1.Inhibitory effects of survivin antisense oligonucleotide on human endometrial carcinoma cells

Objective To investigate the effects of survivin antisense oligonucleotide (ASODN) on human uterine endometrial cancer HEC-1B cell line. Methods The experiment included four groups: blank control group (blank), lipofectin control group (Lip), oligonucleotide (SODN) control group and antisense oligonucleotide (ASODN) test group. Artificially synthesized survivin antisense oligonucleotide was transfected for 48 hours into endometrial cancer cells using liposome after cultivation. The expression of survivin protein was determined by Western blot, and the cellular apoptotic rate was tested by flow cytometric analysis. MTT method was used to assay cellular growth rate. Results In comparing with the blank, Lip and SODN control groups, survivin protein expression was obviously decreased in HEC-1B cancer cells after transfected with survivin ASODN. The apoptotic rate and proliferation inhibition rate in ASODN test group were significantly higher than that in the control groups (P<0.05). There was no significant difference between other control groups (P>0.05). Conclusions The expression of survivin is decreased in endometrial cancer cells after ASODN transfection. Survivin ASODN causes cellular apoptosis and inhibits the proliferation of HEC-1B cells. It may be a useful gene therapy for endometrial cancer.
2.Determination of geniposide and scutellarin in Yinshanlian Granules by HPLC

AIM:To develop a HPLC method for determining scutellarin and geniposide in Yinshanlian Granules(Herba Artemisiae scopariae,Herba Scutellariae barbatae, Fructus Gardemiae,etc). METHODS: The analysis was performed on Diamonsil C_(18)(200 mm?4.6 mm,5 ?m) with mixture of acetonitrile(A) and 0.1% phosphoric acid solution as mobile phase in gradient mode.The concentration of solvent A were 5%,33%,5% and 5% at 0,30,31 and 35 min,respectively.The detection wavelength was set at 240 nm and the column temperature was at 30 ℃.(RESULTS:)The linear calibration curves were obtained in the concentration range of 0.032-0.288 mg/mL for scutellarin and 0.009 6-0.086 4 mg/mL for geniposide.The average recoveries of scutellarin and geniposide were 100.6%(RSD=0.80%,n=9),102.6%(RSD=1.1%,n=9),respectively.CONCLUSION: The method is quick,simple,and reproducible,which can be used to control the quality of Yinshanlian Granules.
3.Relationship between polymorphisms in IGF-1 and IGFBP-3 and non-smoking female lung cancer

Objective:This study aimed to investigate the associations between single-nucleotide polymorphism (SNP) of IGF-1 and IGFBP-3 gene and risk of non-smoking female lung cancer in Chinese population. Methods:Genotyping was performed using the TaqMan method in 287 histologically confirmed non-smoking female lung cancer cases and 281 healthy controls. Results:The geno-type distributions of IGF-1 polymorphisms were significantly different between cases and controls (P<0.001). Analysis of multivariate logistic regression showed that the carriers of the CC genotype exhibited a significantly decreased risk of lung cancer in non-smoking female (adjusted OR=0.28, 95%CI:0.15-0.54). IGF-1 rs1520220 GG genotype may reduce the risk of advanced lung cancer incidence in nonsmoking female (adjusted OR=0.30, 95%CI:0.09-0.96). Log-rank test and Cox regression analyses revealed that variant geno-types of IGF-1 rs2946834 CT/TT had a significantly decreased lung cancer mortality risk compared with the homozygote CC in≥60 age group or patients with a lung tumor>3 cm. Conclusion:The IGF-1 polymorphism was associated with the risk of lung cancer and prognosis among non-smoking female. More rigorous laboratory studies of large sample population and functional studies are warrant-ed to confirm our findings.
4.A new model of anterior spinal cord injury syndrome

Objective To evaluate a new model of anterior spinal cord injury (SCI) syndrome and to explore the pathophysiology of SCI. Methods Fifty-five Wistar rats were randomly divided into a sham-operated group ( 10 rats) and an experimental group (45 rats). A metal hook was fixed in front of the rats'abdomens to compress the ventral part of the spinal cord in the experimental group. According to the degree and time of compression,the rats in the experimental group were divided into light, moderate, heavy injury subgroups. The tilt board test and the Bosso-Beattie- Bresnahan (BBB) locomotor rating scale were used to assess the rats' behavior at the 1st , 7th andl4th days and after 4 and 8 weeks. The latency of somatosensory evoked potential (SEP) and motor evoked potential (MEP) were measured before and 8 weeks after the operation. Results After the operation the gradients in the tilt board test and BBB scores in the experimental subgroups were all significantly lower than in the sham-operated group. There were also significant differences among the 3 severity subgroups. Eight weeks after the operation the average MEP latencies in the experimental subgroups were significantly longer than in the sham-operated group, and there were also significant differences among the 3 severity sub-groups. MEP in the heavy injury subgroup was significantly longer compared with the sham-operated group. Conclusions Obvious behavioral and neuroelectrophysiological changes were observed in the injured rats. Models of different severity could be prepared and reproduced to simulate clinical SCI.
5.Effect observation of 115 children with asthma treated with salmeterol/flutieasone propionate inhalation

Objective To observe the effect of salmeterol/flutieasone propionate on children with asthma. Methods Salmeterol/flutieasone propionate inhalation of different dosages were given according to different illness condition of asthma. Investigate the effects of 180 days inhalation of SM/FP among children. Results After 180 days' treatment, the PEFam and PEFpm of those children of asthma were significantly increased. The lung function has a perceptible improvement. In addition, the daytime and noctural symptom scores and asthma symptom were improved. Moreover, the shore-acting β2 agonist was used with decreasing frequency. The effects of SM/FP inhalation were marvelous after 1 week of treatment and should make a long persisting time alter six months of treatment. Conclusion SM/FP is an effective medicine for children with asthma and should be make more widespread.
6.Electroacupuncture and body-weight supported treadmill training after vertebral fractures

