Objective To explore the imaging features of bone tumor of fibrous tissue origin. Methods 31 patients with bone tumor of fibrous tissue or igin confirmed by pathology were analyzed. All patients were examined by X-ray, of them, 6 by CT in the meantime. Results Non-ossifying fibroma(NOF n=14)occured commonly i n long bone, the imaging findings presented sacculated and expanded bone destruct ion which was encircled by sclerotic borders, bony septum could be observed iusid e the lesion.Ossifying fibroma(OF n=10)occured more in maxillofacial bone, there were circle or elliptical and expansive bone change, and most of them associated with local bone malformation.Depending on the ossified degree, the density of les ion, could be hyperdense,ground-glass opacity, cyst or mixed density, defined borde r,partly associated with sclerotic borders.Desmoplastic fibroma of bone(DF n=1) presented multiple patchy bony erosion at lateral of lower part of femur, and ext ending to articalar facies,ill-defined border, and soft tissue slightly swelling. Fibrosarcoma of bone(FS n=6) occured commonly in long bone, presened bitted-like or greater patchy osseous absorption with soft fissue mass, rarely periosteal rea ction and calcification.58.08% patients(18/31) were accurately diagnosed. The nat ure of 6 patients was undefined.Misdiagnosis was made in 7.Conclusion Most of NOF and OF are of specific imaging fin dings and can be accurately diagnosed before cperation.DF is of slight symptoms with invaded growth and is easily misdiagnosed. In the imaging features of FS are apparently different with various type and histological differentiation,it must be differentiated from other diseases.

Objective:To obtain antibodies against amylin from a 'naive' human Fab fragment antibody phage diasplay library and to analyze the specificity of antigen binding activity. Methods:Panning and screening Fab antibody from the antibody library, the positive clones with well reactivity to amylin were selected after five times selection of 'adsorption-elution-enrichment'.Then the plasmid DNA which was extracted from the clones, was digested with Spe Ⅰ and Nhe Ⅰ to delete gⅢ (about 660 bp).The digested 47 000 bp DNA which was purified after separation of bands from agarose gel was ligated with T4-DNA ligase. The constructed expressing phagemids were transformed to the BL21(DE3)pLysS,soluble Fab was expressed in it by the induction of IPTG and its characteristics and specificity were determined by ELISA and Western blot. Results:Soluble Fab antibodies were expressed in E.coli.According with molecular weight of IgG Fab,protein band of about 47 kD was shown by SDS-PAGE.Western blot using the goat anti human IgG-HRP showed their binding activities.ELISA showed their specificity with amylin antigens and they did not react with bovine serum albumin. Conclusion:The high level expression and identification of the soluble human anti- amylin Fab fragment antibodies has been obtained successfully, which lays a solid foundation for further researching about the biological and pathological activities of amylin.

Chinese Journal of Immunology 2001;0(07):-
Objective:To construct a human Fab fragment phage display library and provide a platform for human antibody preparation. Methods:Peripheral blood lymphocytes were collected from healthy donor. The heavy chain Fd fragment and light chain of human immunoglobulin's genes were amplified by RT-PCR,and then cloned into phagemid pComb3XSS to generate human phage antibody library.Cutting with endonucleases such as SacⅠ,XbaⅠ,XhoⅠand SpeⅠto identify the insertion of the light chain or heavy chain Fd genes.IL-2 and digoxin as the antigen was used to scan the phage antibody library. Results:A phage antibody library of Fab had 8.4?107 members and it's recombinant rate was 70%.Through DNA sequencing of one positive clone,it was showed that its heavy chain belonged to IgG subvariety and its light chain to ? family. Conclusion:The success of constructing a nave human phage antibody library proves the useful of phage display system in human antibody preparation, and it can be used to select, purify and express of amylin Fab antibody.

Objective:
This study’s objective is to assess the efficacy and safety of Pulsed Magnetic Therapy System (PMTS) in improving insomnia disorder.
Methods:
Participants with insomnia disorder were randomly assigned to receive either PMTS or sham treatment for four weeks (n= 153; PMTS: 76, sham: 77). Primary outcomes are the Insomnia Severity Index (ISI) scores at week 0 (baseline), 1, 2, 3, 4 (treatment), and 5 (follow-up). Secondary outcomes are the Pittsburgh Sleep Quality Index at baseline and week 4, and weekly sleep diary-derived values for sleep latency, sleep efficiency, real sleep time, waking after sleep onset, and sleep duration.
Results:
The ISI scores of the PMTS group and the sham group were 7.13±0.50, 11.07±0.51 at week 4, respectively. There was a significant group×time interaction for ISI (F3.214, 485.271=24.25, p<0.001, ηp 2=0.138). Only the PMTS group experienced continuous improvement throughout the study; in contrast, the sham group only experienced a modest improvement after the first week of therapy. At the end of the treatment and one week after it, the response of the PMTS group were 69.7% (95% confidence interval [CI]: 58.6%–79.0%), 75.0% (95% CI: 64.1%–83.4%), respectively, which were higher than the response of the sham group (p<0.001). For each of the secondary outcomes, similar group×time interactions were discovered. The effects of the treatment persisted for at least a week.
Conclusion
PMTS is safe and effective in improving insomnia disorders.

The anterior cruciate ligament (ACL) injury is a common sports injury that has a significant impact on knee function and patients′ mobility. With the popularity of national fitness campaign in China, the incidence of ACL injury is increasing year by year. Currently, there still lacks clinical standards or guidelines on how to choose appropriate treatment methods, surgical plans and rehabilitation protocols for ACL injury. In order to timely reflect the new treatment concept of ACL injury, standardize its diagnosis and treatment and improve the curative effect, the Sports Medicine Society of Chinese Research Hospital Association and the Editorial Board of Chinese Journal of Trauma organized domestic orthopedic and sports medicine experts to formulate the "clinical evidence-based guideline for the diagnosis and treatment of anterior cruciate ligament injury (2022 version)" based on the level of evidence-based medicine and in compliance with the principle of scientificity, practicability and advancement. The present guideline includes 12 recommendations for the diagnosis, treatment and rehabilitation of ACL injury in order to provide guidance and assistance for the clinical diagnosis and treatment of ACL injury in China.
