1.Serum Immunopharmacological Assessment of Effects of Rubia Yunnanensis on Murine Immune Cell Stimulated with Streptococcal Antigens

Objective Serum pharmacological assessment:the effect of Rubia yunnanensis extract on the immune function stimulated by superantigens of streptococcus. Methods Using cell culture technique, mouse spleen lymphocytes and peritoneal macrophages were culture with superantigens of streptococcus and serum of Rubia yunnanensis. Lymphocytes proliferation stimulated by superantigens of streptococcus measured by MTT assay. NO level in mouse peritoneal macrophages affected by Rubia yunnanensis at different concentrations were determined and compared. Results Serum of Rubia yunnanensis significantly inhibit the proliferation of lymphocytes stimulated by superantigens of streptococcus, compared with normal control. NO content of macrophages culture in Rubia yunnanensis extract group were lower than those of control. In these tests, the effect of the extract by alcohol is more obvious than that of extract by water. Conclusions Rubia yunnanensis extract has effect in inhibiting immune function and anti-infect activity, it could be applied in clinical practice for immunoregulation and treatment of psoriasis.
2.Individual dose monitoring and registry for radiation workers in China

Individual dose monitoring and registry for radiation workers plays a significant role in ionizing radiation protection and occupational health management. Since the nationwide implementation of monitoring from the 1980s, individual dose monitoring for radiation workers in China has been going on nearly 30 years, which is of significance in radiation protection optimization, and occupational health and protection for radiation workers. In order to improve individual dose monitoring and registry, to prevent and control occupational radiation diseases, and to help build "Healthy China" program, this paper presents the evolution of individual dose monitoring and registry in China, discusses the key points and difficulties faced, and proposes suggestions.
3.Mitochondrial combined oxidative phosphorylation deficiency type 1 in childrena case report and literature review

Objective To summarize the phenotypic and genotypic characteristics of mitochondrial combined oxidative phosphorylation deficiency type 1 (COXPD1), and to improve the clinicians' awareness of this mitochondrial encephalomyopathy. Methods The clinical characteristics, physical examination, laboratory examination and other data of a child with COXPD1 were analyzed retrospectively. The diagnosis was confirmed by clinical whole exon sequencing and high-precision mitochondrial genome full-length PLUS gene detection, and the phenotype and genotype were analyzed by reviewing relevant literature. Results A one-year and five-month-old boy mainly presented with hyperlactacidemia and abnormal liver function. Clinical whole exon sequencing showed that the child had homozygous variation of c. 688G>A(p.G230S) in the GFM1 gene. Sanger sequencing verified that the variation was respectively inherited from the parents of the child (both were heterozygous) with the autosomal recessive inheritance pattern. The high-precision mitochondrial genome full-length PLUS detection also did not find pathogenic mutations related to clinical phenotypes. The child was diagnosed with COXPD1. After "cocktail" therapy and liver protection therapy, the patient's condition improved. Conclusions The phenotype of COXPD1 is complicated and variable, mainly liver type and brain type. The mutation of GFM1 gene affects mitochondrial translation system function, and early gene detection is helpful for definite diagnosis.
4.Comparison between Lysimachiae Herba and Desmodii Styracifolii Herba in pharmacological activities.

Lysimachiae Herba and Desmodii Styracifolii Herba are common traditional Chinese medicines for treating lithiasis. Both of them have efficacies of clearing heat, diuresis and eliminating calculi. However, there are some differences in their clinic applications. The former is mainly used to treat hepatolithiasis, gallstones, jaundice, stranguria and gout; Whereas the latter is mainly used to treat urinary calculus. In this paper, the pharmacological effects of Lysimachiae Herba and Desmodii Styracifolii Herba on removing calculus, choleresis, anti-inflammation and oxidation resistance were compared and analyzed based on document retrieval. In conclusion, both of them show the preventive and therapeutic effects on kidney stones and gallstones. Particularly, Desmodii Styracifolii Herba has a better effect in treating the kidney stones, while Lysimachiae Herba has a better effect in treating cholesterol gallstones.

