2.The Unscheduled Expression of Cyclin B1/CDK1 Contributes to the Transition of Cell Cycle of Lymphocytes in vivo from G_0 to G_1 Phase

Objective To study the mechanism of unscheduled Cyclin B1 expression at G_1 phase which is usually at G_2/M phase.Methods Human peripheral blood lymphocytes(PBL) from healthy volunteers were firstly activated by PHA and then went into cell cycle.The cells were collected at 0,36,48 and 60 h after activation and divided into two parts:one for Cyclins/DNA muhiparameter assay,and another for Post-sorting Western blot.Results After activation by PHA,Cyclin B1 and CDK1 of lymphocytes were expressed at G_1 phase.Conclusion Unscheduled Cyclin B1/CDKl probably contributes to lymphocytes in vivo into cell cycle.
3.Relationships between the characteristics of the anatomy of the basis nasi and the severity of obstructive sleep apnea hypopnea syndrome

[ABSTRACT]OBJECTIVETo investigate the relationships between the characteristics of the upper airway anatomy, including the basis nasi, and the severity of obstructive sleep apnea hypopnea syndrome(OSAHS). METHODSFifty OSAHS patients and 40 normal subjects received three-dimensional CT scan and poly somnography(PSG). Variables between the two groups were compared. The association between the PSG parameters and the upper airway anatomic features were analyzed. RESULTSThere were significant differences in several CT variables between OSAHS patients and normal subjects(P<0.05), including the minimal lateral airway dimension and the minimal cross-sectional airway area of both velopharynx and glossopharynx, the minimal anterior-posterior airway dimension of velopharynx, and the airway width of basis nasi. The result of logistic regression analysis suggested that the minimal cross-sectional airway area of velopharynx and the airway width of basis nasi were significant predictors of the OSAHS(P<0.05, the values of the odds ratio were 0.978 and 0.589). The correlation analysis suggested that the airway width and the airway area of basis nasi both correlated significantly with several CT variables of pharynx(P<0.05), among these results, the correlation coefficents between these two variables and the mCSA of velopharynx were 0.536 and 0.425 respectively. CONCLUSIONNarrowed basis nasi and velopharynx might be important anatomical features in OSAHS patients. There are correlations between the characteristics of basis nasi and the anatomy of pharyngeal airway.
4.Advances in near infrared photoimmunotherapy of tumor

Near infrared photoimmunotherapy (NIR-PIT) is a highly selective molecularly targeted phototherapy for cancer which is based on injecting a conjugate of IRDye700DX,a water-soluble near-infrared silicon-phthalocyanine dye,and a monoclonal antibody that targets an antigen on the cancer cell surface. Subsequent local irradiation of NIR light causes the rapid and specific tumor cell death. Due to the good clinical translation prospects of NIR-PIT,this paper summarizes the influencing factors,antitumor mechanism,main challenges and recent strategies,which may benefit for its research and clinical application.
5.Risk Factors for Recurrent Colorectal Polyps

The recurrence of colorectal polyps is caused by various factors and leads to the carcinogenesis of colorectal cancer, which ranks third in incidence and fourth in mortality among cancers worldwide. The potential risk factors for colorectal polyp recurrence have been demonstrated in multiple trials. However, an article that pools and summarizes the various results is needed. This review enumerates and analyzes some risk factors in terms of patient characteristics, procedural operations, polyp characteristics, and dietary aspects to propose some effective prophylactic measures. This review aimed to provide a reference for clinical application and guide patients to prevent colorectal polyp recurrence in a more effective manner.
6.Eight-arm polyethylene glycol nanoconjugates for brain targeted delivery of photosensitizer

The aim of this study was to prepare the nanoconjugates for targeted photodynamic therapy of brain cancer by using eight-arm polyethylene glycol(8PEG)as the carrier and cRGD as the targeting ligand, and to investigate the antitumor effect and its mechanism. UV-Vis spectra and confocal microscopy were used for characterization and cellular uptake behavior of nanoconjugates respectively. Alamar Blue assay and Calcein AM/PI staining were applied to investigate the cytotoxocity of nanoconjugates against tumor cells, and tumor spheroid growth curve was used to assess the tumor growth suppression effect. In addition, the generation of reactive oxygen species(ROS), apoptosis and spheroid permeability test was used to reveal the antitumor mechanism of nanoconjugates. The results showed that cRGD-8PEG-IR700 was taken up efficiently by integrin overexpressed U87MG cells, while almost no uptake was found in integrin free NIH/3T3 cells. Remarkable photokilling effect against U87MG cells was only shown in cRGD-8PEG-IR700 group due to the light-induced ROS generation and apoptosis, whereas growth suppression effect was also observed in U87MG spheroids treated with cRGD-8PEG-IR700 plus light owing to the superior penetration ability of targeted nanoconjugates. Hence, tumor-targeted PEG nanoconjugates may provide a promising drug delivery system for photodynamic therapy of cancers.
7.Detection of the Pathogenic Gene Expression in Patients with High Risk Hereditary Cardiovascular Diseases

