1.The use of portable slit lamp in the medical rescue of earthquake casualties

Objective To summarize the experience of examination and treatment of 237 cases of eye diseases with portable slit lamp in earthquake area of Dujiangyan, Sichuan Province, from May 14 to July 8, 2008. Methods 237 cases of the eye diseases were examined and diagnosed. With portable slit lamp, the foreign bodies in 25 cases of conjunctival foreign body and 62 cases of corneal foreign body were removed. Results Of the 237 cases, 39 were diagnosed as conjunctivitis, 25 as conjunctival foreign body, 13 as pterygium, 27 as keratitis, 11 as corneal ulcer, 62 as cornea foreign body, 24 as cornea injury, 7 as hyphema, 16 as iridocyclitis, 5 as glaucoma and 8 as cataract. With portable slit lamp, the foreign bodies in 25 cases of conjunctival foreign body and 62 cases of corneal foreign body were removed in one lump with no corneal perforation or infection. Follow-up examination showed that the wounded cornea repaired pretty well. Conclusions With portable slit lamp, ophthalmologists can not only make a precise diagnosis for the eye diseases in the anterior segment, but also are able to timely to remove foreign bodies in conjunctiva and cornea. Portable slit lamp is a useful and convenient instrument for ophthalmologists in the field of earthquake disaster relief.
2.Effect of platelet-derived growth factor on the expression of ?-smooth muscle actin of cultured human retinal pigment epithelium cells

Objective To investigate the effects of platelet-derived growth factor(PDGF) on the expression of ?-smooth muscle actin(?-SMA) of cultured human retinal pigment epithelium cells(RPE). Methods Cultured human RPE cells of the 4-6 th passages were divided into two groups: Delbecco′s modified Eagle′s medium (DMEM) and 2%DMEM (20 g/L foeta calf serum+DMEM). PDGF (0,1,50 ng/ml) was added to medium.The expression of ?-SMA was detected and quantitatively analyzed by image process of immunofluorescence. Results PDGF stimulated the expression of ?-SMA of human RPE cells.In group of DMEM, The rate of RPE of ?-SMA expression was 40%-50% and the intension of fluorescence was 8 08 without PDGF. After stimulated by PDGF(1 ng/ml,50 ng/ml), the rates were 80% and 90% respectively, and the intension of fluorescence were 12.35 and 17.23. In 2%DMEM group, The rates of RPE of ?-SMA expression were 85% without PDGF, and 95% ,100% respectively treated with PDGF (1 ng/ml,50 ng/ml). The intension of fluorescence was 14.79 without PDGF, and after stimulated by PDGF, they were 16.28 at 1 ng/ml and 21.36 at 50 ng/ml,which was 2.7 times stronger than that in DMEM group without PDGF. Conclusion PDGF could stimulate RPE cells to express ?-SMA.
3.Comparative Research on Removing Organophosphorus Pesticide Residues From Cucumber With Different Soak Water

Objective To evaluate the efficiency of different wash methods for removing organophosphorus pesticide residues from cucumber. Methods The cucumber samples were soaked with tap water, washed rice water, salt solution and detergent solution respectively for 2 to 15 minutes then organophosphorus pesticide residues in the soak water samples were determined with ultrasonic extraction and gas chromatography. Results Soaked in tap water for 2 to 5 minutes was the most effective and convenient way to remove methamidophos and dimethoate with the highest removing rate (about 70%). Conclusion Soaked with tap water for 2 to 5 minutes is more efficient for removing organophosphorus pesticide residues from vegetable.
4.Qualitative research on experience and feelings of mothers of children with hydrocephalus

Objective To describe the lived experience of mothers of children with hydrocephalus. Methods A semi-structured interview was conducted with 14 mothers of children with hydrocephalus. Data were analyzed based on Colaizzi phenomenotogical research method. Results Through the analysis, sorting and refining the theme, the mother's care experience could be divided into three themes: psychological emotional, stress response, social support requirements. Conclusions Nursing staff should have a thorough understanding of the experience to provide medical care and emotional support, improve the ability of the parents care,which could help caregiver adapt to their roles, maintain and promote the health of children patients and their families .
5.Effects of beraprost sodium on serum levels of soluble adhesion molecule-1 and C-reactive protein and urinary albumin excretion rate in patients with early diabetic nephropathy

