2. Detection of bladder cancer by accessing DNA methylation state in urine sediments

Objectives: To assess DNA methylation state of the 13 genes in urine sediments and evaluate their value in diagnosis of bladder cancer. Methods: Ninety-two patients with bladder cancer, 23 patients with non tumorous urinary disease, 6 patients with neurological disease, and 7 healthy volunteers were recruited in the study. The DNA methylation state of tumor-related promoters in urine sediments was determined by Methylation specific PCR method (MSP). Results: The frequency of hypermethylation of 13 genes in 92 cases with bladder cancer was higher than that in 23 cases of bladder diseases, while all the genes were at demethylation state in the 7 healthy volunteers and 6 patients with neurological disease. The incidence of hypermethylation was significantly higher in bladder cancer compared with non tumorous urinary disease (P < 0.05). If DNA hypermethylation was used as a positive indicator, 88.0% (81/92cases) of bladder cancer could be detected. Conclusion: Assessing DNA methylation state in the promoter of tumor-related genes in urine sediments by MSP method is effective for detection of bladder cancer.
3.Novel therapy for Hodgkin lymphoma: reports in the 55th ASH annual meeting

The 55th ASH annual meeting published a special report about Hodgkin lymphoma,which mainly contains management of early-stage Hodgkin lymphoma,novel targeted therapy and the optimal treatment of lymphocyte-predominant Hodgkin lymphoma.The therapy of Hodgkin lymphoma should be individualized,in order to improve both PFS and OS.More clinical trials should be designed on the target therapy and the treatment strategy of nodular lymphocyte-predominant Hodgkin lymphoma.
5.A case of subglottic osteoma.

A 41 years female patient was hospitalized with hoarseness for three months, without cough, expectoration and dyspnea. The symptom had no relief by application of antibiotics. Electronic laryngoscope showed that there were the grey neoplasms under subglottic anteriou commissure with broad base and slightly rough surface, and bilateral vocal cords were normal; throat CT showed subglottic irregular high density and calcification. The subglottal tumor resection was performed under the general anesthesia and laryngoscope CO2 laser assisted. In the operation, we found that the tumor was hard with surface of thin mucosa tissue, and there was white bone tissue under it. The removing tumor was about 0. 8 cm X 0. 6 cm X 0. 3 cm and was diagnosed as subglottic osteoma by pathologic examine. There was no recurrence in three months follow-up.

9.Expression of autophagy related proteins in neonatal rats after early infection induced celebral matter damage

Objective:To explore the effect of infection on autophagy-related proteins,Beclin-1 and LC3,expression in cerebral white matter in newborn rats.Methods: A total of 64 two-day-old Sprague-Dawley(SD) rats were randomly divided into control and experimental groups(n=32 each).At day 2 to 6 after birth,the rats in experimental group were intraperitoneally injected with 0.6 mg/kg of lipopolysaccharide(LPS) once a day to establish a white matter injury induced by infection in neonatal rats while the rats in control group were injected with equal amounts of normal saline.Rats were sacrificed to collect brain tissues at 12 hour,1,3,5 d after model establishment.HE staining was performed to observe the pathological changes.Changes in the expression of Beclin-1 and LC3 in rat white matter were examined by Western blot and RT-PCR.Results: Growth and development of rat in experimental group was slow,cerebral white matter lesions were obvious.Compared with the control group,the experimental group Beclin1 and LC3 protein and mRNA levels in the model after 12 h began to express,1 d reached the peak,and then decreased,each time points were higher than the control group(P<0.05).Conclusion: Early infection in neonatal rats can cause white matter damage;the expression of autophagy-related proteins,Beclin1 and LC3,showed that autophagy may be involved in the pathological process of white matter damage induced by infection.
10.Correlation of Intratumoral Susceptibility Signal Intensity and Apparent Diffusion Coefifcient with Pathological Grading of Brain Glioma

Purpose To analyze the relevance between magnetic susceptibility weighted imaging intratumoral susceptibility signal intensity (ITSS) and diffusion weighted imaging apparent diffusion coefficient (ADC) with different grade gliomas, and to explore the value of ITSS and ADC in the preoperative grading of gliomas. Materials and Methods Fifty-two cases of glioma patients underwent magnetic susceptibility weighted imaging and diffusion weighted imaging scans, all subjects were divided into low-grade glioma group (18 cases) and high-grade glioma group (34 cases) according to brain tumor grading standard, correlation between ITSS grade and ADC values with tumor grade was analyzed, and ROC curve was used to evaluate the diagnostic efficacy of ITSS and ADC values in the grading of brain glioma. Results Difference of ITSS grading was statistically significant (χ2=27.458, P<0.01) among different grade gliomas;difference of ADC values between different grade gliomas was also statistically significant (U=689.000, P<0.01); there was a significant positive correlation (r=0.727, P<0.001) between ITSS grading and pathological grading, ADC values were significantly negatively correlated (r=-0.622, P<0.001) with tumor grading, and ITSS grading was significantly negatively correlated (r=-0.617, P<0.001) with ADC values among different grade brain gliomas. With pathological grading as reference standard, the diagnostic sensitivity, specificity, accuracy, positive predictive value and negative predictive value of ITSS for preoperative glioma grading was 85.29%, 77.78%, 82.69%, 87.88%and 73.68%, respectively, while the diagnostic sensitivity, specificity, accuracy, positive predictive value and negative predictive value of ADC values for preoperative glioma grading was 97.06%, 88.89%, 94.23%, 94.29%and 94.12%, respectively. Conclusion There is a significant correlation between ITSS grading, ADC values and pathological grading of gliomas, the combination use of ITSS and ADC values can significantly improve the preoperative grading accuracy of brain glioma.
