1.Effect of short pulse gastric electrical stimulation on the c-fos expression in nucleus tractus solitarius and hypothalamus and its possible mechanism

Objective To investigate whether enteric nervous system(ENS)involve in mediating the effect of short pulse gastric electrical stimulation on c-fos expression in nucleus tractus solitarius (NTS)and hypothalamus.Methods Fifteen Wistar rats were divided into control group,gastric electrical stimulation(GES)group and ENS free group with 5 rats each.A pair of electrodes was implanted on the gastric serosa.In ENS free group,0.5%benzalkonii chloridum was applied on the serosa during the surgery.A set of short pulse GES with frequency of 20 cpm,pulse width of 300 μs and amplitude of 2 mA was applied via the electrodes for 30 rain in the stimulation group and ENS free group.The c-fos expression in NTS/hypothalamus was detected 1 h after the stimulation.Results The number of neuron with c-fos expression was(71.6±7.4)/HP in NTS group and(63.4±10.8)/HP in ENS free group,both were significantly higher than those in control group(36.4±8.6/HP,P<0.05).The number of neuron with c-fos expression in hypothalamus was higher in GES[(224.2±47.3)/HP]and ENS free groups[(249.1±44.0)/HP]compared with control group[(90.2±47.3)/HP,P<0.05].There was no significant difference in c-fos expression in NTS/hypothalamus between GES group and ENS free group(P>0.05).Conclusions Short pulse gastric electrical stimulation significantly increases c-fos expression in NTS/hypothalamus,which suggests that NTS/hypothalamus may participate in mediating the effect of GES on gastric function.This effect may be not mediated by ENS.
2.Expression of podoplanin-positive stromal cells in esophageal squamous cell carcinoma and its clinical significance

Purpose To study the correlation between the expression of podoplanin-positive stromal cells and clinicopathologic features in squamous cell carcinoma ( SCC) of esophagus, and to explore the role of podoplanin-stromal cells in the progression of SCC of e-sophagus. Methods Immunohistochemical EnVision method was used to detect the expression of podoplanin in the stromal cells of e-sophageal SCC. The correlation between the expression of podoplanin-positive stromal cells and clinicopathologic features in SCC of e-sophagus was analyzed. Results Of 69 cases of SCC, podoplanin-positive stromal cells were seen in 43 and the positive rate was 62. 3%. Lymphatic vessel density in SCCs with podoplanin-positive stromal cell was 45. 72 ± 14. 36, which was higher than that with-out podoplanin-positive stromal cell (21. 23 ± 10. 49) (P<0. 01). Expression of podoplanin in stromal cells in SCCs with lymphatic vessel invasion was higher than those without lymphatic vessel invasion (P<0. 05). There was no correlation between podoplanin-posi-tive stromal cells and age, sex, tumor size, the depth of invasion, differentiation, lymph node metastasis, and clinical stage ( P>0. 05). Conclusion Podoplanin-positive stromal cells may promote the lymphangiogenesis and lymph vessel invasion of esophageal SCC.
3.Clinical evaluation of modified radical neck dissection with internal jugularve in preservation

4.Quality of Life in Patients with Chronic Prostatitis

Objective: To investigate the quality of life of patients with chronic prostatitis. Methods: Two hundred patients were included and tested with WHOQOL-100 . Results: Compared with normal controls, the following aspects had no significant difference, including energy and fatigue, appearance and status, working activity, daily life and environment. There were significant difference to controls in other 6 fields and 20 aspects. The major factors that do harm to QOL include satisfaction on life, income and depression. Conclusion: It should deserve the clinician's attention that the QOL of patients with chronic prostatitis is greatly lower than that of the normal controls.
5.Unexplained rhabdomyolysis: clinical analysis of twenty three cases

Objective To evaluate clinical features, therapeutic regimen and prognosis of unexplained rhabdomyolysis. Method Clinical manifestations, therapeutic regimen and prognosis were recorded in 23 patients,who were admitted to The First Affiliated Hospital of Nanjing Medical University 13 to 27 August,2010.The 23 patients were diagnozed as unexplained rhabdomyolysis. Results The patients all presented myalgia of upper body,like neck,waist and back,maybe with asthenia, nausea,dyspnea,abdominal pain, red urine or changed color of urine. Laboratory examination: obviously step-up of creatine kinase [CK: (4655 ± 2556) U/L( normal: 25 ~ 190U/L) AST:(141 ±78) U/L(normal:10~45 U/L),LDH:(348± 127) U/L(normal: 110~ 250 U/L)]and myoglobin[( Mb > 1000 μg/L (normal: 0 ~ 50 μg/L)]. Therapeutic regimen included treatment of the underlying diseases, volume repletion, alkalization and dealing with the complications. No patients developed acute renal failure.All the patients recovered. Conclusions Rhabdomyolysis is a syndrome with different clinical manifestations.However, early diagnosis, proper treatment could prevent serious complications,and prognosis is good.
6.The study on Occludin in esophageal epithelium in gastroesophageal reflux disease

