MicroRNAs (miRNA), an abundant class of highly conserved small non-coding RNAs, suppress gene expression by binding to the 3' UTR of target mRNAs, which presents an entirely new approach to post-transcriptional regulation of gene expression. MiRNAs have been shown to play key roles in the regulation of diverse biological processes, such as development, inflammation, apoptosis, and tumorigenesis. Several studies have shown the dysregulated miRNAs in various lymphomas including the overexpression of miR-106a and miR-17-92 in T cell lymphoma;upregulated miR-155, miR-221 and miR-21 in diffuse large B-cell iymphoma and follicular cell lymphoma, and these miRNAs associated with subtypes of diffuse large B-cell lymphoma. Overexpression of the miR-17 cluster showed reduced levels of apoptosis, suggesting that the main effect of these miRNAs was to suppress cell death. Although miRNAs can act as both tumour suppressor and oncogenic molecules, the mechanistic explanation for this dysregulation is unclear but maybe linked to the defects in the miRNA biosynthetic machinery. As more evidence accumulates, it is likely that miRNAs will emerge as a new class of epigenetic regulator.

Objective To investigate the cognition and behaviors of sun-protection and the facial photoaging in a Nanjing population, and to analyze the relationships between them. Methods The objects being investigated in Nanjing (n=974) were divided into 10 groups according to age. The ordinary information, knowledge and behavior of sun-protection and Glogau photoageing type of face were studied by questionnaires. The results were analyzed by a logistic regression model to select the related factors to photoageing. Results The risk of skin photoageing increased with age. Most of the 46-65 years old crowds were type Ⅲ photoageing. Most of the objects being investigated had some knowledge and active awareness of sun-protection and could use some ways to protect themselves from sun. However, most people did not use the sunscreen correctly. Those who had higher level awareness and knowledge of sunprotection suffered less risk from skin photoageing. Sunbath without sunscreen for a long time outside activity was a high risk factor of development to skin photoageing. Shade, broad-brimmed hat and sun-protection in autumn were the protective factors. Conclusion Active awareness and correct methods could help prevent skin photoageing. In order to avoid the damages to skin from ultraviolet efficiently, the accurate ways of using sunscreens should be well understood.

Objective To evaluate the expression of epidermal growth factor-like domain 7 (Egfl7) in atherosclerotic plaques and effects of its small interference RNA (siRNA) on angiogenesis gene expression in human endothelial cell line (HUVEC). MethodsEgfl7 expression in atheroscleroticplaquesweredetectedinhumaniliacarteryandmousearteriaeusing immunohistochemistry and immunofluorescence stainings. The siRNA targeting Egfl7 was transfected into HUVEC by lipofectamine with non-transfected cells and unconcerned siRNA as controls. At 0 h,12 h,24 h and 48 h after intervention, the levels of mRNA and protein of Egf17,vascular endothelial growth factor(VEGF),platelet derived growth factor-A (PDGF-A),platelet derived growth factor-B (PDGF-B),vascular cell adhesion molecule(VCAM) and intercellular adhesion molecule (ICAM)were measured by RT-PCR and Western blotting, respectively. ResultsThe expressions of Egfl7 in human iliac artery and mouse arteriae were increased. The expressions of Egfl7 in HUVEC at the levels of mRNA were[(0.14±0.02),(0.09±0.01),(0.02±0.01)]and the levels of protein[(0.71±0.11),(0.39±0.09),(0.07±0.01)]at 12 h,24 h and 48 h after siRNA intervention, respectively, which were decreased as compared with 0 h intervention [(0.31 ±0.05) and (0.93±0.08) ].Other genes such as VEGF,PDGF-A and PDGF-B were reduced or silenced at the levels of protein and mRNA in HUVEC with siRNA longer interventions(all P<0.05).ConclusionsThe expression of Egfl7 in atherosclerotic plaques is increased. The siRNA inhibiting Egfl7 gene expression results in silence of other factors involved in angiogenesis.

Objective To investigate the relationship of serum adiponectin (APN) with left ventricular hypertrophy (LVH) and carotid athcrosclerosis (AS),and to observe the change of serum adiponectin levels in patients with hypertension and obesity. Methods Patients with essential hypertension (50 cases) were divided into simple hypertension group (23 cases) and hypertension with obesity group(27 cases),and 45 normotensives were divided into control group (24 cases) and simple obesity group (21 cases). The levels of serum adiponectin were determined by enzyme immunoassay.Common carotid artery intima-media thickness (CCA-IMT),carotid artery diameter (CADIA) and left ventricular relevant index were measured by color doppler ultrasonography, and left ventricular mass index (LVMI) was calculated. Results In control group, simple obesity group, simple hypertension group and hypertension with obesity group, serum APN levels were decreased by turn,CADIA reduced gradually,LVMI values were increased by turn,CCA-IMT values were increased gradually (F=28.34,10.26,36.52,14.73,all P<0.01).Correlation analysis showed that serum APN levels were significantly negatively correlated with LVMI and CCA-IMT(r=-0.870,-0.710,both P<0.01),and were positively related with CADIA (r=0.742,P<0.00).APN,SBP and BMI were the main influencing factors on left ventricular hypertrophy (β =-0.909,0.126,-0.053).Conclusions The presence of both hypertension and obesity may play a synergistic role in accelerating the process of left ventricular hypertrophy and atherosclerosis.Lower APN levels and/or rising BMI may accelerate occurrence and development of LVH and AS in aged patients with hypertension and obesity.

