1.The use of sodium hyaluronate in cryopreservation of cornea

Objective To assess the effect of cryopreservation of cornea in glycerol,to analyze the protective effect of sodium hyaluronate on corneal endothelial cells(CEC) during preservation,and to find out a convenient and cheap method for the storage of cornea.Methods 20 rabbit corneas were divided into two groups randomly.The corneas of control group were stored directly in pure glycerol at-25℃,while in the experimental group sodium hyaluronate was evenly anointed on the corneal endothelial surface of the cornea before being cryopreserved in glycerol.2 months later,the effect of preservation for both groups was examined with trypan blue combined with alizarin red staining(viability staining) for corneal endothelial cells,electron-microscope examination for preserved corneas,and rabbit experimental penetrating keratoplasty(PKP) with the long-term cryopreserved corneas from both the experimental and control groups.Results CEC viability could be preserved by both methods.CEC trypan blue combined with alizarin red staining(viability staining) showed that the CEC density of cornea in the experimental group was 3 067.9?127.6/mm2,which was higher than that of control group(2 899.9?134.0/mm2).The mortality of CEC in experimental group was 4.2%?3.4%,lower than that in control group(13.8%?2.2%).Suceessful rate of PKP was higher in the experimental group with higher CEC density.Conclusion Cryopreservation with glycerol can preserve the viability of CEC.Sodium hyaluronate can reduce the damage to endothelium in the preserving procedure and can obviously improve the effect of preservation.The improved cryopreservation of cornea with glycerol may be a convenient,economic and effective method for eyebank.
2.Experimental study on transplanted rectal tumor of mice treated by photodynamic therapy combined with 5-FU

Objective To explore the effectiveness of photodynamic therapy(PDT) combined with 5-FU on the treatment of transplanted rectal tumor in mice.Methods The nuke mice transplanted tumor of human rectal cancer HR8348 cells were treated with PDT alone,5-Fu alone or PDT combined with 5-Fu respectively.The volume and weight of the tumors were measured and the histopathologic changes were studied.The level of malonaldehyde(MDA) in the tumors in different groups was analyzed.Results The lethal effect on transplanted rectal tumor was more obvious and the level of MDA was more higher in the PDT combined with 5-Fu group than those of other groups.Conclusion PDT combined with chemotherapy may serve as a method in treatment of tumor.
3.Effects of recombinant human granulocyte-colony stimulating factor therapy on rat pulmonary hypertension and its influence on endothelial progenitor cells

Objective To investigate the effects of recombinant human granulocyte colony stimulating factor (rhG-CSF) therapy on pulmonary hypertension,and its influence on number and functions of circulating endothelial progenitor cells (EPCs) in rats.Methods Eight week old Sprague-Dawlay rats were randomized into model group,treatment group and control group (8 rats in each group).The rats in model group and treatment group were treated with single subcutaneous injection of 1% monocrotaline (50 mg/kg) to induce pulmonary hypertension models,while the control group was treated with phosphate buffered saline.Five days later,the rats in treatment group were administrated with 50 μg/(kg· d) rhG-CSF for 3 days.On day 21,peripheral blood was collected from caudal vein in all groups,and the percentage of EPCs in 100 000 mononuclear cells was evaluated by flow cytometry.Right ventricular systolic pressure was assessed,and the pathological changes of lung tissue and pneumoangiogram were observed by HE staining.Meanwhile,peripheral mononuclear cells collected from caudal vein were separated and cultured in vitro for EPCs.The cell ffunctions as proliferation,adhesion and migration ability were assessed.ANOVA and LSD test were applied as statistical analysis methods.Results (1) The right ventricular systolic pressure of rats in model group was higher than that in the controls [(48.13 ± 2.85) mm Hg vs (27.88 ± 3.04) mm Hg,t=2.016,P<0.01],the lesion of endothelial cells in pulmonary arteriolar was evident,and the vessel wall was thickened.The pulmonary artery pressure of rats in the treatment group [(30.38 ± 2.83) mm Hg] was lower than that in the model group and close to the level of control group (t=0.376,P>0.05) with mild pulmonary pathological changes.(2) The percentage off peripheral blood EPCs in mononuclear cells in the model group was decreased as compared to the control group [(0.016±0.007) % vs (0.031±0.011) %,t=2.617,P<0.01].After administration ofrhG-CSF,the EPCs in treatment group [(0.042±0.013) %] was increased evidently as compared to the model group (t=4.325,P<0.01) and the control group (t =1.942,P<0.05).(3) The proliferation,adhesive and migrated cells of EPCs in model group were 0.49 ± 0.04,(6.93 ± 1.47) cells/HPF and (7.22±1.53) cells/HPF,lower than those in control group [0.68±0.07,(11.05±1.73) cells/HPF and (12.58±2.15) cells/HPF] and treatment group [0.63±0.06,(12.35±1.82) cells/HPF and (12.97±2.84) cells/HPF],the differences were statistically significant(all P<0.05).Conclusions rhG-CSF may be effective in treating pulmonary hypertension through up-regulating the number and function of circulating EPCs in rat model of pulmonary hypertension.
4.Comparative study on serum HMGB1 level in patients with different grades of acute biliary tract infection

