1.The role of peripheral nerve TNF? content in the development of neuropathic pain produced by chronic constriction injury of peripheral nerve in rats

Objective To investigate the changes in TNF? content of sciatic nerve induced by chronic constriction injury (CCI) of sciatic nerve to determine the role of TNFa in the development of neuropathic pain. Methods Eight-four female health SD rats weighing 250-300g were anesthetized with sodium barbiturate. Unilateral sciatic nerve was exposed and ligated at the middle of thigh. Three ligatures (chromic catgut 4.0) were placed around the sciatic nerve and tied. The distance between the two ligatures was about 1 mm. Sham operation was performed on the contralateral thigh. The sciatic nerve was exposed and mobilized but not ligated. The thermal nociceptive threshold was determined by measuring the withdrawal latency of hindpaw placed on a 58℃ hot plate on days 0.5, 1, 3, 5, 7, 9, 11, 13 and 14 after surgery. Animals were sacrificed on days 0.5, 1, 3, 7, 10 and 14 after surgery. Sciatic nerves were removed from both thighs and frozen at - 80℃ for determination of TNF? content. Sciatic nerve from healthy animals was used as control. The percentage of maximal possible response (% MPR) , was determined for each group (CCI, sham operation, control) % MPR= (new withdrawal latency- average baseline latency)/( 15-average baseline latency) . The distribution of TNF? between supernatant and sediment was also determined. Results The average baseline nociceptive threshold (withdrawal latency) was (7.9?0.2)s. There was significant different in %MPR between the two hindpaws on days 1, 3, 5, 7, 9 and 11 after surgery. The TNF? content of sciatic nerve from healthy rats was (40.62? 0.24) pg/mg protein. The TNF? content of the ligated sciatic nerve was elevated abruptly in 12h after ligation, then abruptly declined to a plateau but was still significantly higher than that of sham-operated side on days 1 and 3. There was no significant difference in TNF? content of sciatic nerve between control group and sham-operation group. The relative content of TNF? content in the sediment of ligated sciatic nerve gradually increased and reached the peak on day 7 and then gradually decreased. Conclusion The TNF? content of peripheral nerves plays an important role in the development of neuropathic pain. Membrane-combined TNF? is involved in the process of nerve repairing.

OBJECTIVE:To screen the formulae of norfloxacin zinc(NF-Zn) suspended granules using uniform design METHODS:The uniform design with 5 levels and 4 factors was employed to screen five formulae of NF-Zn and sedimentation rate,granularity,dissolubility,moisture content and taste of these granules were evaluated and scored The total score of each formula was regressed with the 4 factors by computer RESULTS:The results of regression equation suggested that the optimum formula was NF-Zn 0 2g,CMS-Na 0 25g,2%PVP and manitol 0 55g in each packet CONCLUSION:It is effective and convenient to optimize the formula of NF-Zn granules by uniform design
3.Modified Brisson technique for concealed penis in children

Objective To investigate the corrective effect of modified Brisson technique to treat concealed penis in children.Methods From July 2014 to July 2016,selected 12 patients (3-10 years old,mean 7.2 years old) with concealed penis were surgically treated with the modified Brisson technique in the Second Affiliated Hospital of Nanjing Medical University.Their clinical manifestations included the penis buried below the surface of prepubic skin which can be palpated and visualized through pushing the skin around the penis to the pubis and the short foreskin which was not attached to the penile corpus cavernosum.Patients with other conditions such as obesity and penial agenesis that caused buried penis were excluded from the study.The modified Brisson technique consisted of 4 steps.Ventral incision was used to gradually slit the narrow ring of the prepuce and to expose the glans followed by circumfe-rential inner foreskin incision made below the coronary sulcus of penis then the foreskin stripping released the corpus cavernosum.Finally the skin coverage was accomplished by posthioplasty.The operation time,blood loss,complications,postoperative treatments and followed-ups were retrospectively analyzed.Results For all the 12 patients,the operations were successful,lasting 40 to 60 minutes.The bleeding was about 5 to 10 mL and there was no operation-related complications like bleeding and infection of incision.Satisfactory cosmetic outcomes were obtained without retraction and the scars were similar to those of circumcision.All patients had penile lymphedema postoperatively which disappeared within 6 months after operation.Follow-up ranged from 6 months to 1 year,and there were no complications like skin necrosis and contracture or voiding difficulty.Conclusions The modified Brisson technique for concealed penis repair is technically easy and safe.In addition,it provides a good cosmetic appearance,functional outcomes and excellent postoperative satisfaction grades.
4.Curcumin-induced histone acetylation in malignant hematologic cells.

