2.HPLC Determination of Scutellarin and Chlorogenic Acid in Qingkailing Extract

Objective To determine the contents of scutellarin and chlorogenic acid in Qingkailing extract by HPLC.Methods Kromasil C18 column was adopted and the mobile phase consisted of methnol-water-phosphoric acid (20 ∶80 ∶0.2) with the flow rate being 1.0 mL?min-1 and column temperature being 25 ℃.The detection wavelength was 327 nm.Results The linearity of scutellarin was good in the range of 0.090~0.540 ?g,r=0.999 9,and its average recovery rate was 100.3 %(RSD=0.99 %);the linearity of chlorogenic acid was good in the range of 0.196~1.176 ?g,r=0.999 9,and its average recovery rate was 99.9 %(RSD=1.06 %).Conclusion This method is sensitive,simple,accurate,and can be used for the quality control of Qingkailing extract.
4.Changes of epidemiological characteristics of measles in Beijing before and after supplementary immunization campaigns of measles vaccine in 2010.

OBJECTIVETo investigate the changes of epidemiological characteristics of measles in Beijing before and after Supplementary Immunization Campaigns (SIA) (2007-2010 vs 2011-2014) of measles-containing vaccine (MCV) among children aged between 8 months and 14 years in 2010.
METHODSDescriptive epidemiological analysis was conducted on surveillance data of measles cases (clinical cases and laboratory confirmed cases), with the occurrence during 2007-2014, and of outbreaks, with the occurrence during 2009-2014, from National Notifiable Disease Reporting System. MapInfo geographic information system (Version 8.5) was used to illustrate the distribution of measles incidence by district. Annual measles incidence was classified into 5 groups at the same intervals between the upper and lower limits to analyze the morbidity of the different areas.
RESULTSIn total, 7 722 and 3 132 measles cases were reported during 2007-2010 and 2011-2014, with the annual incidence of 11.59 and 3.84 cases per 100 000 population, respectively. Comparing with the results during 2007-2010, total number of measles cases and average annual incidence during 2011-2014 were decreased by 59.4%, and 66.9%, respectively. Among measles cases during 2011-2014, percentage of cases aged 15 years or above were 57.7%(56/97), 62.0%(49/79), 65.5%(370/565), and 71.4% (1 707/2 391), respectively, which increased by years. During 2007-2010, the highest risk age for adults was 20-34, while 2011-2014, 5 years older: 25-39. During 2009-2010, 2011-2012, and 2013-2014, 50.3% (447/889), 30.3% (10/33), and 57.8% (201/348), respectively, of measles cases aged 8-17 months were unvaccinated by MCV. Percentages of measles cases aged 0-7 months, 8 months-14 years, 15-39 years and 40 years or above during 2013-2014, who visited hospitals 7-21 days before disease onset, were 59.8% (238/398), 49.3% (237/481), 32.2% (529/1641), and 37.6% (164/436), respectively. A total of 11 nosocomial measles outbreaks occurred during 2013-2014, which was much higher than that during 2009-2010 (2 nosocomial outbreaks). And universities accounted for the majority of outbreak settings of schools (3/4). All 11 outbreaks among grouped employees during 2009-2012 occurred in factories, restaurants, or large shopping centers, while the largest proportion (6/16) of that kind of outbreaks during 2013-2014 occurred in office buildings.
CONCLUSIONSSIA of MCV in 2010 effectively decreased measles transmission in Beijing. But routine immunization of MCV still needed to be improved. The issue of adult measles has been a prominent problem. Hospitals, office buildings and universities were the focus of prevention of measles transmission.

5.Effect of water stress on growth and accumulation of active components of Isatis indigotica.

OBJECTIVETo study the effect of water stress on the growth and accumulation of active components of Isatis indigotica.
METHODBy pot culture experiment,the characteristic indexes of water requirement, chlorophyll content, transpiration rate, photosynthesis, biomass and effective ingredient indirubin were studied under normal and water stress conditions, respectively.
RESULTWith the increase of the extent and duration of water stress, the quantity of water requirement, chlorophyll content, respiration rate, photosynthesis and biomass of I. indigotica were decreased, while water use efficiency increased.
CONCLUSIONIt is a critical period of water requirement in July, high water content in soil and extreme water stress are not propitious to the accumulation of indirubin, appropriate water stress can stimulate indirubin accumulatio. The harvest of leaves of I. indigotica is optimal in July, while the harvest of root is appropriate in October. Yield and quantity of I. indigotica are improved when the field capacity is between 45% and 70%.

6.Efficacy of targeted monitoring on surgical site infection following caesa-rean section

