Objective To study the relationship between serum iron, serum ferritin and alcoholic fatty liver(AFL), nonalcoholic fatty liver(NFAL). Methods The liver specimens of 97 patients with fatty liver were obtained by 1 second liver biopsy, and were further stained by haematoxylin eosin(HE) and Perl's Prussian. Meanwhile the serum levels of iron and ferritin were detected by atomic absorption spectrum and radioimmunoassay respectively. Results Compare to the control group ((10.5?5.7)?mol/L, (143.3?71.9)ng/ml),the serum levels of iron and ferritin were obviously high in patients with severe NAFL ((21.5 ?11.1 ) ?mol/L , (199.3?72.1)ng/ml) or moderate AFL((20.9? 9.3 )?mol/L,(217.6.0?71.8)ng/ml) and severe AFL ((29.1?6.5) ?mol/L ,(284.7?77.9)ng/ml) ( P
2.The correlation between genetic variation and expression of DLL1 gene and bone marrow suppression after neoadjuvant chemotherapy for breast cancer

Objective:To investigate the effects of two SNP sites of delta-like ligand protein-1 (DLL1) gene rs2738822 (C>T) and rs9459988 (T>G) and gene expression on bone marrow suppression after neoadjuvant chemotherapy for breast cancer.Methods:Breast cancer patients who received neoadjuvant chemotherapy were selected as study subjects, including 90 patients with severe bone marrow suppression and 72 patients with mild bone marrow suppression. Patient’s demographic characteristics and laboratory test indicators were collected. Two SNP sites of DLL1, rs2738822 and rs9459988, were genotyped by capillary electrophoresis and section analysis (SNaPshot) . The relative mRNA expression of DLL1 gene was detected by quantitative reverse polymerase chain reaction (QRT-PCR) method.Results:For The rs2738822 of DLL1 gene, the genotype distribution difference between severe and mild bone marrow suppression groups was statistically significant ( χ2=8.622, P=0.013) . Compared with CC genotype, CT and TT genotype carriers had a higher risk of severe bone marrow suppression, with an OR value of 2.746 (1.335-6.882) and 3.054 (1.282-8.143) , respectively. The dominant model results showed that TT OR CT carriers had a significantly higher risk of severe bone marrow suppression than THOSE with CC genotype [ OR=2.976 (1.231-4.963) ]. For rs9459988, there was no significant difference in genotype distribution between severe bone marrow suppression group and mild bone marrow suppression group ( χ2=2.149, P=0.342) . Results of the dominant model showed that TG or GG carriers had a significantly higher risk of severe bone marrow suppression than TT carriers, with an OR value of 2.046 (1.053-5.611) . The relative mRNA expression level of DLL1 gene was 1.15±0.23 in patients with severe bone marrow suppression, which was significantly lower than that in patients with mild bone marrow suppression (2.64±0.51) ( t=6.381, P<0.001) . For rs2738822, with the increase of T allele, the relative mRNA expression level of DLL1 gene decreased gradually ( P<0.05) . For rs9459988, the relative mRNA expression level of DLL1 gene in patients with mutant allele G was also significantly lower than that in wild-type CC carriers ( P<0.05) . Conclusion:Mutations of DLL1 genes rs2738822 and rs9459988 are related to the occurrence of severe bone marrow suppression after neoadjuvant chemotherapy for breast cancer, and can be used as a genetic marker to predict the degree of bone marrow suppression after neoadjuvant chemotherapy for breast cancer patients.
3.Impact of Expression of DHA AmpC ?-Lactamases Mediated by Plasmid on Antibiotics Susceptibility of Clinical Isolates of Klebsiella pneumoniae

OBJECTIVE To investigate the prevalence of DHA AmpC ?-lactamases mediated by plasmid in Klebsiella pneumoniae in China.METHODS Antimicrobial susceptibility test was conducted by the methods of double agar dilution and ESBLs confirmatory in K-B method according to the criteria of guidelines of CLSI.AmpC ?-lactamases were detected on the basis that AmpC ?-lactamases could be inhibited by 3-aminophenylboronic acid(APB).Gene chip technology and PCR were used to detect ESBLs and AmpC gene.RESULTS Among total 34 isolates of K.pneumoniae 32(94.1%) produced AmpC ?-lactamases and ESBLs.The most common(38.3%) were types DHA and TEM and SHV.MIC50 and MIC90 of all strains to all tested antimicrobial agents were lower than 34 strains tested 0.25?g/ml and 0.5?g/ml.Fourteen strains AmpC and ESBLs were conjugated successfully.CONCLUSIONS DHA AmpC ?-lactamases mediated by plasmid are the most common in K.pneumoniae in General Hospital of PLA of China.The most common(38.3%) are types DHA and TEM and SHV.Fourteen(41.2%) strains can be spreaded by plasmid.

