Objective To describe the burden of caregivers of children with asthma,and to provide reference for the development of family care support program.Methods Qualitative research was used to carry out semi-structured interviews with 13 caregivers of children with asthma in the pediatric outpatient of the First Affiliated Hospital of Guangzhou Medical University.The data collected were analyzed with Colaizzi 7-step analysis method.Results The carcgiver burden was classified into 4 themes,including time and energy exhaustion,heavy economic burden,insufficient knowledge of care and negative emotions.Conclusions The burden of caregivers of children with asthma has a relatively great impact on the daily work and life,even the physical and mental health.Care workers should be concerned about the status of caregiver burden and provide targeted interventions,which could help caregiver adapt to their roles,maintain and promote the health of children patients and their families.
3.Experimental study of the effect of RNAi to silence STAT3 gene expression on the growth of T24 and 5637 bladder cancer cells

Objective Signal transducers and activators of transcription 3 (STAT3) silenced by RNA interference (RNAi) technique were used to induce the apoptosis and growth inhibition in T24 and 5637 bladder cancer cells. Methods Three recombinant plasmids pGenesil-1-shRNA-STAT3 was constructed and transfected into T24 and 5637 cells. The expression of STAT3 gene was detected by RT-PCR and Western blotting. FCM was used to observe the apoptosis in T24 and 5637 cells. Results pGenesil-1-shRNA-STAT3 was successfully constructed, and transfected into T24 and 5637 cells. RT-PCR and Western blot analysis demonstrated that pGenesil-1-shRNA-STAT3 could significantly inhibit the expression of STAT3 in T24 and 5637 cells; FCM results show that it could suppress the growth of 1'24 and 5637 cells. Conclusion pGeneSiI-1-shRNA-STAT3 could significantly inhibit STAT3 expression, suppress the growth of T24 and 5637 cells.
4.Comparison of the proteome of uropathogenic E. coli 132

Objective To study proteome variation between uropathogenic E. coil (UPEC)132, UPEC J96 and non-uropathogenic E. coli K-12 MG1655. Methods Two-dimensional gel electrophoresis(2-DE) was applied to compare the differential expression proteins between UPEC 132, UPEC J96 and non-uro-pathogenic E.coli K-12 MG1655. The differential expression proteins were digested in gel by enzyme. The mass of generated peptides were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). The data obtained from peptide mass fingerprinting (PMF) were re-searched using the internet available database. Results The number of protein spots recognized from UPEC 132 was 466±11, significantly more than that of E. coli K-12 MG1655 (338±15) and UPEC J96 (382±12); there were 298 protein spots shared by the three E.coli strains, 56 protein spots shared by two UPEC strains, and 89 protein spots characterized by UPEC 132. Twenty-two differential expression or significantly increased expression protein spots, involved in virulence factors, metabolism and transportation, regulation of protein synthesis, biological oxidation and unknown functions, were successfully identified by MALDI-TOF-MS. Condusion The proteome from UPEC 132 and non-uropathogenic E. coli K-12 MG1655, or UPEC 132 and UPEC J96 was differentially expressed. It will provide important information on the pathogen-esis of UPEC 132.
5.Treatment outcome comparation of surgery plus radiotherapy and radiotherapy or chemoradiotherapy plus salvage surgery for stage Ⅲ/Ⅳ laryngeal carcinoma