Objective To observe the clinical efficacy of beraprost sodium on early stage of diabetic nephropathy(DN).Methods We measured serum levels of serum soluble intercelluhar adhesion molecule (sICAM)-1 in 27 cases with diabetes without nephropathy and 48 cases with early DN.Patients with early DN were randomly assigned into two treatment groups:the conventional treatment group and the beraprost sodium treatment group.Changes of sICAM-1 and C-reactive protein (CRP) levels and urinary albumin excretion rate (UAER) were measured in the two groups before and after treatment.Results The serum sICAM-1 levels in early DN patients was significantly higher than that of the diabetes without nephropathy group((1385 ± 171) g/ Lvs.(943 ± 167) g/L;t =1.034,P =0.002).There were no significant difference observed on levels of sICAM-1,CRP and UAER between the two treatment groups (P > 0.05).The level of sICAM-1 and CRP in the beraprost sodium treatment group was significantly lower than those before treatment (P < 0.05 or P < 0.01).The symptoms were significantly alleviated in both groups (P < 0.05 or P < 0.01),especially for the beraprost sodium treatment group(P < 0.05 or P < 0.01).Conclusion Patients with early DN have elevated serum sICAM-1 levels.Treatment of beraprost sodium has protective effect on early DN as it significantly decreases the serum levels of sICAM-1 and CRP in patients with early DN.
6.A study on the value of the serial position effect combined with delayed recall of auditory verbal learning test in distinguishing elderly patients with different levels of cognitive impairment

Objective:To explore the diagnostic value of the serial position effect combined with delayed recall of auditory verbal learning test in diagnosing the elderly patients with different levels of cognitive impairment.Methods:A total of 310 subjects were enrolled and divided into three groups: the normal control group(NC group, n=128), the mild cognitive impairment group(MCI group, n=133)and the mild Alzheimer's disease group(mild AD group, n=149)matched for gender, age and education level.Comprehensive neuro-psychological tests were performed on all subjects, and the Auditory Verbal Learning Test-Huashan version(AVLT-H)was used to analyze the serial position effect.Results:There were significant differences in the primacy effect of the second and third time of immediate recall, short-term and long-term delayed recall among the three groups( P<0.05), but no difference was found in the recency effect between the MCI and mild AD groups( P>0.05). The long-term delayed primacy effect combined with the short-term delayed recall showed a better validity to distinguish those elderly subjects with different levels of cognitive impairment than other memorial indexes, and the correlative sensitivity and specificity were 83.21% and 71.43%(NC group vs.MCI group), 85.71% and 92.86%(NC group vs.mild AD group), 66.67% and 75.57%(MCI group vs.mild AD group), respectively. Conclusions:The long-term delayed primacy effect combined with the short-term delayed recall can assist in distinguishing elderly patients with different levels of cognitive impairment.
7.THE EFFECTS OF HYPERTHERMIA ON THE NEUROEPITHELIAL CELLS IN PRIMARY CULTURE

The animal experiment studies have demonstrated that hyperthermia is a strong teratogen to many kinds of animal with high incidence of neural tube defects(NTD).Epidermiological investigation showed that hyperthermia was closely related to the acencephaly and excenphaly in human being,but little was known about the distinct mechanism of NTD induced by hyperthermia.In order to study the developmental mechanism of NTD induced by hyperthermia,we observed the effects of hyperthermia on the neuroepithelial cells in primary culture.The neural tubes of the hamster embryos on 10 d after fertilization were obtained.the neuroepithelia were dissociated and then seeded at the density of 1×106 cells per well.The cells were divided into experimental and control groups randomly.The experimental groups were exposed to 42 C for 20 min,whereas groups exposed to 37 Cserved as control.After treatment of hyperthermia,the cells were incubated continuously at 37 C in a 95%air/5%CO2 humidified incubator,the culture was terminated after different intervals.Phase-contrast microscopy.scanning electron microscopy.transmission electron microscopy,MTT assay,agarose gel electrophoresis analysis,TUNEL detection,immunocytochemistryand image analysis were made.The results of the experimental groups indicated that the floating cells and apoptotic cells increased in number,the neurites of the cells were shortened even disappeared,the survival cells decreased in number,the ultrastructure of the cells demonstrated distinct abnormal changes,the function of mitochondria was impaired,the expressions of both bcl-2 and bax were abnormal.The above results suggest that hyperthermia may induce apoptosis of neuroepithelial cells,duringwhich bcl-2 and bax genes may play important regulatory roles.
8.Effect of natural killer cells expressing KIR2DS1 receptor on efficiently killing dendritic cells in vitro

