1.Levels of urine neutrophil gelatinase-associated lipocalin and kidney injury molecule 1 and its relevant influencing factors in young patients with hyperuricemia

Objective To investigate urine levels of neutrophil gelatinase-associated lipocalin (NGAL) and kidney injury molecule 1(KIM-1), and its relevant influencing factors in young patients with hyperuricemia.Methods Sixty young subjects with hyperuricemia were randomly selected from physical check-up examination population (the hyperuricemic group), and 60 young subjects with normouricemia were randomly selected at the same time from physical check populationexamination (the control group).Urine NGAL and KIM-1, and other biochemical indicators were evaluated.Clinical data, iab tests, testing items and biochemical indicators between the two groups were analyzed by using t test or analysis of variance test.Regression analysis between the above indicators and urinary NGAL/creatinine (cr) and urine KIM-1/cr ratios were carried by using single regression analysis.Analyzed relevant Relevant influencing factors of urinary NGAL/cr and urine KIM-1/cr ratios were analyzed by using multiple linear regression.Results Urine NGAL/cr and urine KIM-1/cr ratios in the hyperuricemic group were higher than the control group (t=22.152 5, t=24.725 5, P<0.01).Single regression analysis showed that serum uric acid level and urine NGAL/cr ratio, KIM-1/cr ratio was positively correlations correlated (r=0.63, P<0.01;r=0.41, P<0.01).Multiple linear regression analysises showed that serum uric acid,systolic blood pressure, HbA1c, serum creatinin and cholesterol level were the independent factors of urine NGAI/cr ratio.Age, blood uric acid level, systolic blood pressure, HbA1c and serum creatinin levels were independent factors of KIM-1/cr ratio.Conclusion There are high levels in of urine NGAL/cr and KIM-1/cr ratios in population with hyperuricemia.Hyperuricemia, high systolic pressure, diabetes mellitus, declined kidney function and hypercholesterolemia are independent factors of urine high NGAL/creatinin ratio.Oldage,hyperuricemia, high systolic pressure, diabetes mellitus and declined kidney function are independent factors of high urine KIM-1/cr ratio.
2.The influence of silencing Ezrin expression by RNA interference on the biological behaviors of breast cancer cell line MCF-7

Objective To study the role of membrane cytoskeleton linker Ezrin in the growth and metastasis of human breast carcinoma cell line MCF-7.Methods Small interfering RNA(siRNA)targeting Ezrin were transfected into MCF-7 cells.RT-PCR and Western blot were used to detect the down-regulation of Ezrin mRNA and protein.MTT method was used to detect the proliferation of the cells.Flow cytometry was used to detect the cell cycle.Transwell assay was used to detect the migration ability of the transfected MCF-7 cells.Results RT-PCR and Western blot results revealed that Ezrin siRNA down-regulated Ezrin expression at both mRNA and protein levels.Down regulation of Ezrin expression decreased the proliferation rate of the cell line(t=4.479,P<0.01).The cell proportion in G2-M phase (t=-5.997,P<0.01)and the invasion ability(t=5.268,P<0.01)decreased significantly.Conclusion Ezrin plays an important role in the process of proliferation,migration,and invasion of MCF-7 cells.
3.Axillary lymph node dissection for breast cancer patients with negative sentinel node biopsy:a sys-temic review

Objective To systematically evaluate the effectiveness and safety of axillary lymph node dissection for breast cancer patients with negative sentinel node biopsy.Methods Literatures in CNKI,PubMed,EMBASE and CBMwere searched from their establishment to December 1,2013.Accord-ing to the inclusion and exclusion criteria,trials of axillary lymph node dissection and sentinel node biopsy for breast cancer were strictly screened and extracted for quality assessment and result analysis.Meta-anal-ysis was conducted by using Revman 5.1 software.Results A total of 10 studies involving 7731 patients were eligible for the final analysis.Because of the large differences in research type,measurement indica-tor,follow-up period and statistical index,subgroup analysis was adopted.Meta-analysis was applied for homogeneous researches and the remaining studies were analyzed with qualitative descriptive analysis.The results showed no significant differences in disease-free survival,overall survival,local recurrence rate,and distant metastasis rate in different follow-up periods.Conclusion For breast cancer patients with single invasive lesion,axillary lymph node dissection is not necessary.More high-quality random control trials and long-term follow-up are required to confirm the conclusions of this systematic review.
4.Transcriptional factor c-Jun regulates Mepe gene expression in osteoblasts

