There was no obvious scar in belly, and all of patients felt no pains after operation. The average inpatient time was shorten, the medical cost decreased. Conclusion:Transvagina endoscopic cholecystectomy is feasible and safety.Comparing to traditional laparoscopic operation,it has obviously superiority.Transvagina endoscopic cholecystectomy should be widely applied in good condition hospitals.
2.Mechanism of the aging phenomenon with passage of human retinal pigment epithelial cell
Chinese Journal of Experimental Ophthalmology 2012;30(1):33-36
BackgroundRetinal pigment epithelial (RPE)cell senescence damage the metabolism of photoreceptor, leading to retinal dysfunction and loss of vision. To understand RPE cell senescence mechanism will contribute to the study of age-related macular degeneration ( AMD).ObjectiveThe present study was to prepare the ageing RPE cell model with passage and explore its potential mechanisms.MethodsThis study was approved by the Ethic Committee of Qingdao University Medical College, and the informed consent was obtained from each gravida.Six human eyeballs were obtained from artificial labor fetusl with the gestational age 16-28 weeks.RPE cells were isolated, cultured and passaged in vitro to establish the cell replicative aging model. The third to twelfth cells were collected to be used to this experiment.Human keratin was used to identify the cells by immunochemistry, and MTT method was utilized to assess the proliferation and viability of different generations of cells as the A490 value. The cellular cycles and transmembrane potential (△ψm)of mitochondrion (△ψm) with passage were detected and compared using Flow Cytometry. Results Cultured and passaged cells showed the hexagon in shape with the melanin in 1-2 generations of cells and presented with the brown staining in cytoplasm for human keratin. The melanin was absent in the third generation cells.Vibrant growth statues were seen from the 3-6 generations cells and thereafter the proliferation ability reduced. The cells of G0/G1 phase were gradually elevated with the passage from 3 - 12 generations with a percentage of 68.40% in the third generation of cells to 87.33% in the twelfth generation of cells, showing a significant difference among various generations ( F =180.43,P =0.00),and that of the sixth, ninth and twelfth generation of cells was significant higher than the third, sixth and ninth generation respectively (t =4.002,P<0.05 ; t=12.885,P<0.01 ;t=16.387,P<0.01 ).MTT assay showed that of RPE cells were significantly declined with the passage ( F =38.77,P =0.00),and the A490 value of the ninth, twelfth generations of cells was considerably lower in comparison with sixth and ninth generation respectively ( t =5.991,11.983,P<0.01 ).From 3 through 12 generations of cells, the staining intensity of rhodamine 123 was gradually decreased ( F =121.68,P =0.00 ),and the staining intensity in the sixth, ninth and twelfth generation of cells was significant lower than that the third, sixth and ninth generation respectively(t=6.918,7.620,11.207,P<0.01 ).Conclusions A replicative aging model can be successfully created by the passage in vitro using human fetal RPE cells. The reduce of transmembrane potential and damage of mitochondria might be one of mechanisms of senescence of RPE cells.

Chinese Journal of Primary Medicine and Pharmacy 2011;18(6):727-728
Objective To evaluate the midterm efficacy of prosthetic disc nucleus ( PDN) replacement for the treatment of lumbar disc herniation. Methods Twenty cases of lumbar disc herniation( including one case of recurrent lumbar disc herniation) were treated with PDN. The twenty cases were followed-up 5.4 ~6.2 years( mean 5.7 years). Functional, and radiographic follow-up examinations,MRI and follow-up records of all patients were reviewed carefully. Results Clinical evaluation at the end of follow-up,there were 10 cases in excellent,6 cases in good,2 cases in fair and 2 cases in poor, and good rate was 80%. One patient accepted the revision operation to remove the PDN because of device migration. One patient accepted the fusion because of adjacent segment disease. The others experienced pain relief, and resumed their normal life and work. The average Oswestry score and VAS get better significantly. Conclusion The PDN was effective in treating patients with lumbar disc herniation. However, the medium term complications of device subsidence should be taken seriously.

