1.Immunoisolation effect of xenografts of microencapsulated human chromaffin cells in rats

Objective To investigate the immunoisolation effect of xenografts of microencapsulated human chromaffin cells (HCCs) .Methods HCCs were microencapsulated with APA microcapsules. Forty-eight SD rats were randomly divided into 3 groups ( n = 16 each) . HCCs (group I ) , empty APA microcapsules (group II ) and microencapsulated HCCs (group III ) were implanted into the anterior chamber of the eyes and the sole of the feet. Seven days after transplantation blood samples were collected for determination of serum IL-2, IgG and IgM concentration. The right eye-balls and left feet were obtained for microscopic examination. 2.A STUDY ON THE VALUE OF LOCAL HYPOTHERMIA THERAPY IN THE TREATMENT OF NEUROSURGICAL SEVERE PATIENTS

Objective To study the effects of hypothermia therapy on inflammation level and safety of patients with severe nervous injury.Methods A total of 82 patients with severe nerve injury were divided into study group and control group by random digits table, 41 patients in each group.The patients in these two groups had no statistical dis-parities in inflammatory agent and intracranial pressure (p>0.05).The patients in the control group were treated by routine therapy and those in the study group were treated by cephalic hypothermia therapy additionally.The two groups were compared in terms of the level of TNFα, IL-6 and IL-8 in the cerebrospinal fluid, prognosis and safety.Re-sults Before treatment there was no significant difference in inflammation level between the two groups (p>0.05). After treatment, the TNFα, IL-6 and IL-8 levels in the study group were significantly lower than those in control group (p<0.01).Before treatment there was no significant difference in ICP between two groups (p>0.05).How-ever, after treatment there was a significant difference in ICP between the two groups in all time points (p<0.01). The GOS score in the study group was significantly lower than that in the control group.There was no statistical dis-parities in complications between the two groups (p>0.0).Conclusion The hypothermia therapy has a great clini-cal efficacy and safety on severe never injury patients, which makes it worth clinical application.
3.Late results of bioprosthesis replacement

Objective: To evaluate the long-term result after bioprosthesis replacement, and analyse the risk factors of the late deaths. Methods: From September 1984 to March 1988, 90 patients underwent bioprosthesis replacement in our hospital. There were 39 males (43%) and 51 females (57%) with an average age of 36.1 years. 78 patients had rheumatic disease, 8 degenerative disease, 3 congenital mitral valve anomaly and 1 Ebstein's anomaly. The preoperatively heart function (NYHA) was grade II in 15, grade III in 54, and grade IV in 21. C/T was 0.51~0.90 (average 0.65?0.10). Results: The operative mortality rate was 7。8%. All survived patients were followed up. The mean follow-up time was 7.8 years (range from 3 months to 15.7 years. 35 patients died during the follow-up period. The causes of death cardiac related deaths in 33 patients (88.6%), noncardiac related death in 3 (8.6%), and unknown in 1 (2.8%). Reoperation was performed in 20patients. At 1, 5 and 10 years after operation, the actuarial survival rate of patients were 92.7%, 80.7% and 57.8%, respectively. Multiple regression analysis showed that preoperatively cardiac function, C/T ratio and EF were risk factors for late deaths. Conclusion: The long-term results with bioprosthesis replacement appear satisfactory.
4.Apoptosis of pancreatic carcinoma mediated by a retroviral vector containing antisense human telomerase gene

Objective To study the apoptosis of pancreatic cancer cells induced by a retroviral vector containing antisense human telomerase RNA gene. Methods Sense and antisense telomerase RNA genes were transfected into Can-pan-2 cell line by electroporation,and selected with G418 to get stable transfection and retrovirus yielding cell lines. We transfected Can-pan-2 cell with concentrated retroviral supernatant,and selected with G418 to get stable transfection cell lines,The expression of the aim gene in Can-pan-2 cell was confirmed through PCR,while TRAP-PCR-ELISA was used to determine the telomerase activity,and immunofluorescence was adopted to determine reproductive activity of the infected cells,flow cytometry(FCM)was used to detect cell apoptosis.ResultsThe titer of sense and antisense recombinant retrovirus reached 0.95?106 CFU/mL and 1.1?106 CFU/mL respectively,PCR demonstrated that the telomerase RNA genes were successfully intergrated into the target cells’ genome,The cell growth and telomerase activity of Can-pan-2 were significantly inhibited and apoptosis occurred after antisense virus infection. Conclusion Antisense human telomerase RNA may inhibite telomerase activity of pancreatic cancer cells and promote apoptosis of the cells.
5.Relative analysis of prognostic factors in primary anorectal malignant melanoma

Objective To investigate the relative analysis of prognostic factors in primary anorectal malignant melanoma (AMM). Method Twenty-seven patients with AMM were treated, Log-rank and COX analysis were used to find out the relationship of ten factors to survival. Results Log-rank analysis found five of ten factors affecting survival, they were age of patient,diameter of tumor, infiltrated depth,pathological staging and metastasis (P<0.05 or<0.01), while COX analysis only found the diameter of tumor and infiltrated depth had close relationship to survival (P<0.05 or<0.01). Conclusion The diameter of tumor and infiltrated depth are the main factors affecting the survival of AMM, and this indicates the importance of early diagnosis and treatment.
6.Exploration of the Teaching Rounds Mode for Tumor Therapeutic Radiology

