1.Effects of large dose Methylprednisolone on level of MCP-1 expression and macrophages infiltration in early spinal cord injured rats

Objective To explore the effects of Methylprednisolone (MP) on level of monocyte chemoattractant protein 1 (MCP-1) expression and macrophages infiltration in early spinal cord injured (SCI) rats.Methods Animal SCI model was established by modified Nystrm method. 72 rats were randomly divided into three groups: SCI group, normal saline control group and MP treatment group. Each group,divided into three subgroups, were sacrificed at 1 d, 3 d, 7 d postinjury. Spinal cord tissues were collected and sections were made to do following examinations: (1)morphological observation by hematoxylin and eosin stain;(2)the expression and distribution of local MCP-1 positive cells, and the infiltration of macrophages by immunohistochemical method. 2.Posterior approach to radical resection for thoracic tumor and spine stabilization and reconstruc-tion in one stage

Objective To discuss total vertebrectomy and spine stability and reconstruction by pos-terior ap proach for thoracic vertebra tumor and pedicle screw system fixation and intervertebral fusion.Methods Eighteen patients of 3males and15females of thoracic total vertebral tumor were operated in this group.The age of the group were from14to58years old,with the average of 23years.There were4aneurysmal bone cysts,2hemangiomas,2osteoblastomas,1neurilemomas,5giant cell tumor,1solitary myeloma and3metastatic thoracic vertebra tumors.The locations of the tumors was T 4 in1case,T 5 in1case,T 6 in2cases,T 8 in4cases,T 9 in3cases,T 10 in4cases,T 11 in2cases and T 12 in1case respectively.Pre-operative Frankel classification was grade A in6cases,B in7cases,C in3cases and E in2cases.Cir-cumferential decompression was performed with total spondylectomy through one-stage posterior ap proach.Anterior verte bral reconstruction was provided by autograft(ribs or ilium)bone or metallic cage;Posterior reconstruction was achieved by pedi cle screws system of CD or TSRH,Scofix fixation and auto graft fusion.Results The mean follow-up was8months,ranging from3months to24months.Twelve of 16cases with neu ro logical dysfunction were recovered completely,1case recovered from Frankel grade A to C,2cases from grade A to D,1case from grade B to D.The patient with neurilemmoma recurred and became ma lig nant after20months of operation and died4months later.One case of metastatic tumor of lung cancer sur vived6months after opera tion.One patient with giant cell tumor recurred.The other patients were still alive in the latest follow-up.There were no instruments breakage and spinal instability during the follow-up.Conclusion If the thoracic total vertebral tumor compress spinal cord se riously,the patients should be indi cat-ed for surgical treatment constructively.Total vertebrectomy followed by one-stage posterior approach re con-struction could relieve the com pression caused by tumor and reconstruct the stabilization of spine effectively.
3.Clinical study of Bryan prosthetic disc in the treatment of ossification of posterior longitudinal ligament of cervical spine

[Objective] To observe the clinical effect of Bryan cervical prosthetic disc in treating ossification of posterior longitudinal ligament(OPLL)of cervical spine.[Method]From October 2005 to June 2007,15 localized OPLL patients were treated surgically by cervical arthroplasty using Bryan disc prosthesis.JOA score and radiological study were performed before and after operation.The range of movement(ROM)of replaced level were investigated postoperatively.[Result]Neurological deficit was alleviated in all patients.All patients were followed up at least 6 months,JOA score increased from an average of 8.5 to 15.8.Replaced segment restored the normal ROM in flexion and extension.[Conclusion]Cervical arthroplasty using Bryan disc prosthesis can achieve good clinical outcome in treating localized OPLL of cervical spine;and the normal ROM can be retained in the replaced level.
4.The technique of double-tunnel with double-bundle for reconstruction of the posterior cruciate ligament with quadriceps tendon-patellar bone under arthroscopy

0.05). Conclusion The procedure of double-tunnel with double-bundle for reconstruction of posterior cruciate ligament (PCL) under arthroscopy can better improve the knee stability at the different flexion degree, and the QT-B is an optimal graft for the double-tunnel reconstruction.
5.Experimental study of motor and somatosensory evoked potentials on spinal cord injury

[Objective]To evaluate the validity and feasibility of somatosensory evoked potentials(SEP) and transcranial magnetic stimulation motor evoked potential(TMS-MEP) in early decompression of spinal cord injury(SCI). [Method]Thirty-two rabbits fell into four groups randomly. The static load model were used to simulate human SCI. According to the compression time, the animals were divided into 4 groups (control,5min, 15min,30min). In this study, somatosensory evoked potentials (SEP) and transcranial magnetic stimulation motor evoked potential (TMS-MEP) were used to assess neurologic function. The EPs were recorded at 5min, 15min, 30min, 60min, 6 hours, 24 hours, 3 days, 7 days post-decompression and before spinal cord injury in injury groups. Latency and amplitude were measured for each wave. Tarlov behaviour score was performed before surgery and after surgery at 1,3,7 days. Histologic studies were performed on part of animals. [Result]Spinal cord compression resulted in a gradual increment in the peak latency and significant decrement in the peak amplitude. After decompression, the recovery of the latency was more obvious than the amplitude in all injury groups, EPs recovered earlier than Tarlov score. [Conclusion]EPs may evaluate the neurologic function of SCI models sensitively and impartially. Dcompression early may promote SCI recovery.
6.Histomorphological observation of PerioGlas implanted in muscle and osseous defects

