1.Expression of polo-like kinase 1 in human renal cell carcinoma and impact of BI2536 on the cell proliferation of renal cell carcinoma A498 cell line

Objective To investigate the expression of Polo-like kinase 1 in renal clear cell carcinoma and tissue adjacent to carcinoma;impact of BI2536 on the cell proliferation of renal clear cell carcinoma A498 cell line.Methods We select 12 cases of renal cancer Specimens of randical surgery between January 2013 and December 2014.The study included 7 male patients and 5 female patients,whose age ranged from 41 to 68 years(mean 49 years).Tumors were single and the location of tumor included 5 in left kidney,7 in right kidney.The size of renal tumor ranged from 2.1 to 6.3 cm.All patients did not have radiotherapy and chemotherapy preoperative who have no local and distant matastasis.The pathological results demonstrated renal clear cell carcinoma in 12 cases.At the same time,we collect the tissue 2 centimeters away as control,which pathological results demonstrated normal tissue.The expression of PLK1 was detected by western bloting in tissues from 12 cases of renal clear cell carcinoma and tissue adjacent to carcinoma and quantitative analysis was made.Cell proliferation of A498 cell line at the different concentrations of BI2536 were assayed by flow cytometry (0,20,40,60μg/L).Results The expression of PLK1 in renal clear cell carcinoma tissue are 0.74 ±0.2 and 0.21 ±0.13.The carcinoma tissue is higher than tissue adjacent to carcinoma (P =0.02).BI2536 act on A498 after 48 hours,the A498 cell number were arrested at G2/M phase at 0μg/L,20μg/L,40μg/L and 60μg/L were (10.28 ± 0.47) %,(14.35 ± 0.85) %,(20.49 ± 0.78) % and (23.90 ± 0.54) %,respectively.The A498 cell number were arrested at G2/M phase increase when we incearse the concentrations of BI2536 at 48 hours (P < 0.05).Conclusions The higher expression of PLK1 in renal clear cell carcinoma and the proliferation of the renal clear carcinoma cell can be repressed by BI2536 both reveal PLK1 may be used as the molecular maker and therapeutics target of renal clear cell carcinoma.
2.Chromatographic fingerprint for Schizonepeta tenuisfolia Briq.

AIM: To study the chromatographic fingerprint for Schizonepeta tenuisfolia Briq. by gas chromatography (GC). METHODS: The GC equipped with FID detector and a Varian CP-Sil 24 CB capillary column was used. The temperature program of column oven was as follows: 50℃, held for 3 min; increased at a rate of 2 ℃/min to 110℃, then at 6℃/min to 250℃, held for 5 min. RESULTS: The analytic method for fingerprint of Schizonepeta tenuisfolia Briq. was developed and validated. The fingerprint for Schizonepeta tenuisfolia Briq. was established. CONCLUSION: The method is simple, precise and reliable. The fingerprint is helpful to the quality control of Schizonepeta tenuisfolia Briq. in Chaijing Injection.
3.Combination of transurethral pneumatic and ultrasonic cystolithotripsy: a clinical analysis of 30cases

The clinical data of 30 patients with vesical calculus of 1.5 cm to 4 cm in diameter,who underwent combination of transurethral pneumatic and ultrasonic cystolithotripsy,were retrospectively analyzed.During the procedure the resectoscopic sheath was inserted into bladder through urethra,then the nephroscope was placed through the sheath.When the stone was located,the ultrasonic and pneumatic devices were used for lithotripsy and the fragments were cleared out.The procedures lasted from 10 to 25 min,all operations were successful.Transurethral resection of prostates (TURP) was performed simultaneously in 28 patients.No serious bleeding,bladder injury or transurethral resection syndrome occurred.One patient with simultaneous TURP developed bladder neck contracture one month after surgery and was later cured by endoresection.No residual chip or stone recurrence was detected.This procedure appears to be safe and highly efficient with low rate of urethrostenosis and is suitable for simotaneous prostate surgery.
4.Correlation of CT features and pathological characteristics with EGFR gene mutations in invasive peripheral pulmonary adenocarcinoma