Objective To investigate the distribution and expression of Oceludin in esophageal epithelium in gastroesophageal reflux disease (GERD).Method Immunohistochemistry was used to examine the distribution and expression of Occludin in esophageal epithelium from 62 cases with GERD and 10 Case with other diseases ag control.Results The Occludin was normal continuous membrane expressed in 10 contrast esophageal epithelium,while in 32 nonerosive reflux disease patients,normal expression Was seen in 4 cases,different degree of abnormal expression was seen in 28 cases,including incomplete membrane expression,cytoplasmic expression and expression depletion respectively,and in 30 reflux esophagitis patients were seen all abnormal expression including incomplete membrane expression,cytoplasmic expression and expression depletion respectively.There was significant difference between nonerosive reflux disease and normal contrast,reflux esophagitis and normal contrast(P<0.01),no difference was seen between reflux esophagitis and nonerosive reflux disease(P>0.05).Concision The number of esophageal epithelial Occludin in GERD patients diminished and abnormally distributed,interfered the integrity of epithelium mucosal barrier,which might be one ofthe pathogenesis of GERD,there is no difference of Occludin expression between reflux esophagitis and nonerosive reflux disease esophageal epithelium.
7.Diagnostic value of HNF-1βin the diagnosis of ovarian clear cell carcinoma

Purpose To study the expression of HNF-1β in different type ovarian carcinoma and to explore the diagnostic value of HNF-1β in the diagnosis of ovarian clear cell carcinoma. Methods Immunohistochemical EnVision method was used to detect the ex-pression of HNF-1βin 27 clear cell carcinomas, 35 high-grade serous carcinomas, 21 endometrioid adenocarcinoma, 10 mucious carci-nomas, 13 metastatic Krukenberg tumors, and 2 transitional carcinomas. Results All of ovarian clear cell carcinomas variably ex-pressed HNF-1β and the positive rate of HNF-1β in ovarian cleat cell carcinoma was 85. 2%. Of 21 ovarian serous carcinomas, the nuclear positive of HNF-1β was 4 and HNF-1βpositive rate was 2. 9%. There was 5 HNF-1βexpression in endometrioid adenocarci-noma and the positive rate of HNF-1β was 23. 8%. The positive rate of HNF-1β in mucious carcinoma and metastatic Krukenberg tumor was 60. 0% and 53. 8%, respectively. No HNF-1βexpression was seen in transtional cell carcinoma. The sensitivity and speci-ficity of HNF-1β in the diagnosis of ovarian clear cell carcinoma was 85. 2% and 76. 5%. Conclusions HNF-1β may be a higher sensitive marker for the diagnosis of ovarian clear cell carcinoma. The diffuse and strong positive of HNF-1βmay have higher specific for diagnosis of ovarian clear cell carcinoma.

9.Content Determination of Baicalin in Pneumonia Ⅰ Mixture by HPLC

OBJECTIVE:To establish a HPLC method for the content determination of baicalin in Pneumonia I mixture.METHODS:The assaying was conducted on a ODS C 18 column with methol-0.06%H 3 PO 4 (43∶57)as mobile phase,the detection wavelength was278nm,the column temperature was25℃and the flow rate was1.0ml/min.RESULTS:The linear concentration range of baicalin was0.1875?g~1.5?g(r=0.9998)with average recovery rate at99.63%(RSD=1.00%,n=6).CONCLUSION:This method is simple,accurate and reproducible,and it can be used for the quality control of baicalin.

AIM: Utilizing mRNA fluorescent differential display RT-PCR in our previous study,we found that the mRNA expression level of an intermediate protein(IF)-desmuslin was dramatically down-regulated in the abnormal veins of patients with primary vein incompetence.In this study,we testified the alteration of desmuslin expressing style at gene transcription and translation levels.METHODS: Specific PCR primers were designed according to the sequence of desmuslin mRNA.The cDNA fragments of desmuslin obtained from differential display were labeled by DIG as specific probes.Then semi-quantitative RT-PCR and Northern blotting techniques were applied to investigate the expression level of desmuslin mRNA in normal and abnormal veins.Simultaneously,the specific single-cloned antibody bestowed by Yuji Mizuno was used to evaluate the amount of DMN protein in the two groups by Western blotting and immunohistochemical techniques.RESULTS: In the abnormal veins isolated from the patients with primary vein incompetence,the expression of desmuslin mRNA was significantly down-regulated,compared with that in control group(semi-quantitative RT-PCR: 0.19?0.05 vs 0.83?0.08,P