BACKGROUND: Proliferation and differentiation of mesenchymal stem cells (MSCs) is associated with platelet concentration in platelet-rich plasma (PRP). Low enrich multiple cannot reach proper effects, but high level had inhibitory effects on osteoanagenesis.OBJECTIVE: To observe the effect of different volume fraction of PRP on dog BMSC proliferation.DESIGN: A cytological in vitro study.MATERIALS: Healthy 12-month male Beagle dogs were supplied by the Experimental Animal Center, Xiangya Medical College,Central South University.METHODS: Dog BMSCs of 5 passage were adjusted to 3×10~8/L, and incubated in a 96-well plate at 200 μL per well. Following 24 hours of routine culture, primary medium and non-adherent cells were discarded. Prepared PRP gel was mixed with serum-free low-glucose DMEM containing penicillin and streptomycin, and then diluted into 5%, 6.25%, 7.5%, 8.75%, 10% volume fraction. 200 μL above-described liquid was added into the 96-well plate, which was subsequently placed in a incubator. We set up a blank control.MAIN OUTCOME MEASURES: MTT was used to investigate effect of different volume fraction of PRP on dog BMSC proliferation.RESULTS: Compared with the blank control group, various volume fraction of PLP could promote dog BMSC proliferation in early stage. With prolonged time, proliferation speed began to increase at day 6 in the 8.75% and 10% PRP groups, entering platform stage. BMSC number was increased rapidly in the 5% and 6.25% PRP groups, especially in the 6.25% PRP group.CONCLUSION: PRP gel could promote BMSC proliferation markedly and proliferation strength of BMSCs was correlated to the density of PRP. BMSC proliferation would be accelerated by the low density of PRP.

BACKGROUND: Platelet-rich plasma (PRP) contains abundant growth factors that were needed for osteanagenesis. Moreover, the proportion of each growth factor formed by an organism, with good synergism.OBJECTIVE: To explore the influence of PRP on osteogenetic activity of canine bone marrow mesenchymal stem cells (BMSCs) after induction in vitro.DESIGN, TIME AND SETTING: The in vitro cytological experiment was performed at the Central Laboratory of Xiangya Hospital of Central South University from June 2007 to February 2008.MATERIALS: Healthy 12-month male Beagle dogs were supplied by the Experimental Animal Center, Xiangya Medical College,Central South University.METHODS: The 3~(rd) generation BMSCs were collected and divided into 4 groups. BMSCs in the control group were incubated in standard medium. BMSCs in the osteogenetic induction medium group were incubated in high-glucose DMEM containing fetal calf serum, dexamethasone, beta-sodium glycerophosphate and vitamin C. BMSCs in the PRP group were incubated in low-glucose DMEM containing 6.25% PRP. BMSCs in the combination group were incubated in high-glucose DMEM containing 6.25% PRP, dexamethasone, beta-sodium glycerophosphate, and vitamin C.MAIN OUTCOME MEASURES: Alkaline phosphatase activities were measured. Expression of collagen type I was examined by immunocytochemical staining. Calcium tuberoses were labeled using modified Von Kossa staining. Expression of osteocalcin mRNA was examined by RT-PCR.RESULTS: Levels of alkaline phosphates of all groups became increased along with time. The alkaline phosphates level of combination group was strongest (P < 0.05). Following 7 and 14 days of induction, type I collagen expressed positively in the osteogenetic induction medium and combination groups, but negatively in the PRP and control groups. Following 14 days, formation of calcium nodules were found in the osteogenetic induction medium and combination groups. Following 7 and 14 days, expression of osteocalcin mRNA were similar between the control and PRP groups (P > 0.05), which was significantly lower than the osteogenetic induction medium and combination groups (P < 0.05). Expression of osteocalcin mRNA was significantly lower in the osteogenetic induction medium group compared with the combination group (P < 0.05).CONCLUSION: PRP gel can effectively promote osteoblastic effect of BMSCs after induction in vitro following induction in osteogenetic medium.