This study investigated the inhibitory effects of curcumin on proliferation of hematological malignant cells in vitro and the anti-tumor mechanism at histone acetylation/histone deacetylation levels. The effects of curcumin and histone deacetylase inhibitor trichostatin A (TSA) on the growth of Raji cells were tested by MTT assay. The expression of acetylated histone-3 (H(3)) in Raji, HL60 and K562 cells, and peripheral blood mononuclear cells (PBMCs) treated with curcumin or TSA was detected by immunohistochemistry and FACS. The results showed curcumin inhibited proliferation of Raji cells significantly in a time- and dose-dependent fashion, while exhibited low toxicity in PBMCs. Curcumin induced up-regulation of the expression of acetylated H(3) dose-dependently in all malignant cell lines tested. In conclusion, curcumin inhibited proliferation of Raji cells selectively, enhanced the level of acetylated (H(3)) in Raji, HL60, and K562 cells, which acted as a histone deacetylase inhibitor like TSA. Furthermore, up-regulation of H(3) acetylation may play an important role in regulating the proliferation of Raji cells.

5.Research on distribution of gastrointestinal peptide glucagon-1 of positive cells in tree shrews

Objective To observe tree shrews gastrointestinal glucagon like peptide 1 ( glp-1 ) immune response positive cells ( EG cells) distribution and the difference of the digestive tract of rats.Methods Immunohisto chemical SABC method to detect tree shrews diges-tive tract (except for oral cavity, pharynx) glp-1 cells positive content.Results EG cells distributed in tree shrews duodenum,ileum and colon,glandular epithelial cells of jejunum,positive material is distributed in the cytoplasm.Tree shrews stomach,rectum did not see EG cell distribution.Ileal immune dyeing results show positive,EG cells of the digestive tract,only the ileum and colon distribution glp-1 more posi-tive, tree shrews compared with the rats in the digestive tract glp-1 significant difference (P<0.01).Conclusion Gastrointestinal tree shrew in duodenum,ileum and colon,jejunum have glucagon positive cells.The rat only have positive cells in the ileum and colon.According to the mechanism of the glp-1 and the distribution of tree shrews gastrointestinal glp-1,tree shrews atherosclerosis ( atherosclerosis,AS) AS the model is not easy to make may be associated with the role of glp-1.

7.Preliminary experimental study on mechanism of intestinal mucosal damage induced by IgG immune complex in mice

Objective:To study the cellular pathology and molecular mechanisms of intestinal mucosal damage induced by IgG immune complex in mice.And to explore the pathogenic mechanism and molecular diagnosis evidence of ulcerative colitis induced by food intolerance in clinical practice.Methods: Six weeks old BALB/c female mice were used to build animal model.All the mice were divided into four groups:the control group(group A),the rabbit intestinal mucosal protein immunized group(group B),the DSS induced group(group C),the rabbit intestinal mucosal protein immunized combined with DSS induced group(group D).After successful establishment of animal model,serum and colon tissues were collected to be performed relevant tests.Results: IgG level in serum and colonic mucosa of group B mice increased.And inflammatory cell infiltration and a small amount of mast cell activation were observed in intestinal mucosa;group C mice showed the typical acute ulcerative colitis:a large number of inflammatory cells infiltration,inflammatory factor levels increased in mucosa and mucosa lamina propria,and mucosal epithelial cells′ tight junction weakened;group D mice manifested both high level of IgG in serum and colonic mucosa and also typical acute ulcerative colitis.Besides,significant mast cell activation was observed in the intestinal mucosa.Conclusion: We infer from the experimental results that IgG immune complexes can induce the damage of intestinal epithelium by mediating activation of mast cells.And during the process,the level of inflammatory cytokines increased in intestinal mucosa and the expression of tight junction protein in epithelial cell decreased.These factors contribute to the promotion of intestinal mucosa damage induced by immune complexes.
8.Clinicopathologic study and review of alveolar soft part sarcoma

