To compare the differences in the measurement of maximum quantity of respiratory oxygen (VO2max) between direct and indirect methods, we selected 30 male students from the College of Physical Education, Hunan Normal University including 15 football players and 15 track and field players in November 2006. Objective To investigate the expression of aquaporin-1 (AQP1) in human peritoneum and its effect on peritoneal dialysis (PD) patients with the different peritoneal transport characteristics.Methods Peritoneal biopsies were obtained from the PD patients (n =30) at catheter insertion.Western blot and immunohistochemical staining were used to investigate AQP1 expression in peritoneal tissues.After catheter insertion,4 week,peritoneal equilibrium test (PET) was used to decide peritoneal transport characteristics.Results Western blot revealed a major band at 28 kDa band.The expression of AQP1 index was 1.49 ±0.67 in the normal subjects,and 1.61 ±0.76 in the PD patients,and there was no significant difference.Positive deposition was distributed in mesothelial cells,endothelial cells of capillaries,and small veins.The index of AQP1 expression in endothelial cells of capillaries in normal subjects and PD patients were 2.12 ±0.18,and 2.50 ±0.81,respectively,and no significant difference was found.The index of AQP1 expression in mesothelial cells of capillaries in normal subjects and PD patients were 1.95 ± 0.67,and 2.23 ± 1.07,respectively,and no significant difference was found.Among PD patients,the level of AQP1 was significantly different.Conclusions The expression of AQP1 was different in the PD patients with different peritoneal transport characteristics.In the high and high average transport groups,the level of AQP1 was lower; the low transport group was higher.It reveals that AQP1 plays a role in the pathogenesis of transcellular water transport.

Objective To investigate the effect of celecoxib on serum biochemical parameters in rheumatoid arthritis patients and its clinical effect and safety assessment.Methods 92 cases of rheumatic arthritis were selected and divided into two groups.46 cases in control group were treated with methotrexate and hydroxychloroquine sulfate, and 46 cases in experitment group were treated on the basis of control group with celecoxib.The serum osteoprotegerin ( OPG) levels and related biochemical parameters, efficacy evaluation by doctors and self evaluation of patients and adverse reactions were compared between two groups.Results After treatment, the serum levels of IL-1, IL-6, CRP, C3, C4, RF, IgA, IgG and IgM levels after treatment were significantly lower than those before treatment in two groups ( P <0.05 ) , OPG serum level after treatment group was lower than that before treatment in experiment group ( P <0.05 ) , but there was no significant difference in control group.The above indexes in experiment group in experiment group were lower than those in control group after treatment.The efficacy evaluation by doctors in experiment group was 86.96%, which was higher than 69.57% in control group(χ2 =4.09,P<0.05), and efficacy self evaluation by patients was 84.78%, which was higher than 65.22% in control group (χ2 =4.70,P<0.05).The adverse reactions in experiment group was 13.04%, which was significantly lower than 45.56% in control group (χ2 =11.79,P<0.01).Conclusion Celecoxib could significantly improve clinical symptoms and physical sign and serum biochemical markers, and reduce adverse reactions in rheumatoid arthritis patients, which is an effective and safety treatment.

Vitamin D deficiency is common among the elderly,which may be an important pathogenic factor in Alzheimer's disease(AD).Vitamin D has multiple functions including immune regulation,anti-oxidation,regulation of neurotrophic factors/acetylcholine neurotransmitters,and clearance of amyloid beta peptide; meanwhile,it may be helpful to avoid hyperparathyroidism.Vitamin D supplementation among the elderly may prevent the loss of neurons and slow down the AD-related cognitive decline,and therefore may play a role in the multi-targeted therapies for AD.

Objective To develop techniques based on polymerase chain reaction(PCR) which can detect 2 of most common deletional ? thalassemia ? 3.7 deletion and ? 4.2 deletion in China accurately and speedily. Methods Two groups of primers were designed and synthesized. PCR conditions were optimized. The PCR pro ducts were analysed by 1.0% agarose gel electrophoresis. The gel was stained by EB and photographed using an UVP gel documentation. Results PCR product of a 1 700 bp DNA fragment with primers A′, B′, C3 indicates the ? 3.7 deletion while a 1 900 bp fragment indicates a normal or wild type of ? globin gene. Occurrence of the 1 700 bp and 1 900 bp simultaneously indicates a heterozygous of the ? 3.7 deletion. Neither of the 2 bands was presented, indicating a homozygous of South East Asia type of deletion (-? SEA ). According to patterns of 1 580 bp and 1 180 bp hand amplified by a PCR with primer G′, E, F′, we detected the ? 4.2 deletion and distinquished its heterozygous and homozygous. Conclusions The 2 PCR based techniques developed in our laboratory are accurate, simple, well reproducible and easy to use for screening of the 2 deletion types of ? thalassemia determinants.

As to a nitrite-oxidizing bacterium screened in our laborator y, the effect of pH, nitrogen sources, carbon sources and sodium chloride on its g rowth was studied in order to obtain high cell density. It showed that the cult ure conditions of nitrite- oxidizing bacterium were 4500mg/L sodium nitrite, 1 .5g/L sodium carbonate, 0~0.5% sodium chloride and 0~0.1% glucose at 28℃~30 ℃, 110 r/min and pH 8.0~8.5. After cultivation for 9 days, the bacteria conc entration reached 4.6?109 MPN/mL and all NO-_2-N in the medium w as converted to NO-_3-N. But the transformation of nitrogen sources was inhibited while the sodium chloride concentration exceeded 0.5% or glucose con centration exceeded 0.1%. According to the test of NO-_2-N conversion with nitrite-oxidizing bacteria in a freshwater aquaculture pond, NO-_2 -N began to decrease on the third day of the moculation of this bacterium and t he concentration dropped from 1.47mol/L to 0.49mol/L after 18 days at 25℃ and pH 8.6.

Objective To investigate the effects of propofol administered at different intervals on acute lung injury induced by endotoxin shock and the mechanism in rats.Methods Seventy-six healthy adult male Wistar rats, weighing 250-290 g were anesthetized with 30 % pentobarbital sodium 30 mg. kg-1 ip. Carotid artery and jugular vein were cannulated for BP monitoring and administration of drugs and fluid. The animals were randomly divided into 5 groups : group A received only normal saline (NS) ( n = 8) ; group B received LPS 8 mg' kg and NS iv ( n =17); group C-E received a bolus of propofol 5 mg .kg-1 iv, 1 h before (group C) or together with (group D) or 1 h after intravenous LPS (group E) followed by propofol infusion at 10 mg . kg-1 . h-1 ( n = 17 in each group). The animals were observed for 5 h after LPS. Blood samples were taken from femoral artery before (baseline) and 1, 3 , 5 h after LPS for determination of PaO2 ,pH, and serum TNF-a and NO levels. The survival rates of the animals in the 5 groups during the 5 h after LPS were compared. The animals were sacrificed and the lungs were removed at the end of 5 hour observation for determination of lung TNF-a and MDA content and myeloperoxidase (MPO) activity and microscopic examination. Results Propofol given 1h before (group C) and simultaneously with LPS (group D) significantly attenuated the decrease in MAP, pH and PaO2 and increase in serum contents of TNF-a and NO and lung TNF-a, MDA contents and MPO activity induced by LPS. The 5 hour survival rate was also significantly higher in group C and D than that in group B. Conclusion Early propofol administration is beneficial during endotoxin-induced shock.
