1.Research progress on bifidobacterium-mediated genetic engineering vaccines against parasites

Parasitosis is one of the most serious infectious diseases endangering human and animal health. Vaccination was an economic and effective measure in prevention and treatment of the disease. Bifidobacterium (Bb) is a kind of important probiotics in intestinal tract of human and other animals. In recent years, with the development of genetic engineering technology, Bb has been selected as a carrier delivery system to construct a series of recombinant Bb, such as in bacteria, viruses, parasites, tumor and other fields. In this review, we reported the preparation and immune effect of several kinds of Bb-mediated genetic engineering vaccines against parasites.
2.A study of PET-CT SUV range for dose verification in carbon ion radiation therapy

Objective The positron generated at the dose deposition site by using high-energy carbon ions to hit the material annihilate with the negative electron in the material to release the gamma photon.The positron-emitting isotope (PEI) distributions in the target volume are activated significantly by carbon ions.Therefore, the mean values of positron emission tomography (PET) activity could be related to the delivered doses to the clinical target volume from carbon ion.This specialty can be used for the image registration fusion of the carbon ion treatment planning computed tomography (CT) and treatment verification PET-CT.After radiation in the almost same decay period, the relationship between the different target volume and the PET-CT SUV of different every single fraction dose can be found, then the range of SUV for the radiation target could be decided.So this PET-CT standardized uptake value (SUV) range can also provide a reference for the correlation and consistency in planning target dose verification and evaluation for the clinical trial.Methods The head phantom was used as a simulation of the real human body, the 1 cc, 4 cc, and 10 cc cube volume target contouring were done in the TPS, the 90 degree fixed carbon ion beams were delivered in different single fraction effective dose of 2.5 GyE, 5 GyE, and 8 GyE.After the beam delivery, later the PET-CT scanning was performed and parameters of scanning followed the trial regulation.The MIM Maestro software was used for the image processing and fusion to determine the maximum, minimum, average, and total values of SUV in the virtual clinical target volumes for the different single fraction dose.Results The results showed that for the same target volume, the SUV range of target had an approximate linear correlation with effective dose of target (P=0.000).The same effective dose for the different target volumes got the same SUV range (P>0.05).Conclusions For the carbon ion treatment plan, the SUV range from image registration and fusion of planning CT and PET-CT after treatment can be used to make an evaluation for accuracy of the dose distribution.And this method also could be used in the hyper-fraction treatment plan.In the SUV range research of different decay periods, the similar method can be performed for the exploration.
3.Effect Observation and Preliminery Mechanism Exploration of Shengji Corium Elephatis Mastic in Treating Chronic Refractory Skin Ulcer

Objective To observe the effect of Shengji corium elephatis mastic in treating chronic refractory skin ulcer, and analyze its preliminary mechanism. Methods Totally 62 patients with chronic refractory skin ulcer in granulation stage were randomly divided into two groups, 32 cases of treatment group were treated with Shengji corium elephatis mastic, and 30 cases of control group were treated with Vaseline gauze. All patients were treated for 4 weeks. The rate of wound healing, wound reduction ratio and wound secretion level of VEGF were observed. Results The cure rate and the total effective rate between the two groups had significant difference (P<0.05). After 2 weeks treatment, the mean wound reduction area of treatment group and control group was 82.31%and 66.32%respectively. After 4 weeks treatment, the mean wound reduction area of treatment group and control group was 90.35%and 78.7%respectively, the difference was statistically significant (P<0.05). After 1, 2 and 3 weeks treatment, the treatment group had significant difference with the control group in wound secretion VEGF level (P<0.05). Conclusion Shengji corium elephatis mastic can promote wound healing of chronic refractory skin ulcer. The possible mechanism is that Shengji corium elephatis mastic promotes the generation of VEGF in wound thus promotes wound repair.
4.Clinicopathologic and expression of TMSG-1 and Cyclin D1 in esophageal squamous cell carcinoma

