1.Different Telomere Fragments Promote Rat's Vascular Smooth Muscle Cell Apoptosis by Silent Information Regulator 1/Cancer Suppressor Gene P53

Objective: To study the influence of different telomere oligonucleotide on rat's thoracic aortic smooth muscle (A7R5 ) cell apoptosis and to clarify weather silent information regulator 1 (SIRT1)/cancer suppressor gene (P53) signaling pathway involved in oligonucleotide induced cell apoptosis. Methods: 3 telomere repeat sequences of TE-12 (TTAGGG)2, TE-24 (TTAGGG)4 and TE-36 (TTAGGG)6 were respectively transfected into rat's A7R5 cells as TE-12 group, TE-24 group and TE-36 group; in addition, blank A7R5 cells were used as Control group. Transfection induced A7R5 cell apoptosis was detected by flow cytometry, mRNA and protein expressions of SIRT1 and P53 in A7R5 cells were examined by RT-PCR and Western blot analysis in different groups. Results: Successful ransfection rates were similar among 3 groups. Compared with Control group and TE-36, TE-24 groups, the highest apoptosis rate of A7R5 cells was found in TE-12 group and the lowest was found in TE-24 group which was still higher than that in Control group, allP<0.05. mRNA and protein expressions of SIRT1 was obviously higher in TE-12 group than the other 3 groups, allP<0.05; mRNA and protein expressions of P53 was obviously lower in TE-24 group than the other 3 groups, allP<0.05. Conclusion: Telomere oligonucleotide sequence may promote rat's A7R5 cell apoptosis; different sequence had various influence and the strongest effect was observed in TE-12 sequence. The above impact might be related to SIRT1/P53 signaling pathway.

Dens in dente is a rare malformation of teeth.This article reports one case of type Ⅲ dens in dente of maxillary bilateral incisors with acute periapical lesion.The case was treated successfully by apexification with Vitapex paste.
3.Senegenin Protects against Ischemia-reperfusion Inj ury by Ameliorating Endoplasmic Reticulum Stress in Mice

Objective To investigate the protective role of senegenin(SEN)in the mouse ischemia-reperfusion injury and its mechanism.Methods Mice were divided at random into control group(the ligature was placed but not ligated for 225 min),is-chemia-reperfusion model group(the anterior descending coronary artery remained ligated for 45 min and then reperfused for 180 min)and SEN treatment group(30 mg/kg SEN was used 10 min before reperfusion).The myocardial infarct size was meas-ured by using Evans blue-TTC staining.Fluorescence assay was employed to detect the activity of Caspase-1 2 and Caspase-3 to assess the myocardial apoptosis.Western blot was used to detect the expression of two endoplasmic reticulum stress (ERS) markers,GRP78 and CHOP,in infarcted myocardia.Results Compared with the control group,the myocardial infarct size was significantly increased,the activities of Caspase-12 and Caspase-3 were increased by 4.71 fold and 3.37 fold,respectively,and the expression levels of ERS markers GRP78 and CHOP were conspicuously elevated in the ischemia-reperfusion model group.Pretreatment with SEN(30 mg/kg)could significantly reduce the myocardial infarct size,the activities of Caspase-12 and Caspase-3 and the expression levels of GRP78 and CHOP when compared with those in the ischemia-reperfusion model group and there was significant difference between the two groups(P<0.05 or P<0.01).Conclusion SEN can protect against the myocardial ischemia-reperfusion inj ury by ameliorating ERS-mediated myocardial apoptosis in mice.
4.Evaluation of therapeutic studies on chemotherapy and/or radiotherapy-induced oral mucositis published in Chinese journals by means of evidence-based medicine