Objective Through comparing the traditional teaching rounds model with innovation model, explore appropriate teaching rounds model for Radiotherapy. Methods From September 2009 to June 2010, 82 students of Kunming Medical University were randomly divided into two groups, 41 students in test group with innovation teaching rounds model, and 41 students in control group with traditional model. The results of the two groups were compared. Results The differences of survey and test scores were statistically significant ( <0.01) between the test and control groups. Conclusion Test group could effectively improve the clinical thinking,practice operating capacity and teaching quality, mobilize the initiative and enthusiasm for learning.The teaching rounds model needs further exploration,improvement and evaluation.
7.Study on teaching reformation necessity of medical mycology for undergraduates of medical laboratory specialty

Objective To study the education of medical mycology for undergraduates of medical laboratory specialty and provide a basis for teaching reformation.Method Setting of mycology related courses of medical mycology for undergraduates in 5 medical schools and 85 inspection and technical personnel's detection of fungi in 81 hospitals were investigated through consultation and questionnaire survey.Results More than 140 class hours for medical mycology were arranged in 5 schools,but as to medical mycology,22 class hours in 1 school and less than 10 class hours in 4 schools,the minimum class hours were 5.Although various numbers of Candida and filamentous fungi could be isolated in hospitals investigated,more than half laboratory workers could not identify penicillium,thermally dimorphic fungi,Zygomycetes and Dematiaceous fungi.Conclusion Education on medical mycology for medical laboratory specialty undergraduates is insufficient and the corresponding teaching lacks such content as medically important pathogenic fungi detection methods and identification characteristics.The hospital technical personnel's fungal identification ability cannot meet the situation of increasing fungal infection involved in clinical medicine,so it is necessary to carry out teaching reformation of medical mycology for undergraduates in laboratory medicine,including adding class hours,increasing course contents and so on.
8.Effects of miRNA-29a on monocrotaline-induced pulmonary hypertension in rats

AIM: To detect the expression of microRNA (miRNA)-29a in pulmonary arteries of monocrotaline (MCT)-induced pulmonary hypertensive rats, and to investigate the effects of miRNA-29a on pulmonary hypertension.METHODS: MCT-induced pulmonary hypertensive model was established in Wistar rats.The expression of miRNA-29a in the lung tissue was determined by qPCR.miRNA-29a was overexpressed in the pulmonary hypertension rats by tail vein injection of miRNA-29a-mimic.Pulmonary artery pressure (PAP) and systemic arterial pressure (SAP) were measured.The morphological changes of the pulmonary arteries were observed by HE staining.The protein levels of p-Akt and p-eNOS were detected by Western blot.RESULTS: The mRNA expression of miRNA-29a was significantly decreased in the pulmonary arteries of MCT-induced pulmonary hypertensive rats.Furthermore, after overexpression of miRNA-29a, PAP was remarkably reduced, while SAP remained unchanged.In addition, the increased thickness of tunica media, the remodeling of pulmonary arteries and the decreased protein levels of p-Akt and p-eNOS in the pulmonary hypertensive rats were dramati-cally changed after miRNA-29a overexpression.CONCLUSION: Overexpression of miRNA-29a ameliorates pulmonary hypertension in rats.These effects may be associated with the activation of PI3K/Akt-eNOS signaling pathway.
9.Inclusion of ginsenoside Rg_3 with hydroxypropyl-?-cyclodextrin

Objective To investigate the inclusion of ginsenoside Rg_3 with hydroxylpropyl-?-cyclodextrin (HP-?-CD) in aqueous solutions, inclusion molar ratio of host and guest molecules, and change of thermodynamic parameters during the inclusion process. Methods The measurements of the inclusion mechanism, inclusion molar of the host and guest molecules, and change of thermodynamic parameters were carried out by the following methods: phase solubility, IR, NMR, and TLC methods, respectively. Results The results indicated that the inclusion compound of ginsenoside Rg_3-HP-?-CD was formed. The content analysis of the inclusion compound showed that the inclusion ratio of ginsenoside Rg_3-HP-?-CD was 1∶1. 10.Effects of AgNO_3 on caducity-resistant from tissue culture of Dedrobium officinale

Objective To solve the caducity question of Dendrobium officinale during the tissue-culture process. Methods Added the different concentration of 0.1 and 2 mg/L AgNO3 into the PLB proliferation medium, buds proliferation medium, and rooting medium separately. The quantity of the ethylene production was to be determined and the situation of the tube seedling growth was to be observed. ResultsAgNO3 can suppress the production of the ethylene, enhance the multiplication speed of PLB and split-up efficiency the clump of bud, and obviously promote the seedling′s growth. It showed that the survival ratio of transplant could enhance as many as two times and display the merit of caducity-resistant. ConclusionAgNO3(1 mg/L) can be taken as the caducity-resistant reagent of D. officinale during the tissue-culture processing.