Objective To study TMSG-1 and Cyclin D1 expressions in esophageal squamous cell carcinoma (ESCC) and their relevance to the clinicopathological data and prognosis. Methods Immunohistochemistry S-P method was used to examine the expressions of TMSG-1 and Cyclin D1 in pathological specimens of 136 cases of ESCC and 13 cases of normal esophageal mucosa. Contrast study among immunohistochemistry, clinicopathological data and prognosis was also analyzed. Results (1)The positive expression rates of TMSG-1 and Cyclin D1 in ESCC were 52.2% and 65.4%, respectively. The expressions of TMSG-1 and Cyclin D1 in ESCC were significantly higher than that in normal esophageal mucosa (P<0.05). (2)The expressions of TMSG-1 and Cyclin D1 were all related to clinical stage , differentiation degree and lymph node metastasis (P < 0.05). (3)The expression of TMSG-1 was negatively correlated to the expression of Cyclin D1 in ESCC (r=-0.386,P=0.000). (4)The expression levels of TMSG-1 and Cyclin D1 were independent risk factors in patients with ESCC (P < 0.05). Conclusions The abnormal expressions of TMSG-1 and Cyclin D1 may cooperatively play a role in initiation and development of ESCC. Co-examination of TMSG-1 and Cyclin D1 expressions in primary tumors may be useful for predicting prognosis of ESCC.
5.Investigation on Purification Process of Intermediate Product ofCaulis Sinomenii fromTong-An Injection

This study was aimed to improve the transfer rate of sinomenine in the intermediate product of Caulis Sinomenii in order to optimize the purification process of intermediate product ofCaulis Sinomenii fromTong-An (TA) injection. The transfer rate of sinomenine and the stability of fingerprints in the intermediate product of Caulis Sinomenii were used as indexes for the investigation on the impact from different pH ofCaulis Sinomenii extract before extraction. The transfer rate of sinomenine was used as an index for the investigation on the impact by different pH of hydrochloric acid created to dry extract solution. The transfer rate of sinomenine was used as an index for the investigation of four separation ways, which included the vacuum filtration, plate and frame filters, high-speed tube separator, and flat direct centrifuge, on the liquid separation of sinomenium acutum acid. The results showed that the pH ofCaulis Sinomenii extract before extraction was 10-11; the transfer rate of sinomenine was the highest in the extraction process and the fingerprints of TA injection was stable. The pH of hydrochloric acid was 2.0-2.5; and the highest transfer rate of sinomenine in acid dissolution process was 92.94%. The high-speed tube separator had the best separation to sinomenium acutum acid-dissolving liquid. The highest transfer rate of sinomenine was 93.34%. It was concluded that the optimized process can effectively improve the transfer rate of sinomenine in the intermediate product ofCaulis Sinomenii. Meanwhile, fingerprints of the product were stable. The process was simple with good repeatability.
6.Cytotoxicity and oxidative damage effect of silica nanoparticles on vascular endothelial cells

Objective To investigate the cytotoxicity of silica nanoparticles on vascular endothelial cells, and to clarify its action mechanism.Methods The 60 nm silica nanoparticle was selected and the invitro cultured human umbilical vein endothelial cells (HUVECs)were used as cell model.The HUVECs were divided into control and silica nanoparticle exposure groups with concentrations of 12.5,25.0,and 100.00 mg·L-1 .MTT assay was used for the determination of cell viability,lactate dehydrogenase (LDH)release assay for membrane integrity,flow cytometry (FCM)for intracellular reactive oxygen species (ROS)content,and real-time PCR assay for intracellular NF-E2-related factor 2 (Nrf2 ), heme oxygenase-1 (HO-1 ), superoxide dismutase 2 (SOD2 ) and glutamate-cysteine ligase catalytic subunit (GCLC)mRNA levels.Results The MTT results showed that the cell viabilities in each silica nnaoparticle exposure group were decreased compared with control group in a dose-dependent manner. Upon the silica nanoparticle exposure for 12 h,the cell viability was declined significantly only in 100 mg·L-1 exposure group compared with control group (P<0.05).When exposured for 24 h,the cell viabilities in 25.0, 50.0,and 100.0 mg·L-1 exposure groups were declined significantly compared with control group (P<0.05). Under the exposure to silica nanoparticle with the same dose, the cell viabilities were decreased along with the elongation of exposure time.LDH assay and FCM showed that except for that in 12.5 mg·L-1 exposure group, both the LDH activities in media and intracellular ROS levels in other exposure groups were increased compared with control group (P<0.05 ). The results of real-time fluorescence PCR showed that the mRNA levels of Nrf2, HO-1,SOD2 and GCLC in 100 mg·L-1 silica nanoparticle exposure group were increased significantly compared with control group (P<0.05).Conclusion Silica nanoparticles have toxicity to vascular endothelial cells,which includes reducing cell viability,membrane integrity destruction,induction of ROS generation,and tranSCriptional regulation of redox-related factors. Oxidative damage is one of the mechanisms of vascular endothelial toxicity mediated by silica nanoparticles.
7.Effects of Different Preparation Methods on Content of Ginsenosides Rg1, Re and Rb1 in Yi-Xin-Shu Tablets by HPLC-ELSD

