Objective:To investigate the effects of granulocyte-macrophage colony-stimulating factor on expressions of CC chemokine receptor 2(CCR2) mRNA and protein in cultured human THP-1 cell lines.Methods:THP-1 cell was stimulated by GM-CSF of different concentrations and time intervals.The mRNA expression of CCR2 was tested by RT-PCR,The protein expression of CCR2 was analyzed by Western blotting.Results:RT-PCR and Western blotting showed that THP-1 cell in control group expressed a low-level of CCR2 mRNA and protein.After stimulated by GM-CSF,the expression of CCR2 mRNA and protein increased in a concentration-and time-dependent manner.Conclusion:GM-CSF could stimulate THP-1 cell to express CCR2 mRNA and protein,in an concentration-and time-dependent manner.

5.Effects of different blood pH of the recipients on ischemia-reperfusion injury to the transplanted lungs in rats

Objective To investigate the effects of different blood pH of the recipients on ischemiareperfusion (I/R) injury to the transplanted lungs in rats. Methods Sixty-four pure bred male SD rats weighing 250-350 g were randomly divided into equal numbers of donors and recipients and the recipient rats were randomly divided into 4 groups(n = 8 each). Both donor and recipient rats were anesthetized with intraperitoneal (IP)ketamine 15 mg/kg and diazepam 6 mg/kg, tracheostomized and mechanically ventilated. In donor rats heparin 1 ml was injected via inferior vena cava and 4 ℃ LDP solution 40 ml was injected via pulmonary artery . The left lung was then removed and placed in 4℃ LDP solution and kept for 4 h. The left lung transplantation was performed using two-cuff-one-stent anastomosis technique. Before the transplanted lung was reperfused, the recipients' blood pH was adjusted to 7.4 (group Ⅰ), 7.2 (group Ⅱ), 7.3 (group Ⅲ) and 7.5 (group Ⅳ) by HCI and NaHCO3 solutions infused via femoral vein. After 2 h reperfusion, the transplants were removed for microscopic examination and determination of W/D lung weight ratio, MPO and SOD activities and MDA, TNF-α and IL-8 contents. Results SOD activity was significantly higher while MPO activity, and MDA, TNF-α and IL-8 contents in the lung tissue were significantly lower in group Ⅲ than in group Ⅰ . There was no significant difference in SOD and MPO activities and MDA, TNF-α, and IL-8 contents in the lung tissue among group Ⅰ , Ⅱ and Ⅳ. There was no significant difference in W/D lung weight ratio among the 4 groups. Microscopic examination showed that the histological damage was attenuated in group Ⅲ compared with the other 3 groups. Conclusion The I/R injury to the transplanted rat lungs can be effectively alleviated by reducing blood pH of the recipients to 7.3.

By tracing of ~(75)Se-Na_2SeO_3 incubating and irrigating of the isolated myocardial slices of rats, the numeric relationship between the free selenics in the serum and the serum sdenics, the sodium sdenite doses injected were observed; the absorption of the frce selenics and the serum selenics was compared; the setenics metabolic rate and quantities of the myocardial tissue in different duration after injecting of selcnite, the effect of selenics concentration to the participating were calculated, It is suggested that there is a very close relationship between the free selenics and the selenium metabolism in the myocardium; the ratio betwen the values of the free selenics and the serum selenics may become a stable indicator of the selenium metabolism; the free selenics in the serum may be where the important mediates of selenium metabolism are.
7.CONSTRUCTION AND IDENTIFICATION OF THE VECTOR FOR CHINESE Hp MUTANT STRAINS DELETING PATHOGENICITY ISLAND

A vector for a mutant Chinese Helicobacter pylori (Hp) strains knock out pathogenicity island (PAI) was constructed, which was the basis to establish a Chinese Hp mutant deleting pathogenicity island. Genetic engineering techniques such as polymerase chain reaction, plasmid extraction, agarose gel electrophoresis, restriction analysis, ligation, preparation of competence cell and transformation were used to make two cloning fragments containing two end regions of PAI and a selectable chloramphenicol resistance marker between them, and then engineering the recombined fragments into pBluescript plasmid. The result showed that restriction analysis demonstrated that the engineering mutant vector had been recombined. It suggested that we constructed an engineering mutant vector which targeted to delete the PAI in Chinese Hp strains. It will be useful for addressing the role of PAI in the pathogenesis of Hp infection.

Objective To explore the clinical significance of detecting serum sCD44V6 in non?- small cell lung carcinoma. Methods Levels of serum sCD44V6 in 42 patients with lung carcinoma and in 30 healthy persons were measured by ELISA. Results There was obvious difference of serum sCD44V6 levels between patients with lung carcinoma and healthy persons. There was no difference between patients with squamous cell carcinoma and with adenocarcinoma. sCD44V6 have the higher positive rate and specificity. Conclusions sCD44V6 would be a good way to non?- small cell lung carcinoma, and could be useful to the diagnosis of early?- stage non?- small cell lung carcinoma.
10.Use of inflammatory markers of induced sputum for assessing the severity of late onset asthma and differential diagnosis

Objective To investigate the relationship between the eosinophil (Eos) percentage and levels of eosinophil cationic protein (ECP) in induced sputum and the severity of late onset asthma(LOA) and differential diagnosis. Methods Fifty-four patients with LOA were selected. The percentage of eosinophil and levels of ECP were measured by Wrigbt′s stain and immuno-CAP system respectively. The lung function was also evaluated. The same indexes were measured in 25 elderly patients with chronic obstructive pulmonary disease (COPD) and 10 healthy elderly subjects as control. Results The percentage of Eos and levels of ECP in induced sputum in patients with LOA were significantly and negatively correlated with FEV_1/FVC (r=-0.68,-0.61, respectively, P
