1.Effects of epigallocatechin-3-gallate on 1-methyl-4-phenylpyridinium ion-induced apoptosis in rat PC12 cells

AIM: To investigate the effects of epigallocatechin-3-gallate(EGCG) on 1-methyl-4-phenylpyridinium ion(MPP+)-induced apoptosis in rat pheochromocytoma(PC12) cells and to explore the relationships between its roles of anti-oxidation,intracellular calcium homeostasis and anti-apoptosis.METHODS: Rat PC12 cells were pretreated with vehicle control or EGCG(10,50,and 100 ?mol/L) for 30 min,then cultured with MPP+(900 ?mol/L) for 24 h.The cell viability and apoptosis were monitored by MTT assay and flow cytometry using Annexin V and PI.The activity of intracellular reactive oxygen species(ROS),contents of superoxide dismutase(SOD) and malondialdehyde(MDA),cytoplasmic Ca2+ density and apoptotic morphology of mitochondria were examined by fluorescent plate-based assays,confocal microscope,and transmission electron microscope,respectively.RESULTS: MPP+ impaired the PC12 cells in a concentration-dependent pattern and induced apoptosis of the cells(31% versus control).Compared with the control,the cells pretreated with EGCG showed markedly higher rate of viability and lower apoptosis.Meanwhile,EGCG pretreatment significantly increased the SOD activity and decreased the levels of MDA and ROS.Interestingly,EGCG also decreased the concentration of cytoplasmic Ca2+ and improved the morphology of mitochondria.CONCLUSION: EGCG exhibits inhibitory effects on MPP+-induced apoptosis in rat PC12 cells,which is possibly associated with increasing the cell ability of anti-oxidation and decreasing the concentration of cytoplasmic Ca2+.

Aim To investigate the antagonistic action of EGCG on apoptosis of rat PC12 cell induced by MPP+.Methods PC12 cells were cultured and the apoptosis induced by MPP+(900 ?mol?L-1)was observed.The cells were randomly divided into 6 groups:blank group without any treatment,MPP+ control group,vitamin E group and EGCG groups(10,50,100 ?mol?L-1).After treatment of drugs,cell viability,leakage of LDH,morphological changes of mitochondria and apoptosis were detected by MTT,Hoechst 33342 staining,transmission electron microscope and flow cytometry,respectively.Results After treatment of cultured PC12 cells with MPP+,cell viability was decreased,leakage of LDH and apoptotic rate were increased,and mitochondria swelling,vacuole and cristae breakage were observed.Vitamin E and EGCG en-hanced cell viability,reduced the leakage of LDH and apoptotic rate,and decreased the damage degree of mitochondria.Conclusions EGCG possesses the ability of inhibiting rat PC12 cell apoptosis induced by MPP+,and its protective action may relate to its function of keeping mitochondria integrality.
3.Bone affinity fluorescein indirectly marks early angiogenesis during endochondral ossification

BACKGROUND: There are various methods to observe and detect early angiogenesis in the process of entochondrostosis, but each holds certain deficiencies.OBJECTIVE: To explore the possibility of tetracycline and alizarin complexone as an indirect marker ofangiogenesis in the process of entochondrostosis.METHODS: New Zealand white rabbit models of bilateral radial bone defects were prepared, followed by β-tricalcium phosphate implantation, and then given the injection of tetracycline and alizarin complexone at 1 and 15 days,respectively. Samples were collected at 28 days, some of which were observed using fluorescence/light microscope after ink perfusion and hard tissue slicing, and the others were decalcified and observed using immunohistochemistry. The uniformity between lumen structures labeled with bone affinity fluorescein and vascular structures marked by immunohistochemistry and ink perfusion was compared.RESULTS AND CONCLUSION: The lumen structure labeled with bone affinity fluorescein was confirmed to be a CD34 positive vascular structure. Under the fluorescence microscope, the bone affinity fluorescein labeled vascular morphology was consistent with ink perfusion-labeled, and black ink lines could be observed in the lumen structures labeled with bone affinity fluorescein after ink perfusion. In addition, the color of the lumen labeled with fluorescein was more gorgeous,three-dimensional structure more vivid, and the vascular evolution process distinguished more easily by different fluorescein colors, exhibiting unique advantages. Therefore, it is available to detect the early angiogenesis in the process of entochondrostosis.
4.Dependability search of human CLC-2 gene associated with idiopathic generalized tonic-clonic seizures in Jinuo people and Han people from Yunnan province