Objective To investigate the clinicopathologic features and differential diagnosis of alveolar soft part sarcoma(ASPS).Methods A case of ASPS was studied by HE stain,light microscopic observation,histochemical stain and immunohistochemical stain,and discussed the disease synthetically with the literature.Results The patient was female,23 years old.The lesion was located in the deep skeletal muscles of left buttocks,microscopically,which consisted of round to polygonal tumor cells,showing alveolus and nest arrangement,arounded by stroma containing sinusoidal capillaries,and there were abundant crystals in cytoplasm of the tumor cells,prominent nucleoli and fewer nuclear fission images in the tumor cells.PAS stain was stongly positive.Immunohistochemical stain showed positive expression of myoglobin in tumor cells,and negative expression of Myosin,Desmin,SMA,Vimentin,S-100 protein,EMA and Syn in them.Conclusion ASPS is a rare malignant soft tissue neoplasm,its histogenesis is uncertain,and diagnosed by pathological examine and treated mainly by operation.
9.Adventitious bud induction and endogenous hormones changing of lamina from Zanthoxylum dissitum in vitro

Objective To establish a regenerative system in vitro and determine the endogenous hormones of Zanthoxylum dissitum. Methods Lamina of Z. dissitum was used as the explant. Callus adventitious bud differentiation was carried out by culturing on MS with different hormones. At the same time,in the course of callus induction,four endogenous hormone of GA3,IAA,ABA,and ZR were determined by ELISA. Results The optimum medium for adventitious bud regeneration of Z. dissitum was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,and the highest induction rate could reach to 61%. During the process of the differentiation of explants callus,the contents of ZR and IAA were correspondingly higher than other endogenous hormones,the content of ABA was always kept in low level during this process,the content of GA3 was kept in the trend of upgrading during the earlier differentiation period. Conclusion It could be considered that ZR and IAA should be the critical factors in the bud induction. According to the results,a proper adding of GA3in the culture medium could improve the differentiation rate of the adventitious bud. On the other side,ABA might be the negative regulation factor.
10.Application of ENCOR vacuum -assisted breast minimally invasive revolving system in the diagnosis and treatment of breast tumors

Objective To study the application effect of ENCOR vacuum-assisted breast minimally invasive polypectomy system in the diagnosis and treatment of breast tumors. Methods Totally 86 breast cancer patients treated in the General Hospital of Yangmei Group from January 2017 to October 2017 were randomly divided into study group and control group,with 43 cases in each group. The study group was treated with ENCOR vacuum-assisted minimally invasive rotational atherectomy system,while the control group was treated by traditional breast excision. The operative conditions, postoperative complications and postoperative recurrence were observed and compared between the two groups. Results The intraoperative bleeding volume,operation time,scar length and postoperative recovery time in the study group were (5.1 ±1.2)mL,(15.9 ±3.5)min,(0.3 ±0.1)cm,(2.7 ±0.4)d,respectively, which in the control group were (12.7 ±2.3)mL,(26.8 ±4.6)min,(3.2 ±1.1)cm and (5.1 ±0.8)d,respectively, the differences between the two groups were statistically significant ( t=15. 579,14. 297,14. 108,13. 597,all P<0. 05). The total incidence rate of postoperative complications in the study group was 11. 63%,which was significantly lower than 30. 23% in the control group (χ2 =5. 187,P<0. 05). During the follow-up of 12 months,in the study group,2 patients (4. 65%) had residual tumor,1 patient (2. 33%) developed recurrence; in the control group, 3 patients (6. 98%) had residual tumor,2 patients (4. 65%) developed recurrence. There was no statistically significant difference between the two groups (χ2 =1. 013,P >0. 05). Conclusion ENCOR vacuum - assisted breast minimally invasive revolving system in the treatment of breast tiny tumors has advantages of less trauma and pain,rapid recovery,etc,it is safe and effective.
