1.Effect of tretinoin gel sheeting on early-stage hyperplastic scars in rabbit ears

Objective To estimate the effect of tretinoin gel sheeting on early-stage hyperplastic scars in rabbit ears,and to evaluate the feasibility to prevent and treat hyperplastic scars with it.Methods The ears of 24 rabbits were used to establish a model of hyperplastic scar according to previously reported methods.Then,the rabbit ears were randomly divided into four groups:control group receiving no treatment,gel sheeting group treated with the vehicle of the tretinoin gel sheeting,0.05% tretinoin group treated with 0.05% tretinoin gel sheeting,0.1% tretinoin group treated with 0.1% tretinoin gel sheeting.All the gel sheetings were topically used twice daily for six consecutive weeks.During the treatment,the size,thickness,color and texture of scars were estimated.After six weeks of treatment,all the scar tissues were resected from the rabbit ears and subjected to hematoxylin and eosin (HE) staining and van Gieson (VG) staining.Statistical analysis was carried out by analysis of variance and rank sum test.Results The scars were deeply colored,thickened,hard and elevated with an uneven surface in the control group,but lightly colored,thinned and soft with the presence of small subcutaneous nodules in the other three groups.The surface of scars in the two tretinoin groups was similar to that of adjacent normal skin,and scaling was observed on the scar surface in the 0.1% tretinoin group.HE and VG staining showed a disarrangement of collagen fibers with the formation of vortex-like structures in the control group.A significant decrease was noted in the number of fibroblasts and microvessels as well as amount of collagen deposition per unit cross-sectional area in the two tretinoin groups compared with the control group and gel sheeting group.Additionally,the collagen fibers were regularly arranged and parallel to the long axis of scars in the two tretinoin groups.The scar hyperplasia index (HI) was 3.173 ± 0.26,2.465 ± 0.19,1.906 ± 0.21 and 1.903 ± 0.23 in the control group,gel sheeting group,0.05% tretinoin group and 0.1% tretinoin group respectively,the fibroblast density (NA) was 5836.7 ± 527.03,4128.73 ± 387.66,3207.59 ± 439.17 and 3200.28 ± 421.48 respectively,and the area density of collagen fiber (AA) was 45.38 ± 5.83,36.57 ± 6.84,28.09 ± 3.82 and 28.07 ± 3.47 respectively.As far as HI,NA and AA were concerned,the control group differed significantly from the other three groups (all P < 0.01),and the gel sheeting group from the two tretinoin groups (all P < 0.01),but no significant difference was observed between the two tretinoin groups (all P > 0.05).Conclusions Topical tretinoin gel sheeting can inhibit scar proliferation at early stage in rabbit ears,and may provide a new choice for the prevention and treatment of hyperplastic scars.
2.Fast Determination of Al Residue in Polyether Ketone Ketone by Graphite Furnace Atomic Absorption Spectrometry

AnewmethodwasdevelopedforfastdeterminationofAlresidueinpolyetherketoneketone ( PEKK) special polymer material by graphite furnace atomic absorption spectrometry ( GF-AAS) . Instead of high temperature ashing, sample was dissolved in concentrated sulfuric acid and diluted by tetrahydrofuran, then was directly analyzed by GF-AAS. Systematical investigation was made on sample preparation, it was found that for 0. 2 g of sample weight, a clear sample solution could be obtained by heating at 220 ℃ with 2 mL of concentrated H2 SO4 for 4 min, and the obtained solution was ready for the subsequent mixing with tetrahydrofuran for sample dilution or dispersion before analysis. Redesign of the GF-AAS temperature program was also made, on the basis of the recommended program a pre-ashing step was added, which greatly helped increasing the analytical precision when the temperature was set at 400 ℃ and the total time was set at 40 s. The detection limits ( 3σ) were 38. 5 ng/g, the RSD for six parallel determinations of real samples was 2. 2%, and the recoveries for spiked tests were 99%-105%, respectively. In comparison with the traditional high-temperature ashing technique, the established method was more concise, faster and less exposed to contamination during sample preparation.
3.Determination of Ultratrace Copper in Barium Phosphate Laser Glass by Inductively Coupled Plasma Mass Spectrometry

With the use of high pure HF and HNO3 reagents, and autoclaves made of high purity Rerfluoroalkoxy ( PFA ) material, a solution sample digestion technique effective for phosphate samples, subjected to high temperature fusion, was established. The whole procedure was concise, fast and of low blank value. Key factors such as the amount and ratios of the reagents, the digestion temperature and time, were systematically optimized, it was found that within 0. 5 h at 150 ℃, only 1. 7 mL of total reagent consumption could lead to a complete sample decomposition. Most importantly, the samples were not required to be ground to fine powder, which greatly reduced the risk of contamination. In addition, an effective liquid_liquid extraction procedure based on 5_nonylsalicylaldehyde oxime as the extractant was established for matrix separation and analyte preconcentration. Under the optimal extraction conditions of 5 mL of 15% extractant, 0. 5% HNO3 of extraction acidity and 20% HNO3 of back_extraction acidity, a matrix separation efficiency of over 99. 999% could be realized and a preconctration factor of 10 could be obtained, which resulted in complete elimination of the matrix_induced interference and great enhancement of the analytical sensitivity. After optimization of the operation parameters of ICP_MS, high signal to background detection of Cu in 20%HNO3 at 840 W of plasma power and low sample uptake rate were realized. The detection limits of 2. 5 ng/g, RSD of 3. 3% for six detections of parallel samples, and the recovery of 94. 3% for spike test were obtained, respectively. The method was finally applied to three real samples analysis, and the results agreed well with the data from laser adsorption loss experiment.

