2.SGI-1776, an imidazo pyridazine compound, inhibits the proliferation of ovarian cancer cells by inactivating Pim-1
Journal of Central South University(Medical Sciences) 2014;(7):649-657
Objective: To investigate the antitumor effect of SGI-1776 on human ovarian cancer HO-8910 cells and its molecular mechanism. Methods: HO-8910 cells were cultured in vitro, and the proliferation inhibitory effects of SGI-1776 were determined by MTT assay and colony formation assay. The effect of SGI-1776 on the distribution of cell cycle phase was observed by flow cytometry with propidium iodide (PI) staining. hTe inhibition rate of migration and invasion were valued by transwell cell assay. Multiple molecular techniques, such as ELISA, Western blot, siRNA and cDNA transfection were used to explore the molecular mechanism. Results: SGI-1776 presented dramatic anti-tumor activity against HO-8910 cells in vitro, inhibited the cells proliferation and colony formation, and attenuated the migration and invasion in a dose-dependent manner, accompanied by cell cycle arrest in G1 phase. SGI-1776 caused the proliferation inhibition with concomitant decrease in Pim-1 kinase activity, down-regulated the expression of Pim-1 protein and and its downstream genes, such as CDK6, pCDK6, CDK4, pCDK4, CDK2 and pCDK2, and increased the expression of P21 and P27. Down-regulation expression of Pim-1 by siRNA followed SGI-1776 treatment resulted in enhanced cell proliferation inhibition rate and attenuated migration/invasion. Up-regulation of Pim-1 by cDNA transfection attenuated SGI-1776-induced cell proliferation inhibition and its migration/invasion. Conclusion: Pim-1 mediates the biological effect of SGI-1776 in human ovarian cancer HO-8910 cells, suggesting Pim-1 might be a novel target for human ovarian cancer.

The Quality-Evaluation Index System ( QEIS ) is constructed which can be used to evaluate clinical teaching of operating room.
5.Advance in study of monitoring acute rejection of pancreas allograft transplant
Chinese Journal of Clinical Pharmacology and Therapeutics 2000;0(02):-
The syndrome of insulin-dependent diabetes mellitus(IDDM)includes not only abnor-mal glucose metabolism but also specific microvascular complications that include nephropathy, retinopathy and neuropathy. In the short term, improvement in the quality of life and possible prevention of further morbidity associated with diabetes makes pancreas trans-plantation an important therapeutic option, particularly when combined with a kidney trans-plant,in appropriately selected diabetic patients. The focus of the early management of the pancreas transplant recipient is the prevention of acute rejection. In this article we review the methods for clinical diagnosis of the early acute rejection due to pancreas transplanta-tion.
6.Effect of hyperoxia liquid preconditioning on malondialdehyde and super oxide dismutase during valve replacement with cardiopulmonary bypass
Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(01):-
Objective To observe the cardioprotective effects of hyperoxia liquid by comparing the changes of blood concentrations of malondialdehyde(MDA)and super oxide dismutase(SOD)in mechanical heart valve replacement. Methods Thirty patients required mechanical heart valve replacement were randomly divided into hyperoxia liquid group(Group HL) and control group 15 cases in each. From the start of skin incision(T_(0)) to(10min) after CPB,patients received 10mL/kg Ringer's injection(control group) and 10mL/kg hyperoxia liquid(Group HL) seperately. The central venous blood samples, which were collected in both groups respectively at skin incision(T_(0)),1h after start of CPB(T_1),2h(T_2) and 24h(T_3) after aorta off-clamping,were used to test the blood concentrations of SOD and MDA.Also a series of clinical data were recorded, such as the rate of heart automatic rebeat after aorta off-clamping,ventricular arrhythmia, usage of dopamine in operation and postoperation.(Results)In both groups, the blood concentrations of MDA and SOD were all within normal range before operation.MDA level at T_(2) was lower in Group HL than that in control(P
7.Suppressing Function of Breast Cancer Metastasis Suppressor 1 in Breast Cancer Metastasis
Chinese Journal of Bases and Clinics in General Surgery 2003;0(03):-
Objective To review the recent studies on the suppressing function of breast cancer metastasis suppressor 1 (BRMS1) in breast cancer metastasis. Methods The recent literatures on the mechanisms of BRMS1 in the breast cancer that were published in and abroad were reviewed and summarized. Results BRMS1, similar to the other anti-metastasis genes, only suppresses the metastasis of breast cancer cells but has nothing to do with the growth of tumor. BRMS1 could suppress metastasis of tumor cells by reestablishing both the homospecific and the heterospecific gap junctional intercellular comminications (GJIC) and by altering the expressions of relevant metastasis genes in the breast cancer. Conclusion Further studies on BRMS1 may be helpful to understand the metastasis of breast cancer, which may provide a new way for the diagnosis and treatment of breast cancer.
8.EFFECTS OF OPEN SURGERY AND ENDOVASCULAR GRAFT EXLUSION FOR ABDOMINAL AORTIC ANEURYSMS ON LIVER AND RENAL FUNCTIONS
Medical Journal of Chinese People's Liberation Army 2001;0(09):-
To compare the effects of open surgery (OS) and endovascular graft exclusion (EVGE) for abdominal aortic aneurysms on the liver and renal functions, the serum total protein, albumin, globulin and the ratio of A/G of 57 patients with alodominal aortis aneurysm were analyzed before and after the two operations. The results showed that the serum total protein, albumin, globalin and the ratio of A/G were signifcantly decreased in the open surgery group, while remarkabe decrease was found in globulin but no significant change in the ratio of A/G in the EVGE group.Creatinine and urea introgen were significantly decreased in the open surgery graup and remained normal in the EVGE group. It is suggested that the effect of EVGE on the liver and renal function is much less than traditional(open) surgery.