Objective To investigate the correlation of CT features and pathological characteristics with epidermal growth factor receptor(EGFR)gene mutations in invasive peripheral pulmonary adenocarcinoma.Methods The amplification refractory mutation system was used to determine EGFR mutations in 1 9 3 surgically resected invasive peripheral pulmonary adenocarcinomas.CT features and pathological characteristics were analyzed retrospectively.Results The total EGFR mutation rate was 62.2% (120/193).Among the features on CT,the maximum tumor diameter (Dmax)in axial plane CT images was significantly smaller in patients with EGFR mutations than that with wild-type EGFR patients [(2.52 ± 1.01)cm vs (3.11 ± 1.34)cm,P<0.05].Receiver operating characteristic (ROC)results indicated that Dmax=2.01 cm was the diagnosis threshold in forecasting EGFR gene mutations,with the sensitivity and specificity of 79% and 64%,respectively.The frequency of EGFR mutations was significantly greater in tumors with ground-glass opacity(GGO)than that without GGO (78.0% vs 56.6%,P<0.05),and in tumors without cystic airspaces than that with cystic airspaces (65.5% vs 40.0%,P<0.05).No correlations were observed between EGFR mutations and other CT features,including GGO/tumor ratio(G/T),lobulation,spiculation,pleural retraction,vascular convergence,air bronchograms,and vacuole signs (P>0.05).Among pathological characteristics,compared with other subtypes,EGFR mutations occurred most frequently in lepidic predominant adeno-carcinomas (77.5% vs 58.2% in other subtypes,P<0.05),and the least frequently in solid predominant adenocarcinomas(26.3%vs 66.1% in other subtypes,P<0.05).The EGFR mutation rate was significantly higher in tumors without lymph node metastases than that with lymph node metastases (66.9%vs 50.9%,P<0.05).Conclusion The CT features and pathological characteristics may be useful indicators to predict EGFR mutation status in invasive peripheral pulmonary adenocarcinoma.
5.Correlation between MSCT findings and pathological classification of thymic neuroendocrine tumors

Objective To study the MSCT appearances of thymic neuroendocrine tumors (NETs)and its correlation with the WHO histological grade.Methods MSCT features of 16 patients with thymic NETs confirmed by pathology were analyzed retro-spectively.The patients were divided into 2 groups according to tumor's grade,i.e.low and intermediate grade,high grade.Results There were 8 patients in low and intermediate grade,8 in high grade.No difference was found among tumor location,size,tumor morphology,calcification,pericardiac thickening,pericardial effusion,pleural thickening,pleural effusion,disappearance of fat line around tumor and mass-pulmonary interface with irregular shape,but significant difference was detected in lymph node metastasis. On enhanced MSCT scanning,statistical difference was detected in linear enhancement of the blood vessels in the tumors,but no difference was found between necrosis and peripheral vessel invasion.Conclusion MSCT findings of different grades in thymic NETs have some characteristics and can be helpful in the grades of predictability.
6.Relationships between the levels of serum bone-specific alkaline phosphatase, cross-linked telopeptide of type Ⅰ collage and bone metastasis in patients with breast cancer

Objective To analysis the relationships between bone markers, bone-specific alkaline phosphatase (BAP) and cross-linked telopeptide of type Ⅰ collage (ICTP), and bone metastasis of breast cancer.Methods A total of 217 patients' serum were collected.The 217 cases were divided into two groups:109 cases with bone metastasis, 108 cases without bone metastasis. Serum BAP and ICTP was measured by ELISA. The relationships between factors of bone metastasis and serum levels of BAP, ICTP were analyzed.Results The levels of serum BAP and ICTP in bone metastases group were significantly higher than those in non-bone metastasis group[BAP:24.8 μg/L(7.60-213.70 μg/L) vs 21.2 μg/L(7.3~68.8 μg/L),ICTP:7.0μg/L(1.4~32.4 μg/L) vs 4.1 μg/L(0.0~15.8 μg/L) (P=0.003,P=0.000)].The level of serum BAP and ICTP in patients with multiple bone metastasis was significantly higher than that in patients with single bone metastasis[BAP:32.3 μg/L(9.A~213.7 μg/L) vs 18.1 μg/L(7.6~60.0 μg/L),ICTP:7.6 μg/L(1.4~32.4 μg/L) vs 4.9 μg/L(1.8~10.5 μg/L),(P=0.001,P=0.010)].The sensibility of BAP and ICTP was 45.0 % (49/109)and 46.8 % (51/109),respectively.The specificity of ICTP and BAP was 83.3 % (90/108)and 84.3 % (91/108),respectively.Joint detection of BAP and ICTP had improved sensibility in the diagnosis of bone metastasis in breast cancer patients. Conclusion Joint detection of serum bone biochemical markers ICTP and BAP have a little values for diagnosing bone metastasis in breast cancer patients.
7.A finite element model of the knee joint for total knee arthroplasty: Construction and biomechanic analysis

