Toll-like receptor (TLR) plays a crucial role in innate immune response and the subsequent induction of adaptive immunity. TLR is expressed on immune cells as well as epithelial cells and tumor cells. When endogenous and exogenous ligands bind with TLR, TLR signaling pathways are activated and a large number of inflammatory cytokines are released, creating an inflammatory microenvironment and leading to tumor formation, invasion and metastasis.
2.Protective effect of glycine on hypoxic injury to murine cardiomyocytes

Objective To investigate the protective effect of glycine (Gly) on hypoxic injury to murine cardiomyocytes and the mechanisms. Methods The survival rate of cardiomyocyte survival was detected by trypan blue exclusion and lactate dehydrogenase (LDH) with an ultraviolet spectrophotometer. Ca 2+ changes in the cardiomyocytes were detected by laser confocal microscopy. Results Glycine markedly improved the survival rate of cardiomyocytes and decreased the release of LDH in cardiomyocytes after hypoxia. The protective effect was in a dose-dependent manner. Glycine could also block calcium overload after hypoxia. Conclusion Glycine has the protective effect on cardiomyocytes through the improvement of survival rate, decrease of LDH release, and blockage of calcium overload after hypoxia.
3.Correlation between transformational leadership style and nurses' job satisfaction in emergency department

Objective To investigate the correlation between transformational leadership style and nurses' job satisfaction in emergency department.Method The survey was done among 168 emergency department nurses from 4 first class hospitals in Guangzhou by the demographic information,multifactor leadership questionnaire and Chinese nurses' job satisfaction scale to investigate the status of transformational leadership style perceived by nurses and nurses' job satisfaction in the emergency departments.Results The score of transformational leadership style perceived by the nurses in the emergency department was (50.60±14.40) and jop satisfaction was (79.80±13.30).The transformational leadership style was positively correlated with nurses' job satisfaction (P<0.01).Conclusions The transformational leadership style is middle-high level.The nurses' job satisfaction is at low level,Nursing managers should play an active role in transformational leadership style to enhance nurses' job satisfaction in emergency departments.

Objective To investigate the effect of hedyotic diffusa willd injection on osteosarcoma MG‐63 cells Bax gene expres‐sion .Methods MTT was be used to detect the inhibition rate of MG‐63 cells by hedyotic diffusa willd injection after 6 ,12 ,24 ,48 h certain concentration ,RT‐PCR was be used to detect the intracellular expression of Bax gene .Results Hedyotic diffusa willd injec‐tion can significantly inhibit the proliferation of MG‐63 cells ,as the concentration was 100 μL/mL ,Bax gene expression was signifi‐cantly increased over time(P<0 .05) .Conclusion Upregulating the expression of Bax gene by hedyotic diffusa willd injection can induced human osteosarcoma M G‐63 cells apoptosis to death .

6.Effect of proanthocyaindin on angiogenesis in rats with ischemic hindlimb

Objective To investigate the effect of proanthcyaindin on angiogenesis of rats with ischemic hindlimb.Methods Twelve male SD rats were randomized divided into control group (n =6) and proanthcyaindin group (PC group,n =6).Lower limb ischemia rat model was establish,rats in both group were administration by oral daily,and PC group was given proanthocyanidins 200 mg/(kg · d),while the control group received the same volume of saline.Forteen days after surgery,ischemic tissues of adductor were collected for several tests including the expression of CD31 in the ischemic muscle tissue and the number of new blood vessels by immunohistochemical staining,hypoxia inducible factor-1 protein expression by western blot,and vascular endothelial growth factor level test by enzyme-linked immunosorbent assay.Results The capillary count showed that the angiogenesis situation of PC group was (69.67 ±3.11)/HP,higher than that in control group((111.00 ± 3.11)/HP,t =13.350,P < 0.0001).The HIF-1 protein expression in PC group was (1.90 ± 0.25),remarkable higher than that in control group (0.54 ± 0.21,t =4.183,P =0.0058).Compared with control group,VEGF level in PC group increased((432.86 ± 13.00) μg/L vs.(326.68 ± 11.08) μg/L,t =6.216,P <0.0001).Conclusion Proanthcyaindin plays a positive role in angiogenesis after ischemia.PC may induce up-regulation of HIF-1 and VEGF,and then promote the formation of endovascular through multiple signaling pathways consequently.
7.Meta-analysis of influential factors of pre-hospital delay of patients with acute ischemic stroke