Objective To observe the effect of electroacupuncture and body-weight supported treadmill training in the treatment of spinal cord injury (SCI) after fractures of the thoracolumbar segment of the spine.Methods Forty-three patients with SCI after fractures of the thoracolumbar segment of the spine were randomly divided into a treatment group (22 cases) and a control group (21 cases). The patients in the treatment group were treated with electroacupuncture and body-weight supported treadmill training, while those in the control group were treated mainly with drugs. Patients were scored with the American Spinal Injury Association (ASIA) assessment and the modified Barthel index (MBI) initially and after 6 months. Results The ASIA and MBI scores of all patients were significantly higher after 6 months than before treatment, but those in the treatment group were significantly higher than those in the control group. Excellent and good ratings were applied to 77% of the treatment group, while only 48% of the control group, a statistically significant difference. Conclusions Electroacupuncture and body-weight supported treadmill training can effectively improve ability in the activities of daily living, sensation and locomotor abilities after thoracolumbar vertebral fractures.
7.Reconstruct bladder function and prevent urinary tract infection of patients with spinal cord injury

Objective To observe the effectiveness of comprehensive rehabilitation for neurogenic bladder of patients with spinal cord injury and prevent urinary tract infection (UTI). Methods Fifty-seven patients with spinal cord injury were randomly divided into two groups: treatment group (29 cases) and control group (28 cases). The patients in treatment group were treated with comprehensive rehabilitation,while those in control group were treated mainly with catheterization and TDP equipment. Scored with Barthel index and the rate of UTI. Results It was shown that the improvement of Barthel index after treatment [(8.93 ± 6.27) scores] and the rate of UTI [13.79%(4/29)] were significantly better than those in control group[(5.16 ± 6.82) scores,67.86%(19/28)], which had significant differences (P< 0.05). Conclusion Comprehensive rehabilitation can reconstruct bladder function and prevent UTI of patients with spinal cord injury effectively.
8.Clinical Study on Tianma Gouteng Decoction Combined with Routine Western Therapy in the Treatment of Early Onset Severe Pre-eclampsia

Objective To observe clinical efficacy of Tianma Gouteng Decoction combined with Western medicine for early onset severe pre-eclampsia. Methods Sixty patients were randomly divided into treatment group with 30 cases and control group with 30 cases. Two groups were both given the routine Western therapy, while the treatment group was added with Tianma Gouteng Decoction. Indicators were observed and compared between two groups including blood pressure, 24 h UAE, D-dimer, fibrinogen and their effects on pregnancy. Results Compared with the control group, the blood pressure, 24 h UAE, fibrinogen, D-dimer, hypoproteinemia, and liver and kidney function were reduced obviously in the treatment group (P<0.05);fibrinogen and D-dimer of control group were not changed significantly;gestational weeks of treatment group were extended (15.2 ± 4.8)d, and neonatal asphyxia was 7 cases;gestational weeks of control group were extended (9.3 ± 2.2)d, and neonatal asphyxia was 11 cases. There was no significant difference about cesarean section rate between the two groups. Conclusion Tianma Gouteng Decoction combined with Western medicine for treatment of early onset severe pre-eclampsia have better clinical efficacy.
9.Analysis on Resistance Status of 279 Strains of Mycobacterium tuberculosis

Objective The purpose of the study was to understand the resistance status of to anti-tuberculosis drug in Yunnan province, and to provide evidence for tuberculosis (TB) control. Methods The proportional method was used in drug sensitivity test according to the guidelines recommending by WHO. Results Of 301 mycobacterial isolates, 279 (92.7%) strains were complex (MTBC) . The total drug resistance rate was 31.5% (88/279),the initial drug resistance rate of was 27.3%, and the acquired drug resistance rate was 44.3%. The drug resistance rates to INH,RFP,EMB and SM were 19.7%, 10.8%, 12.9%and 19.0%, respectively. The multi-drug resistance (MDR) rate was 9.3%. Conclusion The drug resistant rate of is still high in Yunnan province,and TB control is still a long way to go. Early detection and regular treatment should be done for MDR patients. Drug resistance monitoring as an important part of TB control programs should be regular observation and research.
10.Effects of simvastatin on PDGF-BB and serum-induced proliferation of vascular smooth muscle cells and on the expression of tumor suppressor gene PTEN

AIM: To observe the effect of simvastatin on the proliferation of vascular smooth muscle cells(VSMCs) induced by serum and growth factor PDGF-BB and the effect of simvastatin on the expression of PTEN,a important regulator of G 1/S cell cycle transition. METHODS: The DNA synthesis was determined by -TdR incorporation, cell cycle was examined with flow cytometry, the protein level of PTEN was measured by Western blot method. RESULTS: (1)Simvastatin inhibited -TdR incorporation in a dose dependent manner. (2) Flow cytometric DNA analysis revealed that simvastatin induced significantly enhancement of G 0/G 1 phase and decrease in S phase VSMCs.(3)Simvastatin increased protein level of PTEN and mevalonate, a metabolite of HMG-COA, reversed the effect of simvastatin on PTEN protein expression. CONCLUSION: Simvastatin may inhibit proliferation of VSMCs and retarded cell cycle in G 0/G 1 phase by increasing PTEN expression through inhibiting synthesis of mevalonate.