AIM:To investigate the effects of ?v?6 integrin-mediated cell adhesion on 5-fluorouracil (5-FU) induced apoptosis in colon carcinoma cell lines.METHODS: The expression of the ?v?6 integrin in colon carcinoma cell lines HT-29 and WiDr cells was analyzed by flow cytometry. The apoptosis induced by 5-FU and the effects of ?v?6 integrin-mediated cell adhesion on 5-FU induced cell apoptosis were measured by enzyme-linked immunosorbent assay (ELISA) method and acridine orange-ethidium bromide (AO-EB) double fluorescent dye staining.RESULTS: Both the colon carcinoma cell lines HT-29 and WiDr cells expressed the ?v?6 integrin. The percentages of HT-29 and WiDr cells expression were 80.82% and 82.96%. 5-FU induced the apoptosis of colon carcinoma cell lines HT-29 and WiDr. The result of ELISA method displayed that enrichment factor (EF) of HT-29 and WiDr cells planted on fibronectin (FN)-ligand of ?v?6 integrin was lower significantly than the EF of HT-29 and WiDr cells planted on non-integrin ligand polylisin (1.11?0.04 vs 3.68?0.03, 1.09?0.02 vs 3.72?0.02, P
7.Analysis of monitoring data of drinking-water borne endemic fluorosis in Xinzhou of Shanxi province in 2010
Chinese Journal of Endemiology 2012;31(3):321-324
ObjectiveTo master the epidemic situation of drinking-water borne endemic fluorosis in Xinzhou, evaluate the effects of water improvement project to reduce fluoride, and to provide a timely scientific basis for monitoring the disease and for establishment of preventive countermeasures.MethodsAccording to the water fluoride concentration and type of the disease, after stratification 25 endemic villages were selected as survey points.Five water samples were collected in water unimproved monitoring villages according to water well locations of east, west, south, north and center. In monitoring villages with improved water,3 tap water and 1 source water samples were collected, respectively. The fluorine content in water samples was determined according to the “Standard Testing Methods for Drinking Water”(GB/T 5750-2006).All children aged 8 to 12 of the survey villages were examined for dental fluorosis by Dean method.Clinical osteofluorosis of all the resident over the age of 16 was examined,8 villages of these counties were selected,10 men and.women were randomly selected in each village, respectively, and they were examined again by X-ray using “Diagnostic Criteria of Endemic Skeletal Fluorosis” (WS/T 192-2007).Children aged 8 to 12 in 13 villages of chosen villages were selected for urine collection,6 unne samples were collected in each age group and a total of 30 samples were collected, and urinary fluoride was determined by F-ion selective electrode method(WS/T 89-2006).ResultsA total of 56 water samples were tested in water-unimproved villages, the average water fluorine was 2.7 mg/L And 52 water samples were tested in waterimproved villages, the average water fluorine was 1.6 mg/L,water fluoride content > 1.5 mg/L accounted for 46.15% (6/13) in the water-improved village.Incidences of dental fluorosis of children in the water-unimproved villages and water-improved villages were 74.75% (622/832) and 23.67% (241/1018),respectively.The difference was statistically significant(x2 =140,P < 0.01).The prevalence rate of skeletal fluorosis was 30.02%(2803/9335) in water-unimproved villages, and 9.44% (1230/13 022) in water-improved villages. The difference was statistically significant(x2 =1557.75,P < 0.01 ).The mean of urine fluorine in a total sample was 3.31 mg/L,the urinary fluoride concentration of children in water-improved villages was significantly different from that of water-unimproved villages(t =2.27,P < 0.05).ConclusionsBy reducing fluorine and improving drinking water, disease in drinkingwater borne endemic fluorosis areas in Xinzhou has been controlled to a certain degree. However, the disease is still very serious in some endemic areas.Fluoride in drinking water re-increases in some water-improved villages, the disease is also in a rise. So,the work of monitoring and prevention should be strengthened.

Objective To investigate CT features of subconjunctival fat prolapse and choristoma. Methods Thirty-eight patients, including 18 with subconjunctival fat prolapse and 20 with choristoma, were retrospectively analyzed. Transverse plain CT scan was performed in all the patients, and then coronal plane images was reconstructed at CT workstation. Results CT features of subconjunctival fat prolapse revealed a crescent-shaped or horn-shaped fat density mass continuous with the orbital fat in the superior temporal quadrant of the orbit. CT features of choristoma revealed a crescent-shaped or triangle fat density mass incontinuous with the orbital fat in the temporal side. Conclusion Subconjunctival fat prolapse typically occurs in bilateral superior temporal quadrant of the orbit in elderly males. Subconjunctival choristoma occurs in the unilateral temporal side in adolescent. CT is helpful to distinguish subconjunctival fat prolapse and choristoma.