Objective To study the killing effects of the killer cell immunoglobulin-like receptors (KIR)2DS1-mediated natural killer (NK) cells against dendritic cells (DCs) and analysis the expression of chemokine receptor CCR7 after KIR2DS1-positive NK cells interact with the C2 epitope DCs in vitro,and explore the mechanism of alloreactive NK cell to limit graft-versus-host disease (GVHD).Methods NK cells were separated by Rosettesep NK sorting kit from healthy donor peripheral blood and taken as effector cells.The mononuclear cells from peripheral blood of newly diagnosed acute myeloid leukemia (AML) patients,and DCs were induced as target cells.Polymerase chain reaction and sequence specific primers (PCR-SSP) genotyping techniques and direct sequencing SBT were used to detect KIR gene,HLA-Cw of the healthy donors and patients.The cytotoxic activity of KIR2DS1-positive NK cells or KIR2DS1-negative NK cells against DCs were detected by CCK-8 assay.The expression of CCR7 after NK cells interacted with DCs was detected by flow cytometry.Results The purity of NK cells analyzed by flow cytometry was (94.20±1.23) %.Under same target ratio (10∶1),the killing effect of KIR2DS1+ NK cell against DCs was higher than that in KIR2DS1-NK cells (t =3.70,P < 0.05).Under same target ratio (10∶1),the cell lysis in C1/C1 group,KIR2DS1+ NK cells against C2/C2 group DCs (15.06±1.81) % was significantly higher than that against C1/C1 group (10.07±1.03) % and C1/C2 group (8.65±0.93) % DCs (F =56.368,P =0.001).KIR2DS1 + NK cells efficiently acquired CCR7 upon coculture with C2 epitope DCs.Conclusions NK cell expressing the KIR2DS1-activating receptor efficiently kill DCs,prevent GVHD,while enable NK cell acquire CCR7,obtaining migratory properties in vitro.
9.Effects of Panax notoginoside on the expression of TGF-β1 and Smad-7 in renal tissues of diabetic rats.

In order to explore the effects of Panax notoginoside (PNS) on the expression of transforming growth factor β1 (TGF-β1) and Smad-7 in renal tissues of diabetes, a rat model of diabetic nephropathy was set up by intravenous injection of streptozotocin (STZ). Wistar rats were randomly divided into normal group, diabetic control group, group treated by PNS at low-dosage (PL), group treated by PNS at high-dosage (PH) and group treated by catopril (C), respectively. Fasting blood glucose (FBG), renal index, endogenous creatinine clearance rate (C(Cr)) and urinary albumin (UAlb) in 24 h were examined after 6 weeks. Meanwhile, the expressions of TGF-β1 and Smad7 in renal tissues were immunohistochemically dectected. At the end of the sixth week, FBG, renal index, C(cr), UAlb were all elevated significantly in control group (P<0.01). The expression of TGF-β1 protein was increased while Smad7 protein decreased in renal tissue (P<0.01). However, the treatment with PNS reversed the aforementioned changes in renal tissues of diabetic rats. These results indicate that PNS possess a protective effect on the kidney of diabetic rats and it might protect kidney by inhibiting the expression of TGF-β1 protein and enhancing the expression of Smad7 protein.
10.Design and results of preoperative questionnaire in bariatric surgery

Objective To design an appropriate preoperative questionnaire in Chinese bariatric surgery.Methods Bariatric surgery center carried out laparoscopic bariatric operation from January 2010,preoperative questionnaire before the operation were supplied.Till January 2014,a total of 401 patients replied to the survey.The questionnaires were designed through three stages:translated from the Cleveland Clinic (USA) and applied in 30 patients for the first survey; adjustments and changes to the contents of the questionnaire for 90 patients according to Chinese conditions in the second phase; in the third stage,the third edition questionnaire was used in 281 patients.The satisfaction degree of the doctors and patients about the content of the questionnaire was evaluated.Results For the three versions of the questionnaire,the doctor satisfaction rate was 73.3%,86.7% and 96.8%.Patients' satisfaction rate of three versions were 30.0%,40.0%,76.9%,not satisfactory rate was 20.0%,11.1%,2.8%.Conclusions The current preoperative questionnaire includes 11 categories with 101 items of the third edition,doctors and patients satisfaction rate is higher,and it is suitable for Chinese bariatric surgery.