Objective To investigate the main risk factors influencing the pre-hospital delay of patients with acute ischemic stroke (AIS) for preventing methods.Methods A retrieval was done across the epidemiologic studies on pre-hospital delay of AIS patients during 2004 to 2015 using the key words of ischemic stroke,prehospital delay,treatment delay.According to the inclusion and exclusion criteria,the literature was collected and the literature quality was assessed.The data were extracted for meta-analysis by RevMan software.Results Sixteen epidemiologic studies were included.Totally,there were 2,966 cases of prehospital delay and 2,468 cases of non-delay.The pooled OR values and 95% CI by multivariate analysis were as follows:the degree of awareness of stroke was 0.5 (0.3,0.82),emergency medical service (EMS) was 0.49 (0.29,0.85),National Institute of Health Stroke scale (NHISS) score 0.58 (0.36,0.95),outpatient visit as first visit 4.28 (1.44,12.74),distance 0.76 (0.61,0.95),medical history of ischemic stroke 0.37 (0.18,0.79) (with significant differences in OR value,P<0.05).Conclusions The protective factors for pre-hospital delay of acute ischemic stroke patients include:strengthened awareness of stroke,effective EMS,high NHISS score,short distance,medical history of ischemic stroke.The main risk factor is that their outpatient visit is their first visit for medical treatment.Therefore,we need to enhance the health education to the patients and their families about awareness of stroke,improve the use of EMS and build complete and efficient green path for the stroke patient.
8.Effects of arsenic trioxide combined with cisplatin on the growth of human nasopharyngeal carcinoma cells and reversion of RASSF1A hympermethylation.

OBJECTIVE:
To investigate the effect of arsenic trioxide (As2O3) combined with cisplatin on expression of RASSF1A in nude mice with human nasopharyngeal carcinoma xenograft.
METHOD:
The models of human poorly differentiated nasopharyngeal carcinoma in nude mice were established and randomly divided into four groups, control group (NaCl group), As2O3 group, DDP group and As2O3 + DDP group. The expression of RASSF1A mRNA and protein were detected by Real-time RT-PCR and immunohistochemistry respectively. The methylation rate of RASSF1A promoter CpG islands was analyzed by HRM.
RESULTS:
Experimental groups could obviously inhibit the growth of tumor and up-regulate the expression of RASSF1A. The methylation rate of RASSF1A in transplanted tumors in experimental groups was lower than the control group. Especially As2O3 combined with DDP were superior to the single drug use.
CONCLUSION
As2O3 inhibits the growth of human nasopharyngeal carcinoma cell strain CNE2 xenograft in nude mice and increases mRNA expression of RASSF1A. As2O3 inhibits the malignant phenotypes of human nasopharyngeal carcinoma cells and reverses hypermethylation of RASSF1A.

9.Transfecting MIN6 cell line and enhancing cell viability by Reg3? cDNA

Objective To create a MIN6 cell line overexpressing murine Reg3? cDNA and to investigate its cell viability character under low glucose concentrations.Methods The full-length murine Reg3? cDNA was inserted into the plasmid pcDNA3.1(-).Then MIN6 cells were transfected with the Reg3?-pcDNA3.1 and pcDNA3.1 vector alone to establish Reg3?-overexpression and empty vector MIN6 cell line.Reg3? protein in the low glucose concentration cell medium was detected by Western blot.Cell viability was evaluated by an MTT reduction conversion assay.Results Three Reg3?-overexpression MIN6 cells were confirmed by real-time PCR and Western blot.Reg3? expression was barely detectable in the cells transfected with the empty vector alone.In contrast,the levels of Reg3? mRNA and protein in three pcDNA-Reg3?-transfected clones were increased by an average 10-and 6-fold,respectively.Western blot also revealed Reg3? protein release into the culture medium.In MTT cell proliferation assay,compared to vector-transfection alone,three clones of Reg3?-overexpression MIN6 cells exhibited 2-fold increases in cell number over 7 days when cultured in the presence of 10 mL/L fetal bovine serum and 5.5 mmol/L glucose.Conclusion The Reg3?-overexpression MIN6 cells have been established.Reg3? protein was detectable in culture medium,supporting an endocrine action,and Reg3? overexpression promoted islet cell growth.
