2.Screening of Psychrotrophic Bacteria and Their Application to Treatment of Livestock Wastewater

Fourteen psychrotrophic bacteria were isolated from swamp soil collected in Ruoergai plateau wetland,and their generation time and degrading ability of livestock wastewater CODcr was determined.The results showed that the generation time was within 4.9 h to 11.6 h.Based on the generation time,9 psychro-trophic strains(NLJ1,NLJ6,NLJ7,NLJ9,NLJ10,NLJ11,NLJ12,NLJ13 and NLJ14),whose generation time was within 4.9 h to 5.6 h,were chosen to treat livestock wastewater.The results suggested that these 9 strains had different CODcr disposal ability when treating livestock wastewater singly at 6?C for 6 h,and strains NLJ6,NLJ7,NLJ9,NLJ10,NLJ11 and NLJ13 had good ability to degrade livestock wastewater,the CODcr degrading rate was about 60%~70%,hence,they were used as high efficient strains;However,the CODcr degrading rate of the other strains was less than 50%.After inoculating mixture culture of these six strains into the distilled livestock wastewater,after 6 h's treating,the CODcr degrading rate reached to 85.42%.Furthermore,activated sludge collected from Yaan,Dujiangyan and Chengdu were inoculated by the mixture culture of those six strains,and used to treat livestock wastewater for 6 h.The results showed that the average CODcr degrading rate was 81.67%,76.32% and 70.56%,respectively;Variance analysis showed that there was no significant differentiation between each treatment,which revealed that those six psychrotrophic strains had good adaptability to different source of activated sludge.
3.Combination of MGIT liquid culture and Xpert MTB/RIF test in the rapid diagnosis and drug resistance detection in Mycobacterium tuberculosis

Objective To evaluate the feasibility of Mycobacteria growth indicator tube ( MGIT ) liquid culture combined with rifampin resistance test real-time PCR ( Xpert MTB/RIF test) in the diagnosis of tuberculosis.Methods 652 cases of suspected pulmonary tuberculosis from October 2014 to March 2015 in Quzhou People′s Hospital were enrolled. The morning sputum samples were collected for acid-fast staining, L?wenstein-Jensen ( L-J) culture, BACTEC MGIT culture and Xpert MTB/RIF test.Samples with positive results of MGIT liquid culture were subjected to strain identification and drug sensitivity test with fluid method.Methodological comparison between four methods was made and the performance of MGIT liquid culture combined with Xpert MTB/RIF test in the rapid diagnosis and drug resistance detection in Mycobacterium tuberculosis was evaluated by chi-square test. Results Among the samples from 399 confirmed tuberculosis patients, the diagnostic sensitivity of the 4 methods ( acid-fast staining, L-J culture, MGIT culture and Xpert MTB/RIF test) were 17.0%(68/399), 23.8%(95/399), 37.8% (151/399) and 37.3%(149/399) respectively.The sensitivity and specificity of MGIT culture combined with Xpert MTB/RIF test was 39.8%(159/399) and 94.8%(240/253).The sensitivity of MGIT culture combined with Xpert MTB/RIF test was significantly higher than acid-fast staining (χ2 =50.9, P<0.01 ) and L-J culture (χ2 =23.7,P<0.01).The average detection time of MGIT culture was 7.5 days ( smear positive and Xpert MTB/RIF positive), 13.4 days (smear negative and Xpert MTB/RIF positive) and 16.9 days ( smear negative and Xpert MTB/RIF negative) .The sensitivity and specificity of Xpert MTB/RIF test in rifampin resistance detection were 9/9 and 97.3% ( 129/132 ) respectively.The average detection time of fluid method for drug sensitivity test was 8.3 days.Conclusions MGIT liquid culture combined with Xpert MTB/RIF test can detect Mycobacterium tuberculosis and the drug sensitivity rapidly.The method is highly sensitive and specific.It is important for clinical diagnosis and treatment of tuberculosis.

To observe therapeutic effect of continuous rehabilitation exercise combined low‐frequency e‐lectrical stimulation on patients with low extremity dysfunction after ischemic stroke and its influence on patients'lower extremity muscle strength and neurological motor function .Methods : A total of 94 patients with lower ex‐tremity dysfunction after ischemic stroke were randomly and equally divided into electrical stimulation group (re‐ceived low frequency electrical stimulation ) and combined treatment group (received continuous rehabilitation exer‐cise based on electrical stimulation group ).Therapeutic effect after three‐month treatment , lower extremity muscle strength and neurological motor function before and after treatment were evaluated , compared and analyzed be‐tween two groups .Results : After treatment , total effective rate of combined treatment group was significantly higher than that of electrical stimulation group (87. 23% vs.68.09%, P=0.026).Compared with electrical stimu‐lation group after treatment , there was significant reduction in lower extremity muscle strength score [ (1.96 ± 0.45) scores vs .(1. 43 ± 0. 41) scores] , significant rise in scores of Fugl‐Meyer movement scale [FMMS , (24. 54 ± 3.76) scores vs.(29. 98 ± 3. 82) scores] , Barthel index of activities of daily living (ADL ) [BI , (69. 02 ± 6.29 ) scores vs .(79. 46 ± 6. 35 ) scores] , Berg balance scale [BBS , (44.56 ± 5.71 ) scores vs .(52.14 ± 5.79 ) scores] , Holden walking function [ (3.92 ± 0. 46) scores vs .(4.61 ± 0.49) scores] and Tinetti gait analysis [ (6. 57 ± 1.13) scores vs .(8. 06 ± 1. 24) scores] in combined treatment group , P=0.001 all.Conclusion : Continuous rehabilitation exercise combined low frequency electrical stimulation can significantly enhance lower extremity muscle strength , improve neurological motor function , improve ADL in patients with lower extremity dysfunction after ischemic stroke , which is worth extending .
5.Comparative study on distribution of endophytic fungi in Eucommia barks from different habitats.

