1.Discussion on the value of ultrasound gastrointestinal filling angiography in the diagnosis of gastric cancer TNM staging

Objective:To investigate the value of ultrasound gastrointestinal filling angiography in the diagnosis of gastric cancer TNM staging.Methods:A total of 73 patients with pathological diagnosis of gastric cancer who had underwent surgery from June 2014 to March 2016 in our hospital were selected. Taking postoperative pathology results as the gold standard, we observe the diagnostic value of preoperative ultrasonography in TNM staging to compare N staging of EUS angiography and pathological staging of gastric cancer.Results: The overall accuracy rate of EUS filling angiography in diagnosing T staging of gastric cancer was 72.6%, in diagnosing N staging of gastric cancer was 72.6% 84.9%, and in diagnosing M staging of gastric cancer was 100%. Ultrasound T gastric cancer staging, N staging and pathological staging were statistically significant different. Conclusion: Ultrasound gastrointestinal filling angiography has a high diagnostic value in the diagnosis of TNM staging of gastric cancer, in particular for N and M staging, which can be used for preliminary clinical screening.
2.Small interfering RNA targeting Rac1 gene inhibiting rat retinal neovascularization

Objective To evaluate the inhibited effects of small interfering RNA targeting Rac1(Rac1-siRNA)on rat retinal neovascularization in retinae.Methods Retinal vein occlusion was indueedby retinal photodynamic medthod in 25 Sprague-Dawley rats.Rae1-siRNA vector DNA was injected intothe vitrous of one eye of those rats(gene intervention group),and empty vector DNA was injected into thefellow eye(blank control group).Rae1-siRNA vector was injected in other 25 SD rats without retinal veinocclusion(blank intervention group).Two weeks after injection,fluorescein isothiocyanate(FITC)-dextran was perfused into the hearts of all the rats,and the retinal wholemount was made to observe theneovascularization.The numbers of endothelial cells which break through the internal limiting membranewere counted after hematoxylin-eosin staining.Resuits A massive of neovascularization and FITCleakage were found in blank control group.Small part of neovaseularization and a little FITC leakage wereobserved in the gene intervention group.Retinal vessels were normal in blank intervention group.Compared with blank contrast group and blank intervention group.the difference of the mean numbers ofendothelial cells which broke through the internal limiting membrane in the gene intervention group wassignificant(F=47.168,P=0.000).Conclusion Racl-siRNA can inhibit retinal neovascularizationinduced by retinal vein occlusion in rats.
3.Clinical application of carbon nanoparticles suspension in outpatient thyroid microcarcinoma surgery

Objective Recently a lot of studies have shown carbon nanoparticles can protect parathyroid, however, there is no application in outpatient thyroid operation. The article was to investigate the influence of carbon nanoparticles suspension ( CNS) in?jection on the operation and prognosis of outpatient thyroid microcarcinoma(TMC) surgery. Methods Retrospective analysis was conducted on the clinical data of 65 patients who underwent outpatient TMC surgery from January to June in 2015 in our hospital. The patients were divided into 2 groups:33 patients in trial group were injected with CNS, and the other 32 patients in control group under?went conventional surgery. Comparison was made in operation information, pathologic data, postoperative complications, postoperative hospital stay as well as C?reactive protein(CRP) and blood calcium of the first day after operation between the two groups. Results The average number of VI region lymph nodes in trial group(8.21±2.08)was significantly higher than that of control group(P=0.004). Removal of 5 parathyroid glands for mistake was detected in control group, but no removal of parathyroid gland was detected in trial group, showing significant difference (P=0.028). 3 patients in control group had numbness in skin, while none in trial group. The hospital stay of trial group was lowered significantly in comparison to that of control group([31.09±6.62] vs [35.75±7.37], P=0.009). The average C?reactive protein ( CRP ) of the first day after operation in trial group was much lower that of control group([7.17±5.20] vs [9.87±5.60] mg/L, P=0.048). The average blood calcium of the first day in trial group was much higher than that of control group ([2.07±0.17] vs [1.98±0.20] mmol/L, P=0.036). Conclusion Outpatient TMC surgery injected with carbon nanoparticles sus?pension can protect parathyroid gland function, reduce surgical stress and accelerate recovery.
4.Progress on diagnosis and therapy of differentiated thyroid cancer under guidance of precision medicine

