1.Diagnostic Value of Combined Examination of Copeptin and High Sensitivity Cardiac Troponin T in Patients at Early Stage of Acute Myocardial Infarction

Objective: To investigate the diagnostic value of combined examination of copeptin and high sensitivity cardiac troponin T (hs-cTnT) in patients at the early stage of acute myocardial infarction (AMI) . Methods: A total of 272 patients were enrolled in this study, all of them suffered from chest pain and admitted within 4 hours. The patients were divided into 4 groups according to coronary artery angiography (CAG) results. Control group, the patients with normal CAG,n=64, UAP group (unstable angina pectoris),n=50, STEMI group,n=82, NSTEMI group,n=76. All patients received in-hospital observation, plasma levels of copeptin and hs-cTnT were examined at admission and at 6 hours after the chest pain respectively. Results: Within 4 hours of chest pain, combined examination of copeptin and hs-cTnT had the higher sensitivity for diagnosing AMI than a single detection of hs-cTnT with the cut-off point of hs-cTnT ≤ 14ng/L and Copeptin < 14pmol/L. In NSTEMI group, the AUC (area under curve) for combined examination was 0.97 (95% CI 0.88-0.99), AUC for single hs-cTnT detection was 0.75 (95% CI 0.62-0.87),P<0.05. In STEMI group, the AUC for combined examination was 0.97 (95% CI 0.88-0.99), AUC for single hs-cTnT detection was 0.74 (95% CI 0.60-0.88),P< 0.05. The AUC for combined examination of copeptin and hs-cTnT in diagnosing early AMI was 0.912 (95% CI 0.812-0.961) which was higher than single detection of hs-cTnT, AUC 0.851 (95% CI 0.713-0.936), Z=2.553,P<0.05. Conclusion: Combined examination of copeptin and hs-cTnT had the higher sensitivity and accuracy for diagnosing the patients at the early stage of AMI, it may help the risk stratiifcation of chest pain which is valuable in clinical practice.
2.ANTI-APOPTOSIS EFFECT OF NADH ON NORMAL LIVER CELL LINES TREATED WITH CYANIDE

This study was to observe the anti apoptosis effect of NADH on a normal liver cell line inflicted with chemical hypoxia. L02 liver cells were treated with 3mmol/L KCN in the presence of different concentrations of NADH (0~600?g/ml) for 0.5, 2, 4 hours. The percentage of apoptotic cells was assayed with flow cytometry(FCM). Another three groups of experiment were done as following. Group Ⅰ was control. Groups Ⅱ and Ⅲ were treated with 3mmol/L KCN in the presence or absence of NADH (400?g/ml) for 2h. The expression of proteins of Bcl XL, Bcl 2 and Bax in the L02 cells were determined with FCM analysis. The results showed that NADH could obviously prevent L02 cells from apoptotic damage at the concentration of 400?g/ml. it could also significantly upregulate the expression of Bcl 2 and Bcl XL proteins and downregulate the expression of Bax proteins after cyanide challenge( P
3.Effects of repeated treatment with different concentrations of chemotherapy drugs on proliferation and chemo-resistance in human lung cancer cell line

Objective:To explore the variation of tumor cell proteins correlated with cell proliferation and chemo-resis-tance after repeated treatment with chemotherapy drugs.Methods:The human lung cancer cell line(LPET-a-2)was repeat-edly treated with chemotherapy drugs:doxorubicin,etoposide,cisplatin and the combination of the3drugs,and each drug was given at2concentrations.The treatment intervals were recorded.ErbB-2,c-myc,MDR1,MRP,LRP,ref-1and NF-?B were tested by flow cytometry.The cells expressing each protein,the mean and total quantity of each protein after each treat-ment were calculated by different drugs at different concentrations.Results:The levels of every protein decreased along with the time of culture.In high-dose group,every item decreased along with the time of treatment.In low-dose group,there was no rule for the item variation with some decreased and some increased.Conclusion:Low-dose anticancer drugs are easier to induce cell proliferation and chemo-resistance than high-dose one,which suggests that adequate chemotherapy should be given in clinical practice.
4.Analysis on influence factors of medication compliance in elderly patients with hypertension

