1.Propolis decreases the expression of MMP-9 in fibroblasts and monocyte-derived macrophage in vitro

Objective To explore the potential mechanisms of therapeutic effect of propolis in the treatment of diabetic wound. Methods Human fibroblasts(Fb) and monocytic THP-1 cells were routinely cultured at 37℃ in an environment of 95% air and 5% CO2 in RPMI medium containing 11mmol/l glucose, 15mmol/l HEPES and 5% fetal calf serum. To a portion of culturing THP-1 monocytes phorbol myristate actetate (PMA; 2?10 -7mol/L) was added. After 24 hours when approximately 75% of the monocytes became macrophage which attached to the plate. Propolis was added to a culture system of human fibroblasts(Fb), THP-1 monocyte and THP-1 monocyte-derived macrophage(Mac) with different concentration of propolis(10, 50, 100, 200?g/ml) and different glucose(5mmol/L; 25mmol/L), For each experiment the cells were cultured for 48 hours in the presence of Propolis. Then the media were collected and the protein expression of matrix metalloproternase-9 (MMP-9) were determined by gelatin zymography using a MDPF- labeled gelatin as substrate. The media were loaded onto an 8% SDS PAGE gel containing MDPF labeled gelatin (1%) after electrophoresis, the gels were placed on a long wave ultra violet light box and photographed. The area degraded by the MMPs was observed as dark bands on a light background. The MMPs were quantified by measurement of the band area using the Phoretix_1D_Advanced Digital Science Electrophoresis Documentation and Analysis System. Results the high glucose has a tendency to increase the expression of MMP-9, The propolis can decreased the expression of MMP-9, and has a dependent on doses tendency. F test P
4.Influence of lipopolysaccharide on the expression of matrix metalloproteinases and specific tissue inhibitors of metalloproteinases in fibroblasts

Objective To explore the effect of bacterial endotoxin lipopolysaccharide (LPS) on wound healing by observation of change in expression of matrix metalloproteinases (MMPs) and specific tissue inhibitors of metalloproteinases (TIMPs) in fibroblasts after the challenge of lipopolysaccharide. Methods Human dermal fibroblasts were cultured in RPMI medium containing 10% fetal calf serum (FCS). At cellular confluence the medium was changed, and the cells were cultured under serum free conditions with 0.1% bovine serum albumin (BSA) in the presence of LPS (0, 10?g/ml, 100?g/ml and 1000?g/ml). All cultures were maintained at 37℃ in a humidified atmosphere of 5% CO_2 in air. After 48-hour treatment, the medium was collected for measurement of MMPs by fluorescence labeled gelatin zymography and for measurement of TIMPs by fluorescence labeled reverse gelatin zymography. The expressions of MMPs and TIMPs were quantitatively analyzed with the band's area and density from the gel image using the Phoretix software. Results LPS up-regulated the expression of both MMPs and TIMPs in fibroblasts, and expression of TIMPs was higher than that of MMPs. Compared with base level, MMPs respectively increased 1.3, 1.7 and 1.6 times, TIMP-1 respectively increased 3, 4.5 and 3.6 times, TIMP-2 respectively increased 3, 6 and 5.6 times. Conclusion It seems that LPS showed no damaging effect on fibroblasts in regulation of matrix accumulation in wound healing.
5.Changes of PYK2 expression in hippocampal neurons and microglial cells in Kainate acid-induced status epilepticus of rats

Objective To investigate and evaluate the changes of PYK2 expression in hippocampal neurons and microglial cells in Kainate acid-induced status epilepticus. Methods Kainate acid-induced status epilepticus was established in rats, and by using immunostaining, changes of PYK2 expression in hippocampal neurons and microglial cells was investigated. Results Expression of PYK2 in hippocampal pyramidal neurons was markedly decreased after kainate acid-induced status epilepticus. However, 24h after the epileptic onset, a pronounced up-regulation of PYK2 and phosphor-PYK2 immunoreactivities were evident in amoeboid microglial cells. The upregulation of PYK2 and phosphor-PYK2 was in accordance with the morphological changes of the activated microglial cells. Conclusion PYK2 was activated in microglial cells after seizure. Furthermore, the activation of PYK2 in microglial cells after seizure might be related to the morphological and behavioral changes of microglial cells after activation.
6.Phosphorylation of PYK2 and p38MAPK in neurons after focal cerebral ischemia in rats

Objective To investigate and evaluate the changes of phospho PYK2 and phospho p38MAPK expression in neurons after focal cerebral ischemia. Methods Focal ischemia reperfusion model was established in rats, and by using immunostaining, the changes of phospho PYK2 and phospho p38MAPK expression in neurons was observed. Results Faint phospho PYK2 and phospho p38MAPK immunoreactivity were revealed in normal cortical neurons. Fifteen minutes after the ischemia onset, a pronounced upregulation of phospho PYK2 and phospho p38MAPK immunoreactivities were evident in these ischemia attacked neurons. The immunoreactivities of phospho PYK2 and phospho p38MAPK reached its peak at 30min after ischemia, and decreased 60min after ischemia. Conclusion Cerebral ischemia was able to induce neuronal PYK2 phosphorylation. The activation of PYK2 might link ischemia attack to the p38MAPK signaling pathway to initiate the neuronal response to the stress stimuli
7.Characteristic of Multidrug Resistant Organisms in Hospital and Community-acquired Infection

