanwhile, blood pressure and heart rate of patients and side effect of drug were observed.MAIN OUTCOME MEASURES: ① Results of effective evaluation; ②Blood pressure and heart rate before and after treatment; ③ Adverse events and side effect.RESULTS: Totally 30 patients entered the final analysis. ① Among 30 patients with essential tremor, 6 had obvious effect, 11 had effect, 5 had a little effect, 7 had no effect, and 1 was worse. ② Average of blood pressure before treatment was (138.5±14.6)/(84.2±6.4) mm Hg (1 mm Hg=0.133 kPa), and was (130±10)/(79.7±6.3) mm Hg after 1 course. Average heart rate was (77.5±6.4) times/minute before treatment and (75±6.4) times/minute after treatment. The difference was not significant. ③ Among 30 patients, 4 had side effect at various degrees which was accounted for 13.4%; 1 had distress after medication, 1 had agrypnia, 2 had fullness in head and dizziness, and 1 gave up the treatment because of deteriorated tremor.CONCLUSION: Hydrochloric-acid arotinolol can improve tremor symptom of patients obviously and is an effective drug to treat essential tremor with convenient and safe medication.
10.Construction of prokaryotic expression vector of mouse CD25 extracellular domain

Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