Objective To know the medication compliance in elderly patients with hypertension,to analyze the influence factors of medication compliance,and to propose forward intervention countermeasures.Methods The treatment of hypertension in elderly patients was investigated by questionnaire survey.Medication compliance and influence factors were analyzed.Results 166 cases with poor medication compliance,and the rate was 45.0%; 203 cases with good medication compliance,and the rate was 55.0%.It's showed significantly difference on degree of education between the observation group and control group (x2 =12.071,P=0.002).The proportion of peasant and worker was higher in the observation group than control group (78.9% vs.62.1%,x2 =12.817,P=0.002).The smoker (48.8%) and drinker (59.0%) in observation group more than smoker (27.1%) and drinker (31.0%) in control group(x2=18.481,P<0.001; x2=29.115,P< 0.001).The ratio of payout of-pocket for healthcare was higher in the observation group (21.1%) than control group (9.9%) (x2=9.084,P=0.003).The duration of hypertension was longer in the observation group (8.8±4.6) than control group (6.0±4.1) (t=1.806,P=0.036).The income per month was less in the observation group (1850±150) than control group (2150±200) (t=5.799,P< 0.001).There were regression relationship between medication compliance and degree of education,medical security and duration of hypertension.Conclusions According to the influence factors of medication compliance in elderly hypertensive patients,it's useful to improve medication compliance by enhancing health education and designing a personal treatment.
5.Combination with Histamine and LAK/IL-2 Therapy for the Treatment of Metastatic Melanoma

Objective: To investigate the role and mechanism of histamine in regulation of C57BL/6 mice spleen derived LAK activity in vivo. Methods: The C57BL/6 mice carrying B16 pulmonary metastatic melanoma were treated with LAK/IL 2/histamine therapy or LAK/IL 2 therapy with the aim of evaluating both anti tumor responses in vivo. Results: It was found that the addition of histamine effectively potentiated the anti metastatic effect produced by LAK/IL 2 therapy and induced regression of NK resistant B16 pulmonary metastatic melanoma. Survival period was significantly prolonged in mice receiving LAK/IL 2/histamine as compared with LAK/IL 2 therapy alone. The effect of histamine was completely blocked by H 2 R antagonist ranitidine, and mimicked by dimaprit, a H 2 R agonist. Conclusion:Histamine, via specific activation of H 2 R, may be an important regulator of LAK cells activity; histamine and LAK/IL 2 synergistically induce more potent antitumor efficacy in vivo .
6.Intratumor injection of recombinant attenuated salmonella carrying Mycobacterium tuberculosis heat shock protein 70 and herpes simplex virus thymidine kinase genes to suppress murine melanoma growth.

OBJECTIVETo study the effection of suppression murine melanoma growth by Intratumor injection of recombinant attenuated salmonella carrying heat shock protein 70 and herpes simplex virus thymidine kinase genes.
METHODSPlasmids PCMV-mtHSP70-IRES-TK were electro-transferred into salmonella typhimurium SL7207 to construct recombinant salmonella typhimurium. In vivo, Recombinant bacteria were injected into the mouse melanoma and the antitumor effection was observed. The survival period was recorded and safety analysis for this vaccine in each group.
RESULTSIn vivo, the mtHSP70/HSV-tk recombinant bacteria can suppress tumor growth significantly and extend survival. After recombinant Salmonella, 10(9) CFU/mL, was administered as an intratumoral injection, No diarrhea were observed. During therapy, body weight did not change markedly.
CONCLUSIONResults of the animal experiment suggests intratumor injection of recombinant attenuated salmonella typhimurium containing mtHSP70 and HSV-tk genes, has targeting ability against B16 tumor cell and could significantly inhibit tumor growth .

7.Effects of metoprolol tartrate combined with atorvastatin calcium on lipid metabolism, vascular endothelial function and inflammatory factors in patients with essential hypertension

Objective:To investigate the effect of metoprolol tartrate combined with atorvastatin calcium on lipid metabolism, vascular endothelial function and inflammatory factors in patients with essential hypertension.Methods:From January 2015 to December 2018, 80 patients with essential hypertension admitted to the People's Hospital of Dongyang were divided into observation group (40 cases) and control group (40 cases) according to the random digital table method.On the basis of routine treatment, the control group was treated with atorvastatin calcium, while the observation group was treated with atorvastatin calcium and metoprolol.The course of treatment in both two groups was 6 months.The changes of systolic and diastolic blood pressure, lipid metabolism, endothelial function and inflammatory factors were compared before and after treatment.Results:The total effective rate of the observation group (92.50%, 37/40) was higher than that of the control group (70.00%, 28/40) (χ 2=6.646, P<0.05). The systolic blood pressure [(123.15±7.89) mmHg] and diastolic blood pressure [(75.52±3.89)mmHg] in the observation group were lower than those in the control group [(134.29±10.31)mmHg and (82.31±4.04)mmHg] ( t=5.427, 7.657, all P<0.05). The level of serum NO [(69.74±5.46)nmol/L] in the observation group was higher than that in the control group [(57.87±3.67)nmol/L], while ET-1 [(56.48±5.49)mg/L] in the observation group was lower than that in the control group [(73.24±7.18)mg/L], the differences were statistically significant( t=11.411, 11.728, all P<0.05). The serum levels of CRP [(1.18±0.23)mg/L], TNF-α [(0.63±0.18)μg/L] and IL-6 [(84.35±17.28)ng/L] in the observation group were lower than those in the control group [(1.64±0.45)mg/L, (1.23±0.26)μg/L and (128.43±19.86)ng/L] ( t=5.757, 12.000, 10.590, all P<0.05). Conclusion:Metoprolol tartrate combined with atorvastatin calcium is effective in the treatment of essential hypertension, and can improve lipid metabolism, vascular endothelial function, and reduce inflammation.

