1.Analysis of HCV screening results from 2012 to 2014

Objective To study the infection status of hepatitis C virus (HCV) in blood donors .Methods anti‐HCV was detec‐ted by using enzyme linked immunosorbent assay (ELISA) .HCV RNA was detected by using fluorescence quantitative polymerase chain reaction .Results The positive rate of anti‐HCV in blood donors was 0 .07% (109/163 782) .Among 109 positive donors ,80 donors were anti‐HCV positive while only one reagent was used ,and 29 donors were anti‐HCV positive when two reagents were used .80 cases were anti‐HCV positive in first donors and 29 cases were anti‐HCV positive in repeated donors .Among the 80 donors who were anti‐HCV positive while only one reagent was used ,72 samples according with the demand of nucleic acid test were tested by the nucleic acid test ,of whom HCV RNA was negative .Conclusion The positive rate of anti‐HCV in Yancheng donors could be lower than general population .There might be no change of positive rate of anti‐HCV in blood donors during the three years .The positive individuals could be negative in nucleic acid test while only one reagent was used in ELISA test .
2.Effects of Yunnan Baiyao on the hemolytic phenomena and plasma clotting ability of Staphylococcus aureus

Objective To Study the effect of Yunnan Baiyao on the hemolysis and plasma coagulation ability of Staphylococcus aureus.ToseeknewmechanismofYunnanBaiyao′sapplicationonthebasisofanti‐virulence.Methods 0.5MCFbacterialiquidof Staphylococcus aureus standard strains(ATCC29213) were prepared and cultured in blood agar plates which containing Yunnan Baiyao of different concentrations at 37 ℃ for 24 h .Then the diameter of hemolysis rings were measured and recorded .0 .1 MCF bacteria liquids treated with Yunan Baiyao of different concentrations were prepared ,and 20μL of the bacteria liquid were added in‐to 80 μL plasma ,then cultivated at 37 ℃ for 24 h to observe the phenomenon of solidification .Results There was significant differ‐ence among the effects of Yunan Baiyao of different concentrations on the Staphylococcus aureus hemolytic rings(P<0 .05) ,and the drug concentrations inversely proportional to the diameter of hemolysis .Plasma coagulation effect of bacteria was not obvious differ‐ent after the treatment with different concentrations of Yunnan Baiyao ,and not had any regular pattern(P>0 .05) .Conclusion Yunnan baiyao can inhibit the growth of Staphylococcus aureus and its hemolytic ability but has no significant effect on plasma clot‐ting enzyme .
3.Mechanism of L-Ca2+/calpain signal transduction in verapamil inversing resistance of papillary thyroid carcinoma to doxorubicin

[ ABSTRACT] AIM:To investigate the mechanism of L-type calcium channel ( L-Ca2+)/calpain signal transduc-tion pathway in verapamil inversing resistance of papillary thyroid carcinoma to doxorubicin .METHODS:Human papillary thyroid carcinoma TPC-1 cells were cultured for 2 d.For determining the appropriate concentrations and treatment time of verapamil and doxorubicin , a compatibility test was conducted to detect the cell viability by CCK-8 assay.The cells were divided into control group , doxorubicin group , verapamil group and doxorubicin +verapamil group .The techniques of whole-cell patch-clamp was used to record L-Ca2+currents.The protein expression levels of calpain 1 and LC3 were detec-ted by Western blot .RESULTS: Compared with control group , the density of L-Ca2+current decreased in doxorubicin group and verapamil group (P<0.05).Compared with verapamil group , the density of L-Ca2+current decreased in doxo-rubicin+verapamil group (P<0.01).Compared with control group, the expression of calpain 1 decreased in doxorubicin group and verapamil group (P<0.05).Compared with doxorubicin group , the expression of calpain 1 decreased in doxo-rubicin+verapamil group (P<0.05).Compared with control group , the expression of LC3 increased in doxorubicin group and verapamil group (P<0.05).Compared with doxorubicin group , the expression of LC3 increased in doxorubicin +ver-apamil group ( P<0.01) .CONCLUSION:The drug resistance of TPC-1 cells to doxorubicin may be related to the in-crease in autophagic activity .Verapamil further increases autophagic activity of TPC-1 cells, resulting in autophagic death and inversing the resistance of TPC-1 cells to doxorubicin .The mechanism may be involved in L-Ca2+/calpain 1 signal transduction pathway of autophagy .
4.Expression of HIF-1? in rat's radiation-induced oral mucositis

AIM: To detect the mRNA and protein level of HIF-1 alpha in the tissues of rat's radiated mucosa. METHODS: The left buccal mucosa was irradiated and excised. The right buccal mucosa was excised to serve as own control tissue. The mRNA of HIF-1 alpha was determined by using the semi quantitative RT-PCR. SABC method was employed to immunostain and to elucidate the localization, intensity and distribution of HIF-1 alpha protein.RESULTS: A Sprague-Dawley rat's model of radiation-induced oral mucositis (ROM) was successfully established. The results of RT-PCR indicated that the left buccal mucosa expressed HIF-1 alpha mRNA while the right buccal mucosa did not or seldom expressed it. Immunohistochemical analysis of HIF-1 alpha demonstrated that the left side mucosa expressed HIF-1 alpha protein.CONCLUSION: The mucosa of ROM expresses the mRNA and protein of HIF-1 alpha. The expressions of HIF-1 alpha are correlated with the severity of ROM.

To promote the development of extracellular vesicles of herbal medicine especially the establishment of standardization, led by the National Expert Committee on Research and Application of Chinese Herbal Vesicles, research experts in the field of herbal medicine and extracellular vesicles were invited nationwide with the support of the Expert Committee on Research and Application of Chinese Herbal Vesicles, Professional Committee on Extracellular Vesicle Research and Application, Chinese Society of Research Hospitals and the Guangdong Engineering Research Center of Chinese Herbal Vesicles. Based on the collation of relevant literature, we have adopted the Delphi method, the consensus meeting method combined with the nominal group method to form a discussion draft of "Consensus statement on research and application of Chinese herbal medicine derived extracellular vesicles-like particles (2023)". The first draft was discussed in online and offline meetings on October 12, 14, November 2, 2022 and April and May 2023 on the current status of research, nomenclature, isolation methods, quality standards and research applications of extracellular vesicles of Chinese herbal medicines, and 13 consensus opinions were finally formed. At the Third Academic Conference on Research and Application of Chinese Herbal Vesicles, held on May 26, 2023, Kewei Zhao, convenor of the consensus, presented and read the consensus to the experts of the Expert Committee on Research and Application of Chinese Herbal Vesicles. The consensus highlights the characteristics and advantages of Chinese medicine, inherits the essence, and keeps the righteousness and innovation, aiming to provide a reference for colleagues engaged in research and application of Chinese herbal vesicles at home and abroad, decode the mystery behind Chinese herbal vesicles together, establish a safe, effective and controllable accurate Chinese herbal vesicle prevention and treatment system, and build a bridge for Chinese medicine to the world.