0.05). In bone defects PeroGlas granales bond to bone and new bone formation were observed 4 weeks after operation. The bone defects were repa ired by new bone 12 weeks after PerioGlas implantation. Conclusion: PerioGlas may induce new bone formation in osseous defects.
7.Separating cultivating and osteogeneric identifying of bone marrow stromal cells in rabbit

Objective To search a kind of ideal seeded cell of bone tissue engineering. Methods Bone marrow tissue was fetched from rabbit,got bone marrow stromal cells by the method of density centrifuge,observed the conformation,growing feature and osteogeneric capability of the cells during culture in vitro by light microscope,transmission electron microscope and measure of osteogeneric capability. Results Most of the cultivated bone marrow stromal cells were triangle-like or shuttle-like cells. They growed and breeded rapidly,and had osteogeneric capability. They were easy to differentiate to osteoblasts. Conclusion The cultivated bone marrow stromal cells are matured ones,and could become the more ideal seeded cells for bone tissue engineering.
8.An experimental study of the cultivation, identification and differentiation induction of bone marrow stromal stem cells into osteoblasts

Objective To study the model of separation and culture and the growth characteristics and osteogenic capability of bone marrow stromal stem cells (BMSCs). Methods BMSCs were isolated from adult rats using gradient centrifugation and anchoring culture. The passage cells were induced to differentiate into osteoblasts by means of an differentiation induction medium containing dexamethasone,?-glycerophophate and Vitamin C. Alkaline phosphatase (ALP) activity and the ability to form mineralized nodules were examined. Results Colonies of stromal stem cells were formed in the primary culture and contacted one another at the 14th day. In the passage culture, cells became bigger than those in the primary culture and could be subcultured one generation in 5~7 days. After the culture in the differentiation induction medium, the ALP activity was enhanced significantly and the mineralized nodules were formed. Conclusion BMSCs can be obtained easily and have strong proliferation ability and osteogenic capacity. This study suggests that BMSCs can become seeding-cells in bone tissue engineering.
9.Effects of Xuebijing Injection on Blood Coagulation Function and Prognosis of Sepsis Patients

OBJECTIVE:To investigate the effect of Xuebijing injection on coagulation function and prognosis in patients with sepsis. METHODS:50 patients with sepsis were randomly divided into control group and observation group according to the hospi-talization sequence,with 25 cases in each group. Control group received conventional treatment,and observation group was addi-tionally given Xuebijing injection(50 ml added into 5% Glucose solution or 0.9% Sodium chloride solution 250 ml intravenously, bid) on the basis of conventional treatment. Treatment course lasted for 7 d. The coagulation function,APACHE Ⅱscore after 7 days of treatment and 28 d mortality were compared between 2 groups. RESULTS:After 7 d treatment,the coagulation indexes were improved significantly in 2 groups compared with before treatment,and the APACHEⅡscores were decreased significantly in both groups;the observation group were significantly better than the control group,with statistical significance (P<0.05). After followed up for 28 d,the mortality rate of 2 groups were 16% and 28%,there was no statistical significance (P>0.05). CON-CLUSIONS:Xuebijing injection can inhibit blood clotting activation,correct the coagulation disorders,and improve the sepsis se-verity.
10.Dracorhodin perchlorate inhibit high glucose induce serum and glucocorticoid induced protein kinase 1 and fibronectin expression in human mesangial cells.

OBJECTIVETo investigate the effect of dracorhodin perchlorate (DP) on inhibiting high glucose-induced serum and glucocorticoid induced protein kinase 1 (SGK1) and fibronectin (FN) expression in human mesangial cells (HMC), and its mechanism of prevention and treatment on renal fibrosis in diabetic nephropathy (DN) .
METHODThe HMC were divided into normal glucose group (NG group, 5.5 mmol x L(-1) D-glucose), normal glucose +low DP group (NG + LDP group, 5.5 mmol x L(-1) D-glucose +7.5 micromol x L(-1) DP), normal glucose +high DP group (NG + HDP group, 5.5 mmol x L(-1) D-glucose + 15 micromol x L(-1) DP), high glucose group (HG group,25 mmol x L(-1) D-glucose), high glucose +low DP group (HG + LDP group, 25 mmol x L(-1) D-glucose + 7.5 micromol x L(-1) DP)and high glucose +high DP group (HG +HDP group, 25 mmol x L(-1) D-glucose + 15 micromol x L(-1) DP). Each group was examined at 24 hours. The levels of SGK1 and FN mRNA was detected by real-time fluorescence quantitative PCR,and the expression of SGK1 and FN protein was detected by Western blot or indirect immunofluorescence.
RESULTA basal level of SGK1 and FN in HMC were detected in NG group, and the level of SGK1 and FN mRNA and protein were not evidently different compared to that of NG group adding 7.5 micromol x L(-1) DP for 24 hours. On the other hand, the levels of SGK1 and FN mRNA and protein were obviously decreased by adding 15 micromol x L(-1) DP for 24 hours. Compared to NG group, the levels of SGK1 and FN mRNA and protein were increased in HG group after stimulating for 24 hours (P < 0.01). Compared to HG group, the level of SGK1 and FN mRNA and protein were evidently reduced in HG + LDP and HG + HDP groups by adding 7.5 micromol x L(-1) DP and 15 micromol x L(-1) DP for 24 hours (P < 0.01).
CONCLUSIONDracorhodin perchlorate can inhibit high glucose-induced serum and glucocorticoid induced protein kinase 1 (SGK1) and fibronectin(FN) expression in human mesangial cells, and this may be part of the mechanism of preventing and treating renal fibrosis of DN.
