1.Association of peripheral CD4+ CD25+ CD127low/-regulatory T cells and liver pathology in hepatitis B virus-infected individuals

Objective To explore the expression level of CD4+ CD25+ CD127low-regulatory T cells (Treg) in the peripheral blood of hepatitis B virus (HBV)-infected individuals,and its association with liver pathology.Methods Thirty-five HBV-infected individuals and 20 healthy controls were enrolled.The frequencies of peripheral blood CD4+ CD25+ CD127low/-Treg were detected by flow cytometry.HBV-related serological tests as well as ultrasound-guided liver biopsies were performed in HBV-infected individuals.The two groups were compared by t test.Spearman and Pearson correlation analyses were performed.Results The alanine aminotransferase (ALT) level of HBV-infected individuals was (34.57 ± 15.36) U/L,with 25 subjects of ALT<40 U/L.Twenty subjects were hepatitis B e antigen (HBeAg) positive,and 15 were HBeAg negative.The frequency of the peripheral blood CD4+ CD25+ CD127low/-Treg in HBV-infected individuals was (5.59 ±1.50)%,which was significantly higher than that in healthy controls [(4.85±0.95) % ; t=2.191,P=0.033].The level of peripheral blood CD4+ CD25-CD127low/-Treg was positively correlated with HBV viral load (r=0.495,P=0.003).Thirty-one subjects had liver inflammatory score ≤2,with a peripheral blood CD4+ CD25+ CD127low/-Treg level of (5.80 ± 1.44) %,which was significantly higher than that of the rest 4 subjects who had liver inflammatory score >2 [(4.00±0.77)%; t=2.425,P=0.021].Twenty-four subjects had liver fibrosis score <2,with a peripheral blood CD4+ CD25+ CD127low/-Treg level of (6.00±1.44) %,which was significantly higher than that of the rest 11 subjects who had liver fibrosis score ≥ 2 [(4.70 ± 1.24)%; t =2.559,P=0.015].The expression of CD4 + CD25 +CD127low/-Treg in peripheral blood was inversely correlated with liver inflammation (r=-0.539,P=0.001) and fibrosis (r =-0.488,P =0.003) in HBV-infected individuals.Conclusions CD4+ CD25+ CD127low/-Treg is associated with chronic condition and liver damage of hepatitis B.It may play an important role in occurrence,development and prognosis of the disease.
2.A PRELIMINARY REPORT ON THE MITOTIC CHROMOSOMES OF THE MOUSE, MUS MUSCULUS

The present paper is a morphological description of the chromosomes of the mouse, Mus musculus, based on the observations of the metaphase chromosomes of both bone marrow cells and spermatogonial cells. Chromosome morphology is found to be essentially the same as those described by the previous investigators. The systematic measurements of the chromosomes have also been presented.

4. Alleviation of curcumin analog WZ35 on type 1 diabetes-induced renal injury via inhibiting inflammatory response

Objective: To study the alleviation of curcumin analog WZ35 on type 1 diabetic nephropathy (DN). Methods: Mice were treated with a single ip injection of streptozocin (STZ) to induce type 1 diabetes, while the control animals received the same volume of citrate buffer. The curcumin and its analog WZ35 (20 mg/kg) were ig administration for 9 weeks after the diabete obtained. The body weight and blood glucose were monitored every 7 d. Biochemistry analyzer was used to analyze the creatinine (Cr) and urea nitrogen (BUN) levels in serum. The histopathology of kidney tissue was detected by HE staining. RT-qPCR assay was used to evaluate the gene levels of inflammatory cytokines and chemokines. CD68 staining was used to evaluate the macrophages infiltration in kidney tissue. Results: Compared with negative control, the mice in diabetic group showed the reduced body weight, increased blood glucose, high Cr and BUN levels in serum, renal pathological damage, and increased inflammatory gene, and macrophages infiltration in kidney. While the administration with curcumin and its analog WZ35 could obviously attenuate the DN through inhibiting Cr and BUN in serum while had no effect on the body weight and blood glucose. Compared with curcumin, WZ35 could inhibit the gene expression of inflammatory cytokines and chemokines as well as infiltration of macrophages. Conclusion: The curcumin analog could attenuate DN through inhibiting inflammatory response.
5.Localization of intraocular foreign bodies by the combination of CT ocular axial scan and meridian plane reconstruction

AIM: To explore a new compute tomography (CT) for the localization of intraocular foreign bodies (IOFBs).METHODS: After CT ocular horizontal and ocular axial scan,the foreign bodies on the eyeball wall or retinal surface in 26eyes were localized by means of the combination of CT ocular axial scan and meridian plane reconstruction (new method),ocular horizontal scan and ocular axial scan (conventional method Ⅰ) as well as ocular horizontal scan right angle coordination (conventional method Ⅱ) separately. According to the criteria of indirect ophthalmoscope localization and direct observation during operation, the relative accuracies of corresponding points of the foreign bodies on sclera surface along meridian and latitude were measured.RESULTS: In the 26 cases, the mean relative accuracies of corresponding points of foreign bodies on sclera surface along meridian and latitude were 1.53mm、1.64mm (new method), 1.37mm、1.64mm (conventional method Ⅰ) and 2.02mm、2.55mm (conventional method Ⅱ) respectively.There was no statistical difference between the new method and the conventional method Ⅰ, whereas there was statistical difference between the new methods and the conventional method Ⅱ (along meridian: P<0.05; along latitude:P<0.01).CONCLUSION: Compared with the conventional methods,the new method is simpler, more visible and more potential in the clinic.
6.Hypertonic saline resuscitation contributes to early accumulation of circulating myeloid-derived suppressor cells in a rat model of hemorrhagic shock