The incidence of thyroid cancer is increasing recently, and differentiated thyroid cancer accounts for more than 90% of all thyroid malignancies.Although the degree of malignancy of thyroid cancer is lower and prognosis is better, but there is pos-sibility to relapse and metastasis.The concept of precision medicine has paid more and more attention by many surgeons.This paper reviews how to get perfect diagnosis and therapy of differentiated thyroid cancer to reduce risk of relapse and metastasis by application of precision medicine.
5.Adsorption Property of Supported Ionic Liquids for Flavonoids

The effects of adsorption time, solid-liquid ratio, sample concentration, flow rate on the adsorption properties of supported ionic liquids(the N-methylimidazolium functionalized silica, SilprMin) for flavonoids were investigated. The results indicated that the adsorption equilibrium for tested compounds, genistein, luteolin and quercetin, was achieved within 30 min, the adsorption efficiencies of these three compounds were improved with the increase of solid-liquid ratio, and decreased with the increase of their concentrations. Moreover, the adsorption isotherm data of the three tested compounds were in good concordance with the Langmuir model. The saturated adsorption capacity of SilprMim for genistein, luteolin and quercetin was 47.7, 52.5 and 63.2 mg/g, respectively. Adsorption efficiency of SilprMim could reach more than 90% at a sample flow rate of 0.5~1.5 mL/min. Using methanol as eluent, the saturated desorption efficiencies of genistein, luteolin and quercetin were 86.1%, 83.3% and 84.6%, respectively. The elution order of the three tested flavonoids was genistein, luteolin and quercetin. SilprMim had strong adsorption and separation capacity for three tested flavonoids, which was hopeful to be applied in separation and purification of naturally occurring flavonoids.
6.The clinical effects of valsartan on the insulin resistance of type 2 diabetes patients with Ⅰ hypertension

Objective To investigate the effects of valsartan on insulin resistance of type 2 diabetes in patients with Ⅰ hypertension.Methods Newly diagnosed 152 patients of type 2 diabetes with Ⅰ hypertension were randomly divided into treatment group and control group.The treatment group and control group undergone lifestyle changes( diet and adequate exercise) for two weeks.After that,the control group were given Diltiazem,while the treatment group were given valsartan,an angiotensin receptor blocker(ARB) drugs.Both groups were treated for 8 weeks.Fasting plasma glucose (FPG) and fasting plasma insulin (FINS) before and after treatment in both groups were determined.Homeostasis model assessment evaluation of insulin resistance index(HOMA-IR) and insulin sensitivity index (IAI) were carried out and compared between the two groups.Results After 8 weeks of treatment,HOMA-IR in the treatment group was significantly decreased compared with the control group ( P < 0.01 ),while IAI was significantly increased( P < 0.01 ).Conclusion Application of valsartan in type 2 diabetes mellitus with Ⅰ hypertension can significantly improve the level of insulin resistance.
7.Effect of Polymorphisms of the Methylenetetrahydrofolate Reductase Gene (C677T,A1298C and G1793A)on Stillbirth

Objective To determine the relationship between Methylenetetrahydrofolate reductase (MTHFR)(C677T, A1298C and G1793A)polymorphisms and stillbirth.Methods 33 unexplained stillbirth and 582 normal livebirth were col-lected for case-control study from August 2014 to August 2016.The genotyping of C677T,A1298C and G1793A polymor-phisms of the MTHFR gene was tested by using restriction fragment length polymorphism polymerase chain reaction.The relationship between MTHFR polymorphisms and stillbirth susceltibility was analyzed by logistic regression.Results There was no significant difference in the genetype frequencies of A1298C and G1793A of MTHFR polymorphisms between case and control.But there was significant difference in the genetype frequencies of C677T between case and control (CT:P=0.02,χ2=3.67;TT:P=0.02,χ2=3.65).Heterozygous CT and homozygous TT would be high risk factors of stillbirth. C677T was found in one (14.3%)of the cases with early stillbirth and in three (11.5%)of the cases with late stillbirth, having no significant difference.Twin pregnancy and MTHFR C677T increased the risk 8-fold (P<0.001,χ2=13.28)and 3.4(P=0.02,χ2=3.65)for stillbirth,respectively.Conclusion MTHFR C677T polymorphism is high risk factors associ-ated with the susceptibility of stillbirth.MTHFR C677T may be used as a prognostic marker of pregnancy cases predisposed to preterm delivery.
8.Transcriptional factor c-Jun regulates Mepe gene expression in osteoblasts

