1.Protection against Myocardial Ischemia/Reperfusion Injury in Rats by Pretreatment with Paeoniflorin

Objective:To investigate the protective effects of paeoniflorin on myocardial ischemia/reperfusion ( MI/R) injury in rats. Methods:The rat model of MI/R injury was prepared by coronary artery ligation for 30 min followed by 2-hour reperfusion. Then the rats were randomly divided into 5 groups:sham group, model group, paeoniflorin group respectively at high, medium and low dose (20, 10 and 5 mg·kg-1, n=10). Paeoniflorin was respectively injected via tail vein 1 h before the operation and at the beginning of the reperfusion. The CK, LDH and SOD activities and MDA contents in serum of rats were examined, and the area of myocardial in-farction was also calculated after the reperfusion. Results:Paeoniflorin (20 and 10 mg·kg-1 ) could significantly reduce the area of myocardial infarction when compared with the model group (P<0. 05). Paeoniflorin could obviously inhibit the CK and LDH activities (P<0. 05 or P<0. 01) and enhance the SOD activity (P<0. 01) in serum of MI/R rats. The MDA content in paeoniflorin groups at high and medium dose was significantly lower than that in the model group (P<0. 05). Conclusion:Paeoniflorin pretreatment shows effectiveness against MI/R injury, which may be associated with the inhibition of lipid peroxidation.

3.Relationship between the infection of human herpesvirus 6 and oral squamors cell carcinoma

Objective: To study the relationship between human herpes vi rus 6 (HHV-6) infection and oral squamors cell carcinoma. Methods: The serum anti-HHV-6 antibody titers from 16 cases of oral squamors cell carcinoma (OSCC) and 16 control subjects were detected by indirect immunofluores cence assay. HHV-6 DNA in peripheral blood mononuclear cells was amplified by P CR and the specificity was confirmed by Southern-blot hybridization with an int ernal probe oligonuclotide. An immunohistochemical staining was used to detect H HV-6 antigen in OSCC tissues. Results: Positive expression of s erum HHV-6 IgG antibody was found in all the cases of OSCC and in 12 of the 16 controls. With the rapid development of modern science and technology,the post-genomics period has come with the complement of the sequence of body genome including several tens of human genome,the emphasis of life science transfer from instruction genome to the post-genomics period,functional genome.Proteomics is the most important part of it.The technology of proteomics is advanced day by day.Except the classical two dimensional gel electrophoresis,the technology of multi-dimensional liquid chromatography and the technology of isotope coded-affinity tags have been successively developed,as well as protein chip and phage display have been applied extensively.This article simply summarizes the key technologies of proteomics.

Objective:Normal and lung qi deficiency rats of proteomics of lung tissue of rats differentially expressed and mass spectrometry.Methods:A total of 20 SD rats were randomly divided into control group,lung qi deficiency group,with 10 rats in each roup.Lung qi deficiency group model replication using composite method by intratracheal injection of lipopolysaccaride and smoked.All rats in each group before anesthesia 12 hours of fasting,water deprivation,weight after 28 days modeling,Thoracotomy to remove left lung tissue after anesthesia was 10 mg from each rat, 3 rats from each group, using two-dimensional electrophorsis analysis, two-dimensional gel electrophoresis image, peptide mass fingerprinting and electrospray tandem by mass spectrometry to protein identification.Results:2 differntial protin profiles for proteomics analysis,identified 14 protein in rat lung tissue,protein 5 expression, regulation 9 expression.Conclusion:By proteomic analysis,identify the protein have correlation with lung deficiency syndrome.
7.Normal group and artificial simulation of cold environment proteomics of lung tissue of rats variance analysis

Objective:To analyse and discuss the variance and mass spectrometry of proteomics of rats 'lung tissues between groups of normal and simulated cold environments.Methods:A total of 20 SD rats were randomly divided into two groups of normal and simulated cold environments with 10 rats in each group.The temperature of normal environment group was controlled in 21℃ with a fluctuation of 2℃lasting 24 h per day.In contrast, the temperature of simulated cold environment group was controlled in 6℃ with a fluctuation of 2℃lasting 4 h per day while the temperature and humidity for the rest of time were exactly the same as the situation in normal environment group.The preoperative fasting , water-deprivation and weighting were adopted 12 h before anaesthesia to both groups after the continuing 7-day experimentation.Left lung tissues of each rat in both groups were removed by the amount of 10 mg, whereas three random samples from each group were tested by experiments of protein extraction , bidirectional electrophoresis , image analysis and mass spectrometry identification.Results: Given the proteomics analysis of two differential sets , it is indicated that in simulated cold environment group , among nine sorts of protein , four sorts perform rising while five falling compared to that of normal group.Conclusion:There exists a correlation between protein and cold environment based on the results of proteomic analysis .
8.Changes of Electrocardiogram after Acute Stroke

Objective To investigate the feature and mechanism of electrocardiogram (ECG) changes in different onset parts after different types of acute stroke.MethodsA retrospective analysis about ECG was performed according to data of 163 acute stroke patients.ResultsThe abnormal ratio of ECG had no significant difference between hemorrhagic stroke and ischemic stroke. Moreover, the patients with onset areas on basal ganglia and thalamus had a higher rate of abnormal ECG.ConclusionThe patients with acute stroke have a higher incidence of abnormal ECG. The changes of ECG are associate with types and areas of injury.

AIM:To explore the therapeutic effect of a novel Rho kinase inhibitor WAR 5 on the experimental autoimmune encephalomyelitis (EAE) and its possible mechanism.METHODS: Female C57BL/6 mice were randomly divided into EAE group and WAR5 group.EAE model was induced by the application of MOG 35-55 peptide.WAR5 was in-jected intraperitoneally every other day from post-immunization (PI) day 3 to PI day 27.The clinical score and body weight were recorded every other day .On PI day 28, the animals were sacrificed and spinal cords were obtained for HE and mye-lin staining .The splenocytes were isolated and the expression of CD 16/32 and CD206 were analyzed by flow cytometry . The protein extracts from the brains and spinal cords were collected for the measurement of inducible nitric oxide synthase ( iNOS) by Western blotting .RESULTS:The administration of WAR 5 delayed the onset of EAE and attenuated the clini-cal symptoms .The results of the pathological examination revealed that WAR 5 inhibited the infiltration of inflammatory cells and improved myelination in spinal cords , accompanied with the poralization of M 1 macrophages to M2 phenotype in the spleen.WAR5 inhibited the expression of iNOS in the central nervous system , especially in the spinal cords .CON-CLUSION:The therapeutic effect of WAR5 on EAE may be related to the shift of M1 macrophages to M2 phenotype and inhibition of inflammation in the central nerve system .
