2. Content changes regularity of flavonoids in Smilax glabra by QAMS method

Objective: To establish a method of quantitative analysis of multi-components by single-marker (QAMS) for simultaneously determination of neoastilbin, astilbin, neoisoastilbin, isoastilbin, and engeletin in Smilax glabra, and research the tendency of content changes in different growth years by QAMS. Methods: An HPLC method was established to determine the relative correction factors of four other flavonoids by using astilbin as the internal reference standard. Then the method was used to determine the various content of five flavonoids in different growth years and validate the feasibility and accuracy by comparing the content results determined by the external standard method with QAMS. The dynamic change regularity of flavonoids in S. glabra was investigated. Results: A total of 23 samples from five batches in different growth years were simultaneously determined by external standard method and QAMS, the results deviation were all less than 1.0%. The content of five flavonoids in different growth years was different, astilbin decreased with the increase of growth years, neoastilbin, neoisoastilbin and isoastilbin had the reverse trend. Conclusion: The established QAMS method is feasible and accurate for the simultaneous determination of five flavonoids in S. glabra. Growth years have a certain impact on the content of each component.
3. Research progress on ELISA and GICA for aflatoxin determination in Chinese materia medica

Aflatoxins, one of highly toxic and carcinogenic mycotoxins, are widespread in nature including Chinese materia medica (CMM). A simple, fast, high-throughput and on-site detection method is urgent needed for the screening the aflatoxins content in CMM to guarantee its safety. Enzyme linked immunosorbent assay (ELISA) and gold immune chromatograohic assay (GICA) provide an effective technique for the high-throughput detection and on-site single or a few of samples real-time detection of aflatoxin. There have no standard methods though many immunoassays have been established in some studies. The objective of our work is to make a research on the progress of immunoassay for aflatoxin determination and to provide a reference for the standard establishment.
4.Preliminary application of ultrasound real-time elastography in diagnosis of thyroid diffuse disease

Objective To explore the diagnosis value of ultrasound real-time elastography in thyroid diffuse diseases.Methods Eleven elastic quantitative parameters of 123 patients with thyroid diffuse diseases and 42 healthy volunteers obtained from the ultrasound real-time elastography were analyzed with principal components analysis to attain a comprehensive evaluation value which was named stiffness index(SI),reflecting the organization elastic information of different thyroid diffuse diseases.SI among each group was compared by ANOVA.Results Chronic lymphocytic thyroiditis SI was 86.37±20.45,subacute thyroiditis SI was 86.14±19.95,primary hyperthyroidism SI was 54.68±21.69,normal control group SI was 41.34±15.59,respectively.The SI between chronic lymphocytic thyroiditis,subacute thyroiditis,primary hyperthyroidism and normal controls had significant statistical differences (P<0.05),the SI between primary hyperthyroidism and chronic lymphocytic thyroiditis,subacute thyroiditis had significant statistical differences (P<0.05),the SI between chronic lymphocytic thyroiditis and subacute thyroiditis had no significant statistical difference (P>0.05).Conclusions Ultrasound real-time elastography could indirectly reflect the stiffness information of thyroid lension tissue and have a certain degree of diagnosis value in thyroid diffuse disease.
5.Prognostic value of serumal copeptinin patientswith acute exacerbation of COPD complicated with respiratory failure

Objective To analyse the prognostic value of serumal copeptin in patients with acute exacerbation of COPD compli-cated withrespiratory failure.Methods The serumal copeptin were detected in 60 hospitalized AECOPD patients before and after the treatment and in 30 healthy controls.Results Comparing with control groups,the serumal levels was significantly elevated in the patient with AECOPD before thetreatment and declinied two months later(P <0.05).The level of serum copeptin wereincreasing with the increase in the severity of the disease in acertain extent;The serumalcopeptin were positively correlated with lung function and CAT (r=0.058 and 0.821,P =0.00)and were negativelycorrelated with 6MWD(r=-0.652,P =0.00).Conclusion Serum copeptin could as-sess the incidence of acute exacerbations ofchronic obstructive pulmonary disease and its therapeutic effect;Serum copeptin can reflect the se-verity of AECOPD patients in a certain extent,and has certain clinical value to evaluate the quality of life in patients.
6.Long-term efifcacy of standardized speciifc subcutaneous immunotherapy on pediatric persistent allergic rhinitis

OBJECTIVETo observe the long-term therapeutic efficacy of standardized specific subcutaneous immunotherapy on persistent allergic rhinitis in children. METHODSFrom Jan. 2007 to Aug. 2009, 236 children with persistent allergic rhinitis were divided into two groups, which 120 cases underwent standardized house dust mite allergen subcutaneous specific immunotherapy (SCIT), another 116 cases accepted a serious of steroids nasal spray and oral antihistamine (control group). The efficacy of the patients was evaluated by using the visual analog scale (VAS) and Rhino conjunctivitis quality of life questionnaire (RQLQ).RESULTSAfter patients underwent 3-years SCIT, the VAS score was 2.3±0.7, 2.4±0.6, 1.6±0.4, 1.9±0.5, and the RQLQ score was 7.7±1.6, 7.4±1.1, 4.3±0.7, 4.1±0.9, respectively, at the follow-up period of 3 month, 1 year, 3 years and 5 years, and both scores were significantly lower than the score of pre-treatment (P<0.001). Compared the single dust mite allergen positive patients with multiple allergens positive patients, both VAS and RQLQ scores were not significantly changed at each follow-up period after SCIT (P>0.05). No serious adverse events occurred in all treatments.CONCLUSIONThe standardized specific immunotherapy has the long-term efficacy for dust mite positive children with persistent allergic rhinitis. It demonstrated a similar effect in both single dust mite positive patients and multiple allergens positive patients.

10.Interaction of E3 ligase HUWE1 and eukaryotic translation initiation factor eIF4E.

To explore the regulation of eIF4E, we screened the protein interacting with eIF4E from human cDNA library by using yeast two-hybrid system. Several clones interacting with eIF4E were identified. One of them was homologous with HUWE1 (HECT, UBA and WWE domain containing 1, also named as ARF-BP1, HECTH9 or HUWE1). Cell co-immunoprecipitation showed that eIF4E could bind to HUWE1 in mammalian cells. We also found that HUWE1 bearing the HECT domain is necessary for its association with eIF4E.
