1.Effects of FFBHQH on level of expression of caspase-3 protein of middle cerebral artery in ischemia/ reperfusion models

AIM : To investigate the effects of FFBHQH on the level of expression of caspase 3 protein of middle cerebral artery in ischemia / reperfusion model and to study its mechanisms in preventing and treating ischemic apoplexy. METHODS : The model of focal cerebral ischemia / reperfusion in rats was established with the suture occluded method. The effects of FFBHQH on the behavior obstacle of rats after MCAO/R 24, 48, and 72 h were observed, and the levels of expression of caspase 3 protein were observed by immunoassay method in rats treated with FFBHQH after the cerebral ischemia / reperfusion. RESULTS : Different degrees of behavior obstacle appeared in rats after MCAO/R 24,48,72 h, and FFBHQH reduced the behavior obstacle grade. After MCAO/R, the expression of caspase 3 protein increased obviously, but FFBHQH reduced the expression. CONCLUSION : FFBHQH can prevent and treat the ischemic apoplexy, and the mechanisms may relate to the calcium antagonism and the prevention of the apoptosis correlated gene such as caspase 3 after cerebral ischemia.
2.Effect and mechanism of combined therapy using anti-CTLA-4 antibody and doxorubicin on mice bearing breast cancer

Objective To investigate the effect and mechanism of combined therapy using anti-CTLA-4 antibody and doxoru-bicin on mice bearing breast cancer.Methods Balb/c mice inoculated with 4T-1 mouse breast cancer cell were used as tumor mod-els,which were randomly divided into blank group,solvent control group,anti-CTLA-4 antibody only group,doxorubicin only group and combined therapy group.Tumor growth of mice was observed.The ratio of spleen and bone marrow cell subgroup were evaluated.Tumor microenvironment apoptosis and microvessel density (MVD)were evaluated.Results The tumor volume of an-ti-CTLA-4 antibody only group,doxorubicin only group and combined therapy group were lower than those in the rest groups(P <0.05).The tumor volume and mass of combined therapy group was significantly higher than those of anti-CTLA-4 antibody only group,doxorubicin only group (P <0.05).Compared with blank group,solvent control group,CD8 + Tand CD4 + T ratio in anti-CTLA-4 antibody only group,doxorubicin only group,combined therapy group increased with significant difference (P <0.05). The positive cell apoptosis rate of combined therapy group was significantly higher than those of other groups(P < 0.05 ).The MVD of combined therapy group was significantly lower than those of other groups(P <0.05).The positive cell apoptosis rate and MVD of anti-CTLA-4 antibody only group,doxorubicin only group were better than those of blank group,solvent control group. Conclusion Combined therapy using anti-CTLA-4 antibody and doxorubicin could improve the immune,significantly inhibit the growth of tumor,promote cancer cell apoptosis and decrease MVD.
3.Clinical values of nuclear magnetic resonance and B-ultrasonography in diagnoses of gynecological pelvic tumors

Objective To study the clinical values of MRI and B-ultrasonography in the diagnoses of gynecological pelvic tumors.Methods From September 2014 to September 2016,130 patients with gynecological pelvic tumors were randomly selected and examined by MRI and B-ultrasonography respectively.MRI and B-uhrasonography were compared on the misdiagnosis and missed diagnosis rate,detection rate of malignant tumor as well as diagnosis coincidence rate.Results The detection rate of malignant tumor,the misdiagnosis and missed diagnosis rate and the diagnosis coincidence rate of MRI were 96.15%,10.77% and 89.23% respectively,those of B-ultrasonography were 76.92%,24.62% and 75.38% respectively,and there were significant differences between all the rates of MRI and B-ultrasonography (P<0.05).Conclusion MRI diagnoses pelvic tumors effectively,while behaves not so well as B-ultrasonography in cost,timeliness and scan time.
4.Evaluation of Intracoronary Electrocardiogram on Myocardial Ischemia During Percutaneous Transluminal Coronary Angioplasty

The changes of intracoronary electrocardiogram (IC-ECG) and surface electro-cardiogram (S-ECG) were recorded simultaneously during percutaneous transluminal coronary angioplasty(PTCA) in 33 cases of coronary heart diseases with 41 vessel lesions. The results sug-gested that ST-segment change rate on IC-ECG(90. 2%) was higher than that on S-ECG(56%). ST-segment elevation on IC-ECG was earlier and more significant than that on S-ECG. The IC-ECG appeared to be more sensitive than S-ECG for detecting myocardial ischemia. The technique for recording IC-ECG is simple and it can help to perfome the PTCA successfully.
6.Diagnostic value of MSCT combined with CTA in recurrence of hepatocellular carcinoma after transcatheter arterial chemoembolization