Both Patrinia Herba and Patrinia Radix are traditional Chinese herbal medicines. The herbal source and medicinal part of them are confusing in the herbal medicine market of China. To explore the evolution and transition of the herbal source and medicinal part of Patrinia Herba and Patrinia Radix, this paper systematically summarizes the record of the herbal source and medicinal part of them in ancient classics of herbal medicine in China. According to the findings, before Ming Dynasty, Patrinia Herba originated from the radix of the plants with yellow flowers of Patrinia. In Ming and Qing Dynasty, Patrinia Herba originates from the whole plant (including the radix)of the plant with white flowers of Patrinia. In Ming Dynasty, Patrinia Radix, stemming from the radix of the plants with yellow flowers of Patrinia, started to be used as a traditional Chinese herbal medicine, which had the same herbal source with that of Patrinia Herba before Ming Dynasty. Therefore, Patrinia Herba and Patrinia Radix can be seen as the same traditional Chinese herbal medicine, and the genuine of Patrinia Herba should be the radix and the whole herba of P. scabiosaefolia and P. heterophylla.
9.Study on critical quality attributes of Qingjin Huatan Decoction based on serum pharmacochemistry.

Qingjin Huatan Decoction is a classic prescription with the effects of clearing heat, moistening lung, resolving phlegm, and relieving cough. In order to explore the critical quality attributes of Qingjin Huatan Decoction, we identified the blood components of Qingjin Huatan Decoction by ultra-performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-Q-TOF-MS) under the following conditions, chromatographic column: Acquity UPLC BEH C_(18) column(2.1 mm×100 mm, 1.7 μm); mobile phase: 0.1% formic acid acetonitrile(A)-0.1% formic acid in water(B); gradient elution; flow rate: 0.2 mL·min~(-1); column temperature: 30 ℃; injection volume: 5 μL. The electrospray ionization(ESI) source was used to collect data in both positive and negative ion modes under the following conditions, capillary voltage: 3 kV for the positive ion mode and 2 kV for the negative ion mode; ion source temperature: 110 ℃; cone voltage: 30 V; cone gas flow rate: 50 L·h~(-1); nitrogen degassing temperature: 350 ℃; degassing volume flow rate: 800 L·h~(-1); scanning range: m/z 50-2 000. In this experiment, a total of 66 related components of Qingjin Huatan Decoction were identified, including 22 prototype components and 44 metabolites. The results of this study preliminarily revealed the pharmacodynamic material basis of Qingjin Huatan Decoction in vivo, which has provided an experimental basis for the determination of quality markers of Qingjin Huatan Decoction and the development of new drugs.

10.Simultaneous determination of five saponins in Bupleuri Radix by HPLC-DAD dual wavelength method.

Epoxy ether type and isophthalene type saponin are the main saponins of Bupleurum chinense. However,due to the difference of their UV spectrum,there is no quantitative method for simultaneous determination of these two kinds of saponins. In this paper,a dual-wavelength high performance liquid chromatography(HPLC) was developed for simultaneous determination of five saponins in epoxidized ether(saikosaponin a,c,d) and isosorbide type(saikosaponin b1,b2). The mobile phase was eluted with acetonitrile-water(0.1% phosphoric acid) gradient at a column temperature of 30 °C and a flow rate of 1.0 mL·min⁻¹. The detection wavelengths were 208 nm for saikosaponins a,c, and d, and 254 nm for saikosaponins b₁ and b₂. The results showed that the separation of five kinds of saikosaponin was good, with the linear range of 9.70-1 935.00(=0.999 4),8.20-1 380.00(=0.999 3),6.90-1 640.00(=0.999 0),5.25-630.00(=0.999 4), and 5.15-618.00 mg·L⁻¹(=0.999 5), respectively. The average recoveries were 97.70%-100.2% and the RSD was less than 3%(=6). The method is simple,rapid and reproducible. It can be used for the determination of five kinds of saikosaponins in B. chinense.