Objective:To detect pathogenic gene expression in patients with high risk hereditary cardiovascular diseases and explore its predictive value for high risk ventricular arrhythmia .Methods :The expression of pathogenic genes in 30 patients with clinically diagnosed high risk hereditary cardiovascular diseases were detected by using second‐generation high‐throughput sequencing on the basis of gene chip probes associated with high risk hereditary cardiovascular diseases .The patients were divided into two groups by whether the malignant ventricular arrhythmia had happened . And gene expression features of pathogenic genes in the two groups were comparatively analyzed .Results: Among the 30 patients , there were 8 patients (positive rate 26 .67% ) with pathogenic genes .In ten cases with history of high risk ventricular arrhythmia ,the pathogenic gene positive rate was 20% (2/10) .And the pathogenic gene positive rate in control group was 30% (6/20) .The difference between the two groups was not statistically significant .Conclusions :There is a certain positive rate of pathogenic genes in patients with high risk hereditary cardiovascular diseases .However ,further researches are needed to confirm the predictive value of the pathogenic gene expression for high risk ventricular arrhythmia happened in these patients .
8.Evaluation of the clinical characteristics of patients with positional obstructive sleep apnea hypopnea syndrome

Objective To evaluate the clinical characteristics of patients with positional obstructive sleep apnea hypopnea syndrome (P-OSAHS).Methods The study retrospectively analyzed 171 men diagnosed as having OSAHS in Beijing Tongren Hospital,Capital Medical University,during July 2012 to June 2014.That the apnea hypopnea index (AHI) during supine position of ≥ 2 times by the AHI during lateral position was employed as the standard for P-OSAHS.Subjective symptoms,physical examinations,polysomnography (PSG),and three-dimensional computer tomography (3-D CT) were compared between POSAHS group and control group with non-positional OSAHS.Results Of 171 patients with OSAHS,47(27.5%) had P-OSAHS and 124 (72.5%) presented with non-positional OSAHS.There were significant differences between two groups in BMI,neck circumference,AHI,the lowest oxygen saturation,the percentage of time with oxygen saturation below 90% (CT90),and the minimal anterior-posterior airway dimension of the glossopharynx (P < 0.05).The AHI was the only predictive parameter for P-OSAHS (P < 0.01).There were significant differences in the incidence of P-OSAHS between patient with mild or moderate disease (AHI < 30),severe disease (AHI ≥30 but < 60),and extremely severe disease (AHI ≥ 60) (81.8% (18/22) vs45.3% (24/53) vs 5.2% (5/96),P<0.05).Conclusion The severity of OSAHS closely related to the incidence of P-OSAHS,with significant differentee in the incidence of POSAHS between the patients with different severity of OSAHS.
9.Preparation and in vitro evaluation of albumin nanoparticles produced by thermal driven self-assembly

The aim of this study was to prepare albumin nanoparticles by thermal driven self-assembly, and to investigate the formation mechanism, cellular uptake, the kinetics of cellular uptake and intracellular degradation, etc. By measuring the concentrations of thiol, amino and carboxyl groups, the formation mechanism of albumin nanoparticles was revealed. CCK-8 assay was performed to detect the cytotoxicity; inverted fluorescence microscope was used to observe the cellular uptake of the nanoparticles; while the fluorescence resonance energy transfer(FRET)method was applied to investigate the cellular uptake and intracellular degradation kinetics. The drug-loading capacity was investigated using paclitaxel(PTX)as the model drug. The results showed that the albumin nanoparticles produced by thermal driven self-assembly were safe, nontoxic, biodegradable and stabilized by intermolecular disulfide and amide bonds. The drug-loading study indicates that PTX can be highly encapsulated in the nanoparticles. Hence, thermal driven self-assembly method is green and easy to operate, and the albumin nanoparticles can be applied as a new delivery platform for anticancer drugs.
10.Correlation Study between Sphingomyelin and Degree of Coronary Artery Stenosis in Patients with Coronary Atherosclerotic Heart Disease

Objective:To investigate the correlation between the plasma sphingomyelin (SM ) levels and the severity of coro‐nary artery disease in patients with coronary atherosclerotic heart disease(CAD) on admission .Methods:Clinical data of 464 pa‐tients undergoing coronary angiography were retrospectively analyzed ,which included 345 male cases and 119 female cases with mean age of 62 .71 ± 0 .46 .The patients were divided into stenosis group (n=327) and normal group (n=137) according to the results of coronary angiography .The severity of coronary artery stenosis was calculated according to Gensini score .The SM levels of all patients were measured on admission .Results:SM levels in control group and stenosis group were 39 .45 ± 0 .91 mg/dL ,45 .05 ± 0 .78 mg/dL ,respectively ,and the difference between the two groups was statistically significant (P<0 .001) .SM levels were positively correlated with Gensini scores(r=0 .155 ,P=0 .001) .The Gensini scores according to their tertiles of SM levels were 6(0‐38) ,18(2‐50) ,19(0‐49) ,respectively .The differences regarding Gensini scores among the three groups were statistically significant (P=0 .035) .After multiple related risk factors had been corrected ,the multivariate linear regression analysis showed that plasma SM levels were independently associated with Gensini scores (β=0 .143 ,95% CI 0 .148~0 .654 ,P=0 .002) .Conclusions:SM level was significantly associated with coronary angiography Gensini score ,and its level might be an independent risk factor for predicting the degree of coronary stenosis .SM might provide novel therapeutic tar‐get for the treatment of CAD .