Resveratrol is a natural polyphenolic substance with a variety of bioactivities,such as anti-oxidation,anti-tumor and apoptosis induction.This paper summarized the recent research progress of resveratrol induced cancerous cell apoptosis,including human cervical cancer HeLa cells,leukemia cells,hepatoma cells and gastric cancer cells,and analyzed the molecular mechanism behind resveratrol-induced cell apoptosis in tumor.In addition,resveratrol was investigated by blocking the cell cycle,regulating the expressions of related genes and proteins and mitochondria-induced apoptosis pathways as a potential application of cancer drugs to clinical researches.
5.Correlation between metabolic syndrome and early neurological deterioration in patients with minor stroke or high-risk transient ischemic attack

Objective:To investigate the correlation between metabolic syndrome (MetS) and early neurological deterioration (END) in patients with acute minor ischemic stroke (MIS) and high-risk transient ischemic attack (TIA).Methods:Consecutive patients with acute MIS or high-risk TIA admitted to the Second Affiliated Hospital of Xuzhou Medical University between May 2018 and June 2020 were enrolled prospectively. MIS was defined as the National Institutes of Health Stroke Scale (NIHSS) score ≤3, high-risk TIA was defined as ABCD 2 score ≥4, and END was defined as the highest score of NIHSS within 72 h after admission increased by ≥1 compared with the baseline. Multivariate logistic regression analysis was used to determine the correlation between MetS or its component and END. Results:A total of 145 patients with acute MIS or high-risk TIA were enrolled, including 66 males (45.5%), aged 68.28±9.71 years. Fifty-two patients (35.9%) met the diagnostic criteria of MetS, and 46 (31.7%) developed END. Univariate analysis showed that there were significant differences in age, sex, atrial fibrillation, elevated blood glucose, MetS, fasting blood glucose and C-reactive protein between the END group and the non-END group (all P<0.05). Multivariate logistic regression analysis showed that MetS (odds ratio 2.637, 95% confidence interval 1.127-6.169) and high blood glucose (odds ratio 2.672, 95% confidence interval 1.052-6.789) were the independent risk factors for END within 72 h of admission in patients with acute MIS or high-risk TIA. Conclusion:MetS is significantly associated with END in patients with acute MIS or high-risk TIA.
6.Micro RNA is involved in progression of secondary pathological process of traumatic brain injury

Traumatic brain injury (TBI) is a kind of disease in which brain tissue injury and brain dysfunction are caused by external trauma. At present, the pathological mechanism of TBI is not fully clear, and there is no good clinical treatment plan. TBI can be preliminarily divided into primary injury and secondary injury according to the occurrence time. In recent years, a large number of researchers have begun to look for effective biological signaling molecules such as micro RNA (miRNA) to discover and intervene in secondary injury with strong response after TBI as early as possible. MiRNA, as a small non-coding RNA, can play a key role in the pathological process of central nervous system diseases, which is considered as an effective regulator of nerve function under pathophysiological conditions. In this paper, the effects of miRNA on the occurrence, development and recovery of TBI are summarized through the three major mechanisms of nerve cell apoptosis, inflammatory cytokine expression and blood-brain barrier repair, which lays the foundation for researching function of miRNA in TBI.
7.Apolipoproteins as Differentiating and Predictive Markers for Assessing Clinical Outcomes in Patients with Small Cell Lung Cancer.

PURPOSE: The present study aimed to investigate the value of apolipoproteins, including ApoA-1, ApoC-III, and ApoE, in patients with small cell lung cancer (SCLC) as potential biomarkers for diagnosis, prognosis, and cancer progression. MATERIALS AND METHODS: Lung samples were collected from 89 patients with SCLC. Nineteen lung samples from non-small cell lung cancer (NSCLC) patients and 12 normal lung tissues were used as controls. Expression profiles of ApoA-1, ApoC-III, and ApoE in different samples were examined using immunohistochemical methods, and the expression levels were correlated with cancer types, treatment, and outcomes using chi-square and Mann-Whitney tests. RESULTS: Expression of ApoA-1 and ApoC-III in SCLC was significantly different, compared with that in NSCLC and normal lung tissues, and was correlated with recurrence of SCLC. Patients undergoing neoadjuvant chemotherapy before surgery showed significantly reduced expression of ApoA-1 and increased expression of ApoC-III and ApoE. Nevertheless, the expression levels of ApoA-1, ApoC-III, and ApoE were not correlated with SCLC staging. CONCLUSION: ApoA-1 and ApoC-III may be used as differentiating and predictive markers for SCLC. ApoA-1, ApoC-III, and ApoE may be used to monitor the efficacy of chemotherapy.
