1.Cloning and sequence analysis of methyltransferase gene in four species of Huperziaceae

Objective To clone and analyze the coding region of methyltransferase gene from four species of Huperziaceae, the species included Huperzia serrata var.longipetiolata, Phlegariurus minchegensis, Phlegariurus austrosinicus and Phlegariurus petiolatus.Methods The methyltransferase coding region sequences were cloned by RT-PCR with the template of total RNA extracted from four Huperziaceae.Then the sequences were analyzed by means of BLAST and MEGA 5.0 software.ResuIts The coding region sequences of the four species of Huperziaceae were highly similar to the methyltransferase in the database.ConcIusion The methyltransferase genes of the four species of Huperziaceae were cloned in this study.The result will provide a foundation for further elucidate the mechanism of Huperzine A biosynthesis and metabolic pathways in Huperziaceae plants.
2.A comparative study of two reprogramming systems for inducing pluripotent stem cells from human dental origin

Objective To comparatively study the features of two reprogramming systems of induced pluripotent stem cells (iPSCs)from human dental origin.Methods Two kinds of reprogramming system,i.e.STEMCCA lentivirus /feed layer and Sen-dai virus /matrigel were used to induce human stem cells from apical papilla(SCAP)into iPSCs,respectively.The induction efficien-cies,workload of generating iPSCs,aneuploidy karyotype ratio,complexities of eliminating exogenous transcription factors and spe-cific markers expression were compared between these two systems.Results The STEMCCA reprogramming system required to prepare the feeder cell MEF.The reprogramming efficiency was 0.1%.Transcription gene-free iPSCs cells were obtained by the Cre-loxp enzyme digestion technique at the later stage.Sendai virus reprogramming system was feeder-free and the preparation of matrigel was quite simple with unified standard.The reprogramming efficiency was 0.7%,which was much higher than that of STEMCCA system(P <0.05).The exogenous virus and transgenes could be gradually eliminated after several passages of natural subclone.Conclusion The Sendai virus/matrigle reprogramming system is much more applicable for the induction of iPSCs from dental origin than the STEMCCA system.
3.Off -label use of antibacterial agents in Traffic Hospital of Shandong Province in 2016:a cross sectional study

Objective To determine the off -label use of antibacterial agents,we investigated and analyzed the current status about off -label use of antibacterial agents.Methods The random sampling was conducted to select the outpatient prescription including antibacterial agents from January to April in 2016.According to drug instructions,the off -label drug use of prescription was analyzed.Results 1 264 prescriptions involving 58 kinds of drugs were analyzed.The main categories of off -label drug use were no pediatric and elderly information(23.42%), indication(4.1 5%),dosage (6.31 %),dosage range (62.95%)and administration route (3.1 5%).Conclusion The off -label use of antibacterial agents is common in our hospital.It's in need to regulate off -label drug use.
4.Expression and location of YB-1 protein in hepatocellular carcinoma and their clinical significance

ObjectiveTo study the expression levels of YB-1 protein in hepatocellular carcinoma (HCC) tissues,tissues adjacent to tumour and normal liver tissues,and to explore the clinical significance.MethodsTo study the expression levels of YB-1 protein,immunochemistry was carried out on 58 HCC tissues and their corresponding adjacent tissues to tumour and on 20 normal liver tissues.The YB-1 protein with nuclear translocation was detected by Western blot in HCC tissues,tissues adjacent to tumour,normal liver tissues,HCC cell lines including QGY-7701 and SMMC-7721,and a normal hepatic cell line LO2.ResultsPositive signals of YB-1 protein were detected at a high level in HCC tissues (72.4%,42/58) when compared to tissues adjacent to tumour (41.4%,24/58) and to normal liver tissues (35 %,7/20) (P>0.05).The nuclear expression of YB-1 (31%,18/58) was significantly correlated with the pathologicalgrade,tumour size and portal venous invasion (P<0.05).Using Western blot,the YB-1 protein with nuclear translocation was expressed at a higher level in HCC tissues (0.474±0.107) than in tissues adjacent to tumour and in normal liver tissues (P<0.05).Similar results were obtained in HCC cell lines and the normal hepatic cell line (P<0.05).ConclusionThe high expression of YB-1 in HCC and its nuclear translocation may be involved in human HCC progression.YB-1 may be a potential target for HCC treatment in the future.
5.Application of damage control surgery concept application in 22 cases of pancreas trauma

Objective To investigate the application of damage control surgery (DCS) concept in treatment of pancreas trauma. Methods The clinical data of 22 cases of pancreas trauma from January 2009 to June 2016 were analyzed retrospectively, including degree of injury, therapies and effect. Results Following DCS concept, 3 cases were given conservative treatment, and 19 cases were treated by operation, including debridement, hemostasis, suture, simple drainage and preserved pancreas function;21 cases were cured and 1 died;pancrestic fistula occurred in 11 cases, abdominal infection occurred in 6 cases and injured pancreatitis occurred in 1 cases by conservative treatment;false cyst occurred in 1 cases 6 weeks after operation. All patients were followed up for 12-36 months, with an average of (25.1 ± 1.7) months, and No significant impact was seen on the lives or work of 21 patients after surgery. Conclusions Pancreas trauma needs early-stage diagnosis and active treatment. Rational application of dcs concept can reduce the mortality and improve the outcome effectively.
6.Metabolomic study on clinical staging of methamphetamine detoxification by LC-QTOF-MS

