The desired DNA product of KGPcd and KGP-hag was obtained from the total DNA of Porphyromonas gingivalis by PCR with two pairs of gene specific primers. The segment of KGPcd and KGP-hag (about 1.5kb and 1.6kb) was inserted into pGEM-T easy Vector. The double-stranded DNA of the postitive clone was analyzed by restriction endonuclease mapping and DNA sequenceing. The sequences of KGPcd and KGP-hag were consistent with those of the references appeared. The proteins of KGPcd and KGP-hag will be obtained for further study.
2.Determination of dencichine in Sanqi tablet by HILIC .

OBJECTIVETo develop an HILIC method for determination of dencichine in Sanqi tablet and evaluate the quality of Sanqi tablet of different hatches from various manufactures in the market.
METHODThe chromatographic separation was conducted on a Thermo HILIC column (4.6 mm x 250 mm, 5 microm) kept at 25 degrees C with acetonitrile and 0.1% H3PO4 (60:40) as the mobile phase. The flow rate was set at 1 mL x min(-1) and the detection wavelength was set at 213 nm.
RESULTThe contents of dencichine in Sanqi tablet ranged from 1.60 to 4.31 mg x g(-1).
CONCLUSIONThe well established method was successfully applied to determine dencichine in Sanqi tablet. The results demonstrated that this method was simple, accurate and could be applied for quality control of Sanqi as well as its associated preparations.

4.Clinical value of double respiratory navigator-gated high-resolution black-blood coronary artery wall magnetic resonance imaging.

OBJECTIVETo explore the value of phase ordering with automatic window selection(PAWS)and simultaneous multiple volume(SMV)algorithm double respiratory navigator-gated two-dimensional(2DNAV)dual inversion recovery(DIR)fast spin echo(FSE)high-resolution black-blood coronary artery wall magnetic resonance imaging(MRI)and evaluate its advantages and limitations.
METHODSPAWS and SMV 2DNAV DIR FSE high-resolution black-blood MRI was performed in 21 healthy volunteers. The images were evaluated qualitatively by using four grades(grade 0can not evaluate;grade 1bad;grade 2good;grade 3perfect). Images defined as grade 0 and grade 1 were excluded and those defined as grade 2 and 3 were evaluated further. Thickness of proximal(or middle)segment of right coronary artery(RCA)and left anterior descending branch(LAD)were measured. The difference of wall thickness was analyzed by using two-tailed independent sample t-test. P values of less than 0.05 were considered statistically significant.
RESULTSAmong the 38 slice images,31 slices(RCA13 slices,LAD18 slices;grade 214 slices,grade 317 slices)were obtained for further evaluation. The mean thickness of RCA and LAD was(0.94±0.16)and(0.89±0.15)mm,respectively,and the difference was not significant(t=-0.790,P>0.05).
CONCLUSIONPAWS and SMV algorithm 2DNAV DIR FSE high-resolution black-blood MRI has certain clinical value for coronary artery wall imaging.

5.Effects of tetrapod-like zinc oxide whiskers incorporation on antibacterial activity of composite resin.

OBJECTIVETo evaluate effects of incorporating tetrapod-like zinc oxide whisker (T-ZnOw) antibacterial agent on the antibacterial activity of composite resin, compared with that of the silver-based inorganic antibacterial agent.
METHODSThe minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of the two different antibacterial agents against Streptococcus mutans were investigated using the broth dilution test. Then the antibacterial activities of the self-cured composite resin specimens with different incorporating concentrations of the two antibacterial agents were evaluated using direct contact test. And the antibacterial activities of the resin specimens were examined again after 3 months of accelerating aging.
RESULTSThe MIC (MBC) of the T-ZnOw antibacterial agent and silver-based inorganic antibacterial agent were 0.15625 (0.3125) g/L and 0.15625 (0.15625) g/L. When the concentrations of T-ZnOw added to composite resin were 3%, 5%, and 10% (w/w), the antibacterial ratios of the resin specimens were (84.85+/-5.16)%, (94.22+/-3.73)%, and (99.43+/-0.48)% respectively. When the concentrations of the silver-based antibacterial agent added to composite resin were 1%, 3%, and 5%, the antibacterial ratios were (71.01+/-7.67)%, (90.76+/-5.91)%, and (97.19+/-4.10)% respectively. The antibacterial ratios of the resin specimens containing 5% of both antibacterial agents were (89.89+/-5.55)% and (78.79+/-7.81)% respectively after 3 months of accelerating aging.
CONCLUSIONSIncorporation of the T-ZnOw antibacterial agent in composite resin can improve the antibacterial performance of the resin, and the resin exhibits better antibacterial performance than that incorporating silver-based inorganic antibacterial agent after 3 months of aging.

6.Effect of Candida albicans growth on silicone elastomer containing nano-TiO2.