This paper was aimed to study effects of different preparation methods on the content of ginsenosides Rg1,Re and Rb1 in Yi-Xin-Shu (YXS) tablets by HPLC-ELSD.HPLC-ELSD was used as the detection method.The separation and content of ginsenosides Rg1,Re and Rb1 were used as indexes.The influences of three different preparation methods (i.e.,defatted alcohol extraction and butanol extraction,alcohol extraction and butanol extraction,alcohol extraction and butanol extraction ammonia solution washing) on the effect of YXS tablets were studied.Then,the same content determination method was used to compare the influence of alkali washing treatment to ginsenosides Rg1,Re and Rb1 among different batches of Panax ginseng.The results showed that a good separation of ginsenosides Rg1,Re and Rb1 component peak of YXS tablets was achieved by three kinds of separation methods.The separation degree was greater than 1.5.Ammonia solution washing had some effect on ginsenosides Rg1,Re and Rb1 content,which made the content of ginsenosides Rg1,Re and Rb1 be 1.5-1.8 times to those without alkali washing.No effect was shown on the content of ginsenosides Rg1,Re and Rb1 during ammonia solution washing.It was concluded that some other ginsenosides can be transferred into ginsenosides Rg1,Re and Rb1 in YXS tablets solution after ammonia solution washing.
8.Pancreatic serous microcystic neoplasm with atypical clinical and image features

Objective:To summarize the atypical manifestations and treatment strategies of serous microcystic neoplasm of the pancreas.Methods:Review the case data of 11 cases of pancreatic serous microcystic adenoma with atypical preoperative clinical imaging findings admitted to Peking Union Medical College Hospital from July 2008 to October 2019, and summarize their clinical manifestations, CT/MRI features, and preoperative clinical diagnosis, surgical methods, postoperative conditions.Results:The median age of the 11 patients was 50 (46-66) years old, 7 females, and 4 males. There were 8 cases with back pain and 6 cases with weight loss. The preoperative imaging examination found that the lesion was located in the head and neck in 8 cases, with pancreaticobiliary duct dilatation in 7 cases, suspected vascular invasion in 3 cases, and pancreatic parenchymal atrophy in 2 cases, 3 cases showed rich blood supply and solid space, and 2 cases were connected to the pancreatic duct. Preoperative diagnosis of pancreatic malignant tumors accounted for 4 cases, neuroendocrine tumors in 3 cases, solid pseudopapillary tumors in 3 cases, and intraductal papillary mucinous tumors in 1 case. All patients underwent surgical treatment, 3 cases of distal pancreatectomy (2 cases of using Kimura method to protect the spleen), 3 cases of pancreaticoduodenectomy, 1 case of total pancreatectomy, 2 cases of pancreatic head resection with duodenum preservation, 2 cases of local resection. All patients had no early postoperative deaths. There were 2 cases of grade B pancreatic fistula, 2 cases of biochemical leakage, 1 case of postoperative abdominal hemorrhage, and 2 cases of delayed gastric emptying, all of which were cured after active treatment. The median postoperative hospital stay was 18 (7-63) days.Conclusions:Pancreatic serous microcystic neoplasm could be accompanied by atypical features such as pancreatic/bile duct dilation, parenchyma atrophy, or even present vessel invasion. Understanding the atypical clinic and image features would help improving differential diagnosis and treatment. For those with invasive features, surgical exploration should be recommended.
9.Construction and identification of TSOL18 recombinant Lactococcus lactis secretory and non-secretory vaccines of Taenia solium

