Objective To investigate the effects of inhibition of nuclear factor-kappa B (NF-?B) by pyrrolidine dithiocarbamate (PDTC) on tissue pro- and anti-inflammatory cytokine expression in rats with postburn Staphylococcus aureus sepsis and its potential regulating mechanism. Methods Thirty-four male Wistar rats were randomly divided into normal control group (n=6), scald control group (n=6), postburn sepsis group (n=12),and PDTC treatment group (n=10). Tissue samples from the liver, kidneys and lungs were collected to determine tumor necrosis factor-? (TNF-?) and interleukin-10 (IL-10) mRNA expression as well as their protein levels. Objective To study the correlation between TSH decrease and post‐stroke depression .Methods 101 patients with acute brain infarction in our hospital from October 2012 to July 2014 were collected as the study subjects and divided into the TSH decrease group and the TSH normal group according to whether serum TSH levels less than 0 .27 uIU/mL .The two groups were evaluated on admission by the National Institute of Health Stroke Scale (NIHSS) ,at 3 months after discharge by modifies Rankin′s Scale(mRS)and the Hamilton Depression Scale(HAMD)respectively .Results The HAMD scores at 3 months in the TSH decrease group were significantly higher than those in the TSH normal group (16 .04 ± 3 .34 vs .14 .03 ± 3 .47 ,P=0 .000)and the mRS scores NIHSS score were significantly higher than those in the TSH normal (3 .2 ± 1 .1 vs .2 .1 ± 1 .5 ,P<0 .05)) .The ra‐tio of poor prognosis in the TSH decrease group was higher than that in the TSH normal group (89 .3% vs .38 .4% ,P<0 .05) ,in‐dicating that the prognosis in the TSH decrease group was poor .Conclusion The correlation could exist between the decrease of TSH level with post‐stroke depression .In acute brain infarction ,the patient with low TSH level could have the poor prognosis .

To evaluate the effect of delayed treatment with monoclonal antibody to tumor necrosis factor-alpha (TNF-? MAb) on systemic hemodynamics and multiple organ dysfunction following prolonged hemorrhagic shock and resuscitation. Method: Adult male Sprague-Dawley rats were subjected to prolonged hemorrhagic shock (MAP of 4.00-4.66 kPa for 180 rain)followed by resuscitation over 50 min. Purpose:To study the effect of sodium selenate on the viability and proliferation of PC-3 cell, a kind of human prostate cancer cell. Methods:Sodium selenate was administered to PC-3 cells, and MTT assay and ~(3)H-TdR adulterated assay were used to estimate the viability and proliferation of cell. Results:① When cells were treated with Sodium selenate for 24 h, the optical density (A) of middle-dose group decreased significantly, and the A of the high-dose group decreased dramatically. When cells were treated for 24 h, the A of the low-dose group was significantly lower than that of the control group, while the A of the middle-dose and high-dose groups was much lower than control.② When cells were treated for 24 h, the proliferation of middle-dose group decreased, and that of high-dose group decreased markedly. Conclusions:Sodium selenate can inhibit the viability and proliferation of PC-3 cells, and these actions occur in a dose-dependant manner.
5.Effects of Silicone Pessary Treatment on the Symptoms and Life Quality of Pelvic Organ Prolapse Patients

Objective To evaluate the effects of silicone pessary treatment on relieving the symptoms and improving the life quality of the pelvic organ prolapse patients.Methods From November 2005 to March 2009,33 pelvic organ prolapse (POP-Q stage Ⅲ~Ⅳ) patients received silicone pessary coneervative treatment were followed up and required to complete the Pelvic Floor Distress Inventory-20 (PFDI-20) before and after initiating pessary treatment.Results 31 of 33(93.94%) patients finished the follow-up completely.23 of 31(74.19%) patients continually used the pessary.The follow up time was 3 to 17 (10.04±2.57) months.27 patients completed the PFDI-20 questionnaires.Before silicone pessary treatment, PFDI20 total score,POPDI6 (Pelvic organ prolapse distress inventory),CARDI8 (colorectal anal distress inventory 8) and UDI6 (urinary distress inventory 6) scores were (54.16±36.12),(27.78±17.30),(0.61±1.01) and (25.77± 24.10),respectively.After the treatment of pessary,the scores decreased to (20.20±25.98), (4.48±5.88), (0.45±0.84) and (15.28± 21.53),respectively.Except for CARDI8,the PFDI scores decreased significantly after the pessary treatment (P<0.05).Conclusion Silicone pessary is a viable noninvasive treatment for pelvic organ prolapse.It could relieve the symptoms and improve the life quality of protrusion and voiding dysfunction patients.
6.Analysis of mesh related complications after trans-vaginal mesh-augmented pelvic floor reconstruction surgery

Objective To evaluate the complications after trans-vaginal mesh-augmented pelvic floor reconstruction in treatment of pelvic organ prolapse (POP).MethodsFrom February 2007 to October 2009,vaginal mesh procedures were performed on 91 women with POP stage Ⅲ-Ⅳ in Pekiug University Third Hospital.The operative complications were studied.Results Ninety patients underwent successful surgery among 91 patients.Follow-up rate was 94% (85/90) at a median follow-up of 28.4 ( 15 -44) months.One patient underwent intraoperative organ injuries,and 10 patients had postoperation mesh-related complications.The rate of mesh-related complications was 2% ( 2/85 ),2% ( 2/85 ),4% ( 3/85 ),4% ( 3/85 ) on 6,6 - 12,12 -24 and more than 24 months following up,respectively.Seven patients underwent conservative treatment and the symptoms were improved.Three patients underwent the second surgery,and the symptoms were cured or relieved.Conclusion The incidence of mesh-related complications was low,and interventions were effective in vaginal mesh procedure.