At present , methamphetamine has become a major hidden danger in global public health safety. In order to judge methamphetamine addicts and methamphetamine abstainers more scientifically and reliably, this study analyzed the endogenous metabolites in plasma, serum and urine of methamphetamine addicts, methamphetamine abstainers and healthy volunteers by highly sensitive high-throughput liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) analytical instrument. The obtained metabolomic data were processed by univariate analysis (t-test) and multivariate analysis (PLS-DA and OPLS-DA) and eligible potential biomarkers were then screened.The identified biomarkers set enrichment analysis to find the connection between metabolites and metabolic pathways.Multivariate statistical results showed that methamphetamine acute group, recovery group and healthy group were clearly separated.3, 18 and 6 regulated metabolites were identified in serum, plasma and urine, respectively, suggesting that lipid metabolism was abnormal in methamphetamine acute group, and that fatty acid metabolism, sulfate/sulfite metabolism and sex hormone metabolism were abnormal in methamphetamine recovery group.The selected potential biomarkers in this study provide the possibility for scientific judgment of the clinical stage of methamphetamine detoxification.
7.Association between the consumption of milk tea beverage and insomnia symptoms among college students

Objective:
To investigate the association between consumption of milk tea beverages and other sugary beverages with insomnia symptoms among college students, so as to the provide reference for promoting sleep quality among college students.
Methods:
From September to November 2023, a convenience sampling method was used to select 15 440 college students from seven universities in Shanghai, Jiangxi Province, Hubei Province, and Shanxi Province. A self developed questionnaire and the Insomnia Severity Index were used to assess milk tea beverage consumption, other sugary beverages and insomnia symptoms. Univariate analysis of insomnia symptoms among college students was performed using Chi square tests and Cochran-Armitage trend χ 2 tests. Multivariate Logistic regression analysis was conducted for the association between milk tea and other sugary beverage consumption with insomnia symptoms.
Results:
The detection rates of insomnia symptoms among college students was 40.83%. The frequency of consumption of different milk tea and various sugary drinks (juice, tea, milk, sodas,yogurt, functional drinks, coffee) per week was related to the detection trend rate of insomnia symptoms of college students ( χ 2 trend =38.38, 47.66, 74.16, 32.44, 65.78 , 38.71, 35.94, 91.59, P < 0.01 ). Multivariate analysis indicated that compared to individuals with no milk tea consumption, college students engaging in milk tea consumption had a significantly increased risk of insomnia symptoms ( OR =1.24, P <0.05). Students consuming milk tea at low to moderate frequencies (1-3, 4-5 times per week) showed a higher risk of insomnia symptoms compared to those with no consumption ( OR =1.20, 1.54, P <0.05). Furthermore, higher levels of milk tea consumption were significantly associated with insomnia symptoms (1 501-2 999 mL, OR =1.22; ≥3 000 mL, OR =1.36), and the consumption of other sugary beverages were also associated with insomnia symptoms of college students ( OR =1.10-1.55) ( P <0.05).
Conclusions
There is a relationship between consumption of milk tea beverage other sugary beverages with insomnia symptoms among college students. Colleges should regularly conduct dietary health education seminars to improve sleep quality among college students.
8.Preliminary application of three-dimensional visualization in interventional treatment of Budd-Chiari syndrome

Objective To analyze the application of three-dimensional visualization in interventional treatment of Budd-Chiari syndrome (BCS) with the hepatic vein occlusion type.Methods A retrospective study was conducted on 15 patients with BCS of the hepatic vein occlusion type who were admitted to the Affiliated Hospital of Xuzhou Medical University from January 2017 to June 2018.The study included 8 males and 7 females with a mean age of (36.7±14.4) years.All patients were examined with MRI enhanced scanning and the original MRI data of the DICOM format were extracted.The digital data were then extracted and reconstructed by the Mimics software to obtain a three-dimensional visualization model.The hepatic vein was recanalized under the guidance of the three-dimensional visualization model.Results The three-dimensional visualization model was successfully constructed.Of all the 15 three-dimensional visualization models,right hepatic vein ostial stenosis occurred in 4 patients and occlusion in 11 patients.The middle hepatic vein and the left hepatic vein formed a common trunk in 15 patients,and ostial stenosis and occlusion of the common trunk occurred in 3 and 12 patients,respectively.Communicating branches between the right and middle hepatic veins occurred in 6 patients.Accessory hepatic veins occurred in 3 of 15 patients,and the accessory hepatic vein ostium was stenosed in 3 patients.Communicating branches between the accessory hepatic vein and the right hepatic vein occurred in 2 patients,and communicating branches between the accessory hepatic vein and the middle hepatic vein occurred in 1 patient.All patients were treated successfully with interventional treatment without any complications.One,2,and 3 hepatic veins were recanalized in 7,5 and 3 patients,respectively.Conclusion The three-dimensional visualization was valuable in displaying the location,pattern and collateral vessels of the hepatic veins in BCS patients with the hepatic vein occlusion type.It was helpful for accurate preoperative assessment and to make individualized interventional treatment plans.