Objective To compare the treatment outcome of underwent surgery plus radiotherapy and radiotherapy/chemoradiotherapy plus salvage surgery for stage Ⅲ/Ⅳ laryngeal carcinoma,to investigate an optimized treatment for the patients of stage Ⅲ/Ⅳ laryngeal carcinoma.Methods Clinical data from 103 patients with stage Ⅲ (39 cases) or stage Ⅳ (64 cases) laryngeal carcinoma were retrospectively analyzed.The patients were divided into surgery plus radiotherapy group (S±R,46 cases) and radiotherapy/chemoradiotherapy plus salvage surgery group (R±S,57 cases).Overall survival,relapse free survival,and laryngeal preservation rate were used to compare the treatment outcome between two groups.Multivariate regression models were used to analyze the independent factors for survival and laryngeal preservation rate.Results Survival rate was higher in S±R group than in R±S group [2 year overall survival/relapse free survival 74.7 % (34/46) / 72.4 % (33/46) vs 46.4 % (26/57) / 40.9 % (23/57),P < 0.05].Laryngeal preservation rate was higher in R±S group than in S±R group [93.0 % (15/46) vs 32.6 % (53/57),P < 0.05].Multivariate analysis demonstrated that treatment modality and T stage were independent factors for long-term survival,while treatment modality was the only an independent factor for laryngeal preservation rate.Conclusions Surgery plus radiotherapy result in better survival and lower laryngeal preservation rate than radiotherapy/chemoradiotherapy plus salvage surgery in treatment of stage Ⅲ/Ⅳ laryngeal carcinoma.Surgery plus radiotherapy should be the first choice for treatment of locally advanced laryngeal carcinoma.Improvement of the quality of life could be achieved by laryngeal preservation surgery and phonation reconstruction procedures.
6.Ginsenosides from Panax ginseng by supercritical CO_2 reverse microemulsion extraction

Objective To raise ginsenosides yield from Panax ginseng by supercritical CO2 reverse microemulsion extraction.Methods Bis-(2-ethylhexyl)sodium sulphosuccinate(AOT)/ethanol/water/supercritical CO2 reverse microemulsion extraction was carried out to extract gingsenosides.Results The ginsenosides extracting rate by supercritical CO2 reverse microemulsion extraction was 3.2 times that of by ethanol/water/supercritical CO2 extraction in extracting pressure 25 MPa,extracting temperature 45 ℃,extracting time 4 h,and CO2 flow rate 2.0 L/h,respectively.The ginsenosides extracting yield increased with the increasing of the water amount and the extracting pressure,increased first and then decreased with the increasing of AOT concentration and the extracting temperature.The ginsenosides extracting yield with P.ginseng soaked with water before extraction was 1.3 times that with P.ginseng soaked with water during the extraction.Conclusion Combined the experimental results with the theoretic deduction,the mechanism of supercritical CO2 reverse microemulsion extraction is that the polar water pools of reverse microemulsion can make more ginsenosides dissolved.
7.Quantitative structure-activity relationship of synthetic pyrethroids and antibody activity

Objective:To research on the immune recognition mechanism of synthetic pyrethroids and generic specific antibody.Methods:We studied on quantitative structure-activity relationship ( QSAR ) of synthetic pyrethroids and their analogs as well as antibody activity ( IC50:fifty percent inhibition concentration ) using stepwise multiple linear regression method.Based on calculating structure descriptors of synthetic pyrethroids and their analogs , two-demensional QSAR ( 2D-QSAR ) model was established.The main factors affecting antibody activity were screened using 2D-QSAR,and predictive ability of QSAR models were evaluated by the method of leave-one-out( LOO) cross-validation.Meanwhile, the structure parameters of synthetic pyrethroid fragments were calculated and then analyzed using partial least squares ( PLS) assay.And then hologram QSAR ( H-QSAR) model was constructed on molecular substructure and antibody activity.The fragments contribution to antibody activity were illustrated by encoding different colors.Results:Decision coefficent (R2) of 2D-QSAR model and HQSAR model were 0.920 and 0.917 individually,cross-validation coefficient ( Q2 ) of two QSAR models were 0.875 and 0.660 respectively ,which showed two models had good predictive abil-ity.The result from 2D-QSAR model was also obtained that smaller was hydrophobicity of pyrethroids , easier was recognized by antibody.In addition,the optimum HQSAR model was constructed after we tried many combinations of these parameters .The fragment size in optimum HQSAR model was between 4 to 10,a hologram length was 61,optimum principle component was 4,and the fragment type of B/C/Ch was selected.However ,the fingerprint encoded results of synthetic pyrethroids weren′t consistent completely with exper-imental IC50 values.Conclusion:Hydrophobicity of synthetic pyrethroids is the largest correlation factors in antibody recognization .
8.Analysis of the Processing Situation of Marketed Scorpions and Optimization of Salt-water Processing

