2.Clinical analysis of traditional Chinese medicineKuiyu decoction combined with quadruple therapy for peptic ulcer

ObjectiveTo investigate the clinical curative effect of traditional Chinese medicineKuiyu decoction combined with quadruple therapy for peptic ulcer (PU).MethodsThis study was a randomized controlled trial. 82 patients with PU and conformed to the indusive criteria were randomly divided into a control group and a treatment group by the random number table method, with 41 cases in each group. The control group was treated with quadruple therapy, while the treatment group was treated with traditional Chinese medicineKuiyu decoction based on the treatment of the control group. Integral change of main symptoms before and after treatment and the level changes of serum motilin (MTL), somatostatin (SS) of the two groups before and after treatment were compared and clinical total effective rate was evaluated.ResultsThe total effective rate was 95.1% (39/41) in the treatment group and 80.5% (33/41) in the control group. There was statistical difference between the two groups(χ2=4.100,P=0.043). Stomach pain (0.58 ± 0.21vs.0.73 ± 0.28,t=2.744), gasteremphraxis (0.35 ± 0.12vs.0.43 ± 0.14,t=2.778), sour regurgitation integral (0.28 ± 0.09vs.0.34 ± 0.11,t=2.703) and serum MTL level (358.5 ± 81.3 pg/mlvs.422.8 ± 86.9 pg/ml,t=3.460) in the treatment group were lower than those in the control group (P<0.01);the serum SS level (87.5 ± 18.7 pg/mlvs.73.8 ± 17.9 pg/ml,t=3.389) in the treatment group was higher than that in the control group(P<0.01).ConclusionsTraditional Chinese medicineKuiyu decoction combined with quadruple therapy for the patients with PU can promote SS secretion through decreasing MTL, so it can improve the clinical effect.
3.The diagnostic value of musculoskeletal ultrasound in gouty arthritis

Objective To observe the characteristic features of musculoskeletal ultrasound (MSUS) in gouty arthritis (GA) and to assess its value in GA diagnosis.Methods Patients with GA and non-GA who underwent MSUS examination in Chinese PLA General Hospital were enrolled in this study. The synovitis,tenosynovitis,soft tissue swelling,bone erosion,bursitis and ossification were measured by greyscale ultrasound,and the synovial vascularity was measured by power Doppler ultrasound to evaluate the degree of inflammation.Results The ultrasound images of GA were intended to have specific features like tophus,hyperechoic deposition on the surface of the articular hyaline cartilage ( double contour sign ),hyperechoic deposition in the synovium appearing as bright stippled foci,and bright strap in tendons with a significant difference when compared with non-GA group (P < 0.05).The percentage of tenosynovitis and bone erosion in non-GA group was higher than that in the GA group with a significant difference (P <0.05 ). Tophus,double contour sign,bright stippled foci or bright stippled foci in tendons together suggested gout with a specificity of 92.4% and a sensitivity of 60.3%,the AUC of ROC was 0.764 ( P <0.05).Conclusion Tophus,double contour sign,bright foci and bright strap detected by MSUS are of diagnostic value on GA and MSUS may be a promising tool in diagnosing GA.
4.The plasma concentration of omeprazole and metabolites 5′-hydroxy omeprazole, omeprazole sulphone are determinated by HPLC

AIM To set up a method which can determining the blood concentration of omeprazole and its metabolite 5′ omeprazole, omerpazole sulphone in order to study its cinical pharmacokinetics. METHODS The blood concentration of omerpazole was determinated by HPLC. RESULTS Calibrated standard curve of omeprazole in blood is Y=-0 004 499+0 001 909X (r =0 9990), the recoveries of three concentrations 50, 500, 2 000 mg?L -1 are 90 36, 109 62, 108 91%, respectively; and the precisions are 9 86,7 86, 15 52% , respectively. Calibrated standard curve of its metabolite 5′ OH omeprazole in plasma is Y=-0 003 659+0 001 328X(r =0 9970), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 79 42%, 96 49%, 95 04%, respectively; and the precisions are 8 95%, 4 52%, 9 73%, respectively. Calibrated standard curve of its another metabolite omeprazole sulphone in plasma is Y=0 009 248+0 001 765X (r =0 999 2), the recoveries of three concentrations 20, 200, 1 000 mg?L -1 are 94 44%, 105 59%, 104 26%, respectively; and the precisions are 8 72, 8 58, 9 60%, respectively. After 20 mg omeprazole were administered by a voluteer via oral, C max of 5′ OH omeprazole, omeprazole, and omerpazole sulphone were 14 622 7, 408 433 2, 454 363 7 mg?L -1 . CONCLUSION The method is good enough to study pharmacokinetics of omeprazole.
5.Design, synthesis and biological activity evaluation of adenosine analogues.