Objective Few studies are reported on the construction of a finite element model of human complex knee joint using multimodality CT and MRI images.In this study, we developed a finite element model of the knee joint for total knee arthroplasty (TKA) using matched and fused CT and MRI data, hoping to provide a useful tool for the simulation study of knee joint biomechanics of TKA.Methods The CT and MRI image data about an intact knee of a 26-year-old male volunteer were imported into the Mimics software for the establishment of 3D models of bony and soft-tissue structures.A complete knee model was developed following the registration and fusion of the constructed 3D models based on the external landmarks.After the simulated implantation of TKA components, a finite element model of the TKA knee was constructed with the Hypermesh software.Then the finite element model was analyzed following the definition of its material behavior, boundary conditions and loading.Results The finite element model of the TKA knee, which was composed of bones, ligaments, components, polyethylene insert and bone cement, was developed from CT-MRI image registration and fusion and maintained its important spatial relationship among different structures in the TKA knee.The results obtained from the finite element analysis showed the characteristics of stress distribution in the TKA knee.Conclusion The finite element model of the knee joint for TKA can be established by matching and fusing CT and MRI image data, which can be employed as a useful tool for the study of knee joint biomechanics of TKA.
8.Pharmacokinetic study on compatibility of berberine and citrus aurantium extract in rats

Objective UPLC-MS/MS bio-analysis method was developed for the simultaneous determination ofberberine,naringin,hesperidin,and neohesperidin in plasma of rats.Methods UPLC Acquity BEH C18 (50 rmm × 2.1 mm,1.7 μm) column was used,mobile phases were containing 0.05% formic acid and 2 mmol/L ammonium formate in water (A)-containing 0.05% formic acid in acetonitrile (B) as the mobile phase gradient elution;SD rats were randomly divided into oral administration berberine group,Citrus aurantium extract group,and berberine and C.aurantium extract compatibility group.Results UPLC-MS/MS method could be applied to determination of berberine,naringin,hesperidin,and neohesperidin,method validation meets the requirements of biological sample analysis.When rats were administered with berberine and C.aurantium extract compatibility,the plasma concentration of berberine was much more than single dose of berberine group and the bioavailability of berberine was increased.Meanwhile,naringin and neohesperidin can be detected in rat's plasma.Conclusion The bioavailability of flavonoids is significantly improved as well compared to the single dose of C.aurantium extract.This suggests that berberine and C.aurantium extract compatibility has significant drug-drug interaction.
9.Determination of mercury content in yuhong ointment by wet catalytic digestion-atomic absorption spectrometry.

OBJECTIVETo develop a method for determining the content of mercury contained in Yuhong ointment.
METHODThe wet catalytic digestion method was adopted for the pretreatment, and the mercury content in Yuhong ointment was determined by hydride generation-atomic absorption spectrometry (HG-AAS).
RESULTThe mercury showed a good linear relation in the range from 2 to 20 microg x L(-1), with the average recovery of 104.27% and RSD of 3.37%. The RSD for real sample repeated measurement was determined to be 8.4%. The mercury content in Yuhong ointment was detected in range from 0.7 to 1.5 mg x g(-1).
CONCLUSIONThe proposed method is accurate, highly reproducible and it can be used to control mercury content of Yuhong ointment.

10.MSCT findings of airway-invasive pulmonary aspergillosis

Objective To explore the MSCT features of airway-invasive pulmonary aspergillosis.Methods MSCT features of 27 cases of airway-invasive pulmonary aspergillosis confirmed by pathology or clinical experience were analyzed retrospectively.Results The lesions in 27 cases were distributed along the blood vessel and bronchus and located mainly in the upper and middle lung field in 19 cases.Multiple centrilobular nodules and tree-in-bud sign were found in 27 cases and bronchial wall thickening and patchy opacities around the bronchi were in 25 cases.Bronchiectasis was seen in 17 cases,and cavity-like change and inter-cavity separation were in 15 cases.In 22 cases of follow-up,the lesion progressed within 1 week after diagnosised and treatmented in 20 cases and recurred in slow recovery stage in 5 cases.Conclusion MSCT findings of airway-invasive pulmonary aspergillosis are various which may rapidly progress within a short time.MSCT palys an important role in the evolution and follow-up of airway-invasive pulmonary aspergillosis.