Objective To comparatively analyze the difference and characteristics of high mobility group box-1 protein(HMGB1) level with the levels in the patients with different severities of acute biliary tract infection (ABTI) to provide reference for its clinical diagnosis and treatment .Methods One hundred cases of ABTI in our hospital were divided into the mild group (48 cases) ,moder-ate group (29 cases) and severe group (23 cases) according to the severity of the disease .The HMGB1 detection results were com-pared among 3 groups and the differences in different disease types ,sex and age were analyzed .Results (1)The HMGB1 level had statistically significant difference among 3 groups (P<0 .05) ,moreover the mild group< moderate group< severe group ;(2)in the detection results ,the HMGB1 level in the mild group and moderate group had no statistical difference between males and fe-males(P>0 .05) ,but in the severe group ,the HMGB1 level in males was significantly higher than that in females (P<0 .05);(3) the HMGB1 level in the mild group had no statistical difference among difference age groups (P> 0 .05) ,while in the moderate group and severe group ,the HMGB1 level in the patients aged > 60 years old was significantly higher than that in the patients aged ≤60 years old(P<0 .05);(4) in the above 3 groups ,the HMGB1 level in the patients with acute cholecystitis was signifi-cantly higher than that in the patients with acute cholangitis (P<0 .05) .Conclusion The study results analysis indicates that the severe the ABTI disease condition ,the serum HMGB1 level is also accordingly and relatively increased ,in the patients with different severity degrees of ABTI ,the serum HMGB1 level has significant differences in age ,sex and disease type ,which prompts that the HMGB1 level can be used as the laboratory index for predicting and reflecting the ABTI severity and can be paid attention to .
5.Transaction log and its analysis applied to hospital information security

Objective To study transaction log and apply its analysis to hospital information security to prevent data loss and treat abnormal data.Methods The application of transaction log was described in log backup and database recovery,and the optration of log analysis was explored in HIS.Sqlplus and Ftp tools in Oracle were used to analyze the transaction log.Results The tracking and analysis of the log files contributed to making clear the incident,finding program bug,getting back lost data.Conclusion Log backup,database recovery and log analysis were of great importance for hospital information security,and log analysis has to be emphasized on in the future.
6.Airway management of a series of patients with inhalation injury caused by smoke from smoke pot based on grade classification

In this study,we summarized airway management of a series of patients with inhalation injury caused by smoke from smoke pot based on grade classification,which were:establishment of a special team for these classified patients,classifying the patients into four sub-groups including extremely severe,severe,moderate and mild,establishment of a program for trachea management and relevant measures on these classified sub-groups.Key points of management on extremely severe patients were as follows:protective isolation,mechanical ventilation,nursing of extracorporeal membrane oxygenation and pneumothorax and mediastinal emphysema,prevention of tracheo-esophgeal fistula,nursing cooperation of fiberoptic bronchoscopy.Key points of management on severe patients were as follows:disinfection and isolation,rational oxygen therapy,sputum elimination management,observation of illness status,preparation of emergency treatment,prevention and nursing of complications.Key points of management on moderate patients were as follows:ventilation,oxygen uptake,aerosol inhalation,sputum elimination guidance,respiratory function training.For mild patients,there was no special management except ventilation,aerosol inhalation and regular pulmonary function examination.One patient died due to multiple organ failure complicated with massive hemoptysis,four patients recovered with airway scar proliferation and lung fibrosis,and fifty-five patients fully recovered.
7.Fingerprint research on authentication of official and unofficial plants of Rheum L. by RAPD

