Chinese Journal of Analytical Chemistry 2009;37(12):1810-1814
The effects of adsorption time, solid-liquid ratio, sample concentration, flow rate on the adsorption properties of supported ionic liquids(the N-methylimidazolium functionalized silica, SilprMin) for flavonoids were investigated. The results indicated that the adsorption equilibrium for tested compounds, genistein, luteolin and quercetin, was achieved within 30 min, the adsorption efficiencies of these three compounds were improved with the increase of solid-liquid ratio, and decreased with the increase of their concentrations. Moreover, the adsorption isotherm data of the three tested compounds were in good concordance with the Langmuir model. The saturated adsorption capacity of SilprMim for genistein, luteolin and quercetin was 47.7, 52.5 and 63.2 mg/g, respectively. Adsorption efficiency of SilprMim could reach more than 90% at a sample flow rate of 0.5~1.5 mL/min. Using methanol as eluent, the saturated desorption efficiencies of genistein, luteolin and quercetin were 86.1%, 83.3% and 84.6%, respectively. The elution order of the three tested flavonoids was genistein, luteolin and quercetin. SilprMim had strong adsorption and separation capacity for three tested flavonoids, which was hopeful to be applied in separation and purification of naturally occurring flavonoids.

Journal of Medical Research 2009;38(8):55-57
Objective To study the clinical application and its impact factors of tidal flow - volume measurement in infants with bronchiolitis and pneumonia. Methods Thirty infants with bronchiolitis and thirty infants with pneumonia were studied. Tidal breathing flow- volume loops were obtained after the patients were sedated. The tidal volume per kilogram(VT/kg) ,the fraction of tidal volume ex-heled at peak tidal expiratory flow (PTEF) to total tidal volume (VPTEF/VE), and the fraction of exhaled time at PTEF to total expirato-ry time (TPTEF/TE) were measured. Results VT/kg was not significant different between the bronehiolitis group and the pneumonia group. VPTEF/VE and TPTEF/TE were lower in the bronehiolitis group than those in the pneumonia group. Conclusion The measure-ment of tidal breathing flow - volume loops is a good technology, which would be helpful to evaluate airway obstruction in wheezing in-fants. The impact factors should be controlled during the measurement. Otherwise, the results would be affected.

Space Medicine & Medical Engineering 2006;0(01):-
Objective To set up a simple method to measure the SR Ca2+ release function in skeletal muscle strips. Methods The time from peak tension to 75 % relaxation(TR75) during intermittent tetanic contraction(fatigue contraction) of isolated musculus soleus strips was analyzed. It was prolonged rapidly, and then shortened slowly. The ratio of maximal TR75 to initial TR75(R-TR75) indicated a balance between SR Ca2+ release and uptake. If the SR Ca2+-ATPase(SERCA) activity and inhibition extent of SERCA in fatigue contraction were identical,R-TR75 should be an index for SR Ca2+ release function. Results Higher stimulation voltage or 5 mmol/L caffeine perfusion which increased the SR Ca2+ release channel open probability induced an increase of R-TR75 from 2.5 in control to 3.0.On the contrary, inhibition of SR Ca2+ release with 5 mmol/L magnesium sulfate perfusion significantly decreased R-TR75.Inhibition of SR Ca2+ release by metabolites during fatigue contraction was recovered slowly. So R-TR75 at the second fatigued contraction was reduced after 5 or 10 min of recovery, but no significant difference after 60 min of recovery.During intermittent tetanic contraction R-TR75 was increased to be 2.9 times of normal in unloaded soleus. Conclusion The above results suggest that R-TR75 during muscle strip fatigue contraction without any change of SERCA activity can indirectly reflected SR Ca2+ release function. In 2-week unloaded soleus, the enhanced R-TR75 indicates an increase in SR Ca2+ release function.