OBJECTIVETo study the antibacterial effect against Candida albicans of the A-2186 silicone elastomer containing nano-TiO2 in vitro.
METHODSAntibacterial agent of nano-TiO2 was added into A-2186 silicone elastomer with incorporating percentages of 0.5%, 1.0%, 1.5%, and 2.0% (W/W). There was no nano-TiO2 in the control group. The antibacterial effect of the A-2186 silicone elastomer was determined using the film contact method with lighting and without lighting.
RESULTSEither with lighting or without lighting, there were significances between the experiment groups and the control group (P < 0.05). When the incorporating percent was 2.0%, the inhibitory effect was the best among the experiment groups. Without lighting, the inhibitory rate was 53.7% and with lighting, the inhibitory rate was 85.9%.
CONCLUSIONSThe A-2186 silicone elastomer containing nano-TiO2 has antibacterial properties against Candida albicans, which enhances with increases of nano-TiO2 percent in the material. With the same incorporating percentage, the antibacterial effect with lighting is better than that without lighting.

7.Effects of three different zinc oxide incorporation on the antibacterial activity against Streptococcus mutans of composite resin.

OBJECTIVETo investigate the effects of incorporating three different zinc oxide (ZnO) on the antibacterial activity of composite resin.
METHODSThe minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of nano-ZnO, tetrapod-like zinc oxide whiskers (T-ZnOw), micro-ZnO against Streptococcus mutans were examined by the broth dilution test. Then the three different ZnO were added to the powder of one kind of bicomponent self-cured composite resin at 5% respectively, and the antibacterial activities of the resin specimens were evaluated using the membrane covering method before and after 3-month accelerating aging.
RESULTSThe MIC values of the three different ZnO against Streptococcus mutans were 78.13, 312.50 and 1 250.00 microg/mL respectively and the MBC values were 156.25, 625.00, 1,250.00 microg/mL respectively. The antibacterial ratios of the resin specimens incorporating with 5% of the three different ZnO were (93.58+/-5.95)%, (89.42+/-4.11)% and (78.97+/-3.90)% respectively, while after 3-month accelerating aging those were (89.01+/-7.91)%, (84.63+/-4.72)% and (72.27+/-3.89)%.
CONCLUSIONThe three different ZnO could improve the antibacterial activity of the composite resin. The nano-ZnO exhibit the strongest antibacterial activity, while the micro-ZnO weakest. The T-ZnOw presents comparatively strong antibacterial activity although with smaller specific surface area.

8.Renal protective activity of Hsian-tsao extracts in diabetic rats.

OBJECTIVETo investigate the renal protective activity of Hsian-tsao Mesona procumbens Hemsl. water extracts in diabetic rats.
METHODSThirty Sprague-dawley female rats were randomly divided into three groups (n = 10 each), "control group" with intraperitoneal saline injection, "diabetic group" with 60 mg of intraperitoneal streptozotocin injection per kg of body weight and "Hsian-tsao group" with intragastric administration of Hsian-tsao extraction everyday for 4 weeks after intraperitoneal streptozotocin injection. The body weight and blood sugar were measured before and after model induction in the three groups. Thrombospondin-1 (TSP-1) expressions in the kidney were monitored by immunohistochemistry. Kidney ultrastructural changes were also analyzed by using transmission electron microscopy.
RESULTSBefore diabetic model induction, there were no significant differences among the three groups in body weight and blood sugar. Four weeks after the induction of diabetes, the differences became statistically significant. Electron microscopy also revealed disruption of the foot processes of the podocytes and other damages in diabetic group. These damages were significantly less severe in Hsian-tsao group when compared with the diabetic group. TSP-1 expressions in the kidney were significantly increased in both the diabetic group and Hsian-tsao group, but it was relatively lower in Hsian-tsao group than in diabetic group.
CONCLUSIONOur results showed that Hsian-tsao treatment in the diabetic rats effectively prevented the pathological alterations in the kidney and decreased the TSP-1 expression. It was suggested that Hsian-tsao had protective effect on the kidneys of the diabetic rats.

9.Observation on therapeutic effects of red-hot needle therapy on nodules of breast of the type of stagnation of liver-qi and phlegm coagulation.

OBJECTIVETo compare therapeutic effects of red-hot needle therapy and filiform needle therapy on nodules of breast of the type of stagnation of liver-qi and phlegm coagulation.
METHODSSix hundred cases were randomly divided into a treatment group and a control group, 300 cases in each group. The treatment group were treated with red-hot needle pricking at the proliferative parts and Ashi points as main, and the control group with filiform needle therapy.
RESULTSIn the treatment group, 240 cases were cured, 58 cases improved and 2 cases were not cured with an effective rate of 99.3%; and in the control group, 113 cases were cured, 165 cases improved and 22 were not cured with an effective rate of 92.7%, with a significant difference between the two groups (P = 0.001).
CONCLUSIONThe therapeutic effect of red-hot needle therapy is better than that of filiform needle therapy on nodules of breast of the type of stagnation of liver-qi and phlegm coagulation.

10.Emerging role of ER quality control in plant cell signal perception.

The endoplasmic reticulum quality control (ER-QC) is a conserved mechanism in surveillance of secreted signaling factors during cell-to-cell communication in eukaryotes. Recent data show that the ER-QC plays important roles in diverse cell-to-cell signaling processes during immune response, vegetative and reproductive development in plants. Pollen tube guidance is a precisely guided cell-cell communication process between the male and female gametophytes during plant reproduction. Recently, the female signal has been identified as small secreted peptides, but how the pollen tube responds to this signal is still unclear. In this review, we intend to summarize the role of ER-QC in plants and discuss the recent advances regarding our understanding of the mechanism of pollen tube response to the female signals.