OBJECTIVE To investigate the characteristic of multidrug resistant organisms(MDROs) in hospital or community-acquired infection,so that it can approach effective infection management.METHODS Patients infected by Staphylococcus,Enterococcus,Escherichia coli,Klpneumoniae and A.baumanni were investigated in a Hospital from June 2008 to May 2009.RESULTS Among total 929 strains of bacilli from patients,367 were MDROs infection,the detection rate of MDROs,CA-MDROs or HA-MDROs was 39.5%,35.6% and 51.4% respectioety infection;The detection rate of MDROs from HAI patients was significantly higher than that from CAI patients(?2=19.2 P=0.00).There were differences between HA-MDROs and CA-MDROs in different departments.MRSA,ESBLs-kp And PDR-AB were isolated mostly from sputum of patients,and ESBLs E.coli mostly from urine of CAI patients.CONCLUSIONS It is important to analyze the characteristic of HA-MDROs or CA-MDROs in the treatment of infection or use of antibiotics from experience.MDROs control is full of difficulties as a result of high CA-MDROs rates.
8.The value of combined detection with MMP-9 and uPA in prognosis of pancreatic carcinoma

Objective To explore the value of combined detection with MMP-9 and uPA in the progno-sis of pancreatic carcinoma. Methods By immunohistochemistry PV methods,the expression of MMP-9 and uPA was respectively studied in 63 surgical specimens of primary pancreatic carcinoma and the survival time of patients with pancreatic carcinoma was analysed. Results The expressions of MMP-9 and uPA were positively related(r=0. 573,P=0. 000). The expression of MMP-9 and uPA significantly correlated with differentiation (r= -0. 271,P=0. 032;r= -0. 333,P=0. 008),TNM stages(r= -0. 449,P=0. 000;r= -0. 430,P=0. 000)and lymph node metastasis(r=0. 329,P=0. 009;r=0. 400,P=0. 001),separately. The expression of MMP-9 had also a significant correlation with tumer size(r= -0. 297,P=0. 018)and distant metastasis(r=0. 320,P=0. 011). Univariate analysis identified that tumor size(χ2 =8. 766,P=0. 012),differentiation(χ2 =29. 050,P=0. 000),clinical stage(χ2 =24. 940,P=0. 000),distant metastasis(χ2 =12. 846,P=0. 000), lymph node metastasis(χ2 =15. 457,P=0. 000),MMP-9(χ2 =32. 700,P=0. 000)and uPA(χ2 =41. 495,P=0. 000)were significantly associated with prognosis. Kaplan-Meier survival analysis showed that 1-year survival rate of patients with MMP-9 ( -),uPA ( -)were significantly longer than that of the patients with MMP-9( ﹢),uPA( ﹢),respectively(χ2 =32. 700,P=0. 000;χ2 =41. 495,P=0. 000);1-year survival rate of patients with MMP-9( -)/uPA( -)was significantly longer than the others( Log-rank test,χ2 = 54. 892, P=0. 000). COX regression revealed that differentiation(RR=2. 315,P=0. 004),clinical stage(RR=1. 694, P=0. 002),MMP-9(RR=0. 165,P=0. 000)and uPA(RR=0. 244,P=0. 007)was independent prognostic factors in pancreatic carcinoma. Conclusion They may have a synergistic function in the the process of growth and invasion in pancreatic cancer between MMP-9 and uPA,and the posssible mechanism is that uPA activate degradation of MMP-9,which is not favorable to prognosis. Combined analysis of MMP-9 and uPA may lead to a more reliable prognostic estimation,as the beneficial supplement of the differentiation,and clinical stage to judge the prognosis of pancreatic cancer.
9.Disposable Over-shoes Using for Nosocomial Infection Control in Intensive Care Unit

OBJECTIVE To analyze the effect of the disposable over-shoes for the control of nosocomial infection of the intensive care units(ICU). METHODS The effects of the disposable over-shoes for the environment contamination and nosocomial infection control of the surgical ICU were investigated. RESULTS The mean of air bacteria colony counts when disposable over-shoes were worn was lower than that when without their use by healthcare workers (P0.05). The rates of nosocomial infection beteen them were 21.5‰ and 17.1‰,respectively. CONCLUSIONS The use of disposable over-shoes can't improve the environment quality and is not benefit for the control of nosocomial infection of surgical ICU.
10.Effect of comprehensive medical goal appraisal on hand hygiene compli-ance rate of health care workers

Objective To evaluate the comprehensive medical goal appraisal system on hand hygiene compliance rate of health care workers(HCWs).Methods Comprehensive medical goal appraisal system was adopted to inter-vene hand hygiene compliance rate of HCWs in a comprehensive hospital ,hand hygiene compliance rates of HCWs and consumption of instant hand sanitizer per bed-day before (December 2012)and after intervention (January 2013-June 2014)were compared.Results Hand hygiene compliance rate after intervention was higher than before interven-tion (85.17% [18 208/21 379]vs 39.92%[853/2 137]),hand hygiene compliance rate enhanced by 113.35%(χ2 =2 590. 81,P <0.001).Hand hygiene compliance rates of HCWs of different departments,different occupations and different hand hygiene moments were all higher than before intervention (all P <0.001);after intervention ,hand hygiene compliance rate revealed a increased tendency,and has maintained high since October 2013 (>90%),consumption of instant hand sanitizer before and after intervention was 7.24 mL/bed-day(4 200 L/579 841 bed-day)and 10.54 mL/bed-day(9 323.5L/884 489 bed-day)respectively,the consumption after intervention increased by 45.58% compared with that before intervention. Conclusion Comprehensive medical goal appraisal can effectively enhance hand hygiene compliance rate ,and maintains at a high level;the measure can affect hand hygiene behavior of HCWs by hawthorne effect,and is an effective and long-term measure to improve hand hygiene compliance of HCWs.