[ ABSTRACT] AIM:To study the relationship between transcriptional factor c-Jun and Mepe gene expression, and to identify the specific binding site of c-Jun on the promoter of Mepe.METHODS:The expression of c-Jun and Mepe in mouse bone tissue was detected by immunolocalization assay.The mRNA expression of Mepe was determined by real-time PCR when the expression of c-Jun was changed.The techniques of dual luciferase analysis and site-specific mutagenesis were used to measure the effects of c-Jun on the transcriptional activity of Mepe.RESULTS:c-Jun was detected in the nu-cleus of osteocytes, while Mepe was observed in osteocyte cytoplasm.The results of real-time PCR showed that overexpres-sion of c-Jun directly resulted in significantly higher up-regulation of Mepe mRNA.Compared with control group, the tran-scriptional activity of Mepe promoter was increased in osteoblasts which was transfected with pCMV-3Tag-1-c-Jun.Mutation of c-Jun potential binding sites decreased the transcriptional activity of Mepe promoter.CONCLUSION:Mepe gene tran-scription can be up-regulated by c-Jun which binds to the specific sites of Mepe promoter in osteoblasts.
9.Evaluation of left ventricular systolic function with tissue Doppler imaging in patients with type 2 diabetes

Objective To evaluate the clinical application of the tissue Doppler imaging (TDI) in investigating the cardiac longitudinal systolic function in the patients with type 2 diabetes mellitus. Methods Forty-two patients with type 2 diabetes mellitus and 25 volunteers underwent TDI. The long axis myocardial peak systolic velocity (Sm), displacement (Dm) and strain rates (SRs) of 18 segments in different left ventricular walls were measured.Results The Dm and SRs of left ventricular wall in diabetic group dramatically decreased (P<0.01). Meanwhile, the diabetic combined hypertension group had much lower Sm of ventricular middle segments than the control group (P<0.05).Conclusion The stain rate and tissue tracking are sensitive and reliable to evaluate the early local cardiac systolic function in the patients with diabetes mellitus. TDI can be used as an noninvasive method to detect early systolic dysfunction.
10.Detection and Identification of Volatile Constituents from Leaves and Fruits of Piper nigrum by HS-SPME-GC-MS

OBJECTIVE:To establish a method for the detection of volatile constituents from the leaves and fruits of Piper ni-grum. METHODS:HS-SPME-GC-MS was used. The chromatographic conditions:column was HP-5 MS quartz elastic capillaries, carrier gas was high purity helium(99.999%),flow rate was 1.0 mL/min,the inlet temperature was 250 ℃,initial temperature of column was 50 ℃(temperature programmed),split injection with split ratio of 10:1. MS conditions:ionization mode was electron impact ion source,ionization energy was 80 eV,ion source temperature was 230 ℃,quadrupole temperature was 150 ℃,trans-mission line temperature was 280 ℃,electron multiplier voltage was 1588 V,mass scanning range was m/z 30-400. The spectra were retrieved using RTLPEST3. L and NIST08. L,and the relative contents of the volatile constituents were determined by area normalization method. RESULTS:There were 28 volatile constituents in the leaves and 15 in the fruits,respectively accounting for 67.13% and 36.85%. The major volatile constituents of leaves were β-caryophyllene (15.72%),limonene (9.39%),3-carene (9.32%),β-pinene(6.80%),α-terpine(4.98%),etc.,the main volatile constituents of fruits were 1,7,7-trimethyl-2-vinylbicyclo [2.2.1]hept-2-ene(10.45%),espatulenol(8.28%),caryophyllene oxide(4.81%),etc. 5 constituents were owned in both. CON-CLUSIONS:The study basically clears the main volatile constituents from the leaves and fruits of P. nigrum,and verifies existing obvious differences.