Objective To investigate whether the vohage-gated chloride channel CLC-2 gene— CLCN2 is associated with idiopathic generalized tonie-clonic seizures(often called a grand mal seizure, GME)of Jinuo people and Han people from Yunnan province.Methods Three regions,including Intron 2, Exon 5 and Exon 19(Intron 18),of CLCN2 were selected to conduct sequence analysis.The case-control study design was used to detect association between gene polymorphism and idiopathic generalized tonic- clonic seizures of Jinuo people and Han people from Yunnan province.Results No previously reported susceptible mutations were found in Intron 2,Exon 5 and Exon 19 in Jinuo people and Han people from Yunnan province.However we found a single nucleotide polymorphism(SNP)at site 146 of Intron 18. 5.Nosocomial Infections due to Staphylococcus aureus:Clinical Characteristics and Resistance Analysis

OBJECTIVE To investigate the risk factors,clinical characteristics and resistance of Staphylococcus aureus nosocomial infections so as to guide the treatment of S.aureus infection.METHODS To collect clinical materials of S.aureus nosocomial infection and analyze risk factors and clinical characteristics and detect sensitivity of isolated strains to antibacterial agents.RESULTS Severe underlying diseases existed among 73 cases of S.aureus nosocomial infections,82.19 percent of patients had received invasive interventions.Lower respiratory tract was the most common infective site.Seventy-nine strains of S.aureus were isolated,including 66 meticillin-resistant S.aureus(MRSA) and 13 meticillin-sensitive S.aureus(MSSA).S.aureus showed general resistance to many kinds of antibiotic drugs.The resistant rates of MRSA were much higher than those of MSSA(P
6.Study on the association of cytochrome P450 polymorphisms and the risk of esophageal cancer: a meta-analysis

Objective To examine the association between CYP1A1 polymorphisms (MspI and Ile/Val) and esophageal cancer (EC) by systematically reviewing the risk of the original studies. Methods Data from 16 papers (8 for MspI, 14 for Ile/Val) regarding case-control studies on the association of cytochrome P450 polymorphisms and risk of esophageal cancer was analyzed by dominant model (variant genotype vs. wild-type genotype) through meta-analysis. Stratified analysis was carried out according to the pathological types. Results In systematical analysis, CYP1A1 MspI variant genotype (TC+CC) had no association with EC risk (OR=1.17,95%CI: 0.82-1.66). Similar results were observed in esophageal squamous-cell carcinoma(ESCC) (OR=1.17,95%CI: 0.82-1.69) and esophageal adenocarcinoma (EAC) (OR=1.39,95% CI: 0.67-2.09). Individuals with the CYP1A1 Ile/Val variant genotype (Ile/Val + Val/Val) had an increased risk for EC, when comparing with wild type (Iie/Iie ), with an OR of 1.39 (95 %CI: 1.07-1.80). CYP1A1 Ile/Val variant genotype could increase the risk of ESCC (OR=1.43,95%CI:1.07-1.91) but no significant association was found with EAC (OR=1.20,95%CI:0.62-2.30). Conclusion CYP1A1 gene polymorphism Ile/Val might have played a role in the development of ESCC but CYP1A1 MspI polymorphism might not be associated with the susceptibility of EC.
7.Relations between attitude and practice of smoking and the training program regarding tobacco control among community medical staff members in Hangzhou, Zhejiang province

Objective To investigate the relations between training and both the attitude and practice on smoking control among community medical staff members in Hangzhou,Zhejiang province.Methods Three representative districts including Xiacheng,Gongshu and Westlake were chosen from Hangzhou city.Questionnaire survey was applied to collect information from the related community medical staff members.The survey mainly contained three aspects:knowledge,attitude and practice regarding smoking control involved in the community medical activities.Availability and application of the resources on smoking cessation were also studied.Logistic regression analysis was applied to explore the factors associated with the smoking control training programs.Differences of rates between groups were assessed with chi-square statistics.Wilcoxon rank sum test was used to study the relationships among knowledge,attitude and practice related to smoking control programs,targeted to the community medical staff members.Results Eight hundred forty-six community medical workers were involved.Sixty-five percent of the community medical staff members had learned related knowledge on smoking control.Proportion of the community medical staff who had taken lessons on smoking control with 3-10 working years was 1.77 times more than the ones with experience less than two years (OR=1.77,95%CI:1.25-2.51).Eighty-eight point seven percent of the medical staff who had received smoking control training programs were identified with the consciousness that they should advise the patients to quit smoking,comparing to the proportion 81.60% (Z=-2.87,P=0.00) in the control group.In terms of the practice regarding smoking control,data showed that 21.62% of the medical staff who had received smoking control training programs would provide ‘how to quit smoking' to more than 90% of the smoking patients,while the proportion in the control group was 10.65% (Z=-5.68,P=0.00).The use of drugs,traditional Chinese medicine therapy and the smoking cessation hotline rate were all less than 30%.Conclusion The training programs being used on smoking control seemed useful in improving the consciousness and practice towards the smoking control programs during their medical activities among the community medical staff members.
8.Evaluation on the effect of intervention strategies regarding lifestyle-health promotion among community medical staff in Hangzhou city