Objective To construct recombinant non-secretory pMG36e-TSOL18/L.lactis and secretory pMG36e-SP-TSOL18/L.lactis vaccines of Taenia solium. Methods Taking sequence of Taenia solium TSOL18 gene as template, genetic optimization was carried out using lactic acid bacteria as a host system, TSOL18 gene and SP-TSOL18 gene were synthesized using PCR-based accurate synthesis (PAS), through the design of full-length primers, the addition of restriction enzyme cutting sites Sac Ⅰ and Hind Ⅲ and protective bases, and the SPUSP45 secretory signal peptide sequence in the N terminal. TSOL18 gene and SP-TSOL18 gene were cloned into Escherichia coli-L.lactis shuttle expression plasmid pMG36e to construct intracellular expression vector pMG36e-TSOL18 and secreted expression vector pMG36e-SP-TSOL18. The two recombinant plasmids were identified by enzyme digestion and sequencing, and electroporated into L.lactis MG1363 to construct the recombinant pMG36e-TSOL18/L.lactis and pMG36e-SP-TSOL18/L.lactis vaccines of Taenia solium, and they were identified by PCR. Results TSOL18 gene fragment and pMG36e vector fragment were obtained by double digestion with Sac Ⅰ and Hind Ⅲ, which were consistent with the expected results; TSOL18 gene standard sequence was aligned and the matching degree was 100%, and both were inserted into Sac Ⅰ and Hind Ⅲ of pMG36e vector. Our PCR results showed that both recombinant pMG36e-TSOL18/L.lactis and pMG36e-SP-TSOL18/L.lactis were 393 bp gene fragment products. Conclusion The recombinant pMG36e-TSOL18/L.lactis and pMG36e-SP-TSOL18/L.lactis vaccines of Taenia solium are successfully constructed.
10.The application of SilverHawk atherectomy device in the treatment of femoropopliteal occlusive disease

Objective To evaluate the safety and efficacy of SilverHawk directional atherectomy device in the treatment of femoropopliteal occlusive disease. Methods From August 2012 to June 2014,46 patients(58 limbs)with femoropopliteal occlusive diseases in the treatment by SilverHawk directional atherectomy device were analyzed retrospectively . The mean lesion length and degree of diameter stenosisin the femoropopliteal stenoses(52 limbs) were (4.6 ± 2.3) cm and (85.6 ± 11.3)%.The mean lesion length in the femoropopliteal occlusions(6 limbs)was(6.3 ± 3.2)cm. Rutherford score was 3 ~ 5. Mean ABI was 0.45 ± 0.36. Patency was evaluated with color duplex sonography,CTA and DSA postoperatively. Results 46 patients(58 limbs)were recanalizated suc-cessfully via intraluminal approach. The overall technical success rate was 100%. The procedural success rate was 93.10%. Postoperative residual stenosis and ABI were(10.3 ± 6.2)%and 1.05 ± 0.32,which had statistical diff erence compared with preoperative(t=5.83,P=0.02). The average period of follow-up was 22 months. Mean ABI during the follow-up was 0.96 ± 0.15,which had statistical difference compared with preoperative(t = 5.09,P =0.03). The 6-month and 1-and 2-year primary patency rate was 94.83%、91.38%、84.48%,and secondary patency rate was 98.28%、96.55%、93.10%,respectively. Conclusion SilverHawk directional atherectomy device is safe and effective in treament offemoropopliteal occlusive disease ,with satisfactory early-middle results.