ObjectiveTo observe the effect of indomethacin on the migration of breast cancer cell line MCF-7 in vitro and investigate the mechanism involved.MethodsThe migration of MCF-7 cell line stimulated with or without indomethacin were tested using transwell plates consisting upper and lower chambers separated by Millipore polycarbonate membrance filters with 8 μm pore sizes; the levels of chemokine receptor 4(CXCR4),cyclooxygenase(COX-2),epidermal growth factor receptor(EGFR) and vascular endothelial growth factor(VEGF)expression in MCF-7 cell line were detected by flow cytometry,Real-time PCR and ELISA,respectively.Results Indomethacin decreased the migration ability of MCF-7 cell line significandy.CXCR4 membrane expression was significantly reduced in a time-dose dependent manner,and CXCR4,COX-2 and EGFR mRNA levels were significantly downregulated after indomethacin stimulation.However,exposure to indometahcin had no major effect on VEGF production of cells.ConclusionThe downregulation of CXCR4,COX-2 and EGFR expression might be the primary mechanism involved in the inhibitory effect of indomethacin on the migration of MCF-7 cell line.

Objective In order to know first aid awareness and training needs of non-medical college students in Shiyan city,and to provide the basis for an efficient first-aid training.Methods A total of 1063 non-medical colleges in Shiyan city were surveyed by random sampling method.Results 64.61% of students awared of their own lack of knowledge of first aid,only 3.8 % feel rich;based on the first aid knowledge they obtained at present,46.92 % did not hesitate to rescue the stranger.After receiving systematic training,the rate rise to 78.9 %,68.09 % of students worried about their lack of first aid skills were the biggest obstacle for them to implement of rescue;98.3% of the students asked to undergo first aid training,92.27% of students like approach first aid skills was hands-on model.33.03% of students believe that medical schools were the best institutions to undertake emergency training,23.46% of students chose the hospital.Conclusion Non-medical college students in Shiyan city have a bad awareness for firstaid knowledge and a strong desire for training.It is necessary that relevant departments will formulate targeted training programs to improve college students' first-aid response and improve regional emergency level.
6.Inhibitory effects of β-lapachone on the growth and invasion in gastric cancer cells

Purpose To investigate the effects ofβ-1apachone on inhibition of pro1iferation and migration and induction of apoptosis in gastric cancer ce11s in vitro. Methods The ce11 viabi1ity was detected using MTT and co1ony formation assay,the migration abi1ity was determined using scratch assay method,and the apoptosis was examined using f1ow cytometry. Meanwhi1e,the expression of biomarkers of pro1iferation,EMT markers andapoptosiswere detected using Western b1ot ana1ysis. Results β-1apachone cou1d significant1y inhibit the pro1iferation of SGC-7901 and AGS gastric cancer ce11s( P<0. 05),and down-regu1ate the expression 1eve1s of Skp2 and DEK pro-teins. β-1apachonecou1d a1so inhibited the invasion and moti1ity of gastric cancer ce11s via down-regu1ating the expression 1eve1s of MMP-2/9 and Ezrin proteins and up-regu1ating the epithe1ia1 markers. In addition,β-1apachone enhanced the apoptosis of gastric canc-er ce11s,down-regu1ation of BCL-2/Bax ratio and up-regu1ation of activated Caspase-3/8/9. Conclusions β-1apachone can effective1y inhibit the pro1iferation and induce the apoptosis of gastric cancer ce11s,and inhibit the migration of gastric cancer ce11s via MMPs and EMT pathways.
7.Effect of insulin treatment on cognitive function of patients with Alzheimer′s disease and type 2 diabetes mellitus

Objective To investigate the therapeutic effect of insulin combined with oral hypoglycemic agents on cognitive function of patients with Alzheimer′s disease(AD) and diabetes mellitus(DM).Methods We retrospectively reviewed 135 cases of AD patients with type2 DM(T2DM) admitted to our hospital between March 2011 and March 2013. According to drug use , they were randomly divided into two groups , oral hypoglycemic drug group ( group A, 65 cases ) and insulin combined with oral hypoglycemic drug group (group B,70 cases).After 12 months of follow-up, the Mini Mental State Examination (MMSE) score by Folstein et al and Clinical Global Impression Scale (CGI) were used to assess their cognitive function .Results After 12 months of treatment , the MMSE score decreased significantly in group A ( P=0.000), but not in group B(P=0.281).Also, CGI-C scores showed significant deterioration for all domains after 12 months in group A vs group B(P=0.001).Conclusion Insulin therapy can effectively slow down the decline of cognitive function in patients with AD .
8.Effect of Intervention of Somatosensory Interaction (Kinect) on Post-stroke Depression