Object In order to identify the resources of official and unofficial plants in Rheum L.at molecular level, a random amplified polymorphic DNA (RAPD) analysis was performed. Methods The amplification program of polymerase chain reaction (PCR) was optimized. Arbitrary decamer primers of 121 based on the individual selecting method, and 64 based on the BSA method were screened. Results Four primers among all screened primers were found to be useful to amplify specific strips to the official plants of Rheum L. Conclusion This method is reliable, accurate, quick and reproducible for authenticating the official and unofficial plants of Rheum L. at molecular level. Moreover, the experiment results indicate that it is more effective to screen the primers using BSA method than using individual selecting method.
8.Epidemiological characteristics and meteorological factors in childhood Mycoplasma pneumoniae infection in Guilin

Objective To study the epidemiology of mycoplasma pneumonia (MP) infection in children and its relationship to meteorological conditions in Guilin. Methods Total of 1 342 hospitalized children with respiratory tract infections from Janu-ary 2011 to December 2012 were recruited, and the serum levels of mycoplasma pneumoniae antibody Ig (MP-Ig) were measured. Meanwhile, monthly meteorological conditions were collected. The relationships between the MP-Ig detection rates and meteoro-logical conditions were analyzed by multiple linear regression analysis. Results The total detection rate of MP-Ig was 23.2%with positive rates as 5.51%, 28.99%, 37.31%and 44.97%for 0-1, 1-3, 3-6 and>6 year-old groups respectively (P<0.01). The positive rate of female was higher than that of male with signiifcant difference (30.08%vs 19.05%, P<0.01). The positive rate of spring, summer, autumn and winter were 18.89%, 17.39%, 23.83%and 33.05%individually (P<0.01). Negative correlation was found be-tween detection rate of MP-Ig and monthly mean air temperature (P=0.001). Conclusion MP is one of the commonest pathogens of respiratory tract infection in children and mostly seen in elder female children with a higher incidence in winter. The main me-teorological factor is the low temperature that affects MP prevalence.
9.Clinical effect of conversion to Rapamycin on chronic allograft nephropathy

Objective To study the effect and safety of conversion from calcineurin inhibitors to rapamycin in kidney transplantation recipients with chronic allograft nephropathy.Methods In 82 kidney transplant recipients enrolled in this study,72 cases were diagnosed as having chronic allograft nephropathy by biopsy.Recipients (SRL group) were administered with rapamycin after withdrawal of calcineurin inhibitors.The doses of CNI in other recipients (non-SRL group) were not changed.Renal function,proteinuria,blood pressure,blood fat,hepatic function and hemogram were observed for 24 months in each group.Results During the follow-up period,serum creatinine level was dropped significantly in SRL group (P<0.05),but it was increased in non-SRL group (P<0.05).SRL group showed increased proteinuria,serum cholesterol and triglycerides (P<0.05),and reduced Plt (P<0.05).According to the renal function before conversion,the recipients who were administered rapamycin divided into four groups.In group A (Scr < 120 μmol/L),there was no significant difference in diverse variables before and after conversion.In group B (Scr 120-200 μmol/L and Banff Ⅰ-Ⅱ),renal function was improved,and proteinuria alleviated.In group C (Scr 120-200 μmol/L and Banff > Ⅱ),and group D (Scr >200 μmol/L),renal function was damaged to varying degrees and proteinuria was deteriorated.Conclusion It is safe and effective for patients with chronic allograft nephropathy to convert from calcineurin inhibitors to rapamycin.