[ ABSTRACT] AIM:To study the relationship between transcriptional factor c-Jun and Mepe gene expression, and to identify the specific binding site of c-Jun on the promoter of Mepe.METHODS:The expression of c-Jun and Mepe in mouse bone tissue was detected by immunolocalization assay.The mRNA expression of Mepe was determined by real-time PCR when the expression of c-Jun was changed.The techniques of dual luciferase analysis and site-specific mutagenesis were used to measure the effects of c-Jun on the transcriptional activity of Mepe.RESULTS:c-Jun was detected in the nu-cleus of osteocytes, while Mepe was observed in osteocyte cytoplasm.The results of real-time PCR showed that overexpres-sion of c-Jun directly resulted in significantly higher up-regulation of Mepe mRNA.Compared with control group, the tran-scriptional activity of Mepe promoter was increased in osteoblasts which was transfected with pCMV-3Tag-1-c-Jun.Mutation of c-Jun potential binding sites decreased the transcriptional activity of Mepe promoter.CONCLUSION:Mepe gene tran-scription can be up-regulated by c-Jun which binds to the specific sites of Mepe promoter in osteoblasts.
5.Constitution of lymphoid neoplasm in Xinjiang:analysis of 645 cases

Purpose To study the pathologic types and subtypes distribution of 645 cases of lymphoma in Xinjiang. Methods Clini-cal data of lymphoma from April 2008 to march 2013 in the people’s hospital of Xinjiang Uygur autonomous region in Xinjiang were col-lected and reviewed, including morphological, immunohistochemical and clinical characteristics. According to the WHO classification all the cases were reappraised. Results Of the 645 cases, 558 cases (86.51%) were non-Hodgkin’s lymphoma (NHL) and 82 ca-ses (12.71%) were Hodgkin’s lymphoma (HL). Among NHL cases, 448 cases (80.29%) were B-cell neoplasms and 110 cases (19.71%) were T/NK-cell neoplasms. The commonest subtype was diffuse large B-cell lymphoma (258 cases, comprising 40% of all lymphomas) and extranodal NK/T-cell lymphoma (41 cases, comprising 6.36% of all lymphomas) in B cell lymphoma and T/NK cell lymphoma. Burkitt’s lymphoma and lymphoblastic leukemia/lymphoma were predominanly Uyghurs, but mantle cell lymphoma, follicu-lar lymphoma and extranodal marginal zone lymphoma of mucosa associated lymphoid tissue were predominanly Hans. The most com-mon subtypes of Hodgkin’s lymphoma are mixed cellularity, nodular sclerosis and lymphocyte-rich classical Hodgkin’s lymphoma. Sub-types distribution of Hodgkin’s lymphoma has a certain difference in the different ethnic groups, the age of onset did not present twin peaks and the highest proportion was children. Conclusion The lymphoid neoplasms of Xinjiang displayed some ethnic features simi-lar to those reported in literature as well as other regions of China, whereas the distribution of some subtypes showed some differences.
6.Ethical Thinking on the Assisted Reproductive Technology Treatment in Senility Infertile Women in China

Young women affected by education and career delay the childbearing period,which results in reduced fertility.With the two-child policy completely opening,more and more elderly women have demands for pregnancy and the need of assisted reproductive technology (ART) increases day by day.However,the ART treatment faces with a series of medical and ethical problems.For example,the pregnancy rate of aged women is low but the abortion rate is high;and the incidence of pregnancy complications and fetal chromosomal abnormality increases significantly.In this regard,it can solve these problems through limiting the women's age,choosing the donated eggs,strengthening the counseling for elderly infertile women before ART treatment,paying attention to the psychological problems and applying the genetic screening technology before implantation.
7.Exploration of the oral health education experimental teaching for oral health education reform.

OBJECTIVEThis study aimed to improve students' ability in practical and theoretical courses of oral health education and to promote students' learning interest and initiative.
METHODSFourth-year students of the oral medical profession from 2006 to 2008 at Weifang Medical University were chosen as research objects for oral health education to explore the experimental teaching reform. The students were divided into test and control groups, with the test group using the "speak out" way of teaching and the control group using the traditional teaching method. Results of after-class evaluation of the test group, as well as final examination and practice examination of the two groups, were analyzed and compared.
RESULTSAfter-class evaluation results of the test group showed that the "speak out" teaching method was recognized by the students and improved students' ability to understand oral health education. The final examination and practice examination results showed that the score of the test group was higher than that of the control group (P < 0.01).
CONCLUSION"Speak out" teaching methods can improve students' ability for oral health education, in accordance with the trend of teaching reform.

