1.Sero-pharmacological Studies on Guyanding in Promoting the Proliferation of Vascular Endothelial Cells

【Objective】To observe the effect of Guyanding(GYD)decoction,a prescription with the actions of tonifying kidney,strengthening Qi and activating blood on proliferation of vascular endothelial cells(VEC).【Methods】New Zealand rabbits were randomized into blank control groups and GYD decoction groups in the dosages of 2.5,5 and 10g/kg respectively.GYD in the dosage of 10g/kg was given to rats for 7 successive days,and blood sampling was collected 0.5,1,1.5,2,2.5,3,3.5 and 4 hours after the last administration to prepare serum containing GYD.Then human umbilical vein endothelial cells were cultured with serum containing GYD,and the effects of GYD-containing serum at different time points on the proliferation of VEC were observed by methylthiazolyltetrazolium(MTT)assay.GYDcontaining serum collected 2h after the last administration was partially inactivated and its effect on VEC proliferation was compared with that of non-inactivated GYD-containing serum.Meanwhile,the effect of non-inactivated GYD-containing serum on VEC proliferation was observed 12,24 and 36 hours after the culturing.GYD in three different dosages was given to rats for 7 successive days,and serum containing GYD collected 2h after the last administration was prepared to observe the effect of GYD in different doses on VEC proliferation.【Results】Serum containing high-dose GYD collected 0.5~4h after the administration had obvious effect in promoting VEC proliferation(P
2.Effect of Rhizoma Drynariae Total Flavones on Expression of Bone Morphogenetic Protein 2 and Transforming Growth Factorβ1 in Distraction Osteogenesis

Objective To investigate the effect of total flavones of Rhizoma Drynariae on femur distraction osteogenesis in the rabbits. Methods Thirty-two healthy rabbits were randomly divided into treatment group and control group, 16 rabbits in each group. The femoral fracture was treated with unilateral femoral distraction and was fixed with a self-made distraction instrument. After 7-day intermittent period, the fractured femur was distracted at a rate of 1 mm/d, twice a day for 10 continuous days. The treatment group was fed with total flavones of Rhizoma Drynariae from the first post-operative day to the end of the experiment. And then all of the animals were sacrificed after fixation for 28 days. The bone specimens were used for histological observation and immunohistochemical detection. Results The area of mature bone in the newborn bone tissue of the treatment group was increased, and osteoblasts number and the percentage of trabecular bone area were significantly higher than those of the control group . The bone morphogenetic protein-2 (BMP-2) and transforming growth factor-β1 (TGF-β1) were stained brown deeply, the staining degree being stronger than that of the control group. Conclusion Rhizoma Drynariae total flavones can effectively accelerate the formation and maturation of newborn bone tissue during bone distraction.
3.Comparison of imaging features between focal nodular hyperplasia and hepatocellular carcinoma on MR diffusion weighted imaging

Objective To investigate the imaging features of focal nodular hyperplasia and hepatocellular carcinoma on DWI. Methods The data of patients with histopathologically confirmed FNHs and HCCs between August 2008 and November 2010 were collected. A total of 24 patients with 26 FNH lesions and 36 patients with 39 HCC lesions were included in our study. All patients underwent breath-hold DWI with b = 500 s/mm2 and dynamic contrasted-enhanced (DCE) MRI. The imaging findings of FNHs and HCCs were retrospectively analyzed and compared. The signal intensity (SI) of the lesions on DWI were classified as iso-, slightly high, high SI and the distribution of SI between FNHs and HCCs was compared with Fisher exact test. ADC value and lesion-to-liver ADC ratio of FNHs and HCCs were measured and compared by using independent sample t test. ROC was performed to assess the diagnostic value of ADC value and lesion-liver ADC ratio in the characterization FNHs versus HCCs. Results Of 26 FNHs,23 manifested as isointensity or slightly high SI on DWI, but most 25 out of 39 HCCs showed high SI. The distribution of SI between FNHs and HCCs had significant difference ( P = 0. 000). The mean ADC value and lesion-liver ADC ratio for FNHs [ (1.76 ± 0. 62 ) × 10-3 mm2/s and 1.06 ± 0. 18, respectively ] were significantly higher ( P = 0. 001, P = 0. 000, respectively ) than those for HCCs [ ( 1.26 ± 0. 46 ) × 10-3mm2/s and 0. 79 ±0. 12, respectively]. The area (Az) under the ROC for the ADC value and lesionliver ADC ratio for the differentiation of FNHs versus HCCs were 0. 79 ± 0. 05 and 0. 85 ± 0. 05,respectively, with no significant difference (P =0. 270). The specificity of the two measures was 69. 23% and 97.44%, respectively, with significant difference (P = 0. 001 ). Conclusion FNH shows isointensity or slightly high SI with relatively higher ADC value and lesion-liver ADC ratio than those of HCCs on DWI,which is characteristic for its diagnosis and differentiation.