5.Surgical treatment of Tibia plateau fracture:a clinical analysis of 32 cases

Objective To investigate the clinical effects of surgery treatment of the Tibia plateau fracture.Methods Thirty-two cases diagnosed Tibial plateau fracture were performed internal fixation and bone grafting surgery treatment.Results According to Rasmussen Evaluation,there were the gifted 22 cases(68.8%)excellent,6 cases(18.8%)good,2 cases(6.3%)ordinary,2 cases(6.3%)poor.The good rate Was 87.6%.Conclusions Surgery is effective to trot Tibial plateau fracture.A stable internal fixation,as far as possible,reduces the blood supply to fracture block interference and protects joint subsidiary structure to correct joint function rehabilitation treatment.
6.Cartilage tissue engineering of TGF-β3 combined with IGF-1-induced bone marrow mesenchymal stem cells derived chondrocytes

Objective To investigate the effect of IGF-1 on the TGF-β3 induced chondrogenesis of MSCs encapsulated in alginate beads and its application in cartilage tissue engineering.Methods MSC chondrogenesis in alginate beads was induced by TGF-β3 and/or IGF-1.Collagen type Ⅱ,aggrecan and Sox-9 expression was evaluated by immunostaining,RT-PCR and Western blot,respectively.Scanning electron microscope and laser confocal microscope were used to observe the differentiated chondrocytes when cultured on the chitosan-based scaffold.Results TGF-β3 with IGF-1 induced MSCs in alginate beads to express the higher level of collagen type Ⅱ,aggrecan and Sox-9 than any other growth factor alone (P<0.05).The correlation coefficient between Sox9 and collagen type Ⅱ or aggrecan was 0.95 and 0.91,respectively.The chitosan-based scaffold supported the cell's adhesion,migration and proliferation.Conlusion IGF-1 enhances the TGF-β3-induced MSC chondrogenesis via upregulating Sox9 expression.The chitosan-based scaffold is biocompatible with the differentiated chondrocytes.
7.Study on the genetic susceptibility of HLA-DRB1,-DQB1 alleles in colorectal neoplasm of hubei Han Chinese

Objective To investigate the genetic susceptibility of HLA DRB1, DQB1 alleles to colorectal neoplasm in Hubei Han Chinese. Methods 54 patients with colorectal neoplasm and 136 healthy controls were examined for HLA DRB1 and HLA DQB1 genotypes, which were typed by using polymerase chain reaction/sequence specific primer (PCR/SSP) technique and allele sequence analysis.SAS(6.12 for win) was used for statistics. Results Compared with the control, the allele frequency (AF) of HLA DRB1*0901 (0.2315 vs 0.1397, P =0.033) and DRB1*080X(0.009 3 vs 0.080 9, P =7.11?10 -3 ) was significantly increased or decreased, respectively, in colorectal neoplasm of Hubei Han Chinese. And there was not significantly different between the patients with the control in the AF of HLA DQB1 alleles. Conclusion The results indicate that HLA DRB1*0901 allele is the positive association to , but DRB1*080X is the negative association to the patients of colorectal neoplasm among Hubei Han Chinese. The nucleotide sequences of these two alleles were identified by allele sequence analysis, approach to the corresponded allele sequence(exon 2)of genbank.There is not association HLA DQB1 alleles with the patients.
8.Study on association of HLA-DRB1 alleles with gastric carcinoma and Helicobacter pylori infection