Objective To explore the changes of the amount and function of Vα24~+Vβ11~+,CD161~+Vα24~+NKT cells in the peripheral blood of systemic lupus erythematosus (SLE) patients. Methods The amount of Vα24~+Vβ~+,CD16~+Va24~+NKT cells in the peripheral blood of 30 SLE cases and 30 healthy persons were detected by flow cytometry. Results Vα24~+Vβ11~+,CD161~+Vα24~+NKT cells in the peripheral blood of SLE patients were significantly lower than that of normal control (P < 0.05). Vα24~+Vβll~+ and CD161~+Va24~+NKT cells in the pe-ripheral blood of SLE patients positively correlated with the level of the complement (C3,C4) and negatively correlated with SLE disease ac-tivity index (SLEDIA) and ESR. Conclusion Activated NKT cells decreased in the peripheral blood of SLE patients. Vα24~+Vβ11~+ and CD161~+Vα24~+NKT cells might be involved in the pathogenesis and development of SLE.

Objective To investigate the potential of AFP-targeted ultrasmall superparamagnetic particles of iron oxide (USPIO) molecular probe in specific detection of hepatocellular carcinoma (HCC) with MRI.Methods The targeted probe was synthesized by conjugating AFP antibody with modified USPIO.Two groups treated with AFP-USPIO and USPIO were set up in the study. The HepG2 cells were incubated with AFP-USPIO or USPIO (100 μg/ml) respectively with the dosage of 50 μ1,100 μl or 150 μl for 4 hours, followed by MR imaging in vitro. The signal-noise ratio (SNR) of the cells on T2-weighted image (T2WI) was measured. The rat models with orthotopic HCC were divided into two groups with 5 rats for each group at random.Pre-and post-contrast enhanced (after 1 hour) MR imaging were performed with caudal vein injection at a dosage of 20μg/ml.The contrast noise ratio (CNR) on T2WI and the difference of CNR between pre-and post-enhancement or between both groups were calculated. The relationship of SNR or CNR with the iron particles in cells or tumors was confirmed by Prussian blue iron staining. Results Cytology experiment showed the SNR in both groups was decreased with the increase of the dosage of AFP-USPIO or USPIO,indicating statistically significantdifference in SNR among three different doseage groups (P<0.05).Prussian blue iron staining showed that the iron particles in cells were increased with the increase of AFP-USPIO dosage, and was negatively correlated with SNR (P=0.00,r=-0.926).However,the iron particles were less in cells in USPIO group. The CNRs of liver tumors in Wistar rat of pre-and post-AFP-USPIO injection were 2.05±0.88 and 0.96±0.31 respectively, indicating a significant difference (P=0.028,t=3.380).However,the CNRs in USPIO group,2.25±1.50 and 2.57±1.49,showed no statistical difference (P=0.275,t=1.263).The CNR after enhancement also had a statistical difference between both groups(P=0.042,t=3.487).Pathological results confirmed more iron particles in tumor tissues in AFP-USPIO group, whereas less in USPIO group. Conclusion AFP-USPIO molecular probes can initiatively target to the HepG2 cells and the liver cancer of rat models expressing AFP,which may help to achieve the specificity of MR imaging in the diagnosis of HCC.

China Oncology 1998;0(01):-
Background and purpose:Few reports demonstrate the relation between the expression of RIZ1 which has been found to be a tumor suppressor gene recently and leukemia. This study investigated the effect of RIZ1 expression on the apoptosis of human myeloid leukemic cell lines. Methods:The expression of RIZ1 mRNA was observed in human myeloid leukemic cell lines AML193, KG-1, KG-1a, K562 and Ery-1 by reverse transcription polymerase chain reaction assay. RIZ1 was forced to express in the expression-lacking cell lines by transfecting pRIZ1 RH containing full length of RIZ1 cDNA to the cell lines. As controls, the cell lines were transfected with pcDNA3.1. The apoptotic cells were determined by using the annexin V/propidium iodide stain 24 h after transfection. Results:Among the 5 cell lines, no expression of RIZ1 mRNA had been detected in AML193 and low expression in K562. The forced expression could be found in both cell lines 24 h after transfection of pRIZ1 RH, accompanied by obviously increased apoptotic rates which were (22.7? 0.7)% in AML193 and (28.6?1.2)% in K562 compared to controls (11.7%?1.6% and 9.0%?0.8%, respectively) (P