N6-(2-Hydroxyethyl) adenosine, HEA (1), an active ingredient isolated from cultured mycelia of cordyceps species which is a famous traditional tonic in China, showed brain protective, sedative hypnotic activity in pharmacological tests. In order to explore novel non-benzodiazepine sedative-hypnotic agents, HEA was treated as the lead compound. Twenty three target compounds were designed and synthesized. Their chemical structures were characterized by 1H NMR, MS and elemental analysis. Pharmacological test in vivo showed that target compounds 8, 4, 13 were more active than HEA on locomotor and gasping activities of mice. Structure-activity relationships showed that the ribose moiety at N-9 position of adenine base was critical for activity.
6.Effect of phytohormones and elicitors on growth and anthraquinone production of Cassia obtusifolia hairy roots

Objective To study the effects of phytohormones and elicitors on the growth and anthraquinone production of Cassia obtusifolia hairy roots. Methods Blades and petioles of C. obtusifolia〖WTBZ〗 were infected with Agrobacterium rhizogenes LBA9402 and the in vitro culture system of the hairy root was established. Results Hairy roots were induced by A. rhizogenes LBA9402. The results showed that the addition of 0.5% NAA to the hairy roots increased the content of five kinds of anthraquinones by four times, and Aspergillus niger elicitor had no much effect and JA had slightly inhibitory effect on root growth, but both of them significantly promoted the anthraquinone accumulation, the anthraquinone yield increased more than two times with the treatment of 2.0 mL elicitor/50 mL medium and by 73% with the treatment of 10 ?mol/L jasmin acid (JA), respectively. JA had little inhibitory effect on the growth of fairy roots. Conclusion The fundation has been established for further oplimizing the proper cultural system for C. obtusifolia hairy roots and regulating the secondary melabolite.
8.Effect of various media on quality of hairy root clones of Rheum palmatum

Objective To study the effect of optimized media and clones on growth and anthraquinone yield in hairy root cultures of Rheum palmatum.Methods The four monoclones and four media were optimized statistically for studying the effect on the biomass accumulation and five anthraquinones yield in hairy root cultures of R.palmatum.Results Significant differences in biomass were shown among various clones and media,in which monoclone DH7a was the highest and WP medium was the best.Chrysophanol and physcion were predominant among five free anthraquinones in hairy root cultures of R.palmatum.Notable differences of total five free anthraquinones yield were observed among different clones, in which DH7a was the highest.Reciprocity analysis showed that the highest yield of five free anthraquinones is in the combinations of DH7a with B5 medium.DH5a less and DH5a the least with WP medium,DH7a and B5 medium were predominent for the yield of above five free anthraquinones with exception of DH5c which has no significant difference in four media.Conclusion The study lays the foundation for mass production of anthraquinone by hairy root culture of R.palmatum.
9.Differential gene expression on PBMCs in the high sensitive patients with uremia

%85, entered into the high sensitive group (30 cases), and the patients with negative PRA into control group. The fresh blood was collected, and PBMCs was collected by Ficoll method. Total RNA were extracted by one-step technique and purified. The total RNA in high sensitive group were labeled with Cy5-dUTP, and control group with Cy3-dUTP, then the cDNA probe was labeled by reverse transcript way. High throughout gene chip ((16 920)) was hybrided and scanned. Cy3/Cy5 image files were copied. Then fluorescent signal value of gene expressing was obtained, and differential expression genes were sifted. RESULTS: Among the differential expression 877 genes, there were up-regulated 88 genes and down- regulated 789 genes. The mechanism of high sensitive status in human immune system was analyzed by some function-known genes which coded NY-REN-55 antigen, CD100, defender against cell death 1, breast cancer resistance protein, transcriptional repressor, death domain containing protein, cyclophilin-33A, rapamycin-binding protein, heat shock protein 40, interferon-alpha receptor and STAT inhibitor-2. CONCLUSIONS: The effect of PBMCs in high sensitive status of human immune system in patients with uremia may be associated with recognition of auto-antigen,signal conduction, aggregation and differentiation of B lymphocyte, anti-apoptosis and resistance of immunosuppressant. 10.Determination of Norcantharidin using high performance liquid chromatography

ABSTRACT AIM To determine Norcantharidin in its tables. METHODS A PHLC method was adopted, using Polaris C18 column(5?,4.6?250 mm) and UV detector at 211 nm wavelength. The mobile phase is consisted of water: methanol(85 : 15 V/V), adjust the pH to 3.1 with phosphoric acid. RESULT Excellent liner relationship was ob- tained from the range of 25 to 1 000 mg ? L-1. The minimal limit of Norcantharidin was 0.2 mg ? L-1. Recovery was 100.84% and RDS was 1.335%. COUCLUSION Practice suggested that this method was rapid, accuracy and suitable for Norcantharidin determining.