Objective: To analyze the processing situation of 124 batches of scorpions and optimize the salt-water processing.Methods: The character, extract, moisture, total ash, acid insoluble ash, amino acids content and heavy metals in the marketed scorpions were determined.Using extract and amino acids contents as the indices, the salt-water processing was optimized.Results: The results of the above determinations showed that the quality difference was very obvious for the marketed scorpion, and the extract and amino acids contents were the highest in scorpion boiled in 2% sodium chloride aqueous solution for 5 min.Conclusion: The item of extract test should be revised in the current standard, and the items such as moisture, total ash, acid insoluble ash, amino acids content and heavy metals should be taken into consideration in order to control the quality of scorpion more comprehensively.The best salt-water processing of scorpion is boiled in 2% sodium chloride aqueous solution for 5 min.
9.Antidepressant activities of flavonoids from Glycyrrhiza uralensis and its neurogenesis protective effect in rats.

Adult rats chronic unpredictable stress model of depression (CUS) was adopted to elucidate the antidepressant pharmacological activity and related neurogenesis protective effect of the total flavonoids extract (licorice flavonoids, LF) from the Glycyrrhiza uralensis Fisch. cultivated locally in Ningxia. The rats were exposed to 9 kinds of unpredictable sequence of stressors and were given flavonoids (300 mg x kg(-1), 100 mg x kg(-1) and 30 mg x kg(-1)) for 28 days. The antidepressant effect was elucidated by open field test, forced swimming test and tail suspension test. The level of serum corticosterone was detected by radioimmunoassay. 5'-Bromo-2'-deoxyuridine (BrdU) labeling experiments was employed to study the neurogenesis protective activities. The flavonoids can increase the sum of line crosses and number of rears, and decrease the number of fecal boli produced in the open field test of the CUS rats. Also the flavonoids can decrease the immobility time in forced swim test as well as in the tail suspension test. In addition, the flavonoids (300 mg x kg(-1)) can decrease the serum corticosterone level of the CUS rats, and increase the number of the new born BrdU positive progenitor cells at the subgranular zone (SGZ) of dentate gyrus (DG) region in hippocampus. The results demonstrated that the total flavonoids extract from the cultivated Glycyrrhiza uralensis Fisch. could produce the anti-depressive effect on chronic unpredictable stress of depression model rats and its mechanism may be associated with its neurogenesis protective effect.
10.Application of comfortable nursing in postoperative patients with liver cancer

Objective To investigate the clinical effect of comfortable nursing intervention on postoperative patients with liver cancer. Methods A total of 66 postoperative patients with liver cancer who were treated in our hospital from November 2015 to August 2017 were selected as experimental subjects. Patients were divided into observation group and control group by digital odd-even method. The patients in the control group(n=33)were given the routine nursing method, and the observation group(n=33) were given the comfortable nursing method. The postoperative pain degree, the scores of self-rating anxiety scale(SAS)and self-rating depression scale (SDS) between the two groups were compared. Results Compared with the control group, postoperative pain degree of patients with liver cancer was significantly relieved in the observation group (P<0. 05). Compared with the control group, SAS score of postoperative patients with liver cancer reduced significantly in the observation group (P<0. 05). Compared with the control group, SDS score of post-operative patients with liver cancer reduced significantly in the observation group(P<0. 05). Conclusion Appropriate comfortable nursing interventions for postoperative patients with liver can relieve postoperative pain, reduce postoperative SAS score and SDS score adequately, so that can improve for rapid recover and prognosis for postoperative patients with liver cancer.