Objective To investigate the association of HLA DRB1 alleles with gastric carcinoma and H. pylori infection. Methods HLA DRB1 alleles polymorphism in 63 patients with gastric adenocarcinoma and 136 normal controls were tested with the polymerase chain reaction/sequence specific primer(PCR/SSP) technique. The status of H. pylori infection was determined by histology of gastric biopsy specimens and /or by ELISA. Results Both HLA DRB1 0901 and HLA DRB1 12 were positively associated, and HLA DRB1 15 was negatively associated with gastric adenocarcinoma, but no significant associations were found between patients with and without above HLA DRB1 alleles with regard to patients' age at presentation, sex ratio, tumor site (distal vs proximal), TNM staging, histological grading of tumor or status of H. pylori infection. Conclusions Our results suggest that HLA DRB1 0901, DRB1 12 and DRB1 15 are associated with gastric adenocarcinoma, but are not associated with the clinical features of gastric adenocarcinoma and the status of H. pylori infection.
9.Effect of myopic defocus on static visual acuity and dynamic visual acuity

Background Researches have suggested that the defocus can induce the change of static visual acuity,but whether it produce influence on dynamic visual acuity is not clear.Objective This study was to investigate the impact of myopic defocus on static visual acuity and dynamic visual acuity and explore the essential difierence between static visual acuity and dynamic visual acuity. Methods Forty volunteers were enrolled in this trial.including 20 adults with the age of 27.4±1.64 years and 20 children with the age of 11.70+1.49 years.All the eyes of subjects received regular examined to excluded the eye disease with the best corrected vision of ≥1.0 D,astigmatism of ≤0.75 D and anisometropia <1.50 D.+1.00 D,+1.50 D,+2.00 D,+2.50 D slasses were ware respectively for the defocus on the foundation of full correction.Dynamic visual acuity was inspected by using selfmade DVA-I training software.and static visual acuity wag tested by static visual acuity chart (Precision Vision,CAT.NO.2125).This clinical trial complied with the Helsinki Declaration and obtained the approval of Ethic Committee of Wenzhou Medical College.Written informed consent was received from each individual prior to the protocol. Results The dynamic and static visual acuities were gradually decreased with the elevation of defocus (F=506.907,P=0.000).No significant differences were found between static visual acuity and dynamic visual acuity in adult or children at various defocus(P>0.05).The regression linear analysis showed that a positive correlation between static visual acuity with defoeus(R2=0.819,t=26.72,P=0.000) or dynamic visual acuity with defoeus(R2=0.826,t=27.42,P=0.000).The slope and intercept between defocus with static visual acuity were steeper than that between defocus and dynamic visual acuity (slope:F=34.18,P=0.000;intercept:F=1005.56,P=0.001). Conclusion The effect of defocus on static visual acuity is different from that of dynamic visual acuity.It could be speculated that dynamic visual acuity is related not only to refractive systems but also other factor.
10.Sequential changes of HIF-1αprotein and mRNA in hypoxic bovine retinal microvessel endothelial cells

● AIM: To investigate the sequential changes of HIF-1 α Protein and mRNA in hypoxic bovine retinal microvessel endothelial cells.● METHODS: The bovine retinal microvessel endothelial cells were cultured in normoxic and CoCl2-induced hypoxic conditions respectively. Expressions of HIF-1 α Protein were measured with immunohistochemical staining, and RT-PCR was used to determine the HIF-1 α mRNA.● RESULTS: HIF-1 α began to increase 1h after hypoxia,and reached the peak at 4h. After 16h, it declined significantly. Compared with the normoxic group, the expression of HIF-1 α protein in the hypoxic groups had significant difference (P<0.01), and HIF-1 α mRNA expression was unchanged under hypoxia.● CONCLUSION: HIF-1 α participates in the hypoxic procedures in retinal microvessel endothelial cells, and hypoxia induce time-dependent changes of HIF-1 α protein expression, which is not modulated on the transcription level. Analysis of HIF-1 α expression revealed a temporal and spatial changes with regard to the hyperoxic repression, indicating that HIF-1 may play a major role in the development of retinopathy of prematurity and other ischemic retinal disorders such as diabetic retinopathy.