Objective:The non targeted high-throughput urine metabolomics technology was used to study the pathogenesis of APP/PS 1 double transgenic mice and the mechanism of action of Gouteng san.Methods:5-month-old APP/PS 1 double transgenic mice were test with Morris water maze for spatial learning ability.Then we employed the non targeted high-throughput urine metabolomics technology to study the pathogenesis of APP/PS1 double transgenic mice based on the metabolic network.The focus investigation of the key pathways and the observation of the treatment by Morris water maze and metabolic level have been used after spatial learning ability damaged confirmed.Results:The comparison between APP/PS1 double transgenic mice and normal mice suggested that a significant longer was existed in former,which was call-back by Gouteng san.With the non targeted high-throughput urine metabolomics analysis and pathway focused analysis,we found certain signals from metabolic profiling,which was identified to be 6 biomarkers associated with learning and memory function by mass spectrometry analysis or authoritative database.Respectively,they were taurine,pteroylglutamic acid,neopterin,glutaurine,2-oxoglutarate and dihydroneopterin.They were mainly related to taurine metabolism and folate metabolism and represented an effective callback.Conclusion:Gouteng san possess a favorable effect on learning and memory ability of APP/PS1 double transgenic mice,6 biomarkers may be a potential target for the pathogenesis of APP/PSI double transgenic mice and provide experimental basis for the study of Gouteng san.
8.Effects of amyloid ? on kinesin superfamily gene expression in hippo campus of rats

AIM: To investigate the effect of amyloid ? on the expres sion of kines in superfamily(KIF) genes in hippocampus of SD rats. METHODS: Th e model of Alzheimer's disease was established by injection of amyloid ? to bra in ventricle of rats. The learning impairment was verified by water maze test. The levels of mR NA for five kinesin superfamily genes, KIF1A, KIF2, KIF3A, KIF4, and KIF5A, in h ippocampus of rats, were estimated by RT-PCR. RESULTS: Two weeks after amyloid ? injection, the rats showed significant learning impairments. I n happocampus, the expression of KIF genes increased after amyloid ? injection. CONCLUSION: The increase in KIF gene expression may reflect th e excitotoxity of amyloid ? to neurons.
9.Construction of human ScFv phage display library against ovarian tumor.

In order to construct a single chain fragment variable (ScFv) phage display library against ovarian tumor, by using RT-PCR, the human heavy chain variable region genes (VH) and light chain variable region genes (VL) were amplified from lymphocytes of ovarian tumor patients and subsequently assembled into ScFv genes by SOE. The resulting ScFv genes were electrotransformed into E. coli TG1 and amplified with the co-infection of helper phage M13KO7 to obtain phage display library. The capacity and titer of the resulting library were detected. The phage antibody library with a capacity of approximately 3 x 10(9) cfu/microg was obtained. After amplification with helper phage, the titer of antibody library reached 5 x 10(12) cfu/mL. Human ScFv library against ovarian tumor was constructed successfully, which laid a foundation for the screening of ovarian tumor specific ScFv for the radioimmunoimaging diagnosis of ovarian tumor.
10.Factors influencing therapy decision in patients with severe pelvic organ prolapse

Objective To investigate the factors influencing therapy decision of surgery or pessary in patients with severe pelvic organ prolapse (POP). Methods Totally 419 cases ofⅢtoⅣdegree POP patients were studied retrospectively. Patients were divided into surgery and pessary groups according to their own choice. Clinical characters were compared such as age, body mass index (BMI), age of onset and disease duration, POP stage, complications. Results 67.5%(283/419) patients were in the surgical group and 32.5%(136/419) patients in the pessary group. Patients in surgical group had higher BMI [(25.1 ± 3.5) versus (23.8±2.6) kg/m2], elder age of onset [(62±12) versus (57±11) years old], longer disease duration [(5± 8) versus (11±11) years] and higher POP staging of middle compartment and less cardiac disease [20.1%(57/283) versus 30.9% (42/136)] than those in pessary group, all had significant difference (P<0.05). Logistic regression analysis on the above factors showed a statistically significant difference between two groups, BMI, disease duration and POP staging of middle compartment were independent factors (OR=1.141, 0.932, 1.389;all P<0.01). Conclusions Patients with higher BMI, higher POP staging of middle compartment and less cardiac disease tended to choose surgery. Patients with younger age of onset and longer disease duration tended to choose pessary. Factors as age, POP staging of anterior and posterior compartment, history of POP surgery, complicated with hypertension and diabetes, showed no influence on treatment choice.