Background Hemorrhagic shock is usually associated with complicated immune and inflammatory responses,which are sometimes crucial for the prognosis.As regulators of the immune and inflammatory system; proliferation,migration,distribution and activation of myeloid-derived suppressor cells (MDSCs) are intimately linked to the inflammation cascade.Methods In a model of severe hemorrhagic shock,thirty-five rats were randomly divided into control,sham,normal saline resuscitation (NS),hypertonic saline resuscitation (HTS),and hydroxyethyl starch resuscitation (HES),with seven in each group.M DSCs were analyzed by flow cytometric staining of CD11b/c+Gra+ in peripheral blood mononuclear cells (PBMC),spleen cell suspensions,and bone marrow nucleated cells (BMNC).Simultaneously,the expressions of arginase-1 (ARG-1) and inducible nitric oxide synthase (iNOS) mRNA in MDSCs were evaluated by quantitative reverse transcription-polymerase chain reaction (qRT-PCR).Results In the early stage after hemorrhagic shock,fluid resuscitation and emergency treatment,the MDSCs in the PBMC of NS,HTS and HES groups markedly increased,and MDSCs in BMNC of these groups decreased accordingly,significantly different to the control group.In hemorrhagic shock rats infused with HTS at the early resuscitation stage,MDSCs in PBMC increased about 2 and 4 folds,and MDSCs in BMNC decreased about 1.3 and 1.6 folds,as compared to the sham group respectively,with statistically significant difference.Furthermore,compared to the NS and HES groups,the MDSCs in PBMC of HTS group increased 1.6 and 1.8 folds with statistically significant differences; the MDSCs decrease in BMNC was not significant.However,there was no statistically significant difference in MDSCs of spleen among the five groups.In addition,compared to the control,sham,NS and HES groups,the ARG-1 and iNOS mRNA of MDSCs in PBMC,spleen and BMNC in the HTS group had the highest level of expression,but no statistically significant differences were noted.Conclusions In this model of rat with severe and controlled hemorrhagic shock,small volume resuscitation with HTS contributes to dramatically early migration and redistribution of MDSCs from bone marrow to peripheral circulation,compared to resuscitation with NS or HES.
7.MESH diagrams of Chinese in Beijing and its preliminary application in practice.

OBJECTIVETo establish the normal MESH diagrams of Chinese in Beijing, and to build a computerized MESH analysis system for orthodontic practice.
METHODSTwenty-eight subjects with normal occlusion were selected in Beijing and their lateral cephalograms were taken at the age of thirteen and eighteen, respectively. Individual MESH diagrams were then established for each subject mainly according to Moorrees' method from the cephalograms orientated in estimated natural head position. Male and female normal MESH diagrams were created. A computerized MESH analysis system was also developed.
RESULTSThe normal MESH diagrams of Chinese in Beijing, thirteen and eighteen years old respective, were established. The computerized MESH analysis system was constructed and used in orthodontic patients.
CONCLUSIONSMESH analysis is a proportional analysis method. It can show the results directly, succinctly and holistically. It is also a favorable complement and amendment to the commonly used angle and linear X-ray analysis methods.

8.Effect of parallel axipetal centesis in vascular access setting-up

Objective To compare the parallel axipetal centesis with traditional centisis in hemodialysis efficiency, and discuss the practicability and effectiveness.Methods 30 patients with maintenance hemodialysis were involved. Parallel axipetal centesis and traditional centesis were applied by means of self control and hemodialysis efficiency was compared.Results Effective blood flow could be achieved with both centesis methods. There was no significant difference of the rate of descend in blood urea, creatine, uric acid and phosphorus and Kt/V between the two groups(P>.05).Conclusions Parallel axipetal centesis has numerous advantages compared to traditional centesis, and the hemodialysis efficiency remains high. Thus the parallel axipetal centesis is an ideal new-style centesis for hemodialysis.
9.The effects of TENS on metatarsus plantarflexion and inversion in stroke patients

Objective To evaluate the effects of TENS on metatarsus plantar flexion and inversion in stroke patients,and to explore its mechanism.Methods Thirty-two stroke patients with gastrocnemius spasticity were randomly divided into a control group (n=16) and a TENS group (n=16).All patients were treated with foot sup- ports,neurodevelopmental and manipulation therapies.In addition,the TENS group received TENS on the anterior tibialis,peroneus longus and brevis muscles.All patients were assessed in terms of their Chinese stroke scale(CSS) and H reflex scores before and after therapy.Results Compared with those in the control group,the H reflex scores in the TENS group were obviously decreased,while H reflex latency was prolonged and H/M was reduced. Gait in the TENS group was evidently improved.Conclusion TENS is an effective therapy to decrease gastrocnemi- us spasticity and to improve the gait of stroke patients.