Objective To observe the effect of somatosensory interactive technology of Kinect on post-stroke depression (PSD). Methods 60 patients with PSD were divided into control group (n=30) and experimental group (n=30), who accepted routine rehabilitation and Kinect in addition, respectively. They were assessed with Zung's Self-rating Depression Scale (SDS) before and 4 weeks after intervention. Results The scores of SDS was (43.25±10.11) in the experimental group, which was less than that in the control group (55.67±8.80) after intervention (P<0.01). Conclusion Rehabilitation with somatosensory interactive technology of Kinect may improve the efficiency on PSD.
9.Mechanisms of miR-503 inhibiting radio-resistance of esophageal squamous cell carcinoma cells by targeting ERCC1

Objective: To investigate the mechanism of miR-503 modulates radio-resistance of esophageal squamous cell carcinoma (ESCC) by targeting excision-repair cross-complementing 1 (ERCC1). Methods: The expression level of miR-503 in radio-resistant ESCC tumor tissues and KYSE140 and KYSE140R cells was detected by qPCR. The miR-503 mimic, miR-503 inhibitor or si-ERCC1 was transfected into KYSE140 and KYSE140R cells.After radiation treatment, the colony formation assay and CCK-8 assay were used to detect the proliferation of KYSE140R cells. Flow cytometry was used to detect apoptosis of KYSE140R cells. WB was used to detect changes in protein expression of ERCC1. Dual luciferase reporter gene assay was used to validate the targeting relationship between miR-503 and ERCC1. Results: The expression level of miR-503 was down-regulated in radio-resistant tissues and ESCC cell lines (all P<0.01). Over-expression of miR-503 significantly inhibited cell proliferation and promoted apoptosis of KYSE140R cells (all P<0.01). Dual-luciferase reporter assay validated that ERCC1 was a target gene of miR-503, and miR-503 negatively regulated the expression of ERCC1. Over-expression of miR-503 significantly down-regulated the expression of ERCC1 in KYSE140 and KYSE140R cells (both P<0.01), inhibited cell proliferation (both P<0.01), but significantly increased apoptosis rate (all P<0.01); knockdown of ERCC1 exhibited a similar effect, while knockdown of both ERCC1 and miR-503 reversed the above effects. Conclusion: Over-expression of miR-503 up-regulated the radio-sensitivity of KYSE140R cells by targeting ERCC1.
10.Latent profile analysis and influencing factors of health emergency literacy among emergency department nurses

Objective:To investigate the current situation of health emergency response literacy among emergency nurses, analyze the potential profile of emergency nurses′ health emergency response literacy and explore the influencing factors of different categories, so as to provide reference for formulating targeted and specific training intervention programs.Methods:From September to October 2022, 386 emergency nurses from 5 tertiary A-level hospitals in Shandong Province were selected as the research objects by convenient sampling method. A cross-sectional survey was conducted by using the general information questionnaire, the Literacy Self-assessment Scale of Public Health Emergency Response In Nurses, the Chinese version of Modified Self-leadership Questionnaire, and Nurse Perceived Professional Value Scale. The latent profile analysis was carried out with the results of 3 explicit indexes of the Literacy Self-assessment Scale of Public Health Emergency Response In Nurses. At the same time, the influencing factors of different latent profiles of health emergency literacy were explored by univariate analysis and ordered multi-classification Logistic regression.Results:The health emergency literacy of emergency department nurses was divided into three latent profiles: high health emergency literacy type accounting for 45.4% (175/386), medium health emergency literacy type accounting for 41.5% (160/386), and low health emergency literacy type accounting for 13.1% (51/386). Ordinal Logistic regression showed that the main emergency work post was pre hospital first aid, participated in public health emergency rescue operations, self leadership, and professional value were protective factors of health emergency literacy (all P<0.05). Age≤25 years old and junior college degree were risk factors of health emergency literacy (both P<0.05). Conclusions:Nursing managers can implement personalized training and intervention strategies according to different demographic characteristics of the health emergency literacy of emergency nurses, develop self leadership improvement plans from individual and organizational factors, and actively cultivate their sense of professional value, in order to comprehensively improve the health emergency literacy of emergency nurses.