Objective To investigate the effect of workplace intervention programs related with health-promoting lifestyles among community medical staff members in the communities of Hangzhou city.Methods Three representative districts including Xiacheng,Gongshu and Westlake were chosen and all the community medical staff was involved in the study.Questionnaire survey was applied to collect the information from all the community medical staff.Three aspects including 1,environmental support and policies on healthy lifestyles 2,lectures and activities related to healthpromoting programs provided,and 3,peer effect on health promotion,were studied.Chi square test and logistic stepwise regression analysis were applied.Results Intervention strategies on smoking,diet and physical exercises were analyzed respectively.It was indicated that none of the lectures,regulations or individual counseling regarding smoking cessation was associated with the decrease of smoking behavior.Medical staff that taking part in the lectures on balanced diet showed higher frequencies of vegetable/fruit intake,with the frequencies of vegetables/fruits consumption on 4 or more days/per week appeared 1.74 times more than that in the control group.As for the physical exercise intervention was concerned,the peer effect was found to be obvious.Medical staff that taking exercise with colleagues tended to do more exercises with frequency in taking moderate or high intensity exercises 5 or more days/per week as 1.62 times than that of the control group.Data also indicated that regardless of intervention programs on diet or on physical activities,the combination effect could statistically improve the healthy behavior on diet or physical exercise.Conclusion Intervention programs on both balanced diet and physical exercises were preliminarily found to be associated with enhanced health promotion behavior for workplace health promotion strategies but need to be verified through further intervention studies of the same sort.
9.A Study on Association of Nutrition Training and Nutrition Intervention Consciousness,Intervention Behaviors among Community Medical Staff in Hangzhou City

Objective To know the status of nutrition education of community medical staff and analyze its association with nutrition intervention consciousness and behaviors.Methods Three representative districts including Xiacheng,Gongshu and Westlake were selected and 846 community medical staff was involved in this study.Questionnaire investigation was applied to collect the information from mainly three aspects including training,consciousness and practice.Logistic stepwise regression analysis and Cochran -Mantel -Haenszel test were used.Results Most of the community medical staff was female (78.49%).78.95% of the staff had received the training of nutrition knowledge and its main influencing factor was working time.The Association was observed between the nutrition training and the preparation of nutrition consultation. The more training received,the higher self -assessment would be (P <0.05 ).Meanwhile,as the types of training increased,the awareness and attitude of medical staff was improved remarkably,such as knowing risks of unhealthy dietary,asking the dietary habit of the patients routinely,and advising patients to establish a good dietary habit to their children (all P <0.05 ).Nutrition training also had a positive association with nutrition intervention behavior ,such as implementing more measurement of blood pressure, weight, and blood cholesterol for community outpatients, communicating with them in the importance of balanced diet and proper nutrition,and giving weight loss suggestion to obese patients (all P <0.05).Conclusion Training on nutrition knowledge and intervention skills of community medical staff should be emphasized and reinforced in the future,in order to promote the effect on chronic diseases prevention of community -based intervention strategies.
10.Combined anterior and posterior surgery for treatment of cervical fracture-dislocation in patients with ankylosing spondylitis.

OBJECTIVETo discuss the pathological characteristics of cervical spinal fracture complicating ankylosing spondylitis (AS) and the effect of combined anterior and posterior operation.
METHODSEighteen AS patients with traumatic cervical fracture-dislocation were treated operatively from January 2000 to January 2006. The symptom duration of AS was 14.5 years on average. Three cases had undergone osteotomy in lumbar spine. There were 4 cases of Grade A, 3 cases of Grade B, 9 cases of Grade C and 2 cases of Grade D according to Frankel's score. There were 15 cases of Grade III dislocation and 3 cases of Grade II. All patients underwent surgical procedures by combined anterior and posterior approach.
RESULTSThere were 4 anterior-posterior procedures, 8 anterior-posterior-anterior procedures and 6 posterior-anterior procedures. Seven patients had one stage operation and 11 cases underwent two stage surgeries. There was certain extent of neurological improvement in 14 incomplete paraplegic patients, but no improvement in 4 complete paraplegic patients. The follow-up period was 21.2 months on average and the time for bone fusion was 3.6 months. There were 4 complications during operation and a long-term complication in follow-up.
CONCLUSIONSThe study suggests that anterior combined with posterior approach makes the spine stable and relieves the pressure immediately. It is a reasonable surgical strategy for treatment of cervical spinal fracture-dislocation with AS.