Objective:To investigate the diagnostic value of multi-slice spiral CT (MSCT) combined with CT angiography (CTA) in the recurrence of hepatocellular carcinoma (HCC) after transcatheter arterial chemoembolization (TACE).Methods:97 HCC patients who received TACE therapy in the 942 Hospital from January 2017 to January 2019 were enrolled in the study. MSCT, CTA and MSCT combined with CTA were performed in all subjects to compare the efficacy of different examination methods in diagnosing postoperative recurrence.Results:MSCT results showed that 45 new lesions were detected in 30 of the 97 patients, with a diameter of 0.4-1.8 cm, with an average of (0.92±0.31)cm. In arterial phase, 32 lesions were significantly enhanced, 10 were mildly enhanced and 3 were not significantly enhanced. In the venous phase, there were 20 high-density shadows, 18 isodensity shadows and 7 low-density shadows. In the delay period, there were 24 shadows with high density or slightly high density, 10 with clear edge of low density, and 11 with unclear edge of low density. The results of CTA showed that there were 47 vessels in 32 cases of 97 patients, including 18 cases of hepatic artery variation and 29 cases of blood supply artery of lateral hepatic branch. Taking pathological diagnosis as the gold standard, the sensitivity, specificity and accuracy of MSCT combined with CTA in the diagnosis of HCC recurrence after TACE were 97.50% (39/40), 96.49%(55/57), 96.91% (94/97), which were higher than 75.00% (30/40), 77.19% (44/57), 76.29% (74/97) of MSCT and 80.00% (32/40), 80.70% (46/57) and 80.41%(78/97) of CTA respectively (all P<0.05) . Conclusions:MSCT combined with CTA has a better efficacy in the diagnosis of TACE recurrence in HCC patients, which is worthy of clinical application.
7. Bacteriostatic effect of ε-polylysine and its effect on urine lead level

OBJECTIVE: To evaluate the bacteriostatic effect of ε-polylysine( ε-PL) on four common putrefactive bacteria including Staphylococcus aureus,Enterococcus faecalis,Pseudomonas aeruginosa,and E. coli and its effect on urine lead level. METHODS: Broth dilution method was used for the determination of the minimum inhibitory concentration( MIC) ofε-PL on the four kinds of putrefactive bacteria; the inhibitory effects of ε-PL with final mass concentration of 40. 000 mg / L on the urine sample were observed; graphite furnace atomic absorption spectrometry was used for determining the lead level in the 40. 000 mg / L( mass concentration) ε-PL solution and the urine lead level in normal healthy groups; the bacteriostatic effects of ε-PL and nitric acid were compared. RESULTS: The MIC of ε-PL on Staphylococcus aureus,Enterococcus faecalis,Pseudomonas aeruginosa,and E. coli was 40. 000 mg / L. There was no bacterial growth in the urine sample with40. 000 mg / L( mass concentration) ε-PL when urine was kept at room temperature for 24 hours to 15 days. The lead level was < 2. 0 μg / L in the 40. 000 mg / L( mass concentration) ε-PL solution. When the ε-PL with final mass concentration of 40. 000 mg / L and the nitric acid with a volume fraction of 1. 0% were respectively used as the antiseptics,the descending rates of the lead levels in the urine samples were similar,and after the urine sample was preserved for 15 days,the descending rates of the urine lead were both smaller than 10. 0% after be stored for 15 days. CONCLUSION: ε-PL can substitute nitric acid as a new natural preservative for preservation of samples for urine lead determination.
8.A retrospective cohort study on postoperative radiotherapy and postoperative chemoradiotherapy for soft tissue sarcomas

Objective:To analyze the differences in clinical outcomes and toxicities between postoperative radiotherapy alone and postoperative radiochemotherapy for soft tissue sarcoma (STS), as well as the related factors affecting clinical prognosis of STS patients.Methods:Retrospective analysis of patients diagnosed with primary STS admitted to Zhejiang Cancer Hospital from May 2012 to May 2019 was performed, who received adjuvant radiotherapy after surgery, combined with or without postoperative chemotherapy. A total of 100 patients were enrolled and divided into postoperative radiotherapy group ( n=52) and postoperative radiochemotherapy group ( n=48). The median follow-up time was 65 months (24-124 months). The local recurrence-free survival (LRFS), distant metastasis-free survival (DMFS), overall survival (OS), and treatment-related toxicities were recorded in two groups. The survival rate was calculated by Kaplan-Meier analysis. Log-rank test was used for univariate analysis, and Cox model was used for multivariate analysis. Results:In multivariate analysis, the maximum tumor diameter was an independent predictor of local tumor recurrence ( HR=4.80, 95% CI=1.16-19.85, P=0.031), distant metastasis ( HR=4.67, 95% CI=1.53-14.26, P=0.007) and OS ( HR=4.10, 95% CI=1.35-12.48, P=0.013). In addition, the degree of myelosuppression in patients in postoperative radiochemotherapy was significantly higher than that in their counterparts in postoperative radiotherapy group ( P<0.001). Conclusions:In the limited number of patients, radiochemotherapy has no advantages over radiotherapy alone in distant metastasis or survival rate. Besides, it increases toxicities, but the overall tolerability is favorable. It is necessary to conduct prospective randomized studies in a large population and subgroup analysis of histological subtypes, aiming to obtain results with better reference value.
