Objective To investigate the effect of Rumo Shengji San on promoting tissue regeneration of skin defect after repair with tissue-engineered skin. Methods The dermal scaffolds were reconstructed by collagen sponge in which human skin fibroblasts were transplanted inside and human keratocytes were transplanted on the surface. The tissue-engineered skin was transplanted into the wound surface of defected skin in naked mice.Rumo Shengji San was applied externally once every other day after transplantation for 2 days. The animals were randomly divided into two groups:external application group and control group,4 mice in each group. The differentiation tissue of the tissue-engineered skin was sampled 9 days after transplantation. The epidermis thickness and dermal capillary proliferation in wound of defected skin were observed with HE staining, and double-labeled immunofluorescence method was used to test dermal laminin(Ln) and typeⅠcollagen protein expression. Results Rumo Sheng Jji San promoted the development of epidermis after transplantation for 9 days. The number of capillaries in dermis was increased, and the expression of Ln and typeⅠcollagen protein was promoted in external application group than that in the control group. Conclusion Rumo Shengji San can promote tissue regeneration of skin defect after repair with tissue-engineered skin.

BACKGROUND:Mesenchymal stem cells are found to have the immunoregulatory activities and a potential application prospect in the treatment of autoimmune diseases.
OBJECTIVE:To explore the mechanism of transplanting mesenchymal stems cells on the treatment of multiple sclerosis.
METHODS:The mouse mesenchymal stems cells were prepared, and injected into the al ogenic and syngenic normal mice, to detect the frequency of CD4+CD25+Foxp3+T cells in the spleen, thymus, and lymph nodes by flow cytometry, and to detect the Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes by reverse transcription-PCR.
RESULTS AND CONCLUSION:Transplantation of mesenchymal stem cells on normal mice led to a significant up-regulation of CD4+CD25+Foxp3+T cells, Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes both in the al ogenic and syngenic transplant groups. Transplantation of mesenchymal stem cells may be an available method in the treatment of autoimmune diseases, and CD4+CD25+Foxp3+T cell, Foxp3, transforming growth factor-β1, and interleukin-10 may be involved in this process.

OBJECTIVE:To determine the resistance of clinically isolated bacteria to meropenem METHODS:The resistance of 150 clinically isolated strains to meropenem and several other antibiotics was detected by K-B method and the drug resistant spectra to E coli were compared between meropenem and imipenem RESULTS:The sensitivity of 130 clinical isolates to meropenem was 100% No cross resistant strains of E coli was found to meropenem and imipenem CONCLUSION:Meropenem is highly sensitive to the clinical isolates, and its antibacterial spectrum is similar to that of imipenem

Objective:To investigate perioperative management of anesthesia for septic shock. Methods:Anesthesia performed on an elderly patient with septic shock who underwent emergency surgery in Beijing Hospital was analyzed and summarized, with a literature review. Results:Etomidate, Ketamine, cis Atracurium and Remifentanil are preferred for anesthesia induction, and Remifentanil and Sevoflurane are the first choices for anesthesia maintenance.Combined application with Dexmedetomidine may improve patients' prognosis. For septic shock patients with new-onset atrial fibrillation, β-blockers are preferred for perioperative anti-arrhythmia.If necessary, propafenone or amiodarone can be used for cardioversion.Perioperative ultrasound evaluation may be used to guide perioperative fluid therapy and vasoactive drug administration for septic shock in the future. Conclusions:Anesthesiologists should place a high value on and have a good command of the main aspects of perioperative management of anesthesia for septic shock.

Objective To assess the safety and efficacy of preoperative intensity-modulated radiotherapy(IMRT) with oral capecitabine in patients with locally advanced mid-low rectal cancer using a simultaneous integrated boost (SIB) of tomotherapy. Methods Total 16 patients with resectable locally advanced mid-low rectal cancer (patients with T3 to T4 and/or N ± rectal cancer) were enroll in current study.Patients were received IMRT to 2 dose levels simultaneously (55 and 47.5 Gy in 25 fractions) with concurrent capecitabine 825 mg/m2 twice daily,5 days/week.Total mesorectal excision was performed at 8 to 9 week after the completion of chemoradiation. The primary end point included side effect, the rate of sphinctersparing, postoperative complication and pathological complete response rate (pCR) were observed.Side effects were scored using the National Cancer Institute Common Terminology Criteria for Adverse Events version 3.0.Results All patients were received chemoradiotion therapy without any break.Tomotherapy showed superiority with respect to target coverage, homogeneity and conformality. Two patients refused to perform radical surgery because of almost complete primary tumor regression and complete symptom relief after neoadjuvant therapy.Fourteen patients underwent surgical resection and 11 patients (78.6%) underwent sphincter-sparing lower anterior resection.Four patients(28.6%) had a pathological complete response. The incidence of grade 1-2 hematologic,gastro-intestinal toxicities were 62.5% (10/16) and 18.8% (3/16).The incidence of grade 3 skin toxicities were 68.8%(10/16).Grade Ⅳ side effect was not observed.Surgical complications (incisional infection on thirteen after surgery) were observed in 1 patient. Conclusion Preoperative simultaneous integrated boost of tomotherapy with concurrent oral capecitabine is safe and well tolerated in patients with a promising local control. However,a larger number of patients and a long follow-up are required to assess its potential superiority.

Objective To examine the mechanism by which miR-155-5p regulates sex determining region Y-boc 1(SOX1) expression in breast cancer cell line MCF-7,and to investigate the effect of miR-155-5p on the invasion and metastasis of these cells. Methods After up-regulation or down-regulation of the expression of miR-155-5p in MCF-7 cells, the transwell assay was performed to detect the invasive and migrating ability of MCF-7 cells;SOX1 mRNA expression and protein levels were detected by real-time PCR and Western blot, respectively;immunofluorescence was used to test the distribution and levels of SOX1 pro-tein,and dual luciferase reporter experiments to discover the binding of miR-155-5p to the 3′-UTR region of SOX1 mR-NA.Results Up-regulating the expression of miR-155-5p could enhance the migrating and invasive ability of MCF-7 cells and simultaneously decrease the mRNA and protein levels of SOX1(P <0.05).On the contrary, inhibition of miR-155-5p depressed the invasion and migration of MCF-7 cells and increased the expression of SOX1(P < 0.05).Dual luciferase reporter experi-ments confirmed that miR-155-5p could bind to the specific sequence in the 3′-UTR of SOX1 mRNA and play a role of regula-tor.Conclusion miR-155-5p promotes the invasion and migration of breast cancer cell line MCF-7 via inhibiting the expression of SOX1.