Objective:To evaluate the quality and the effectiveness of therapeutic studies on chemotherapy and/or radiotherapy-induced oral mucositis published in Chinese journals by means of evidence-based medicine. Methods:Relevant papers were searched in Chinese Biological Medical Disc (CBMDisc)database and China National Knowledge Infrastructure(CNKI) database. Papers using randomized controlled trials (RCT) were identified and analyzed according to the international standards of evidence-based medicine. Results:There were totally 98 articles focusing on therapeutic studies of chemotherapy and/or radiotherapy-induced oral mucositis and only 22 of them met RCT criteria. Study design, diagnostic standards, therapeutic effects and adverse effects decribed in those 22 papers were fully evaluated by means of evidence-based medicine. Conclusions:Research quality of the analyzed papers could not meet clinical needs and Meta analysis could not been done due to their low quantity and poor quality. However, as far as the published articles concerned, GM-CSF showed a good short-term treatment effect.
5.The expression and clinical significance of NKp44+NK cells in the peripheral blood and synovial fluid of patients with rheumatoid arthritis

Objective To investigate the clinical significance of NKp44+NK cells in the peripheral blood and synovial fluid of rheumatoid arthritis (RA) patients.Methods The proportions of NKp44+NK cells in the PB of 20 active and 15 remission patients with RA and 20 healthy individuals were detected using flow cytometry.The proportions of NKp44+NK cells in the SF of 10 active patients were detected.Multiple nonparametric test was used to compared the difference of NKp44+NK cells proportions.Clinical data including DAS28,anti-CCP antibody and RF were collected.The relationship between NKp44+NK cells and clinical data was analyzed by Spearman correlation analysis.Results The proportions of NKp44+NK cells in PB of active and remission patients and healthy individuals were (1.480±4.750)%,(0.540±0.590)%,(0.000±0.000)%respectively.The proportion of NKp44+NK cells in PB of active patients was significantly higher than that of remission patients and healthy individuals (x2=46.708,P=0.000).The proportion of NKp44+NK cells in the SF of ten active RA patients was significantly higher than matched PB.There was positive correlation between NKp44+NK cells proportion in PB and SF of active patients and DAS 28 and anti-CCP antibody level.There was no correlation between NKp44+NK cells proportion in PB,SF and RF.Conclusion There is a marked increase in the proportion of NKp44+NK cells in PB and SF of RA patients.Moreover,NKp44+NK cells are positively correlated with DAS28 and anti-CCP antibody,suggesting that NKp44+NK cells may be correlated with disease activity and severity.
6.Research of anti-keratin monocional antibody on animal's psoriasis

Objective In order to explore the mechanisms of the anti-keratin monoclonal antibody 5G5(MAbSG5)in psoriasis treatment.Methods The different groups of mouse were fed with 5G5.The effect on regulating keratinocyte proliferation was observed.Results The experimental study revealed that MAb5G5 has the function of restraining the karyokinesis of vagina epithelium and accelerating the formation of stratum granulosum in rat squamous epidermis.Conclusion The mechanism of MAb5G5 in psoriagis treatment may be related to regulate keratinocyte proliferation.

9.Experimental Study on 99mTc-MIBI Imaging Evaluating Multidrug Resistance of Mice Lewis Lung Cancer

Objectives 99mTc-methoxyisobutylisonitrile (MIBI) SPECT imaging technology was used to observe the condition of tumor cell in-taking imaging agent in the C57BL/6J mice Lewis lung cancer model before and after using Ginsenoside Rg3 (short for Rg3).We aimd so as to discuss the feasibility of using this method to evaluate tumor multidrug resistance (MDR) status.Methods Mice Lewis lung cancer models were randomly divided into Rg3 group and the control group.After applying Rg3,semi-quantitative analysis was made on 99mTc-MIBI SPECT imaging and region of interest (ROI) to observe the multidrug resistance state of tumor and then the results were compared with the detection results of flow cytometry.Results The tumor intake ratio (T/N) difference between the control group and the Rg3 group in imaging,imaging before applying Rg3 and imaging after applying Rg3 were separately 15,60 and 120 min.The differences were statistical significant (P < 0.01).The eliminate indexes (WR) of the control group and Rg3 group were positively related to P-gp protein expression positive cells detected by flow cytometry (P < 0.05).Conclusions 99mTc-MIBI imaging is negatively related to P-glycoprotein (P-gp) expression in mice Lewis lung cancer cells,which can clearly show the multidrug resistance state of tumors and dynamically monitor the effect of Rg3 on multidrug resistance reversion of mice Lewis lung cancer.