8.Consistency of gene amplification and its expression at protein level of human epidermal growth factor receptor 2 in gastric carcinoma

Objective To investigate the consistency of gene amplification and its expression at protein level of human epidermal growth factor receptor 2 (HER2) in gastric carcinoma.Methods From 2010 to 2012,120 gastric cancer specimens of patients with gastric cancer were collected,of which 100 were surgical specimens and 20 were specimens from biopsy under gastroscope.The protein expression of HER2 in 120 specimens was detected by immunohistochemistry (IHC).According to the results of IHC,the positive parts of HER2 expression of IHC slices were developed into tissue microarrays for fluorescence in situ hybridization (FISH) to test the gene amplification of HER2.The different parts with different color intensity of focal (+ + +) (≤ 10% tumor cell strongly staining) specimens detected by IHC detection were compared with the results of FISH.Kappa test was performed for statistical analysis.Results Among 120 gastric cancer specimens,the results of IHC indicated that 77 specimens were positive with different staining intensity including 16 strong positive (+++),six focal positive (+++),37 moderate positive (++) and 18 weak positive (+).The positive rate of HER2 protein expression detected by IHC was 18.3% (22/120).The results of FISH showed 41 specimens were positive and the rate of gene amplification was 34.2%.Among which,21 were moderate positive (++) detected by IHC,15 were strong positive (+ + +) and five were focal positive (+ ++).The positive rate of HER2 was 35.0% (42/120) with IHC and FISH combined detection.The consistent rate of IHC and FISH was 91.6 % (76/83).Kappa coefficient was 0.960 (P<0.01).In five positive specimens detected by FISH and which were focal positive (+ + +) by IHC,the different parts with different color intensity were compared with the results of FISH and gene amplification was found in all specimens.Conclusion Tissue microarray technology is consistent with IHC in HER2 detection in gastric cancer specimens and could help to improve the detection rate.
9.PTEN protein loss is associated with an increased risk of recurrence in Chinese patients after prostatectomy for clinically localized prostate cancer

Background and purpose:Loss of the tumor suppressor phosphatase and tensin homolog deleted on chromosome 10 (PTEN) is one of the most common somatic genetic aberrations in prostate cancer in Western countries and is frequently associated with tumor progression and poor prognosis. This study aimed to investigate the frequency of PTEN protein loss in Chinese prostate cancer patients and to determine its association with the biochemical recurrence of prostate cancer.Methods:The data from 225 diagnosed localized prostate cancer patients with radical prostatectomy from 2006 to 2011 were collected retrospectively, including patient’s age at diagnosis, prostate-speciifc antigen (PSA) level at diagnosis, Gleason score, clinical stage, surgical margin, and time to biochemical recurrence or not. This study performed PTEN protein immunohistochemistry on tissue microarrays, which were made from 225 Chinese prostate cancer patients mentioned above, treated by radical prostatectomy with one case including 2 cancer spots and 2 adjacent normal gland spots. Correlations of PTEN loss with clinicopathological features were analyzed usingχ2 test. Kaplan-Meier survival model and Cox proportional hazards regression model were used to evaluate the predictive role of PTEN protein expression and patient characteristics for biochemical recurrence. Results:PTEN protein loss was observed in 15% of the patients and was associated with increased preoperative PSA levels (P=0.03) and old age (P=0.009). In univariate Kaplan–Meier analysis, the factors associated with the biochemical recurrence of prostate cancer included PSA levels (P=0.000 4), Gleason sum (P=0.019 8), and PTEN status (P=0.013 1). In multivariable Cox regression analysis, PTEN expression (HR=0.536, P=0.044), PSA levels (HR=1.879, P=0.001), and Gleason score (HR=1.361,P=0.03) were signiifcant in predicting biochemical recurrence of prostate cancer.Conclusion:PTEN protein loss is associated with an increased risk of recurrence, independent of known clinicopathological factors.
10.Discussion on Egg Freezing with Non-medical Factors from the Perspective of Medical Ethics

By expounding the definition and application status quo of egg freezing with non-medical factors, this paper ethically analyzed the conflict between egg freezing with non-medical factors and assisted reproductive technology standard management. After systematic evaluation on effectiveness, safety, economy and ethics, it sug-gested that the egg freezing with non-medical factors required careful consideration.