Objective To understand the occurrence of surgical site infection(SSI)following caesarean section,analyze risk factors,implement intervention measures,and evaluate intervention efficacy. Methods All puerperas who underwent caesarean section in the obstetric department of a hospital from January to December 2013 were mo-nitored,investigation were performed in two stages:evaluation stage(January-June,2013)and intervention stage (July-December,2013). Targeted intervention and clinical intervention were combined to intervene the risk factors. Occurrence of SSI,length of hospital stay,and hospitalization expense before and after intervention were compared. Results A total of 1 593 patients with caesarean section were monitored,31(1.95%)had SSI,incidence of SSI in evaluation stage and intervention stage were 3.40% and 0.95% respectively;incidence of SSI before and after inter-vention was significantly different(χ2= 12.02,P<0.01). Univariate analysis on evaluation stage showed that risk factors for SSI in patients with caesarean section were duration of operation≥1 hour,body mass index≥26 kg/m2,intraoperative blood loss ≥300 mL,underlying diseases,premature rupture of membranes,and without antimicro-bial prophylaxis(all P<0.05). In evaluation stage,648 patients received post-operative antimicrobial prophylaxis for>48 hours(n= 395,60.96%);in intervention stage,945 patients received post-operative antimicrobial prophy-laxis for<24 hours(n= 776,82.12%),different time distribution of post-operative antimicrobial prophylaxis in two stages after intervention was compared,difference was statistically significant(χ2= 673.26,P<0.01). The mean length of hospital stay of 31 SSI patients were(13.83±3.26)days,non-SSI patients were(7.06±1.66) days,difference was statistically significant(t= 7.86,P<0.01);the average hospitalization expenses for patients with SSI were(9 044.77±2 649.19)yuan,non-SSI patients were(6 254.73±638.52)yuan,difference was statis-tically significant(t= 4.344,P<0.01).Conclusion Intervention measures for risk factors of SSI after caesarean section can effectively reduce the incidence of SSI in caesarean section.
7.The role of CT and MRI fusion reconstruction-guided 3D printing navigation template for puncture during sacral neuromodulation: a clinical randomized controlled trial

Objective:To evaluate the safety and effectiveness of the accurate puncture during sacral neuromodulation (SNM) guided with 3D printing navigation template based on reconstruction techniques using fusing sacral CT and MRI images.Methods:Totally 42 patients operated with SNM were selected in Renji Hospital, School of Medicine, Shanghai Jiaotong University from July 2016 to August 2017. The patients were randomly divided into control group ( n=22) and experimental group ( n=20) using random number table. The conventional cross-positioning technique under X-ray was used for puncture during SNM in the control group. While in the experimental group, the sacral CT and MRI images were fused for reconstruction and design of the navigation template, printed by 3D technique for the puncture in SNM. The times of punctures, the average time for puncture operation, the time of intraoperative testing of the stimulator device, the minimum onset voltage of the stimulator, the X-ray radiation dose, postoperative curative effect (rate of secondary transformation) and the incidence rate of complications were compared between the two methods using independent-simple t test or χ 2 test. Results:Compared to control group, fewer times of punctures, shorter time needed for puncture operation, shorter time of intraoperative testing of the stimulator, smaller radiation dose and minimum effective voltage were found in the experimental group ( P<0.05). There were 15 and 16 patients who completed the secondary transformation in the control group and experimental group, and there was no significant difference between the two groups (χ2=0.757, P=0.384). There were 3 cases of complications in the control group, including 2 cases of infection and 1 case of bleeding, while no complications in the experimental group. Conclusions:CT and MRI images fusion reconstruction-guided 3D printing navigation template can help perform accurate and safe punctures in SNM. Compared to conventional puncture positioned under X-ray, it can effectively improve the puncture efficiency, and reduce the radiation dose in the operation.
8.Outbreak investigation of a campus bacterial dysentery by water source pollution in Lhasa City

Objective:
To investigate risk factors and the nature of the outbreak of bacillary dysentery in Lhasa City, so as to customize targeted prevention and control measures.
Methods:
Using on site epidemiological investigation, the suspected, probable and confirmed cases of bacillary dysentery in one school and one kindergarten in Lhasa City from June 26 to July 1, 2022 were collected, and additional cases were identified through interview from school staffs and family members, reviewing morning examination records and tracking records of school illness related absence. A case control study was conducted to investigate suspicious meals and drinking raw water, and polymerase chain reaction (PCR) was performed to detect Shigella line nucleic acid fragment in the patients feces, anal swabs, retained food, and terminal water.
Results:
A total of 55 cases were found in two schools, with the prevalence rate of 15.41% in total and 16.71% in students ( n =53), 7.5% in staff ( n =2). The epidemic curve was suggestive of a point source exposure. The prevalence rate among students who walk to school and students who live in the school showed no difference (16.10%,17.09%)( χ 2=0.05, P >0.05), and the prevalence rate was higher among elementary school students than kindergarten students (19.83%,6.67%)( χ 2=7.13, P <0.05). Case control comparisons showed a direct association between drinking raw water and morbidity in the case and control groups during June 24-26 ( OR=4.01, 95%CI =1.75-9.19, P < 0.05). A total of 23 fecal Shigella nucleic acid positives were detected from the two schools, two from the end water in front of the cafeteria door, and two from sludge in the sewage pipe around the wellhead.
Conclusions
The outbreak of bacillary dysentery is caused by the contamination of the pipe network water. Health administrative departments should improve the supervision and management of drinking water health safety, and schools should strengthen the management of water supply facilities for effectively prevent of waterborne infectious disease outbreaks.
10.Combination of AD5-10 and epirubicin in treating rheumatoid arthritis.

OBJECTIVETo investigate the mechanism of anti-death receptor 5-10 (AD5-10) combined with epirubicin in treating rheumatoid arthritis (RA).
METHODSWe detected the cell viability of the fibroblast-like synoviocytes (FLS) from RA patients with MTT. The expression level of apoptosis signaling pathways protein, p53, and p21 were evaluated with Western blot.
RESULTSWe found that epirubicin, at different doses, could enhance the effect of AD5-10 on FLS, promoting the apoptosis of FLS. The expression levels of caspase-3, -8, -9, c-FLIP, Bcl-2, p53, and p21 in the FLS changed after epirubicin treatment.
CONCLUSIONEpirubicin may coordinate with AD5-10 in inducing FLS apoptosis through affecting the levels of p53, p21, c-FLIP, and Bcl-2.