A total of 152 strains of endophytic fungi were isolated from the barks of Eucommia ulmoides in three regions (Lueyang country, Zunyi country, Cili country). Based on morphological characteristics and analysis of ITS sequences, these strains were identified into 8 genera. Thereinto Phomopsis, Diaporthe and Alternaria were common genera to Eucommia barks from different sites. But the dominant genus was different: Alternaria was the dominant genus in the barks from Cili country, and Phomopsis was the dominant genus from Zunyi country, then Diaporthe was the one from Lueyang country. According to the similarity coefficient, the composition of the endophytic fungi was distinctly different between the barks from three sites. The diversity and species richness in Lueyang country and Cili country were found higher than those in Zunyi country. The evenness of endophytic fungi was 0.936 5 in Lueyang county, which was higher than 0.737 1 or 0.641 0 in Cili county or Zunyi county, respectively. After phylogenic analysis and calculating the genetic distances of typical strains belong to Phomopsis and its perfect stage--Diaporthe, there was very high genetic diversity in the two genera from our study. In conclusion, the community structure and diversity of endophytic fungi were significant different in Eucommia barks from the three habitats.

7.Advances in the study of gene chip technology for the investigation of the mechanisms underlying cerebral ischemia and anti-cerebral ischemia agents.

With the development of molecular biology, genome science becomes an important subject currently. Characterized by high-throughput, high-integration, high-parallelism, miniaturization and automation, it is the integrated study of gene properties on a large scale. Stroke, an important cerebral vascular disease, is one of the threats to human health. The utilization of microarray study for the pathogenesis of stroke, not only reveals the essentials of the disease in the overall level of genes, but also contributes to the detection of therapeutic targets and the development of novel drugs for stroke. Referring to our own work, this discussion focuses on the progress of the mechanisms underlying experimental cerebral ischemia investigation in vivo as well as anti-cerebral ischemia agents by gene chip technology.

9.Assessment of static and dynamic balance in hemiparetic stroke patients

ObjectiveTo discusse the relationship between static balance in laboratory approaches and dynamic balance in clinical assessment and identify the value of static and dynamic balance at functional outcome in hemiparetic stroke patients. MethodsNineteen stroke subjects were assessed in this study. The static balance was measured by postural sway test, the dynamic balance was measured by Berg balance scale(BBS)and Time up to go test(TUGT),the outcome was measured by FIM and 10m maximum walking speed(MWS). The level of association between the parameters of postural sway test and clinical variables were examined with Pearson's correlation coefficients. ResultsThe parameters of postural sway test was significantly negative related to BBS(r=-0.705--0.475,P<0.05);The parameters of postural sway close-eye test was significantly positive related to TUGT(r=0.508-0.583,P<0.05);The parameters of postural sway test was no related to FIM and MWS (r=-0.048--0.296;r=-0.404--0.01,P>0.05);BBS was significantly positive related to FIM and MWS(r=0.752;r=0.700,P<0.001). TUGT was significantly negative related to FIM and MWS(r=-0.600,P<0.01;r=-0.817,P<0.001).ConclusionClinical and laboratory balance assessments are related and that dynamic rather than static balance measures are valid indicators of functional outcome performance in hemiparetic stroke patients.
10.Influence of Dangguibuxue Decoction on protein expression of NF-κBp65 activated by ox-LDL in RAW264.7 cells

Objective To investigate the possibility and mechanism of Dangguibuxue Decoction in early prevention of the development of atherosclerosis (AS) based on the study of the effect of Dangguibuxue Decoction on the signal transduction iter of nuclear transcription factor-kappa Bp65 (NF-κBp65) in monocyte activated by oxidized low-density lipoprotein (ox-LDL) in RAW264.7 cells. Methods The rabbits were randomly divided into the control group and medicated serum group and then given respectively normal saline and Dangguibuxue Decoction orally for preparing the medicated serum. The suspension of RAW264.7 cells was vaccinated into culture flask and cultured for 24 hours, and then divided into the blank control group, ox-LDL group, inhibitor group and medicated serum group. Except the blank control group all other groups were given 100 mg/L ox-LDL to activate RAW264.7 cells. After incubation the cells were collected. The expression of NF-κBp65 was detected by using immunohistochemistry assay, and the quantity of activated protein in RAW264.7 cells was detected by using immunoblotting technique. Results In ox-LDL group there were a lot of brown-yellow positive granules observed in the cytochylema and nucleus of RAW264.7 cells, and less in the medicated serum group. The ratio of protein band gray scale was 1.288 3±0.013 9 in the ox-LDL group and 0.918 9±0.015 3 in the medicated serum group. There was a significant difference statistically between two groups (P<0.05). Conclusion Dangguibuxue Decoction can down-regulate the protein expression and activation of NF-κBp65 in RAW264.7 cells stimulated by ox-LDL. It may be one of the mechanisms of Dangguibuxue Decoction for antagonizing AS that the signal transduction iter of NF-κBp65 was blocked.
